Inhibition of EGFR attenuates EGF-induced activation of retinal pigment epithelium cell via EGFR/AKT signaling pathway

AIM:To explore the effect of epidermal growth factor receptor(EGFR)inhibition by erlotinib and EGFR siRNA on epidermal growth factor(EGF)-induced activation of retinal pigment epithelium(RPE)***:Human RPE cell line(ARPE-19 cells)was activated by 100 ng/mL *** and EGFR siRNA were used to intervene EGF *** viability,proliferation,and migration were detected by methyl thiazolyl tetrazolium(MTT)assay,bromodeoxyuridine(BrdU)staining assay and wound healing assay,***/protein kinase B(AKT)pathway proteins and N-cadherin,α-smooth muscle actin(α-SMA),and vimentin were tested by Western blot *** was also determined by immunofluorescence ***:EGF treatment for 24h induced a significant increase of ARPE-19 cells’viability,proliferation and migration,phosphorylation of EGFR/AKT proteins,and decreased total EGFR *** suppressed ARPE-19 cells’viability,proliferation and migration through down regulating total EGFR and AKT protein *** also inhibited EGF-induced an increase of proliferative and migrative ability in ARPE-19 cells and clearly suppressed EGF-induced EGFR/AKT proteins phosphorylation and decreased expression of N-cadherin,α-SMA,and vimentin ***,EGFR inhibition by EGFR siRNA significantly affected EGF-induced an increase of cell proliferation,viability,and migration,phosphorylation of EGFR/AKT proteins,and up-regulation of N-cadherin,α-SMA,and vimentin ***:Erlotinib and EGFR-knockdown suppress EGF-induced cell viability,proliferation,and migration via EGFR/AKT pathway in RPE *** inhibition may be a possible therapeutic approach for proliferative vitreoretinopathy(PVR).

AIM:To explore the effects of hepatocyte growth factor(HGF)on retinal pigment epithelium(RPE)cell ***:The human adult retinal pigment epithelial cell line-19(ARPE-19)were treated by HGF or mesenchymalepithelial transition factor(MET)inhibitor SU11274 in *** viability was detected by a Cell Counting Kit-8 *** proliferation and motility was detected by a bromodeoxyuridine incorporation assay and a wound healing assay,*** expression levels of MET,phosphorylated MET,protein kinase B(AKT),and phosphorylated AKT proteins were determined by Western blot *** MET and phosphorylated MET proteins were also determined by immunofluorescence ***:HGF increased ARPE-19 cells’viability,proliferation and migration,and induced an increase of phosphorylated MET and phosphorylated AKT ***11274 significantly reduced cell viability,proliferation,and migration and decreased the expression of MET and AKT ***11274 suppressed HGF-induced increase of viability,proliferation,and migration in ARPE-19 ***,SU11274 also blocked HGF-induced phosphorylation of MET and AKT ***:HGF enhances cellular viability,proliferation,and migration in RPE cells through the MET/AKT signaling pathway,whereas this enhancement is suppressed by the MET inhibitor ***-induced MET/AKT signaling might be a vital contributor of RPE cells survival.

High-entropy alloys(HEAs),with a new alloying concept,could possess many unique mechanical and functional *** current work investigated whether one such alloy offers potential for bearing surfaces under dry *** dry,reciprocating sliding wear characteristics of AlCoCrFeNiTi0.5alloy were investigated under various applied loads and sliding *** electron microscopy(TEM) and scanning electron microscopy(SEM) were utilized to characterize internal structure and wear surfaces of the alloy,*** is found that the AlCoCrFeNiTi0.5alloy preserves better wear resistance than Fe77Ni23solid solution alloy,Ti-46Al-2Cr-2Nb intermetallic alloy,or a wear-resistant steel AISI52100,especially under higher *** wear rate increases slowly with the applied loads increasing and keeps steady under different sliding *** wear mechanisms are abrasive wear,adhesive wear and oxidative *** nano-sized Fe-Cr solid solution and Al-NiTi rich intermetallic phase precipitated in the dendritic regions and the formation of oxidation play important roles in the good wear resistances of this high-entropy alloy.

