TL1A Promotes Fibrogenesis in Colonic Fibroblasts via the TGF-β1/Smad3 Signaling Pathway

Objective Intestinal fibrosis is a refractory complication of inflammatory bowel disease(IBD).Tumor necrosis factor ligand-related molecule-1A(TL1A)is important for IBD-related intestinal fibrosis in a dextran sodium sulfate(DSS)-induced experimental colitis *** study aimed to explore the effects of TL1A on human colonic *** A trinitrobenzene sulfonic acid(TNBS)-induced experimental colitis model of LCK-CD2-TL1A-GFP transgenic(Tg)or wild-type(WT)mice was established to determine the effect and mechanism of TL1A on intestinal *** human colonic fibroblast CCD-18Co cell line was treated concurrently with TL1A and human peripheral blood mononuclear cell(PBMC)*** proliferation and activation of CCD-18Co cells were detected by BrdU assays,flow cytometry,immunocytochemistry and Western *** metabolism was tested by Western blotting and real-time quantitative polymerase chain reaction(RT-qPCR).Results The level of collagen metabolism in the TNBS+ethyl alcohol(EtOH)/Tg group was greater than that in the TNBS+EtOH/WT *** growth factor-β1(TGF-β1)and p-Smad3 in the TNBS+EtOH/Tg group were upregulated as compared with those in the TNBS+EtOH/WT *** proliferation of CCD-18Co cells was promoted by the addition of human PBMC supernatant supplemented with 20 ng/mL TL1A,and the addition of human PBMC supernatant and TL1A increased CCD-18Co proliferation by 24.4%at 24 ***1A promoted cell activation and increased the levels of COL1A2,COL3A1,and TIMP-1 in CCD-18Co *** of CCD-18Co cells with TL1A increased the expression of TGF-β1 and *** TL1A promotes TGF-β1-mediated intestinal fibroblast activation,proliferation,and collagen deposition and is likely related to an increase in the TGF-β1/Smad3 signaling pathway.

Prophylactic effects of Edible Lilium-Egg yolk compound powder on depression-like behaviors in rats exposed to chronic unpredictable mild stress: A role of gut-brain axis

The incidence rate and harmfulness of depression are increasing yearly， and the research on the antidepressant effect of regulating the intestinal microecology balance has attracted widespread attention. The purpose of this research was to study the preventive effects and the regulatory mechanism of edible Lilium-egg yolk compound powder(Chinese abbreviation， BHT) on gut-brain crosstalk in depressive rats induced by chronic unpredictable mild stress(CUMS) through the "microbiota-gut-brain" axis. Results showed that infusion of BHT for 21 days ameliorated the morphology of the hippocampus and colon， increased the rate of sucrose preference， reduced the immobility time of forced swimming， increased the expression level of anti-inflammatory cytokine Interleukin-10 (IL-10) in serum， up-regulate the expression level of plasma neurotransmitter 5-hydroxy tryptamine (5-HT)， down-regulate the expression of inflammatory factors tumor necrosis factor-α (TNF-α)， interleukin-6 (IL-6)， and interleukin-1β (IL-1β) and neurotransmitter glutamic acid (Glu) in serum， and decrease the expression levels of lipopolysaccharide (LPS)， diamine oxidase (DAO) and lactate dehydrogenase (LD) in plasma. The underlying mechanism may be to reduce the activation of microglia (lower levels of CD11b， IBa1 protein and gene expression)， regulate hippocampal GR/BDNF/NPY signal pathway (higher levels of GR， BDNF， NPY protein and gene expression)， enhance intestinal mucosal barrier function (higher levels of ZO-1， Occludin protein and gene expression)， regulate the composition and structure of intestinal flora(Analysis results of LEfSe combined with random forest showed that Prevotella and Paraprevotella might be biomarkers of BHT intervention in depression)， and it is related to the improvement of glutathione metabolism， and other metabolic pathways. Type 2 diabetes mellitus(T2DM)is a metabolic disease caused by a glycolipid metabolism disorder and isletβ-cell ***-80-I is a biologically active water-soluble polysaccharide isolated from sweet corncob,an agricultural *** hypoglycemic effects of SCP-80-I on T2DM mice and its mechanisms were investigated in this ***-80-I was found to significantly reduce blood glucose and lipid deposition levels in T2DM mice,as well as decrease serum leptin and increase adiponectin ***,real time-polymerase chain reaction(RT-PCR)and Western blotting results revealed that SCP-80-I could regulate the expression of several glycolipid metabolisms and insulin secretion genes and proteins,including 5'-AMP-activated protein kinase(AMPK),carnitine palmitoyltransferase I(CPTI),and acetyl coenzyme A carboxylase(ACC)in the liver and AMPK,sirtuin1(Sirtl),peroxisome proliferator-activated receptorycoactivator-1(PGC-1α),and uncoupling protein 2(UCP2)in the *** have a hypoglycemic effect,SCP-80-1 regulated glycolipid metabolism and islet cell function in the liver by regulating the AMPK/AC C/CPT1 signaling pathway and the AMPK/Sirt1/PGC-1αand AMPK/Sirtl/UCP2 signaling *** findings improve our understanding of polysaccharides derived from sweet corncob and the use of SCP-80-I in the production of hypoglycemic foods.

