Mechanisms inactivating the gene for E-cadherin in sporadic gastric carcinomas

AIM： To study the role of CDH1/E-cadherin （E-cad） gene alteration profiles including mutation, loss of heterozygosity （LOH）, promoter polymorphism and hypermethylation in mechanisms of CDH1 inactivation in gastric carcinoma （GC）. METHODS： Specimens were collected surgically from 70 patients with GC. Allelotyping PCR and detection of LOH, denaturing high pressure liquid chromatography and DNA sequencing, restriction fragment length polymorphism analysis, methylation specific PCR, and immunohistochemical staining were used. RESULTS： Promoter polymorphism was not a major mechanism of E-cad inactivation. Only one truncating mutation was found in a diffuse type tumor （3%）. Both LOH and promoter hypermethylation were major mechanisms of E-cad inactivation, but interestingly, there was a negative association between the fraction of allelic loss （LOH） in tumors and hypermethylation of CDH1. Therefore LOH and hypermethylation were two different tumorigenic pathways involved in GC. CONCLUSION： Given the findings that somatic mutation was extremely low and the relationship between LOH and hypermethylation was inverse, any two combinations of these three factors cannot fulfill the classical two-hit hypothesis of CDH1 inactivation. Thus, other mechanisms operating at the transcriptional level or at the post-translational level might be required to induce E-cadherin inactivation.

Alleviating effects of dehydration under no hyperthermia on the immunomodulatory response to the polysaccharide fraction from fu-ling （Poria cocos） in male collegiate wrestlers

Background It is well known that dehydration can impair bodily functions. To evaluate the impact of hydration status under ambient environmental temperature on the immune system, 25 male collegiate wrestlers were recruited to undergo an experimental dehydration *** Thirteen subjects had controlled diets with individual energy requirements to prevent body mass loss and restricted water intake to cause 4.52% dehydration; they formed the dehydrated group （DE）. These subjects developed a urine specific gravity of about 1.030 in 84 hours. Twelve other subjects had no water restriction and maintained their total body weight comprised the euhydrated group （EU）. Peripheral blood monocytes （PBMNC） were isolated after dehydration to perform immune response testing by being incubated with a polysaccharide fraction from fu-ling, Poria cocos （polysaccharide fraction from Poria cocos, PCPS, 1-30 μg/L）, to prepare a conditioned medium termed conditioned medium of PBMNC stimulated by PCPS （PCPS-MNC-CM）. More PCPS （25 μg/L） was needed in the DE group to prepare the PCPS-MNC-CM, which was assayed with a growth inhibitory curve for treated U973 *** The treated U937 cells, incubated together with PCPS-MNC-CM from the DE group, exhibited a much lower nitroblue tetrazolium （NBT） positive value of （63.7±4.7）%. The concentration of interferon-gamma （IFN-Y）, interleukin （IL）-1β and tumor necrosis factor （TNF）-α in PCPS-MNC-CM from subjects after dehydration was much lower than in the CM from the EU *** The immune response to PCPS in the DE group was lower than in normally hydrated subjects.

BACKGROUND Cystic artery pseudoaneurysm is a condition rarely encountered by clinicians;this,its etiology and presentation as well as appropriate treatments are not well *** it is treated by removal of the diseased gallbladder and cystic artery,such surgery can be difficult and risky if acute inflammation with bleeding occurs,and not every patient can tolerate the *** SUMMARY An 81-year-old man complained of epigastric pain and tarry stool passage that lasted for 3 *** had a medical history of poor cardiopulmonary *** computed tomographic scan of abdomen showed cystic artery pseudoaneurysm and dilatation of *** of high adverse outcomes related to general anesthesia,the patient was successfully managed with endovascular embolization for this cystic artery pseudoaneurysm and percutaneous drainage for the distended *** A patient with cystic artery pseudoaneurysm may quickly deteriorate with the occurrence of concurrent arterial bleeding and *** report presents the case of a patient who did not undergo surgery due to multiple cardiopulmonary comorbidities and whose condition was managed successfully with embolization and biliary *** embolization and biliary drainage may provide an alternative option to manage this complicated condition.

