Virus-induced gene silencing (VIGS) is a recently developed technique for characterizing the function of plant genes by gene transcript suppression and is increasingly used to generate transient loss-of-function assays. Here we report that the 2mDNA1, a geminivirus satellite vector, can induce efficient gene silencing in Nicotiana tabacum with Tobacco curly shoot virus. We have successfully silenced the β-glucuronidase (GUS) gene in GUS transgenic N. tabacum plants and the sulphur desaturase (Su) gene in five different N. tabacum cultivars. These pronounced and severe silencing phenotypes are persistent and ubiquitous. Once initiated in seedlings, the silencing phenotype lasted for the entire life span of the plants and silencing could be induced in a variety of tissues and organs including leaf, shoot, stem, root, and flower, and achieved at any growth stage. This system works well between 18-32°C. We also silenced the NtEDS1 gene and demonstrated that NtEDS1 is essential for N gene mediated resistance against Tobacco mosaic virus in N. tabacum. The above results indicate that this system has great potential as a versatile VIGS system for routine functional analysis of genes in N. tabacum.

BackgroundFew studies have compared change in the health-related quality of life （HRQL） following treatment of non-ST-elevation acute coronary syndrome （NSTE-ACS） with either percutaneous coronary intervention （PCI） or coronary artery bypass grafting （CABG）. This study is tocompare changes in HRQL six months after hospital discharge between NSTE-ACS pa-tients who underwent either PCI or *** HRQL was assessed using the Seattle angina questionnaire at admission and six months after discharge in 1012 consecutive patients with NSTE-ACS. To assess associations of PCI and CABG with HRQL changes, logistic regression models were constructed treating changes in the score of each dimension of the Seattle angina question-naire as dependent *** Although both the PCI and CABG groups experienced angina relief and other improvements at 6-month follow-up （P〈0.001）, the CABG relative to PCI group showed more significant improvements in angina frequency （P= 0.044） and quality of life （P= 0.028）. In multivariable logistic analysis, CABG also was an independent predictor for both im-provement of angina frequency （OR： 1.62, 95%CI： 1.09-4.63,P= 0.042） and quality of life （OR： 2.04, 95%CI： 1.26-6.92,P= 0.038） relative to *** In patients with NSTE-ACS, both PCI and CABG provide great improvement in disease-specific health status at six months, with that of CABG being more prominent in terms of angina frequency and quality of life.

With ever-increasing depth of coal mine and the continuous improvement of mechanization, heat damage has become one of the major disasters in coal mine exploitation. Established the temperature prediction models suitable for different kinds of tunnels through analysis of the heat of shafts, roadways and working faces. The average annual air temperature prediction equation from the inlets of shafts to the working faces was derived. The formula was deduced using combine method of iteration and direct calculation. The method can improve the precision of air temperature prediction, so we could establish the whole pathway air temperature prediction model with high precision. Emphasizing on the effects of leakage air to air temperature of working face and using the ideology of the finite difference method and considering the differential equation of inlet and outlet at different stages, this method can significantly improve the accuracy of temperature prediction. Program development uses Visual Basic 6.0 Language, and the Origin software was used to fit the relevant data. The predicted results shows that the air temperature generally tends to rapidly increase in the air inlet, then changes slowly on working face, and finally increases sharply in air outlet in the condition of goaf air leakage. The condition is in general consistent with the air temperature change tendency of working face with U-type ventilation system. The software can provide reliable scientific basis for reasonable ventilation, cooling measures and management of coal mine thermal hazards.

