Deciphering the molecular mechanisms of Simiaowan in the treatment of hyperuricemia: in vivo and in silico approaches

Background:Simiaowan(SMW),a well-known traditional Chinese medicine,has been employed to treat hyperuricemia(HUA)and gout for ***,the bioactive components and underlying mechanisms have not been *** objective of this study was to identify the active components and potential mechanisms of SMW by integrating pharmacological experimentation,serum pharmacochemistry,network pharmacology and molecular ***:HUA rats modelling by high-fat/high-sugar diet and potassium oxonate/adenine oral administration were used to evaluate the pharmacodynamic effects of ***-Q-Exactive-MS/MS was employed to detect the bioactive components present in SMW-containing *** pharmacology and molecular docking were utilized to elucidate the potential targets and underlying ***:SMW effectively ameliorated HUA rats via the inhibition of uric acid(UA)production,promotion of UA excretion,improvement of lipid and glucose metabolic abnormalities,antioxidant,anti-inflammatory and anti-insulin resistance effects.A total of 73 compounds detected in SMW-containing serum were identified as potential active components,with alkaloids,flavonoids,organic acids,and terpenoids emerging as the primary active *** 203 corresponding targets were obtained as SMW anti-HUA/gout targets,which mainly participated in apoptosis,insulin resistance,TNF,PI3K-Akt,HIF-1,NF-κB,MAPK,IL-17 and TLR signaling *** docking indicated that active compounds(***,phellodendrine,quercetin,formononetin,ferulic acid)had superior binding abilities to the key targets(*** carrier family 22 member 12(URAT1),solute carrier family 22 member 6(OAT1),ATP-binding cassette sub-family G member 2(ABCG2),solute carrier family 2,facilitated glucose transporter member 9(GLUT9),xanthine dehydrogenase/oxidase(XDH),transcription factor p65(RELA),toll-like receptor 4(TLR4),prostaglandin G/H synthase 2(PTGS2),caspase-3(CASP3),insulin(INS)).Conclusion:S

The formation of new and functional cardiomyocytes requires a 3-step process:dedifferentiation,proliferation,and redifferentiation,but the critical genes required for efficient dedifferentiation,proliferation,and redifferentiation remain *** our study,a circular trajectory using single-nucleus RNA sequencing of the pericentriolar material 1 positive(PCM1^(+))cardiomyocyte nuclei from hearts 1 and 3 days after surgery-induced myocardial infarction(MI)on postnatal Day 1 was reconstructed and demonstrated that actin remodeling contributed to the dedifferentiation,proliferation,and redifferentiation of cardiomyocytes after *** identified four top actin-remodeling regulators,namely Tmsb4x,Tmsb10,Dmd,and Ctnna3,which we collectively referred to as *** expressed changes of 2D2P,using a polycistronic non-integrating lentivirus driven by Tnnt2(cardiac-specific troponin T)promoters(Tnnt2-2D2P-NIL),efficiently induced transiently proliferative activation and actin remodeling in postnatal Day 7 cardiomyocytes and adult ***,the intramyocardial delivery of Tnnt2-2D2P-NIL resulted in a sustained improvement in cardiac function without ventricular dilatation,thickened septum,or fatal arrhythmia for at least 4 *** conclusion,this study highlights the importance of actin remodeling in cardiac regeneration and provides a foundation for new gene-cocktail-therapy approaches to improve cardiac repair and treat heart failure using a novel transient and cardiomyocyte-specific viral construct.

The properties of the triplet excited state of [60]fullerene-containing cyclic sulphoxide have been investigated by time-resolved absorption spectroscopy. Transient absorption bands of [60]fullerene-containing cyclic sulphoxide showed two decay-components, which were attributed to triplet excited states of different spin multiplicity. The properties of photoexcited states of [60]fullerene-containing cyclic sulphoxide are also reported.

Formation of disinfection byproducts from accumulated soluble products of oleaginous microalga after chlorination

When microalgae are simultaneously applied for wastewater treatment and lipid production, soluble algal products （SAP） should be paid much attention, as they are important precursors for formation of disinfection byproducts （DBPs）, which have potential risks for human health. Chlorella sp. HQ is an oleaginous microalga that can generate SAP during growth, especially in the exponential phase. This study investigated the contribution of SAP from Chlorella sp. HQ to DBP formation after *** predominant DBP precursors from SAP were identified with the 3D excitation-emission matrix fluorescence. After chlorination, a significant reduction was observed in the fluorescence intensity of five specific fluorescence regions, particularly aromatic proteins and soluble microbial by-product-like regions, accompanied with slight shifting of the peak. The produced DBPs were demonstrated to include trihalomethanes and haloacetic acids. As the algal cultivation time was extended in wastewater, the accumulated SAP strengthened the formation of DBPs. The trend for DBP formation was as follows： chloroform〉dichloroacetic acid〉trichloroacetic acid.