Plant RNA-binding proteins:Phase separation dynamics and functional mechanisms underlying plant development and stress responses

RNA-binding proteins(RBPs)accompany RNA from synthesis to decay,mediating every aspect of RNA metabolism and impacting diverse cellular and developmental processes in *** RBPs undergo phase separation along with their bound RNA to form and function in dynamic membraneless biomolecular condensates for spatiotemporal coordination or regulation of RNA *** evidence suggests that phase-separating RBPs with RNA-binding domains and intrinsically disordered regions play important roles in plant development and stress ***,we summarize the current knowledge about how dynamic partitioning of RBPs into condensates controls plant development and enables sensing of experimental changes to confer growth plasticity under stress conditions,with a focus on the dynamics and functional mechanisms of RBP-rich nuclear condensates and cytoplasmic granules in mediating RNA *** also discuss roles of multiple factors,such as environmental signals,protein modifications,and N6-methyladenosine RNA methylation,in modulating the phase separation behaviors of RBPs,and highlight the prospects and challenges for future research on phase-separating RBPs in crops.

Clinical efficacy and changes of serum VEGF-A,VEGF-B,and PLGF after conbercept treating neovascular agerelated macular degeneration

AIM:To evaluate the clinical efficacy and systemic safety profile of conbercept in clinical practice on vascular endothelial growth factor(VEGF)-A,VEGF-B,and placental growth factor(PLGF)levels after intravitreal injections for the neovascular age-related macular degeneration(AMD).METHODS:Thir ty-five patients(35 eyes)with neovascular AMD received intravitreal injections of conbercept treatment with pro re nata *** visual acuity(BCVA)and central retinal thickness(CRT)were detected before the intravitreal injection and at 1,3,and 12mo after conbercept *** levels of serum VEGF-A,VEGF-B,and PLGF were measured by enzyme-linked immunosorbent assay before the injection and 1 and 12mo after conbercept ***:At baseline,the mean BCVA score was 39.89±14.64 *** mean BCVA scores were 51.03±15.78,56.71±14.38,and 52.49±10.16 letters at 1,3,and 12mo after conbercept treatment,and the BCVA improvements were all significant,respectively(P0.05).CONCLUSION:Conbercept intravitreal injection leads to BCVA and CRT improvement,however,it does not significantly affect systemic serum VEGF-A,VEGF-B,and PLGF levels at 1 and 12mo after intravitreal injection treating neovascular AMD.

The gasdermin(GSDM)family has garnered significant attention for its pivotal role in immunity and disease as a key player in *** recently characterized class of pore-forming effector proteins is pivotal in orchestrating processes such as membrane permeabilization,pyroptosis,and the follow-up inflammatory response,which are crucial self-defense mechanisms against irritants and *** have been implicated in a range of diseases including,but not limited to,sepsis,viral infections,and cancer,either through involvement in pyroptosis or independently of this *** regulation of GSDM-mediated pyroptosis is gaining recognition as a promising therapeutic strategy for the treatment of various *** strategies for inhibiting GSDMD primarily involve binding to GSDMD,blocking GSDMD cleavage or inhibiting GSDMD-N-terminal(NT)oligomerization,albeit with some off-target *** this review,we delve into the cutting-edge understanding of the interplay between GSDMs and pyroptosis,elucidate the activation mechanisms of GSDMs,explore their associations with a range of diseases,and discuss recent advancements and potential strategies for developing GSDMD inhibitors.

BL03HB: a Laue microdiffraction beamline for both protein crystallography and materials science at SSRF