Aerodynamic roughness length is an important physical parameter in atmospheric numerical models and microme- teorological calculations, the accuracy of which can affect numerical model performance and the level of micrometeorological computations. Many factors influence the aerodynamic roughness length, but formulas for its parameterization often only con- sider the action of a single factor. This limits their adaptive capacity and often introduces considerable errors in the estimation of land surface momentum flux （friction velocity）. In this study, based on research into the parameterization relations between aerodynamic roughness length and influencing factors such as windrow conditions, thermodynamic characteristics of the sur- face layer, natural rhythm of vegetation growth, ecological effects of interannual fluctuations of precipitation, and vegetation type, an aerodynamic roughness length parameterization scheme was established. This considers almost all the factors that af- fect aerodynamic roughness length on flat land surfaces with short vegetation. Furthermore, using many years＇ data recorded at the Semi-Arid Climate and Environment Observatory of Lanzhou University, a comparative analysis of the application of the proposed parameterization scheme and other experimental schemes was performed. It was found that the error in the friction velocity estimated by the proposed parameterization scheme was considerably less than that estimated using a constant aero- dynamic roughness length and by the other parameterization schemes. Compared with the friction velocity estimated using a constant aerodynamic roughness length, the correlation coefficient with the observed friction velocity increased from 0.752 to 0.937, and the standard deviation and deviation decreased by about 20% and 80%, respectively. Background:Although overnight fasting is recommended prior to endoscopic retrograde cholangiopancreatography(ERCP),the benefits and safety of high-carbohydrate fluid diet(CFD)intake 2 h before ERCP remain *** study aimed to analyze whether high-CFD intake 2 h before ERCP can be safe and accelerate patients’***:This prospective,multicenter,randomized controlled trial involved 15 tertiary ERCP centers.A total of 1330 patients were randomized into CFD group(n=665)and fasting group(n=665).The CFD group received 400 mL of maltodextrin orally 2 h before ERCP,while the control group abstained from food/water overnight(>6 h)before *** ERCP procedures were performed using deep sedation with intravenous *** investigators were blinded but not the *** primary outcomes included postoperative fatigue and abdominal pain score,and the secondary outcomes included complications and changes in metabolic *** outcomes were analyzed according to a modified intention-to-treat ***:The post-ERCP fatigue scores were significantly lower at 4 h(4.1±2.6 vs.4.8±2.8,t=4.23,P<0.001)and 20 h(2.4±2.1 vs.3.4±2.4,t=7.94,P<0.001)in the CFD group,with least-squares mean differences of 0.48(95%confidence interval[CI]:0.26-0.71,P<0.001)and 0.76(95%CI:0.57-0.95,P<0.001),*** 4-h pain scores(2.1±1.7 vs.2.2±1.7,t=2.60,P=0.009,with a least-squares mean difference of 0.21[95%CI:0.05-0.37])and positive urine ketone levels(7.7%[39/509]vs.15.4%[82/533],χ^(2)=15.13,P<0.001)were lower in the CFD *** CFD group had significantly less cholangitis(2.1%[13/634]vs.4.0%[26/658],χ^(2)=3.99,P=0.046)but not pancreatitis(5.5%[35/634]vs.6.5%[43/658],χ^(2)=0.59,P=0.444).Subgroup analysis revealed that CFD reduced the incidence of complications in patients with native papilla(odds ratio[OR]:0.61,95%CI:0.39-0.95,P=0.028)in the multivariable ***:Ingesting 400 mL of CFD 2 h before ERCP is safe,with a reduction in post-ERCP fatigue,

Developing technologies that can be applied simultaneously in battery thermal management(BTM)and thermal runaway(TR)mitigation is significant to improving the safety of lithium-ion battery *** phase change material(PCM)with nonflammability has the potential to achieve this dual *** study proposed an encapsulated inorganic phase change material(EPCM)with a heat transfer enhancement for battery systems,where Na_(2)HPO_(4)·12H_(2)O was used as the core PCM encapsulated by silica and the additive of carbon nanotube(CNT)was applied to enhance the thermal *** microstructure and thermal properties of the EPCM/CNT were analyzed by a series of characterization *** different incorporating methods of CNT were compared and the proper CNT adding amount was also *** preparation,the battery thermal management performance and TR propagation mitigation effects of EPCM/CNT were further investigated on the battery *** experimental results of thermal management tests showed that EPCM/CNT not only slowed down the temperature rising of the module but also improved the temperature uniformity during normal *** peak battery temperature decreased from 76℃to 61.2℃at 2 C discharge rate and the temperature difference was controlled below 3℃.Moreover,the results of TR propagation tests demonstrated that nonflammable EPCM/CNT with good heat absorption could work as a TR barrier,which exhibited effective mitigation on TR and TR *** trigger time of three cells was successfully delayed by 129,474 and 551 s,respectively and the propagation intervals were greatly extended as well.

