The pseudospectrum and spectrum (in)stability of quantum corrected Schwarzschild black hole

In this study, we investigate the pseudospectrum and spectrum(in)stability of quantum corrected Schwarzschild black ***, we use the hyperboloidal framework to cast the quasinormal mode(QNM) problem into an eigenvalue problem associated with a non-selfadjoint operator, and then the spectrum and pseudospectrum are depicted. Besides, the invariant subspace method is exploited to improve the computational efficiency for pseudospectrum. The investigation into the spectrum(in)stability entails two main aspects. On the one hand, we calculate the spectra of the quantum corrected black hole, then by the means of the migration ratio, the impact of the quantum correction effect on the Schwarzschild black hole has been *** results indicate that the so-called “migration ratio instability” will occur for small black holes with small angular momentum number l. In the eikonal limit, the migration ratios remain the same for each overtone. On the other hand, we study the spectrum(in)stability of the quantum corrected black hole by directly adding some particular perturbations into the effective potential,where perturbations are located at the event horizon and null infinity, respectively. There are two interesting observations under the same perturbation energy norm. First, perturbations at infinity are more capable of generating spectrum instability than those at the event horizon. Second, we find that the peak distribution can lead to the instability of QNM spectrum more efficiently than the average distribution.

An easy-to-use site-directed mutagenesis method with a designed restriction site for convenient and reliable mutant screening

Site-directed mutagenesis (SDM) has been a very important method to probe the function-structure relationship of proteins. In this study, we introduced an easy-to-use, polymerase chain reaction (PCR)-based SDM method for double-stranded plasmid DNA, with a designed restriction site to ensure simple and efficient mutant screening. The DNA sequence to be mutated was first translated into amino acid sequence and then the amino acid sequence was reversely translated into DNA sequence with degenerate codons, resulting in a large number of sequences with silent mutations, which contained various restriction endonu-clease (RE) sites. Certain mutated sequence with an appropriate RE site was selected as the target DNA sequence for designing a pair of mutation primers to amplify the full-length plasmid via inverse PCR. The amplified product was 5′-phosphorylated, cir-cularized, and transformed into an Escherichia coli host. The transformants were screened by digesting with the designed RE. This protocol uses only one pair of primers and only one PCR is conducted, without the need for hybridization with hazardous isotope for mutant screening or subcloning step.

Detection of Antibody to Hepatitis Delta Virus in Human Serum by Double Antigen Sandwich ELISA

A simple rapid detection of antibody to hepatitis delta virus (anti-HDV) in human serum was developed by using double antigen sandwich ELISA. HDV gene fragment encoding HDAg was isolated from a Chinese patient infected with HDV by RT-PCR, and a high-efficient expression HD-PQE31 strain was constructed with the fragment. We obtained high titer and good quality hepatitis delta virus protein purified by Ni-NTA metal-affinity chromatography, which was identified by Western blot and ELISA, then we set up the double antigen sandwich ELISA for detection of anti-HDV in human serum, and the performance of the sandwich ELISA was evaluated in terms of specificity and sensitivity. Results were: 1) The purified HDAg protein’s purity was 90%, and its ELISA titer was 1/100 000. 2) 42 anti-HDV positive sera were detected and showed that the sensitivity of sandwich ELISA was higher than that of competitive ELISA (t=2.44, psensitivity, It can be used in routine laboratories to diagnose the HDV infection.

Enhanced degradation of tetracycline by gas-liquid discharge plasma coupled with g-C_(3)N_(4)/TiO_(2)

