AIM To investigate micro(mi)R-34 a-antagonizing circular(circ)RNA that underlies hepatocellular *** The effect of circ RNA on mi R-34 a was recognized by the mi RNA response element(MRE), and validated by the dual-luciferase reporter assay. Its association with hepatocellular steatosis was investigated in Hep G2-based hepatocellular steatosis induced by free fatty acids(FFAs; 2:1 oleate:palmitate) stimulation. After normalization of the steatosis-related circRNA by expression vector, analysis of mi R-34 a activity,peroxisome proliferator-activated receptor(PPAR)α level, and expression of downstream genes were carried out so as to reveal its impact on the mi R-34 a/PPARα regulatory system. Both triglyceride(TG) assessment and cytopathological manifestations uncovered the role of circRNA in miR-34 a-dependent *** Bioinformatic and functional analysis verified circRNA_0046366 to antagonize the activity of mi R-34 a via MRE-based complementation. In contrast to its lowered level during FFA-induced hepatocellular steatosis, circ RNA_0046366 up-regulation abolished the mi R-34 a-dependent inhibition of PPARα that played a critical role in metabolic signaling pathways. PPARα restoration exerted transcriptional improvement to multiple genes responsible for lipid metabolism. TGspecific lipolytic genes [carnitine palmitoyltransferase 1 A(CPT1 A) and solute-carrier family 27 A(SLC27 A)] among these showed significant increase in their expression levels. The circ RNA_0046366-related rebalancing of lipid homeostasis led to dramatic reduction of TG content, and resulted in the ameliorated phenotype of hepatocellular *** Dysregulation of circ RNA_0046366/mi R-34 a/PPARα signaling may be a novel epigenetic mechanism underlying hepatocellular steatosis. circ RNA_0046366 serves as a potential target for the treatment of hepatic steatosis.

The multi-layer cylindrical helicoidal fiber structure(MCHFS)exists widely in biological materials such as bone and wood at the *** typically function as reinforcing elements to enhance the toughness of *** this study,we establish a shear lag-based pullout model of the cylindrical helicoidal fiber(CHF)for investigating interlayer stress transfer and debonding behaviors,with implications regarding the underlying toughening mechanism of *** on the shear lag assumptions,analytical solutions for the stress and displacement fields of the MCHFS during the pullout are derived by considering the CHF as a cylindrically monoclinic material and verified through the 3D finite element *** is found that the helical winding of CHF results in both axial and hoop interlayer shear *** the helical winding angle and the elastic moduli of the fiber and matrix have significant influences on interlayer stress *** work reveals a new interlayer stress transfer mechanism in the MCHFS existing widely in biological materials.

Background Impaired exercise capacity is one of the most common clinical manifestations in patients with chronic heart failure （CHF）. The severity of reduced exercise capacity is an indicator of disease prognosis. The aim of the current study was to investigate the association between left heart size and mass with exercise capacity. Methods A total of 74 patients were enrolled in the study, with 37 having congestive heart failure （left ventricular ejection fraction （LVEF）〈0.45） and the other 37 with coronary heart disease （by coronary angiography） serving as the control group （LVEF 〉0.55）. Echocardiography and cardiopulmonary exercise test were performed. The multiply linear regression model was used to evaluate the association between echocardiogrphic indices and exercise capacities. Results The study showed that left ventricular end diastolic / systolic diameter （LVEDD/LVESD）, left atrial diameter （LAD） and left ventricular mass index （LVMI） were significantly enlarged in patients with chronic heart failure compared with controls （P 〈0.01）. The VO2AT, Peak VO2, Load AT, and Load Peak in chronic heart failure patients were also significantly reduced compared with controls （P 〈0.05）, VENCO2 slope was increased in patients with chronic heart failure （P 〈0.01）. Multivariate linear regression analysis indicated that the patients＇ exercise capacity was significantly associated with the left heart size and mass, however, the direction and/or strength of the associations sometimes varied in chronic heart failure patients and controls. Load AT correlated negatively with LVEDD in chronic heart failure patients （P=0.012）, while Load AT correlated positively with LVEDD in control patients （P=0.006）. VENCO2 slope correlated positively with LAD （B=0.477, P 〈0.0001） in chronic heart failure patients, while the VENCO2 slope correlated negatively with LAD in control patients （P=0.009）. Soil acidifications become one of the main causes restricting the sustainable development of agriculture and causing issues of agricultural product *** order to explore the effect of different acidification on soil cadmium(Cd)availability,soil pot culture and hydroponic(soil potting solution extraction)were applied,and non-invasive micro-test technique(NMT)was *** three different soil acidification processes were simulated,including direct acidification by adding sulfuric acid(AP1),acid rain acidification(AP2)by adding artificial simulated acid rain and excessive fertilization acidification by adding(NH_(4))_(2)SO_(4)(AP3).The results showed that for direct acidification(AP1),DTPA-Cd concentration in field soils in Liaoning(S1)and Zhejiang(S2)increased by 0.167-0.217 mg/kg and 0.181-0.346 mg/kg,respectively,compared with control *** soil pH decreased by 0.45 units in S1,the Cd content of rice stems,leaves and roots increased by 0.48 to 6.04 mg/kg and 2.58 to 12.84mg/kg,respectively,When the pH value of soil S1 and S2 decreased by 0.20 units,the average velocity of Cd^(2+)at 200μm increased by 10.03-33.11 pmol/cm~2/sec and 21.33-52.86pmol/cm^(2)/sec,respectively,and followed the order of AP3>AP2>*** summary,different acidification measures would improve the effectiveness of Cd,under the same pH reduction condition,fertilization acidification increased Cd availability most significantly.

The aims of the study were to determine the distribution of inhibin and its α subunit in some rat tissues by an immunohistochemical method of streptavidin-biotin complex （SABC） and to assess the transport mechanism of inhibin by investigating the localization of inhibin and α subunits in the central nervous system （mainly in hypothalamus and pituitary） of ovariectomized rats. Investigations on the extragonadal tissues of ovariectomized rats showed positive expression of inhibin and its α subunit in heart, kidney, spleen, pancreatic gland cells, but no positive reaction sites were seen in lung, liver, submaxillary gland, and adrenal gland. After injection with inhibin α subunit fragment or inhibin extract, positive reaction sites were observed in hypothalamus and pituitary of ovariectomized rats by SABC. Inhibin and its subunit was present in a wide variety of nonreproductive organs and tissues, and its expression was tissue specific, which indicated that inhibin might play a role in regulating tissue function through autocrine/paracrine mechanisms. Inhibin dimer and α subunit could be transported through the BBB by the method of “separation and reconstruction”.