为了增强网络传输数据的安全性和稳定性,提出了一种激光通信网络空间恶意节点识别方法。在解析节点通信方式的基础上,明确节点平均包转发延时、转发率和丢包率的属性矢量。然后利用函数极值计算节点隶属度,提取恶意节点入侵特征。结合历史恶意入侵数据,运用二维熵识别不同类别的恶意节点,筛选出恶意节点的差异特征并获得恶意节点识别结构。利用识别分数获得恶意节点空间特征向量,对所有差异特征的二维熵做最优解处理,明确识别临界值,进而实现对恶意节点的识别。实验表明,上述方法能够精准识别出恶意节点,保障了激光通信网络空间运行和用户隐私信息的安全。

Community-based question answer（CQA） makes a figure network in development of social network. Similar question retrieval is one of the most important tasks in CQA. Most of the previous works on similar question retrieval were given with the underlying assumption that answers are similar if their questions are similar, but no work was done by modeling similarity measure with the constraint of the assumption. A new method of modeling similarity measure is proposed by constraining the measure with the assumption, and employing ensemble learning to get a comprehensive measure which integrates different context features for similarity measuring, including lexical, syntactic, semantic and latent semantic. Experiments indicate that the integrated model could get a relatively high performance consistence between question set and answer set. Models with better consistency tend to get a better precision according to answers.

This study was conducted to examine the responses of earthworms to soil organic matter and litter at different decomposition stages and their contributions in litter decomposition processes in southern subtropical areas of China. Two plantations were selected as the study sites: Site I was dominated by the exotic endogeic earthworm species Ocnerodrilus occidentalis; Site II was dominated by epigeic species Amynthas corticis. After the fallen litter and earthworms were removed or expelled, four treatments were set up as: reserving the top soil (0–5 cm, equal to H layer) (H), removing the top soil and adding fresh litter (Le), removing the top soil and adding semi-decomposed litter (Li), and a control with no top soil nor any litter (CK). Five randomized blocks that were enclosed with nylon nets on the top were set up in each site, and then the four treatments were arranged randomly in each block. After 2–3 months, earthworms were collected using the formalin method. The results showed that Ocnerodrilus occidentalis preferred Treatment H though it was found in Treatments Le and Li as well; Amynthas corticis preferred Treatment Li though sometimes it also appeared in Treatment H; and Amynthas sp., another epigeic species, was mainly present under Treatment Le and only appeared in Treatment H occasionally. These findings confirmed that earthworm species belonging to different ecological groups had different responses to organic matter at different decomposition stages. The impacts of earthworm communities dominated by O. occidentalis mainly appeared at the later periods of litter decomposition.

AIM： To construct the recombinant lentivirus expression plasmid, pLenti6/V5-NT4 p53（N 15）-antennapedia （Ant）, and study its effect on HepG2 cells. METHODS： Plasmid pLenti6/V5-NT4 p53（N15）-Ant was constructed incorporating the following functional regions, including signal peptide sequence and proregion of neurotrophin 4, N-terminal residues 12-26 of p53 and 17 amino acid drosophila carrier protein, Ant. Hepatocellular carcinoma （HepG2） cells were used for transfection. 3-[4,5-climethyl-thiazol-2yl]-2,5 diphenyl tetrazolium bromide （MI-I） assay, lactate dehydrogenase （LDH） release assay, transmission electron microscopy （TEM） and flow cytometric analysis （FCM） were employed to investigate the effects of LV-NT4（Si）- p53（N15）-Ant in vitro on HepG2 cells. In vivo experiment was also performed to investigate the inhibitory effect of LV-NT4（Si）-p53（N15）-Ant on tumor growth in nude ***： LV-NT4（Si）-p53（N15）-Ant significantly suppressed the growth of HepG2 cells. MTT assay showed that the growth of HepG2 cells was mucj more significantly inhibited by LV-NT4（Si）-p53（N15）-Ant than by LV-EGFP. The inhibition rate for HepG2 cell growth in the two groups was 46.9% and 94.5%, respectively, 48 h after infection with LV-NT4（Si）-p53（N15）-Ant, and was 33.9% and 95.8%, respectively, 72 h after infection with LV-NT4（Si）-p53（N15）-Ant （P 〈 0.01）. Light microscopy and TEM showed morphological changes in HepG2 cells infected with LV-NT4（Si）-p53（N15）-Ant, but no significant changes in HepG2 cells infected with LV-EGFR Changes were observed in ultra-structure of HepG2 cells infected with LV-NT4（Si）-p53（N15）-Ant, with degraded membranes, resulting in necrosis. LDH release from HepG2 cells was analyzed at 24, 48, 72 and 96 h after infection with LV-NT4（Si）-p53（N15）-Ant and LV-EGFP, which showed that LDH release was significantly higher in LV-NT4（Si）-p53（N15）-Ant treatment group （682 IU/L） than in control group （45 IU/L, P 〈 0.01）.
