FOXO1-miR-506 axis promotes chemosensitivity to temozolomide and suppresses invasiveness in glioblastoma through a feedback loop of FOXO1/miR-506/eTS1/FOXO1

To explore the role of forkhead box protein O1(FOXO1)in the progression of glioblastoma multiforme(GBM)and related drug resistance,we deciphered the roles of FOXO1 and miR-506 in proliferation,apoptosis,migration,invasion,autophagy,and temozolomide(TMZ)sensitivity in the U251 cell line using in vitro and in vivo *** viability was tested by a cell counting kit-8(CCK8)kit;migration and invasion were checked by the scratching assay;apoptosis was evaluated by terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNeL)staining and flow *** construction of plasmids and dual-luciferase reporter experiment were carried out to find the interaction site between FOXO1 and *** was done to check the protein level in tumors after the in vivo *** found that the FOXO1-miR-506 axis suppresses GBM cell invasion and migration and promotes GBM chemosensitivity to TMZ,which was mediated by ***1 upregulates miR-506 by binding to its promoter to enhance transcriptional ***-506 could downregulate e26 transformation-specific 1(eTS1)expression by targeting its 3'-untranslated region(UTR).Interestingly,eTS1 promoted FOXO1 translocation from the nucleus to the cytosol and further suppressed the FOXO1-miR-506 axis in GBM ***,both miR-506 inhibition and eTS1 overexpression could rescue FOXO1 overactivation-mediated TMZ chemosensitivity in mouse *** study demonstrated a negative feedback loop of FOXO1/miR-506/eTS1/FOXO1 in GBM in regulating invasiveness and ***,the above axis might be a promising therapeutic target for GBM.

Relation Between the eventual Continuity and the e-property for Markov-Feller Semigroups

We investigate some relations between two kinds of semigroup regularities, namely the e-property and the eventual continuity, both of which contribute to the ergodicity for Markov processes on Polish *** precisely, we prove that for Markov-Feller semigroup in discrete time and stochastically continuous MarkovFeller semigroup in continuous time, if there exists an ergodic measure whose support has a nonempty interior,then the e-property is satisfied on the interior of the support. In particular, it implies that, restricted on the support of each ergodic measure, the e-property and the eventual continuity are equivalent for the discrete-time and the stochastically continuous continuous-time Markov-Feller semigroups.

Cancer immunotherapy,mainly including immune checkpoints-targeted therapy and the adoptive transfer of engineered immune cells,has revolutionized the oncology landscape as it utilizes patients’own immune systems in combating the cancer *** cells escape immune surveillance by hijacking the corresponding inhibitory pathways via overexpressing checkpoint *** checkpoints,such as CD47,CD24,MHC-I,PD-L1,STC-1 and GD2,have emerged as essential checkpoints for cancer immunotherapy by functioning as“don’t eat me”signals or interacting with“eat me”signals to suppress immune *** checkpoints link innate immunity and adaptive immunity in cancer *** ablation of these phagocytosis checkpoints,as well as blockade of their signaling pathways,robustly augments phagocytosis and reduces tumor *** all phagocytosis checkpoints,CD47 is the most thoroughly studied and has emerged as a rising star among targets for cancer ***47-targeting antibodies and inhibitors have been investigated in various preclinical and clinical ***,anemia and thrombocytopenia appear to be formidable challenges since CD47 is ubiquitously expressed on ***,we review the reported phagocytosis checkpoints by discussing their mechanisms and functions in cancer immunotherapy,highlight clinical progress in targeting these checkpoints and discuss challenges and potential solutions to smooth the way for combination immunotherapeutic strategies that involve both innate and adaptive immune responses.

Graph Signal Reconstruction from Low-Resolution Multi-Bit Observations

Low hardware cost and power consumption in information transmission, processing and storage is an urgent demand for many big data problems, in which the high-dimensional data often be modelled as graph *** paper considers the problem of recovering a smooth graph signal by using its low-resolution multi-bit quantized observations. The underlying problem is formulated as a regularized maximum-likelihood optimization and is solved via an expectation maximization scheme. With this scheme, the multi-bit graph signal recovery(MB-GSR) is efficiently implemented by using the quantized observations collected from random subsets of graph nodes. The simulation results show that increasing the sampling resolution to 2 or 3 bits per sample leads to a considerable performance improvement, while the energy consumption and implementation costs remain much lower compared to the implementation of high resolution sampling.

Performance analysis and multi-objective optimization based on a modified irreversible Stirling cycle

The efficiency of the actual Stirling engine is much lower than the ideal Carnot cycle *** obtain more precise efficiency for Stirling engines,this paper proposes a modified Stirling cycle with a more accurate heat transfer process in Stirling engines based on a thermodynamic function called available *** finite-time thermodynamic method is used to analyze the model performance under constant heat source temperature,finite temperature difference heat transfer,and incomplete regenerative processes.A new polytropic process is introduced to model the heat transfer between the working fluid and external heat sources in which only heat above ambient temperature is converted into technical *** regenerator is divided into numerous smaller heat reservoirs with individual temperature to analyze the incomplete regenerative *** expressions of the output power and thermal efficiency are obtained based on the modified irreversible Stirling cycle,and the effects of irreversible losses are analyzed to evaluate the performance of the proposed *** indicate that the efficiency of the modified cycle is much lower than that of the ideal Stirling cycle with an isothermal *** the increase of the average heat transfer temperature difference,there exists an optimum value for the power of the irreversible *** optimum power of the model was obtained for varying thermodynamic parameters by optimizing the average heat transfer temperature difference between the hot and cold *** optimize the irreversible model,the multi-objective optimization analysis is carried out based on NSGA-Ⅱ,which results in an optimized output power of 40.87 kW and an optimized thermal efficiency of 44%.

