Phase separation of GRP7 facilitated by FERONIAmediated phosphorylation inhibits mRNA translation to modulate plant temperature resilience

Changes in ambient temperature profoundly affect plant growth and performance.Therefore,the molecu-larbasis of plant acclimation to temperature fluctuation is of great interest.In this study,we discovered that GLYCINE-RICH RNA-BINDING PROTEIN 7(GRP7)contributes to cold and heat tolerance in Arabidopsis thaliana.We found that exposure to a warm temperature rapidly induces GRP7 condensates in planta,which can be reversed by transfer to a lower temperature.Cell biology and biochemical assays revealed that GRP7 undergoes liquid-liquid phase separation(LLPS)in vivo and in vitro.LLPS of GRP7 in the cyto-plasm contributes to the formation of stress granules that recruit RNA,along with the translation machinery component eukaryotic initiation factor 4E1(elF4E1)and the mRNA chaperones COLD SHOCK PROTEIN 1(CSP1)and CSP3,to inhibit translation.Moreover,natural variations in GRP7 affecting the residue phos-phorylated by the receptorkinase FERONIA alter its capacity to undergo LLPS and correlate with the adap-tation of some Arabidopsis accessions to a widertemperature range.Taken together,ourfindings illustrate the role of translational control mediated by GRP7 LLPS to confer plants with temperature resilience.

The albumin-bilirubin(ALBI)score,which was proposed to assess the prognosis of patients with hepatocellular carcinoma,has gradually been extended to other liver diseases in recent years,including primary biliary cholangitis,liver cirrhosis,hepatitis,liver transplantation,and liver injury.The ALBI score is often compared with classical scores such as the Child-Pugh and model for end-stage liver disease scores or other noninvasive prediction models.It is widely employed because of its immunity to subjective evaluation indicators and ease of obtaining detection indicators.An increasing number of studies have confirmed that it is highly accurate for assessing the prognosis of patients with chronic liver disease;additionally,it has demonstrated good predictive performance for outcomes beyond survival in patients with liver diseases,such as decompensation events.This article presents a review of the application of ALBI scores in various non-malignant liver diseases.

Molecular investigation into the transformation of recalcitrant dissolved organic sulfur in refinery sour water during stripping process

Refinery sour water primarily originates from the tops of towers in various units and coker condensate,and cannot be discharged directly to a wastewater treatment plant due to high levels of chemical oxygen demand(COD) and organic sulfur contents. Even after the recovery of H2S from the sour water by the stripping process, the effluent still contains a high concentration of dissolved organic sulfur(DOS), which can have a huge bad influence. While chemical composition of dissolved organic matter(DOM) in refinery wastewater has been extensively studied, the investigation of recalcitrant DOS from sour waters remains unclear. In the present study, chemical composition of sour water DOMs(especially DOS) was investigated using fluorescence spectroscopy(excitation-emission matrix, EEM) and mass spectrometry,including gas chromatography-mass spectrometry(GC-MS) and high-resolution Orbitrap MS. The GC-MS and EEM results showed that volatile and low-aromaticity compounds were effectively removed during the stripping process, while compounds with high hydrophilicity and humification degree were found to be more recalcitrant. The Orbitrap MS results showed that weak-polar oxygenated sulfur compounds were easier to be removed than oxygenated compounds. However, the effluent still contained significant amounts of sulfur-containing compounds with multiple sulfur atoms, particularly in the form of highly unsaturated and aromatic compounds. The Orbitrap MS/MS results of CHOS-containing compounds from the effluent indicate that the sulfur atoms may exist as sulfonates, disulfide bonds, thioethers. Understanding the composition and structure of sour water DOS is crucial for the development of effective treatment processes that can target polysulfide compounds and minimize their impact on the environment.