A Laue microdiffraction beamline (BL03HB) was constructed at the Shanghai SynchrotronRadiation Facility (SSRF). This beamline featu res two consecutive focusing points in twodifferent sectors within its end station, the first dedicated to protein crystallography and theother tailored to materials science applications. Based on a superbend dipole magnet with amagnetic field of 2.29 T, a two-stage focusing design was implemented with two sets of Kirkpatrick- Baez mirrors to achieve a micro white beam as small as 4.2 um X4.3 pμm at the firstsector and 09 pum X 1.3 pum at the second sector in the standard beamline operation mode at *** X-ray microbeam in the two sectors can be easily switched between monochromatic andwhite beams by moving a four-bounce monochromator in or out of the light path, respectively. Inthe protein crystallography sector, white-beam Laue microdifraction was dermonstrated tosuccessully determine the structure of protein crystals from only a few images of diffraction data .collected by a Pilatus 2 M area detector. In the materials science sector, the white -beam Lauediffraction was collected in a reflection geometry using another Pilatus 2 M area detector, whichcould map the microstructural distribution on the sample surface by scanning the samples. Ingeneral, the BL03HB beamline promotes the application of Laue microdiffraction in both proteincrystallgraphy and materials science. This paper presents a comprehensive overview of theBL03HB beamline, end station, and the first commission results.

Incorporating contextual evidence to improve implicit discourse relation recognition in Chinese

The discourse analysis task,which focuses on understanding the semantics of long text spans,has received increasing attention in recent *** a critical component of discourse analysis,discourse relation recognition aims to identify the rhetorical relations between adjacent discourse units(e.g.,clauses,sentences,and sentence groups),called arguments,in a *** works focused on capturing the semantic interactions between arguments to recognize their discourse relations,ignoring important textual information in the surrounding ***,in many cases,more than capturing semantic interactions from the texts of the two arguments are needed to identify their rhetorical relations,requiring mining more contextual *** this paper,we propose a method to convert the RST-style discourse trees in the training set into dependency-based trees and train a contextual evidence selector on these transformed *** this way,the selector can learn the ability to automatically pick critical textual information from the context(i.e.,as evidence)for arguments to assist in discriminating their *** we encode the arguments concatenated with corresponding evidence to obtain the enhanced argument ***,we combine original and enhanced argument representations to recognize their *** addition,we introduce auxiliary tasks to guide the training of the evidence selector to strengthen its selection *** experimental results on the Chinese CDTB dataset show that our method outperforms several state-of-the-art baselines in both micro and macro F1 scores.

CO_(2)flooding in shale oil reservoir with radial borehole fracturing for CO_(2)storage and enhanced oil recovery

This study introduces a novel method integrating CO_(2)flooding with radial borehole fracturing for enhanced oil recovery and CO_(2)underground storage,a solution to the limited vertical stimulation reservoir volume in horizontal well fracturing.A numerical model is established to investigate the production rate,reservoir pressure field,and CO_(2)saturation distribution corresponding to changing time of CO_(2)flooding with radial borehole fracturing.A sensitivity analysis on the influence of CO_(2)injection location,layer spacing,pressure difference,borehole number,and hydraulic fractures on oil production and CO_(2)storage is *** CO_(2)flooding process is divided into four *** in layer spacing will significantly improve oil production rate and gas storage ***,serious gas channeling can occur when the spacing is lower than 20 *** the pressure difference between the producer and injector,the borehole number,the hydraulic fracture height,and the fracture width can also increase the oil production rate and gas storage *** analysis shows that layer spacing and fracture height greatly influence gas storage and oil *** outcomes are expected to provide a theoretical basis for the efficient development of shale oil reservoirs in the vertical direction.

Topological Stability and Entropy for Certain Set-valued Maps

In this paper,the dynamics(including shadowing property,expansiveness,topological stability and entropy)of several types of upper semi-continuous set-valued maps are mainly considered from differentiable dynamical systems points of *** is shown that(1)if f is a hyperbolic endomorphism then for eachε>0 there exists a C^(1)-neighborhood U of f such that the induced set-valued map F_(f,U)has theε-shadowing property,and moreover,if f is an expanding endomorphism then there exists a C^(1)-neighborhood U of f such that the induced set-valued map F_(f,U)has the Lipschitz shadowing property;(2)when a set-valued map F is generated by finite expanding endomorphisms,it has the shadowing property,and moreover,if the collection of the generators has no coincidence point then F is expansive and hence is topologically stable;(3)if f is an expanding endomorphism then for eachε>0 there exists a C^(1)-neighborhood U of f such that h(F_(f,U,ε))=h(f);(4)when F is generated by finite expanding endomorphisms with no coincidence point,the entropy formula of F is ***,the dynamics of the set-valued maps based on discontinuous maps on the interval are also considered.
