Comprehensive cultivation of the broiler gut microbiota guides bacterial isolation from chickens

Chicken gut microbiota plays an important role in maintaining their physiological health. However, the cultivability of chicken gut microbiota is not well understood, limiting the exploration of certain key gut bacteria in regulating intestinal health and nutritional metabolism. This study aimed to examine the cultivability of chicken cecal microbiota and to provide guidance for future chicken gut microbiota cultivation. A total of 58 different culture conditions were applied to culture broiler cecal microbiota, and the culture-dependent(CD;pooled colonies form each plate) and culture-independent(CI; broiler cecal contents) samples were collected for 16S r RNA gene sequencing and microbial analysis. The CD methods detected higher microbial richness(3,636 vs 2,331 OTUs) than CI methods, and the recovery rates of bacterial OTUs and genera reached 43.6% and 68.9%, respectively. The genera of Bacteroides(19.9%), Alistipes(11.0%) and Barnesiella(10.7%) were highly abundant detected by CI methods, however, there occupied a small proportion(<1.0%) of total cultured microbiota in CD methods. We then developed reference figures and tables showing optimal cultivation conditions for different gut bacteria ***, 81 different lactic acid bacteria strains covering 5 genera were isolated, and 15 strains had less than 97.0% similarity to known bacteria in the national center for biotechnology information(NCBI) online database. Overall, this study provides preliminary guidance in culturing specific gut microbiota from chickens, which will contribute to future studies to characterize the biological functions of key microbes in chicken nutritional metabolism and health.

Perioperative neurocognitive disorder(PND) is a significant neurological complication in aging perioperative patients that impacts postoperative cognition. PND is currently diagnosed through cognitive function testing, which is limited by its subjectivity and time requirements. Thus, the identification of biomarkers to assess PND onset is a priority to identify at-risk individuals and enable interventions and treatments to patient outcomes. This article synthesizes expert perspectives on brain aging and PND, presents the latest clinical evidence on PND biomarkers(imaging, electroencephalography, and molecular biomarkers), and delves into the relationship between PND and other age-related cognitive disorders. Thorough review of PND research identified several biomarkers with high sensitivity and specificity,offering a solid scientific foundation to predict and diagnose PND. These biomarkers not only enhance diagnostic accuracy for clinicians but also provide opportunities for earlier intervention and more effective treatment, potentially enhancing patient outcomes and quality of life.

Because of weak dissipation effects, swells generated by fierce storms can propagate across an entire ocean basin;therefore, observing swell generation and decay and retrieving storm characteristics from a swell by satellite remote sensing are possible. In this study, based on the dispersion relation and geometrical optics principle, we used SAR wave mode data from 2003 to 2010 provided by GlobWave to track swells with peak wavelengths of more than 300 m to locate a storm-generated far-traveling swell and present the swell field related to this "static" origin. Through a comparison with ECMWF wind datasets, we conducted validations and explored some conditions that cause misjudgments in swell origins. Finally, we obtained the spatiotemporal distribution characteristics of satellite-observed swell origins(i.e., the fierce wind condition) and their evolution. This work can be used as a reference for wave models, providing early swell warnings, determining air-sea surface interactions, and determining global climate change.

The uranyl ion(UO22+) poses high risks to human health and the environment, hence its detection and monitoring is of utmost significance. However, the development of an ultra-sensitive, high-efficiency and convenient approach for on-site detection of UO22+ remains a challenge. Herein, a reliable and reusable surface-enhanced Raman spectroscopy(SERS)-based microfluidic biosensor was developed for rapid detection of UO22+ in real samples. The detection protocol involved the reaction of 5′-Rhodamine B(RhB)-labeled double-stranded DNA for UO22+-specific DNAzyme-cleavage reaction in a U-shaped microchannel. Then, the reaction products were delivered into three parallel samples for high-throughput tests by SERS biochips,where 3 D ZnO-Ag mesoporous nanosheet arrays(MNSs) were modified with a single-stranded DNA(ssDNA). The ssDNAwas sequence-complementary with the 5′-RhB-labeled cleaved-stranded DNA(csDNA) from the reaction products. By the hybridization of ssDNA and csDNA, the signal probe RhB was fixed close to the surface of the ZnO-Ag MNSs to enhance the Raman signal. The limit of detection for UO22+ with the microfluidic-SERS biosensor was 3.71×10-15 M. An over 20,000-fold selectivity towards UO22+ response was also achieved in the presence of 15 other metal ions. The high-throughput microfluidicSERS biosensor operated well for practical UO22+ detection, with excellent recoveries in contaminated river and tap water from95.2% to 106.3%(relative standard deviation(RSD)<6.0%, n=6). Although the SERS-based microfluidic biosensor developed in this study was deployed for the detection of UO22+, the reusable and high-efficiency system may be expanded to the detection of other analytes on-site.