Cross-Modal Consistency with Aesthetic Similarity for Multimodal False Information Detection

With the explosive growth of false information on social media platforms, the automatic detection of multimodalfalse information has received increasing attention. Recent research has significantly contributed to multimodalinformation exchange and fusion, with many methods attempting to integrate unimodal features to generatemultimodal news representations. However, they still need to fully explore the hierarchical and complex semanticcorrelations between different modal contents, severely limiting their performance detecting multimodal falseinformation. This work proposes a two-stage detection framework for multimodal false information detection,called ASMFD, which is based on image aesthetic similarity to segment and explores the consistency andinconsistency features of images and texts. Specifically, we first use the Contrastive Language-Image Pre-training(CLIP) model to learn the relationship between text and images through label awareness and train an imageaesthetic attribute scorer using an aesthetic attribute dataset. Then, we calculate the aesthetic similarity betweenthe image and related images and use this similarity as a threshold to divide the multimodal correlation matrixinto consistency and inconsistencymatrices. Finally, the fusionmodule is designed to identify essential features fordetectingmultimodal false information. In extensive experiments on four datasets, the performance of the ASMFDis superior to state-of-the-art baseline methods.

Biomimetic porous silicon oxycarbide ceramics with improved specific strength and efficient thermal insulation

Considering the challenge of aerodynamic heating,the development of high-performance insulating ce-ramic materials with lightweight and low thermal conductivity is crucially important for aerospace vehi-cles to achieve flight at high speed for a long time.In this work,macro-porous silicon oxycarbide(SiOC)ceramics with directional pores(DP-SiOC)(mean pore size of 88.1μm)were prepared using polysiloxane precursors via freeze casting and photocrosslinking,followed by pyrolysis.The DP-SiOC samples were lightweight(density∼0.135 g cm^(-3))with a porosity of 90.4%,which showed good shapability through the molding of polysiloxane precursors.The DP-SiOC samples also exhibited an ultra-low thermal con-ductivity of 0.048 W(m K)^(-1)at room temperature,which can also withstand heat treatment at 1200°C for 1 h.In addition,scaffolds with triply periodic minimal surfaces(TPMS)were fabricated using digital light processing(DLP)printing,which was further filled with polysiloxane precursors for increasing the strength of DP-SiOC.The TPMS scaffolds filled with macro-porous SiOC ceramics(TPMS-DP-SiOC)showed good integration between TPMS and macro-pore structures,which had a porosity∼75%and high specific strength of 9.73×10^(3)N m kg^(-1).The thermal conductivity of TPMS-DP-SiOC samples was 0.255 W(m K)^(-1)at room temperature.The biomimetic TPMS-DP-SiOC ceramics developed in this study are likely used for thermal protection systems.

Application of immune checkpoint inhibitors and microsatellite instability in gastric cancer

In this editorial we comment on the article by Li published in the recent issue of the World Journal of Gastroenterology.We focus specifically on the application of immune checkpoint inhibitors(ICIs)and microsatellite instability(MSI)in gastric cancer(GC).The four pillars of GC management have long been considered,including surgery,chemotherapy,radiotherapy and targeted therapy.However,immunotherapy has recently emerged as a“fifth pillar”,and its use is rapidly expanding.There are four principal strategies for tumor immunotherapy:ICIs,tumor vaccines,adoptive immunotherapy and nonspecific immunomodulators.Of them,ICIs are the most advanced and widespread type of cancer immunotherapy for GC.Recent breakthrough results for ICIs have paved the way to a new era of cancer immunotherapy.In particular,inhibition of the PD-1/PD-L1 axis with ICIs,including nivolumab and pembrolizumab,has emerged as a novel treatment strategy for advanced GC.Unfortunately,these therapies are sometimes associated with often subtle,potentially fatal immune-related adverse events(irAEs),including dermatitis,diarrhea,colitis,endocrinopathy,hepatotoxicity,neuropathy and pneumonitis.We must be aware of these irAEs and improve the detection of these processes to prevent inappropriate discharges,emergency department revisits,and downstream complications.Recent studies have revealed that MSI-high or mismatch-repair-deficient tumors,regardless of their primary site,have a promising response to ICIs.So,it is important to detect MSI before applying ICIs for treatment of GC.

The Enhanced Period Map and Equivariant Deformation Quantizations of Nilpotent Orbits

In a previous paper,the author and his collaborator studied the problem of lifting Hamil-tonian group actions on symplectic varieties and Lagrangian subvarieties to their graded deformation quantizations and apply the general results to coadjoint orbit method for semisimple Lie groups.Only even quantizations were considered there.In this paper,these results are generalized to the case of general quantizations with arbitrary periods.The key step is to introduce an enhanced version of the(truncated)period map defined by Bezrukavnikov and Kaledin for quantizations of any smooth sym-plectic variety X,with values in the space of Picard Lie algebroid over X.As an application,we study quantizations of nilpotent orbits of real semisimple groups satisfying certain codimension condition.