Povacoat affecting solid-state polymorphic changes of indomethacin after co-evaporation from different types of solvents via conventional and microwave drying techniques

The effect of Povacoat on the solid state characteristics and polymorphic transformation of γ-form of indomethacin(γ-IMC) after solvent co-evaporation via two drying methods was investigated by thermoanalytical and FTIR spectroscopic studies. The types of solvents(water,ethanol, and water:ethanol(1:9 ～ 2:8, v/v)), weight ratios of Povacoat:IMC(4:1 ～ 4:10, w/w),and drying methods(air-drying and microwave-drying) were selected as variables in this study. A physical mixture of Povacoat and γ-IMC was also carried out by neat grinding *** results indicate that there was no interaction between Povacoat and γ-IMC or no polymorphic change of γ-IMC after the neat co-grinding process. Since Povacoat was only soluble in water or <30% ethanol aqueous solution but γ-IMC was only soluble in organic solvents,different solvents and drying methods might significantly influence the solid-state characteristics of IMC formed in the presence of Povacoat. It was found that the amorphous IMC was formed in the Povacoat/IMC evaporates after dissolving both Povacoat and γ-IMC in water:ethanol(mixed ratio=1:9 and 2:8, v/v) and co-evaporation via microwave-drying, whereas the α-IMC was formed in the Povacoat/IMC evaporates via the air-drying method. In addition, the predominate α-IMC formation was only found in all the Povacoat/IMC evaporates with different weight ratios prepared by previously dissolving in water:ethanol(1:9, v/v) and co-evaporation via air-drying, but the amorphous IMC formation after co-evaporation via microwave-drying was obtained except the 4:10(w/w) weight ratio. The thermal effect of microwave irradiation and the presence of Povacoat might be responsible for the amorphous IMC formation in the Povacoat/IMC evaporates.

Carbon neutrality has been proposed as a solution for the current severe energy and climate crisis caused by the overuse of fossil fuels, and machine learning(ML) has exhibited excellent performance in accelerating related research owing to its powerful capacity for big data processing. This review presents a detailed overview of ML accelerated carbon neutrality research with a focus on energy management, screening of novel energy materials, and ML interatomic potentials(MLIPs), with illustrations of two selected MLIP algorithms: moment tensor potential(MTP) and neural equivariant interatomic potential(NequIP). We conclude by outlining the important role of ML in accelerating the achievement of carbon neutrality from global-scale energy management, unprecedented screening of advanced energy materials in massive chemical space, to the revolution of atomicscale simulations of MLIPs, which has the bright prospect of applications.

A Program of Art Education Exploring Life Issues --By Two Nudity Artworks

The purpose of this essay is to design an art education program based on post-modernist concepts to lead students to contemplate the meaning of life and build their insight about visual imagery of artworks, by looking at two nudity artworks--Edward Munch＇s ＂Puberty＂ and Frida Kahlo＇s ＂The Broken Column＂. The program will look at metaphors and the significance of life in these two artworks. Over and above exploring relative philosophical, aesthetic, historical, hermeneutic, and semiotic domains, the program will be designed in such a way that the intrinsic quality of art will be emphasized. The research method is a visual narrative approach, including image interpretation. Its purpose is to encourage students to consider how they experience life, the emotional elements of life, as well as the spiritual progression, physiological development, anxiety and suffering of life, etc.. The program will also provide students with diverse information and potential ways of thinking about and discussing the issues of life and art by using questions in a learning list. This program will be employed and executed over two hours of class time. It could become the general education teaching material or art curriculum.

Fabrication and study of supercapacitor electrodes based on oxygen plasma functionalized carbon nanotube fibers

Dry-spun Carbon Nanotube(CNT)fibers were surface-modified by atmospheric pressure oxygen plasma functionalization using a well controlled and continuous *** fibers were characterized by scanning electron microscopy(SEM),Raman spectroscopy,and X-ray Photoelectron Spectroscopy(XPS).It was found from the conducted electrochemical measurements that the functionalized fibers showed a 132.8% increase in specific capacitance compared to non-functionalized ***-adsorption test and the obtained Randles-Sevcik plot demonstrated that the oxygen plasma functionalized fibers exhibited increased surface *** was further established by Brunauer-Emmett-Teller(BET)measurements that the surface area of the CNT fibers was increased from 168.22 m^2/g to 208.01 m^2/g after plasma *** pore size distribution of the fibers was also altered by this *** improved electrochemical data was attributed to enhanced wettability,increased surface area,and the presence of oxygen functional groups,which promoted the capacitance of the *** supercapacitors were fabricated from the oxygen plasma functionalized CNT fiber electrodes using different electrolyte *** devices with functionalized electrodes exhibited excellent cyclic stability(93.2% after 4000 cycles),flexibility,bendability,and good energy *** 0.5 m A/cm^2,the EMIMBF4 device revealed a specific capacitance,which is 27% and 65%greater than the specific capacitances of devices using EMIMTFSI and H2SO4 electrolytes,*** practiced in this work plasma surface processing can be employed in other applications where fibers,yarns,ribbons,and sheets need to be chemically modified.

