Ginsenoside Rd Induces Differentiation of Myeloid Leukemia Cells via Regulating ERK/GSK-3β Signaling Pathway

Objective: To investigate the role of ginsenoside Rd（GRd） in acute myeloid leukemia（AML） cell differentiation. Methods: AML cells were treated with GRd（25, 50, 100 and 200 μg/mL）, retinoic acid（RA, 0.1g/L） and PD98059（20 mg/mL） for 72 h, cell survival was detected by methylthiazolyldiphenyl-tetrazolium bromide and colony formation assays, and cell cycle was detected by flow cytometry. Cell morphology and differentiation were observed by Wright-Giemsa staining, peroxidase chemical staining and cellular immunochemistry assay, respectively. The protein expression levels of GATA binding protein 1（GATA-1）, purine rich Box-1（PU.1）, phosphorylated-extracellular signal-related kinase（p-ERK）, ERK, phosphorylated-glycogen synthase kinase-3β（p-GSK3β）, GSK3β and signal transducer and activator of transcription 1（STAT1） were detected by Western blot. Thirty-six mice were randomly divided into 3 groups using a random number table: model control group（non-treated）, GRd group [treated with 200 mg/（kg·d） GRd] and homoharringtonine（HTT） group [treated with 1 mg/（kg·d） HTT]. A tumor-bearing nude mouse model was established, and tumor weight and volume were recorded. Changes of subcutaneous tumor tissue were observed after hematoxylin and eosin staining. WT1 and GATA-1 expressions were detected by immunohistochemical staining. Results: The cell survival was inhibited by GRd in a dose-dependent manner and GRd caused G0/G1cell arrest（Plicited upregulation of p-ERK, p-GSK-3β and STAT1 expressions in cells, and reversed the effects of PD98059 on inhibiting the expressions of peroxidase, GATA-1 and PU.1（P<0.05）. After GRd treatment, tumor weight and volume of mice were decreased, and tumor cells underwent massive apoptosis and necrosis（P<0.05）. Video portrait segmentation(VPS), aiming at segmenting prominent foreground portraits from video frames, has received much attention in recent years. However, the simplicity of existing VPS datasets leads to a limitation on extensive research of the task. In this work, we propose a new intricate large-scale multi-scene video portrait segmentation dataset MVPS consisting of 101 video clips in 7 scenario categories,in which 10843 sampled frames are finely annotated at the pixel level. The dataset has diverse scenes and complicated background environments, which is the most complex dataset in VPS to our best knowledge.Through the observation of a large number of videos with portraits during dataset construction, we find that due to the joint structure of the human body, the motion of portraits is part-associated, which leads to the different parts being relatively independent in motion. That is, the motion of different parts of the portraits is imbalanced. Towards this imbalance, an intuitive and reasonable idea is that different motion states in portraits can be better exploited by decoupling the portraits into parts. To achieve this, we propose a part-decoupling network(PDNet) for VPS. Specifically, an inter-frame part-discriminated attention(IPDA)module is proposed which unsupervisedly segments portrait into parts and utilizes different attentiveness on discriminative features specified to each different part. In this way, appropriate attention can be imposed on portrait parts with imbalanced motion to extract part-discriminated correlations, so that the portraits can be segmented more accurately. Experimental results demonstrate that our method achieves leading performance with the comparison to state-of-the-art methods.