The MYC2-PUB22-JAZ4 module plays a crucial role in jasmonate signaling in tomato

Jasmonates(JAs),a class of lipid-derived stress hormones,play a crucial role across an array of plant physiological processes and stress responses.Although JA signaling is thought to rely predominantly on the degradation of specific JAZ proteins by SCF^(COI1),it remains unclear whether other pathways are involved in the regulation of JAZ protein stability.Here,we report that PUB22,a plant U-box type E3 ubiquitin ligase,plays a critical role in the regulation of plant resistance against Helicoverpa armigera and other JA responses in tomato.Whereas COI1 physically interacts with JAZ1/2/5/7,PUB22 physically interacts with JAZ1/3/4/6.PUB22 ubiquitinates JAZ4 to promote its degradation via the 26S proteasome pathway.Importantly,we observed that pub22 mutants showreduced resistance to H.armigera,whereas jaz4 single mutants and jaz1 jaz3 jaz4 jaz6 quadruple mutants have enhanced resistance.The hypersensitivity of pub22 mutants to herbivores could be partially rescued by JAZ4 mutation.Moreover,we found that expression of PUB22 can be transcriptionally activated by MYC2,thus forming a positive feedback circuit in JA signaling.We noticed that the PUB22-JAZ4 module also regulates other JA responses,including defense against B.cinerea,inhibition of root elongation,and anthocyanin accumulation.Taken together,these results indicate that PUB22 plays a crucial role in plant growth and defense responses,together with COI1-regulated JA signaling,by targeting specific JAZs.

Plant RNA-binding proteins:Phase separation dynamics and functional mechanisms underlying plant development and stress responses

RNA-binding proteins(RBPs)accompany RNA from synthesis to decay,mediating every aspect of RNA metabolism and impacting diverse cellular and developmental processes in eukaryotes.Many RBPs undergo phase separation along with their bound RNA to form and function in dynamic membraneless biomolecular condensates for spatiotemporal coordination or regulation of RNA metabolism.Increasing evidence suggests that phase-separating RBPs with RNA-binding domains and intrinsically disordered regions play important roles in plant development and stress adaptation.Here,we summarize the current knowledge about how dynamic partitioning of RBPs into condensates controls plant development and enables sensing of experimental changes to confer growth plasticity under stress conditions,with a focus on the dynamics and functional mechanisms of RBP-rich nuclear condensates and cytoplasmic granules in mediating RNA metabolism.We also discuss roles of multiple factors,such as environmental signals,protein modifications,and N6-methyladenosine RNA methylation,in modulating the phase separation behaviors of RBPs,and highlight the prospects and challenges for future research on phase-separating RBPs in crops.

Effects of rafting on the meso deformation and fracture behaviour of a Ni-based single crystal superalloy at room temperature: In-situ observation and simulation

Microstructural rafting of Ni-based single-crystal(SC) superalloys is inevitable at elevated temperatures during long-term service with mechanical loading, which significantly affects the mechanical behaviour of the material. In this study, the effects of rafting on the mesodeformation and fracture behaviour of a Ni-based SC superalloy under cyclic and tensile loads were investigated using in situ scanning electron microscopy(SEM), digital image correlation(DIC), and crystal plasticity finite element method(CP-FEM) simulations. The results indicated that the tensile strength decreased significantly in the rafted specimens. In the cyclic tests, both the virgin and rafted specimens showed an increase in the maximum shear strain with cycle number. The interaction of cross-slip bands was captured by in situ SEM-DIC around the micro-notch of the virgin specimens during the tensile test, while a more homogeneous local deformation field was observed in the rafting specimens. In addition,the fracture behaviour was strongly influenced by the rafting morphology. The crack exhibited instantaneous and long-range fracture features along the octahedral plane as it propagated in the rafting specimen, whereas it deflected over a short distance between the crystallographic planes at an early stage in the virgin specimen, which is consistent with the CP-FEM results.Furthermore, the CP-FEM results for the crack initiation direction on(111) dominant slip plane were consistent with the in situ SEM observations.