Retinoic acid alleviates rotavirus-induced intestinal damage by regulating redox homeostasis and autophagic flux in piglets

Rotaviruses(RV) are a major cause of severe gastroenteritis, particularly in neonatal piglets. Despite the availability of effective vaccines, the development of antiviral therapies for RV remains an ongoing challenge. Retinoic acid(RA), a metabolite of vitamin A, has been shown to have anti-oxidative and antiviral properties. However, the mechanism by which RA exerts its intestinal-protective and antiviral effects on RV infection is not fully understood. The study investigates the effects of RA supplementation in Duroc × Landrace × Yorkshire(DLY) piglets challenged with RV. Thirty-six DLY piglets were assigned into six treatments, including a control group, RA treatment group with two concentration gradients(5and 15 mg/d), RV treatment group, and RV treatment group with the addition of different concentration gradients of RA(5 and 15 mg/d). Our study revealed that RV infection led to extensive intestinal architecture damage, which was mitigated by RA treatment at lower concentrations by increasing the villus height and villus height/crypt depth ratio(P < 0.05), enhancing intestinal stem cell signaling and promoting intestinal barrier functions. In addition, 15 mg/d RA supplementation significantly increased NRF2 and HO-1 protein expression(P < 0.05) and GSH content(P < 0.05), indicating that RA supplementation can enhance anti-oxidative signaling and redox homeostasis after RV challenge. Additionally,the research demonstrated that RA exerts a dual impact on the regulation of autophagy, both stimulating the initiation of autophagy and hindering the flow of autophagic flux. Through the modulation of autophagic flux, RA influence the progression of RV infection. These findings provide new insights into the regulation of redox hemostasis and autophagy by RA and its potential therapeutic application in RV infection.

Effects of Diesel Concentration on the Thermal Conductivity,Specific Heat Capacity and Thermal Diffusivity of Diesel-Contaminated Soil

High energy consumption is a serious issue associated with in situ thermal desorption(TD)remediation of sites contaminated by petroleum hydrocarbons(PHs).The knowledge on the thermophysical properties of contaminated soil can help predict accurately the transient temperature distribution in a remediation site,for the purpose of energy ***,such data are rarely reported for PH-contaminated *** this study,by taking diesel as a representative example for PHs,soil samples with constant dry bulk density but different diesel mass concentrations ranging from 0% to 20% were prepared,and the variations of their thermal conductivity,specific heat capacity and thermal diffusivity were measured and analyzed over a wide temperature range between 0℃ and 120℃.It was found that the effect of diesel concentration on the thermal conductivity of soil is negligible when it is below 1%.When diesel concentration is below 10%,the thermal conductivity of soil increases with raising the ***,when diesel concentration becomes above 10%,the change of the thermal conductivity of soil with temperature exhibits the opposite *** is mainly due to the competition between soil minerals and diesel,because the thermal conductivity of minerals increases with temperature,whereas the thermal conductivity of diesel decreases with *** analysis results showed that,compared with temperature,the diesel concentration has more significant effects on soil thermal *** of the diesel concentration,with the increase of temperature,the specific heat capacity of soil increases,while the thermal diffusivity of soil *** addition,the results of a control experiment exhibited that the relative differences of the thermal conductivity of the soil samples containing the same concentration of both diesel and a pure alkane are all below 10%,indicating that the results obtained with diesel in this study can be extended to the family of PHs.A theo