Carbon source is an important factor of biological treatment systems, the effects of which on their temporal community assembly patterns are not sufficiently understood currently.In this study, the temporal dynamics and driving mechanisms of the communities in three parallel bioreactors for low-concentration coking wastewater(CWW) treatment with differing carbon source concentrations(S0 with no glucose addition, S1 with 200 mg/L glucose addition and S2 with 400 mg/L glucose addition) were comprehensively studied. Highthroughput sequencing and bioinformatics analyses including network analysis and Infer Community Assembly Mechanisms by Phylogenetic bin-based null model(iCAMP) were used. The communities of three systems showed turnover rates of 0.0029~0.0034 every 15days. Network analysis results showed that the S0 network showed higher positive correlation proportion(71.43%) and clustering coefficient(0.33), suggesting that carbon source shortage in S0 promoted interactions and cooperation of microbes. The neutral community model analysis showed that the immigration rate increased from 0.5247 in S0 to 0.6478in S2. The iCAMP analysis results showed that drift(45.89%) and homogeneous selection(31.68%) dominated in driving the assembly of all the investigated microbial communities.The contribution of homogeneous selection increased with the increase of carbon source concentrations, from 27.92% in S0 to 36.08% in S2. The OTUs participating in aerobic respiration and tricarboxylic acid(TCA) cycle were abundant among the bins mainly affected by deterministic processes, while those related to the metabolism of refractory organic pollutants in CWW such as alkanes, benzenes and phenols were abundant in the bins dominated by stochastic processes.

BACKGROUND:Postpartum posttraumatic stress disorder(PTSD)can occur in women who give birth after emergency admission.The identification of risk factors for this condition is crucial for developing effective preventive measures.This retrospective study aimed to explore the incidence and risk factors for postpartum PTSD in women who give birth after emergency admission.METHODS:Medical records of women who gave birth after emergency admission were collected between March 2021 and April 2023.The patients’general conditions and perinatal clinical indicators were recorded.The puerperae were divided into PTSD group and control group based on symptom occurrence at six weeks postpartum.Multivariate logistic regression analysis was performed to identify risk factors.RESULTS:A total of 276 puerperae were included,with a PTSD incidence of 20.3% at six weeks postpartum.Multivariate logistic regression analysis identified emergency cesarean section(odds ratio[OR]=2.102;95%confidence interval[CI]:1.114-3.966,P=0.022),admission to the emergency department after midnight(12:00 AM)(OR=2.245;95%CI:1.170-4.305,P<0.001),and cervical dilation(OR=3.203;95%CI:1.670–6.141,P=0.039)as independent risk factors for postpartum PTSD.Analgesia pump use(OR=0.500;95%CI:0.259–0.966,P=0.015)was found to be a protective factor against postpartum PTSD.CONCLUSION:Emergency cesarean section,admission to the emergency department after midnight,and cervical dilation were identified as independent risk factors for postpartum PTSD,while analgesic pump use was a protective factor.These findings provide insights for developing more effective preventive measures for women who give birth after emergency admission.

Facile preparation of Fe/N-based biomass porous carbon composite towards enhancing the thermal decomposition of DAP-4

Fe/N-based biomass porous carbon composite(Fe/N-p Carbon) was prepared by a facile high-temperature carbonization method from biomass,and the effect of Fe/N-p Carbon on the thermal decomposition of energetic molecular perovskite-based material DAP-4 was studied.Biomass porous carbonaceous materials was considered as the micro/nano support layers for in situ deposition of Fe/N precursors.Fe/Np Carbon was prepared simply by the high-temperature carbonization method.It was found that it showed the inherent catalysis properties for thermal decomposition of DAP-4.The heat release of DAP-4/Fe/N-p Carbon by DSC curves tested had increased slightly,compared from DAP-4/Fe/N-p Carbon-0.The decomposition temperature peak of DAP-4 at the presence of Fe/N-p Carbon had reduced by 79°C from384.4°C(pure DAP-4) to 305.4°C(DAP-4/Fe/N-p Carbon-3).The apparent activation energy of DAP-4thermal decomposition also had decreased by 29.1 J/mol.The possible catalytic decomposition mechanism of DAP-4 with Fe/N-p Carbon was proposed.

