Cytokines have been implicated in pancreatic β-cell destruction leading to type 1 *** to interleuken-1β（IL-1β）of pancreatic β-cells induces expression of inducible nitric oxide syn-thase（iNOS）and cyclooxygenase-2（COX-2）.Subsequent formation of nitric oxide（NO）and prostaglandinE2（PGE2）may impair β-cell *** NOS inhibitor NG-monomethyl-L-arginine（L-NMMA）,wehave further investigated the relation between NO formation and COX-2 ***-1β stimulated theformation of NO and PGE2 by pancreatic β-cells.L-NMMA completely inhibited IL-1β-induced NO forma-tion and attenuated PGE2 ***-2 gene transcription level and protein expression were deter-mined by real-time PCR,Western blot and luciferase analysis.L-NMMA inhibited IL-1β-induced promoteractivity,gene transcription and protein expression of COX-2 in pancreatic β-***,we concludedthat NO-affected COX-2 activity is directly linked to COX-2 gene transcription and protein expression inpancreatic β-*** identification of a novel interaction of NO on the COX signaling pathway in β-cellsprovides a strategy of intervention for further evaluating the role of NO and PGE2 in autoimmune diabetes.

Peroxisome proliferator-activated receptor (PPAR) isoforms (α and γ) are known to beexpressed in pancreatic islets as well as in insulin-producing cell *** of PPAR have been shoWn toenhance glucose-induced insulin secretion in rat pancreatic ***,their effect on insulin secretionis still *** understand the molecular mechanism by which PPAR7 exerts its effect on glucose-induced insulin secretion,we examined the endogenous activity of PPAR isoforms,and studied the PPARyfunction and its target gene expression in INS-1 *** found that:(1)endogenous PPARγ was activatedin a ligand-dependent manner in INS-1 cells;(2)overexpression of PPARy in the absence of PPARγ ligandsenhanced glucose-induced insulin secretion,which indicates that the increased glucose-induced insulin secretionis a PPARγ-mediated event;(3.the addition of both PPARγ and retinoid X receptor (RXR) ligands showed asynergistic effect on the augmentation of reporter activity,suggesting that the hetero-dimerization of PPAR7and RXR is required for the regulation of the target genes;(4)PPARs upregulated both the glucose transporter2 (GLUT2) and Cbl-associated protein (CAP) genes in INS-1 *** findings suggest an importantmechanistic pathway in which PPARγ enhances glucose-induced insulin secretion by activating the expressionof GLUT2 and CAP genes in a ligand-dependent manner.

目的:研究脱氧胆酸对人胆管癌细胞凋亡和增殖的影响;以及该影响的细胞内信号转导途径。方法:用四甲基偶氮唑蓝[3.(4,5鄄dimethylthiazole鄄2yl)鄄2,5鄄diphenylte鄄trazoliumbromid,MTT法]比色法和流式细胞仪技术(flowcytometry)检测脱氧胆酸对QBC93.细胞增殖和凋亡的影响。用荧光素酶报道基因技术检测在脱氧胆酸作用前后核转录因子(nuclearfactorkappaB,NF鄄κB)活性的改变。再将NF鄄κB的天然抑制因子IκB转染人胆管癌细胞系QBC93.,观察用IκB抑制NF鄄κB后肿瘤细胞的增殖和凋亡的情况。结果:脱氧胆酸能抑制胆管癌细胞的增殖、促进其凋亡,这一抑制作用的机制是通过抑制NF鄄κB实现的,用NF鄄κB的抑制因子IκB作用于QBC93.细胞,所起的作用与脱氧胆酸相似。结论:脱氧胆酸能通过抑制NF鄄κB的活性,抑制QBC93.细胞的增殖,并促进其凋亡。

Aberrant DNA methylation is the first epigenetic marker associated with chemotherapy resistance. However, the acquisition of drug-induced aberrant DNA methylation during chemotherapy is poorly understood, although steroid hormones and receptors might be involved. Breast cancer is a highly hormone-dependent cancer;approximately 65% of tumors express estrogen receptor alpha（ERα）, which is strongly associated with drug resistance. We used two ERα-positive drug-resistant breast cancer cell lines, MCF-7/PTX and ZR-75-1/PTX, to determine a role for ERα in drug-induced global DNA methylation in relation to drug resistance. Exposure of these cancer cells to paclitaxel elevated ERα expression and increased ERα binding to the promoters of DNMT1 and DNMT3.b, but not DNMT3.a, resulting in enhanced gene expression and increased global DNA methylation. The ERα/DNMT pathway was directly involved in the drug responses of the cells, as knockdown of either DNMT1 or DNMT3.b partly reversed ERα-mediated paclitaxel resistance, while estrogen exposure increased DNMT1 and DNMT3.b expression and enhanced multi-drug resistance. Our finding strongly support the idea that ERα could epigenetically regulate the drug resistance of breast cancer cells by directly promoting DNMT1 and DNMT3.b expression to mediate anticancer-drug-induced global DNA hypermethylation. The present study uncovers an intrinsic link between ERα and anticancer-drug induced epigenetic instability of breast cancer cells and could be exploited to develop new strategies for breast cancer treatment.

It's a challenge for detecting the therapeutic targets of a polypharmacological drug from variations in the responsed networks in thedfferentiated populations with complex diseases,as stable coronary heart ***,in an adaptive,3.-center,randomized,double-blind trial invoving 920 patients with moderate symptomatic stable angina treated by 14-day Danhong injection(DHI),a kind of polyphamacological drug with high quality control,or placebo(0.9%saline),with 76-day following-up,we firstly confrmed that DHl couldincrease the proportion of patients with clinically significant changes on angina-frequency assessed by Seattle Angina Questionnaire(ASAQ-AF220)(12.78%at Day 3.,95%confidence interval[C]5.86-19.71%,P=0.0003.13.82%at Day 6C0,95%CI 6.82-20.82%,P=0.0001and 8.95%at Day 90,95%CI 2.06-15.85%,P=0.01).We also found that there were no significant differences in new-onset major vascularevents(P=0.8502)and serious adverse events(P=0.9105)between DHl and *** performing the RNA sequencing in 62 selectedpatients,we developed a systemic modular approach tp identfy differentilly expressed modules(DEMs)of DHI with the Z_(summay)valueless than 0 compared with the control group,calculated by weighted gene co-expression network analysis(WGCNA),and sketched out thebasic framework on a modular map with 25 functional modules targeted by ***,the effective therapeutic module(ETM),defined as the highest corelation value with the phenotype alteration(SAQ-AF,the change in SAQ-AF at Day 3. from baseline)calculatedby WGCNA,was identifed in the population with the best effect(ASAQ-AF240),which is related to anticoagulation and regulation ofcholesterol *** assessed the modular flexbility of this ETM using the global topological D value based on Euclidean distance,which is corelated with phenotype alteration(r^(2):0.8204,P=0.019)by linear *** study identified the ant-angina therapeuticmodule in the effective population treated by the multi-target

Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73. in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. As there were obvious heterogeneity in autoantibodies and clinical manifestation in different phenotypes of the disease, we should take more factors into consideration when identifying type 1 diabetes mellitus.