Production of Functional Materials Derived from Regenerated Silk Fibroin by Utilizing 3D Printing and Biomimetic Enzyme-induced Mineralization

Critical-sized bone defects, commonly encountered in clinical orthopedic surgery, present a significant challenge. One of the promising solutions is to prepare synthetic bone substitute materials with precise structural control, mechanical compatibility, and enhanced osteogenic induction performance, nevertheless the successful preparation of such materials remains difficult. In this study, a two-step technique,integrating an extrusion-based printing process with biomimetic mineralization induced by alkaline phosphatase(ALP), was developed. Initially,a pre-cured hydrogel of regenerated silk fibroin(RSF) with a small quantity of hydroxypropyl cellulose(HPC) and ALP was prepared through heating the mixed aqueous solution. This pre-cured hydrogel demonstrated thixotropic property and could be directly extruded into predetermined structures through a 3D-printer. Subsequently, the 3D-printed RSF-based materials with ALP underwent biomimetic in situ mineralization in calcium glycerophosphate(Ca-GP) mineralizing solution, utilizing the polymer chains of RSF as templates and ALP as a trigger for cleaving phosphate bonds of Ca-GP. The resulting 3D-printed RSF-mineral composites including hydrogel and sponge possessed adjustable compression modulus of megapascal grade and variable hydroxyapatite content, which could be controlled by manipulating the duration of the mineralization ***, these 3D-printed RSF-mineral composites demonstrated non-cytotoxicity towards rat bone marrow mesenchymal stem cells. Therefore, they may hold great potential for applications involving the replacement of tissues characterized by osteoinductivity and intricate structures.

The as-cast amorphous Ti_(48)Zr_(27)Cu_(6)Nb_(5)Be_(14)composites,comprising in situ formedβ-Ti ductile crystalline precipitates,were prepared by water cooled copper mold suction ***,the semi-solid composites were obtained after the as-cast composites were treated by semi-solid isothermal *** microstructure evolution and kinetics of the composites were *** show that the microstructures of both the as-cast and semi-solid composites comprise ofβ-Ti crystal phases and amorphous matrix *** and after treatment,the crystals evolve from fine granular or fine dendritic crystals to coarse *** the treatment temperature increasing or the time prolonging,the average crystal size gradually increases and the surface morphology of the crystals gradually becomes *** studying the microstructural evolution and dynamics during the isothermal treatment process,it is found that the final morphology ofβ-Ti crystals is influenced by the isothermal treatment temperature and time(t),and theβ-Ti evolution rate increases with an increase in treatment *** addition,a linear relationship was observed between the size of cubicβ-Ti crystals(D^(3))and t;the growth kinetics factor K is 3.8μm^(3)·s^(-1).As the K value closes to 4μm^(3)·s^(-1),it is inferred the morphology evolution ofβ-Ti crystals is a coarsening behavior controlled by the diffusion of solute elements.

With the ongoing advancements in sensor networks and data acquisition technologies across various systems like manufacturing,aviation,and healthcare,the data driven vibration control(DDVC)has attracted broad interests from both the industrial and academic *** shaping(IS),as a simple and effective feedforward method,is greatly demanded in DDVC *** convolves the desired input command with impulse sequence without requiring parametric dynamics and the closed-loop system structure,thereby suppressing the residual vibration *** on a thorough investigation into the state-of-the-art DDVC methods,this survey has made the following efforts:1)Introducing the IS theory and typical input shapers;2)Categorizing recent progress of DDVC methods;3)Summarizing commonly adopted metrics for DDVC;and 4)Discussing the engineering applications and future trends of *** doing so,this study provides a systematic and comprehensive overview of existing DDVC methods from designing to optimizing perspectives,aiming at promoting future research regarding this emerging and vital issue.