The population of non-alcoholic fatty liver disease(NAFLD)patients along with relevant advanced liver disease is projected to continue growing,because currently no medications are approved for treatment.Fecal microbiota transplantation(FMT)is believed a novel and promising therapeutic approach based on the concept of the gut-liver axis in liver disease.There has been an increase in the number of pre-clinical and clinical studies evaluating FMT in NAFLD treatment,however,existing findings diverge on its effects.Herein,we briefly summarized the mechanism of FMT for NAFLD treatment,reviewed randomized controlled trials for evaluating its efficacy in NAFLD,and proposed the prospect of future trials on FMT.

lipid metabolism play an essential role in occurrence and development of asthma,and it can be disturbed by phthalate esters(PAEs)and organophosphate fame retardants(OPFRs).As a chronic infammatory respiratory disease,the occurrence risk of childhood asthma is increased by PAEs and OPFRs exposure,but it remains not entirely clear how PAEs and OPFRs contribute the onset and progress of the disease.We have profiled the serum levels of PAEs and OPFRs congeners by liquid chromatography coupled with mass spectrometry,and its relationships with the dysregulation of lipid metabolism in asthmatic,bronchitic(acute infammation)and healthy(non-infammation)children.Eight PAEs and nine OPFRs congeners were found in the serum of children(1–5 years old)from Shenzhen,and their total median levels were 615.16 ng/m L and 17.06 ng/m L,respectively.Moreover,the serum levels of mono-methyl phthalate(MMP),tri-propyl phosphate(TPP)and tri-n-butyl phosphate(TNBP)were significant higher in asthmatic children than in healthy and bronchitic children as control.Thirty-one characteristic lipids and fatty acids of asthma were screened by machine-learning random forest model based on serum lipidome data,and the alterations of infammatory characteristic lipids and fatty acids including palmitic acids,12,13-Di HODE,14,21-Di HDHA,prostaglandin D2 and Lyso PA(18:2)showed significant correlated with high serum levels of MMP,TPP and TNBP.These results imply PAEs and OPFRs promote the occurrence of childhood asthma via disrupting infammatory lipid and fatty acid metabolism,and provide a novel sight for better understanding the effects of plastic additives on childhood asthma.

Panonychus citriMcGregor(Acari:Tetranychidae),a destructive citrus pest,causes considerable annual economic losses due to its short lifespan and rapid resistancedevelopment.MicroRNA(miRNA)-induced RNA interference is a promising approachfor pest control because of endogenous regulation of pest growth and development.Tosearch for miRNAs with potential insecticidal activity inP.citri,genome-wide analysis ofmiRNAs at different developmental stages was conducted,resulting in the identificationof 136 miRNAs,including 73 known and 63 novel miRNAs.A total of 17 isomiRNAsand 12 duplicated miRNAs were characterized.MiR-1 and miR-252-5p were identified asreference miRNAs forP.citriandTetranychus urticae.Based on differential expressionanalysis,treatments with miR-let-7a and miR-315 mimics and the miR-let-7a antagomirsignificantly reduced the egg hatch rate and resulted in abnormal egg development.Over-expression or downregulation of miR-34-5p and miR-305-5p through feeding significantlydecreased the adult eclosion rate and caused molting defects.The 4 miRNAs,miR-let-7a,miR-315,miR-34-5p,and miR-305-5p,had important regulatory functions and insectici-dal properties in egg hatching and adult eclosion.In general,these data advance our un-derstanding of miRNAs in mite biology,which can assist future studies on insect-specificmiRNA-based green pest control technology.

Investigation on occupant presence and appliance operation schedules for university campus in south China sub-tropical area

Building occupant presence during varying periods is crucial to the performance studies of buildings and city regions.However,the understanding of the building occupancies on the university campus remains limited.To address this gap,our study employs field measurements,payment records,course arrangements,and building access systems to depict the occupancy patterns of the canteen,dormitory,library,and teaching and lab buildings during weekdays and weekends.We found that the occupancy rates across different buildings are somehow interrelated,given that the total number of occupants on campus is generally constant.Notably,dormitory occupancy rates tend to be low during the morning and afternoon course hours,which inversely correlates with the high occupancy rates in the teaching and lab buildings during these periods.Similarly,canteens experience surges in occupancy during meal times,which coincide with a decrease in library usage.Moreover,we established appliance operation schedules for dormitories through surveys and on-site investigations.Water dispensers and electronic devices were identified as the primary energy consumers for both male and female occupants,with desk-top fans and hairdryers being significant energy users for male and female occupants,respectively.These findings are essential for energy studies within a campus setting,underlining the importance of considering occupant behaviors on a regional scale.