Diverticulosis of the jejunum with intestinal obstruction: A case report

A diagnosis of intestinal diverticulosis is difficult to make pre-operatively because the clinical symptoms are usually non-specific. We report the case of a 70-year-old man who had suffered from three episodes of intestinal obstruction in 1 year. He experienced dull pain and a sensation of fullness over the whole abdomen. The symptoms did not improve after conservative *** presumptive diagnosis was intestinal obstruction, and an exploratory laparotomy found diverticulosis of the proximal jejunum, with an adhesion band formed from the base of one diverticulum. Strangulation of a segment of the jejunum resulted from the internal herniation caused by the band. The band was removed and the proximal jejunum segmentally resected. His postoperative course was uneventful.

Image Reconstructions from Limit Views and Angle Coverage Data for a Stationary Multi-Pinhole SPECT System

The goal of this paper is to investigate different reconstruction methods for solving the limited angle problem in reconstructing the projection data from a stationary multi-pinhole system based on a three-head clinical single photon emission computed tomography （SPECT） camera. Three iterative recon- struction approaches were studied including maximum likelihood expectation maximization （MLEM）, maximum a posteriori expectation maximization （MAPEM） with a smoothing prior, and an alternating optimization schemes from MLEM and total variation （TV） optimization. A three-headed multipinhole SPECT was simulated with apertures of nine 1-mm pinholes each, and covered scanning volume of 6-mm diameter. The reconstructions were optimized for various iterations based on visual inspections, and finally 20 iterations were applied for each method. For both MLEM-TV and MAPEM-TV, various initial reconstructions before TV optimization were studied. The smoothing parameter for MAPEM and the gradient descent constant for TV were also investigated through visual comparison. The preliminary results showed the 3 reconstruction methods generated compatible images, and can restore the images from projection data suffering limited angular sampling. However, MLEM was noisy for low-count and highly limited angle data, and thus suitable smoothing in MAPEM alleviated this problem, initial reconstructions were necessary for better edge enhancement in TV. The conclusion is that TV might be potential in producing more edge-enhanced images if all parameters were optimized.

Extensive retroperitoneal and right thigh abscess in a patient with ruptured retrocecal appendicitis:An extremely fulminant form of a common disease

As a disease commonly encountered in daily practice, acute appendicitis is usually diagnosed and managed easily with a low mortality and morbidity rate. However, acute appendicitis may occasionally become extraordinarily complicated and life threatening. A 56-year-old man, healthy prior to this admission, was brought to the hospital due to spiking high fever, poor appetite, dysuria, progressive right flank and painful swelling of the thigh for 3 d. Significant inflammatory change of soft tissue was noted, involving the entire right trunk from the subcostal margin to the knee joint. Painful disability of the right lower extremity and apparent signs of peritonitis at the right lower abdomen were disclosed. Laboratory results revealed leukocytosis and an elevated C-reactive protein level. Abdominal CT revealed several communicated gas-containing abscesses at the right retroperitoneal region with mass effect, pushing the duodenum and the pancreatic head upward, compressing and encasing inferior vena cava, destroying psoas muscle and dissecting downward into the right thigh. Laparotomy and right thigh exploration were performed immediately and about 500 mL of frank pus was drained. A ruptured retrocecal appendix was the cause of the abscess. The patient fully recovered at the end of the third post-operation week. This case reminds us that acute appendicitis should be treated carefully on an emergency basis to avoid serious complications. CT scan is the diagnostic tool of choice, with rapid evaluation followed by adequate drainage as the key to the survival of the patient.