为探究青海省不同气候分区(东部农业区、柴达木盆地、环青海湖区、三江源区)水面蒸发的变化特征及其影响因子,基于FAO-56 Penman-Monteith(PM)公式,利用青海省50个国家气象站点2018-2022年的气象资料,建立多元回归模型计算青海省各气候区水面蒸发量,分析不同时间尺度上青海省不同气候分区水面蒸发的变化特征,并利用通径分析方法对各气候区水面蒸发的影响因子进行剖析。结果表明:水面蒸发年际变化范围为2.58~2.92 mm/d,4个气候区均在2022年达到最大值;季节变化表现为夏季>春季>秋季>冬季;月变化最大值均出现在7月,为柴达木盆地>东部农业区>环青海湖区>三江源区。东部农业区,气象因子对水面蒸发的综合决定能力排序为VPD>R_(n)>n>WS,VPD对水面蒸发变化的直接作用最大(决策系数为0.75),其次是WS,R_(n)主要通过n路径对水面蒸发变化产生间接作用(间接作用系数为0.47);柴达木盆地,各因子决策系数排序为VPD>R_(n)>RH_(mean)>WS>T_(min),R_(n)对水面蒸发变化的直接作用最大(决策系数为0.50),其次是T_(min),VPD对水面蒸发的间接作用最大(间接作用系数为0.51),通过R_(n)路径间接影响水面蒸发,WS通过T_(min)路径间接影响水面蒸发(间接作用系数为0.18),RH_(mean)对水面蒸发的影响最小且为负效应;环青海湖区,VPD、R_(n)和WS是驱动水面蒸发变化的最主要因子且起直接作用,决策系数分别为0.62、0.69和0.05,表明R_(n)对水面蒸发变化促进作用比VPD明显;三江源区,VPD对水面蒸发的直接影响最大(决策系数为1.05),其次是WS。T_(mean)可通过VPD、R_(n)和RH_(mean)路径间接影响水面蒸发(间接作用系数为0.76),R_(n)和n分别通过T_(mean)和R_(n)路径对水面蒸发变化产生间接作用(间接作用系数分别为0.48和0.69),VPD、R_(n)、WS、T_(mean)和n对水面蒸发均有增进作用,而RH_(mean)对水面蒸发的影响最小且对水面蒸发变化有抑制作用(决策系数为-0.001)。总体而言,VPD和R_(n)是影响青海省水面蒸发的主导因子。

Significant progress has been made in machine learning with large amounts of clean labels and static ***,in many real-world applications,the data often changes with time and it is difficult to obtain massive clean annotations,that is,noisy labels and time series are faced *** example,in product-buyer evaluation,each sample records the daily time behavior of users,but the long transaction period brings difficulties to analysis,and salespeople often erroneously annotate the user’s purchase *** a novel setting,to our best knowledge,has not been thoroughly studied yet,and there is still a lack of effective machine learning *** this paper,we present a systematic approach RTS both theoretically and empirically,consisting of two components,Noise-Tolerant Time Series Representation and Purified Oversampling ***,we propose reducing label noise’s destructive impact to obtain robust feature representations and potential clean ***,a novel learning method based on the purified data and time series oversampling is adopted to train an unbiased *** analysis proves that our proposal can improve the quality of the noisy data *** experiments on diverse tasks,such as the house-buyer evaluation task from real-world applications and various benchmark tasks,clearly demonstrate that our new algorithm robustly outperforms many competitive methods.