Numerical investigations of the interactions between bubble induced shock waves and particle based on OpenFOAM

The presence of particles and the shock waves generated by the cavitation bubbles can significantly affect the safety and the performance of hydrodynamic *** the present paper,the shock waves generated by cavitation bubble collapsing near the particle are numerically investigated based on the OpenFOAM together with the numerical schlieren for the shock wave *** numerical results reveal that the stand-off distance is one of the paramount factors affecting the interactions between the particle and the shock *** different kinds of shock waves(e.g.,bubble-inception,jet formation,particle reflected and jet-split shock waves)are observed during the bubble collapsing near the *** stand-off distance smaller than 0.5 or larger than 1.1,the maximum pressure at particle surface generated by the bubble growth can surpass those of the collapse stage.

An integrative mathematical model of microwave ion thruster for mHz-frequency thrust noise analysis

Many high-precision space missions need thrusters to produce thrust with low noise to compensate for disturbances and ensure satellite platform stability. Microwave ion thruster is characterized with a wide thrust range and potential for these missions. A cost-effective and accurate mathematical model is crucial for mHz-frequency thrust noise analysis and feedback controller design. The Particle-In-Cell(PIC) and global models are two common simulation tools. The PIC model is characterized with high accuracy but huge computation cost, which is difficult to analyze long-time performance characteristics. Now, the global model is only used for the discharge chamber with low accuracy and cannot reflect ion extraction properties. In this paper, an integrative mathematical model is built for a 1-cm microwave ion thruster and can reflect ion beamlet divergence and impingement on the Accelerator Grid(AG). Simulation results show good agreement with experiments at 0.06 sccm. However, the model demonstrates worse consistency with experiments when the flux increases to 0.1 sccm, which may be because the influence of neutral gas on the electron Cyclotron Resonance(eCR) is not considered in the model. A long-time(1000 s) simulation is conducted with this model under 35 μN. It takes 3 hrs, and the thrust noise reaches 1 μN/Hz^(0.5) at 1 mHz.

Design and Gait Planning of a Worm-inspired Metameric Robot for Pipe Crawling

The earthworm has been attracted much attention in the research and development of biomimetic robots due to their unique locomotion mechanism,compact structure,and small motion *** paper presents a new design and prototype of a worm-inspired metameric robot with a movement pattern similar to that of *** robot consists of multiple telescopic modules connected in series through joint *** telescopic module mimics the contraction and elongation motion modes of the earthworm segments.A kinematic and dynamic analysis is conducted on the telescopic module,and an input torque calculation method is provided to ensure sufficient friction between the robot and the pipe *** gait modes of the prototype robot for straight and turning locomotion are introduced,and these modes are extended to robots constructed by different numbers of telescopic *** addition,a method is proposed to increase the friction between the robot and the pipe wall in the aforementioned gait modes without changing the robot structure,thereby improving the robot’s motion ability in *** theoretical model of gait modes has also been validated through gait *** findings of this paper would provide a useful basis for the design,modeling,and control of future worm inspired robots.

Multi‐object tracking in autonomous driving is a non‐linear *** better address the tracking problem,this paper leveraged an unscented Kalman filter to predict the object's *** the association stage,the Mahalanobis distance was employed as an affinity metric,and a Non‐minimum Suppression method was designed for *** the detections fed into the tracker and continuous‘predicting‐matching’steps,the states of each object at different time steps were described as their own continuous *** conducted extensive experiments to evaluate tracking accuracy on three challenging datasets(KITTI,nuScenes and Waymo).The experimental results demon-strated that our method effectively achieved multi‐object tracking with satisfactory ac-curacy and real‐time efficiency.

Wellbore-heat-transfer-model-based optimization and control for cooling downhole drilling fluid

To address the two critical issues of evaluating the necessity of implementing cooling techniques and achieving real-time temperature control of drilling fluids underground in the current drilling fluid cooling technology,we first established a temperature and pressure coupled downhole heat transfer model,which can be used in both water-based and oil-based drilling ***,fourteen factors,which could affect wellbore temperature,were *** on the standard deviation of the downhole temperature corresponding to each influencing factor,the influence of each factor was *** influencing factors that can be used to guide the drilling fluid's cooling technology were drilling fluid thermal conductivity,drilling fluid heat capacity,drilling fluid density,drill strings rotation speed,pump rate,viscosity,ROP,and injection *** nondominated sorting genetic algorithm was used to optimize these six parameters,but the optimization process took 182 *** these eight parameters'influence rules with the nondominated sorting genetic algorithm can reduce the optimization time to 108 ***,the downhole temperature has been demonstrated to increase with the inlet temperature increasing linearly under quasi-steady *** this law and PID,the downhole temperature can be controlled,which can reduce the energy for cooling the surface drilling fluid and can ensure the downhole temperature reaches the set value as soon as possible.