Electrochemical co-deposition of reduced graphene oxide-gold nanocomposite on an ITO substrate and its application in the detection of dopamine

The simultaneous deposition of rGO and gold nano structures has been achieved by electrodeposition from mixed solutions containing graphene oxide(GO)and a gold *** electron microscope(SEM),Raman spectroscopy and atomic force microscopy(AFM)have been employed to reveal the morphology,uniformity and practical stability of the nanocomposite films on the indium tin oxide(ITO)*** AFM data showed heights of tens of nanometers of the nanocomposite,suggesting that multilayers of rGO with gold nanoparticles had been formed as a result of the electrochemical *** pulse voltammetry(DPV),as a widely used analytical technique,has been carried out on the rGO-Au/ITO electrode for the quantitative detection of dopamine(DA).The detection limit(S/N=3)for the determination of DA was evaluated as 0.6μM.

Elastoplastic pipe-soil interaction analyses of partially-supported jointed water mains

Water distribution networks are essential components of water supply systems. The combination of pipe structural deterioration and mechanics leads to the failure of pipelines. A physical model for estimating the pipe failure must include both the pipe deterioration model and mechanics model. Winkler pipe-soil interaction (WPSI), an analytical mechanics model developed by Rajani and Tesfamariam (2004), takes external and internal loads, temperature changes, loss of bedding support, and the elastoplastic effect of soil into consideration. Based on the WPSI model, a method to evaluate the elastic and plastic areas was proposed in the present study. An FEM model based on pipe-soil interaction (PSI) element was used to verify the analytical model. Sensitivity analyses indicate that the soft soil, long pipe and high temperature induced the axial plastic deformation more likely, which, however, may not occur in normal scenarios. The soft soil, pipes in small diameters, long unsupported bedding are prone to form flexural plastic area. The results show that the pipes subjected to the same loads have smaller stresses in the elastoplastic analysis than elastic analysis. The difference, however, is slight.

Modeling method and preliminary model of Asteroid Toutatis from Chang'E-2 optical images

Shape modeling is fundamental to the analysis of dynamic environment and motion around asteroid. Chang＇E- 2 successfully made a flyby of Asteroid 4179 Toutatis and obtained plenty of high-resolution images during the mis- sion. In this paper, the modeling method and preliminary model of Asteroid Toutatis are discussed. First, the optical images obtained by Chang＇E-2 are analyzed. Terrain and silhouette features in images are described. Then, the modeling method based on previous radar model and preliminary information from optical images is proposed. A preliminary polyhedron model of Asteroid Toutatis is established. Finally, the spherical harmonic coefficients of Asteroid Toutatis based on the polyhedron model are obtained. Some parameters of model are analyzed and compared. Although the model proposed in this paper is only a preliminary model, this work offers a valuable reference for future high-resolution models.

Anti-inflammation Effects of Sinomenine on Macrophages through Suppressing Activated TLR4/NF-kB Signaling Pathway

Novel bake hardening mechanism for bainite-strengthened complex phase steel

In the automobile industry,stamping and paint baking processes are used to strengthen components,and this not only saves costs,but also further reduces the bodyweight of *** this work,the bake hardening mechanism of the complex phase(CP)steel CP980 was explored by comparing it with that of DP1180,which has a clear bake hardening mechanism and a carbon content similar to that of *** analyzing the bake hardening response and the microstructural changes of the two steels,we found that the bake hardening process of CP980 was divided into three *** the first two stages,the carbon atoms diffused into dislocations to form Cottrell atmosphere pinning dislocations,and excess car-bon atoms formed carbon clusters or low-temperature carbide pinning dislocations that were similar to *** the third stage,the dislocation acted as rapid channels for carbon diffusion,and fine cementite gradually formed when the carbon clusters gathered at the dislocations as precursors,resulting in pre-cipitation *** novel bake-hardening(BH)mechanism is crucial for obtaining a comprehensive understanding of the strain-baking behavior of advanced high-strength steals(AHSS).