Plasma-catalysis is considered as one of the most promising technologies for antibiotic degradation in *** the plasma-catalytic system,one of the factors affecting the degradation effect is the performance of the photocatalyst,which is usually restricted by the rapid recombination of electrons and holes as well as narrow light absorption *** this research,a photocatalyst g-C_(3)N_(4)/TiO_(2) was prepared and coupled with gas-liquid discharge(GLD)to degrade tetracycline(TC).The performance was examined,and the degradation pathways and mechanisms were *** show that a 90%degradation rate is achieved in the GLD with g-C_(3)N_(4)/TiO_(2) over a 10 min *** the pulse voltage is conducive to increasing the degradation rate,whereas the addition of excessive g-C_(3)N_(4)/TiO_(2) tends to precipitate agglomerates,resulting in a poor degradation *** redox properties of the g-C_(3)N_(4)/TiO_(2) surface promote the generation of oxidizing active species(H2O2,O3)in *** quenching experiments showed that·OH,hole(h^(+)),play important roles in the TC degradation by the discharge with g-C_(3)N_(4)/TiO_(2).Two potential degradation pathways were proposed based on the *** toxicity of tetracycline was reduced by treatment in the ***,the g-C_(3)N_(4)/TiO_(2) composites exhibited excellent recoverability and stability.

In-situ electrochemical study on the eff ects of Fe(Ⅲ)on kinetics of pyrite acidic pressure oxidation

Fe(Ⅲ)has been proved to be a more eff ective oxidant than dissolved oxygen at ambient temperature,however,the role of Fe(Ⅲ)in pyrite acidic pressure oxidation was rarely discussed so *** this paper,in-situ electrochemical investigation was performed using a flow-through autoclave system in acidic pressure oxidation *** results illustrated that increasing Fe(Ⅲ)concentrations led to raising in redox potential of the solution,and decreased passivation of pyrite caused by deposition of elemental *** of Fe(Ⅲ)at pyrite surface was a fast reaction with low activation energy,it was only slightly promoted by rising ***,the oxidation rate of pyrite at all investigated Fe(Ⅲ)concentrations increased obviously with rising temperatures,the anodic reaction was the rate-limiting step in the overall *** energy of pyrite oxidation decreased from 47.74 to 28.79 kJ/mol when Fe(Ⅲ)concentration was increased from 0.05 to 0.50 g/L,showing that the reaction kinetics were limited by the rate of electrochemical reaction at low Fe(Ⅲ)concentrations,while,it gradually turned to be diffusion control with increasing Fe(Ⅲ)concentrations.

Clinicopathological study on TTV infection in hepatitis of unknown etiology

AIM: To investigate the state of infection, replication site, pathogenicity and clinical significance of transfusion transmitted virus (TTV) in patients with hepatitis, especially in patients of unknown etiology. METHODS: Liver tissues taken from 136 cases of non-A non-G hepatitis were tested for TT virus antigen and nucleic acid by in situ hybridization (ISH) and nested-polymerase chain reaction (PCR). Among them, TT virus genome and its complemental strand were also detected in 24 cases of autopsy liver and extrahepatic tissues with ISH. Meanwhile, TTV DNA was detected in the sera of 187 hepatitis patients by nested-PCR. The pathological and clinical data of the cases infected with TTV only were analyzed. RESULTS: In liver, the total positive rate of TTV DNA was 32.4% and the positive signals were located in the nuclei of hepatocytes. In serus, TTV DNA was detected in 21.4% cases of hepatitis A-G, 34.4% of non-A non-G hepatitis and 15% of healthy donors. The correspondence rate of TTV DNA detection between liver tissue with ISH and sera with PCR was 63.2% and 89.3% in the same liver tissues by ISH and by PCR, *** double-strand probes and single-strand probes designed to detect TTV genome, the correspondence rate of TTV DNA detected in liver and extrahepatic tissues was 85.7%. Using single-strand probes, TTV genome could be detected in liver and extrahepatic tissues by PCR, but its complemental strands (replication strands) could be observed only in livers. The liver function of most cases infected with TTV alone was abnormal and the liver tissues had different pathological damage such as ballooning, acidophilia degeneration, formation of apoptosis bodies and focus of necrosis, but the inflammation in the lobule and portal area was mild. CONCLUSION: The positive rate of TTV DNA among cases of hepatitis was higher than that of donors, especially in patients with non-A non-G hepatitis, but most of them were coinfected with other hepatitis viruses.
