Integration of gut microbiota and metabolomics analysis to unravel the neuroprotective effects of Hydroxy-α-sanshool on Alzheimer's Disease

Alzheimer's disease （AD） is a common neurodegenerative disease， and oxidative stress induced by Aβ deposition is the key cause of neuronal apoptosis in AD patient. Hydroxy-α-sanshool （HAS） is the predominant flavour substances in the pericarp of Zanthoxylum bungeanum Maxim. （Rutaceae family）， which is a known condiment food and herbal medicine in China. We attempt to comprehensively elucidate the pathway and related mechanisms of HAS therapy on AD through in vivo and in vitro experiments. The results showed that HAS could inhibit cell apoptosis， suppress the reactive oxygen species （ROS）， and increase the antioxidative enzymes in Aβ1-42induced HT22 cells. In vivo， HAS attenuated the learning and memory impairment in senescence-accelerated mouse prone 8 （SAMP8） mice， and protected neuronal cells in hippocampus. Interestingly， the 16S rRNA gene sequencing results showed HAS could restore the richness and diversity of the intestinal microbiota， reduce the abundance of Lactobacillus， and increase the abundance of Bateroides and Parabacteroides. Moreover， the contents of 10 metabolites were significantly changed after HAS treatment， which was beneficial to treat AD. In conclusion， HAS could inhibit apoptosis in nerve cells by attenuating Aβ induced oxidative stress level， while it can also influence the metabolic pathways and affect the gut microbiota composition of AD mice.

Effect of low-temperature ausforming on bainitic transformation and mechanical properties in ultrahigh-strength bainitic steel

Low-temperature ausforming(LT-AF)prior to bainitic transformation leads to a noticeable acceleration of bainitic transformation kinetics;however,the effect of LT-AF on the retained austenite(RA)features and the resulting mechanical properties is still ***-AF was applied to ultrahigh-strength bainitic steel before *** deformation behavior and the resulting dislocation substructures were investigated by thermomechanical simulator and transmission electron microscopy(TEM).The planar dislocation structures produced during deformation at 350℃ accelerate the bainitic transformation kinetics during isothermal *** effect of LT-AF on the bainitic transformation kinetics and the features of RA was elucidated via dilatometer measurement,TEM,scanning electron microscopy,and X-ray *** is observed that LT-AF not only retains more RA content but also facilitates improved RA *** trend is mainly due to the large amounts of planar dislocations in RA and bainitic laths inherited from undercooled austenite caused by LT-AF,the decrease in bainitic sheaves size,and the increase in filmy RA content compared to the sample without ausforming.A large fraction of filmy RA with high stability and the refinement of bainitic sheaves obtained by LT-AF remarkably enhance the strain hardening capacity and achieve significantly better ductility compared to the directly austempered sample.

Fluorinated metal-organic framework for methane purification from a ternary CH_(4)/C_(2)H_(6)/C_(3)H_(8)mixture

Herein we present a fluorinated metal-organic framework of{(Me_(2)NH_(2))[Ni_(3)(μ_(3)-OH)(CF_(3)-BPDC-CF_(3))_(3)(tpt)]}_(n)(1)constructed from 2,2'-bis(trifluoromethyl)biphenyl-4,4'-dicarboxylic(CF_(3)-BPDC-CF_(3)^(2-))and 2,4,6-tri(4-pyridyl)-1,3,5-triazine(tpt)ligands,which is developed for separating propane(C_(3)H_(8))and ethane(C_(2)H_(6))from natural *** 1 preferentially adsorbs C_(3)H_(8)and C_(2)H_(6)over CH_(4)demonstrated by gas adsorption *** presence of trifluoromethyl groups on the biphenyl-4,4'-dicarboxylic ligands facilitates the highly polarized micropore environments for compound 1,thus providing suitable micorpores for capturing the C_(3)H_(8)and C_(2)H_(6)molecules with larger polarizabilities and sizes compared to CH_(4)*** dynamic mixture breakthrough experiments showed that compound 1 can separate C_(3)H_(8)and C_(2)H_(6)from the ternary CH_(4)/C_(2)H_(6)/C_(3)H_(8)mixtures efficiently,endowing compound 1 with excellent methane purification ability.

The risk model construction of the genes regulated by H3K36me3 and H3K79me2 in breast cancer

Abnormal histone modifications(HMs) can promote the occurrence of breast cancer. To elucidate the relationship between HMs and gene expression, we analyzed HM binding patterns and calculated their signal changes between breast tumor cells and normal cells. On this basis, the influences of HM signal changes on the expression changes of breast cancer-related genes were estimated by three different methods. The results showed that H3K79me2 and H3K36me3 may contribute more to gene expression changes. Subsequently, 2109 genes with differential H3K79me2 or H3K36me3 levels during cancerogenesis were identified by the Shannon entropy and submitted to perform functional enrichment analyses. Enrichment analyses displayed that these genes were involved in pathways in cancer, human papillomavirus infection, and viral carcinogenesis. Univariate Cox, LASSO, and multivariate Cox regression analyses were then adopted, and nine potential breast cancer-related driver genes were extracted from the genes with differential H3K79me2/H3K36me3 levels in the TCGA cohort. To facilitate the application, the expression levels of nine driver genes were transformed into a risk score model, and its robustness was tested via time-dependent receiver operating characteristic curves in the TCGA dataset and an independent GEO dataset. At last, the distribution levels of H3K79me2 and H3K36me3 in the nine driver genes were reanalyzed in the two cell lines and the regions with significant signal changes were located.

Exact analytical solutions are good candidates for studying and explaining the dynamics of solitons in nonlinear *** further extend the region of existence of spin solitons in the nonlinearity coefficient space for the spin-1 Bose-Einstein *** types of spin soliton solutions can be obtained,and they exist in different *** analysis and numerical simulation results indicate that three types of spin solitons are stable against weak *** non-integrable properties of the model can induce shape oscillation and increase in speed after the collision between two spin *** results further enrich the soliton family for non-integrable models and can provide theoretical references for experimental studies.

A Possible X-ray Quasi-periodic Oscillation in the Narrow-line Seyfert 1 Galaxy Mrk 142

A possible quasi-periodic oscillation(QPO) at frequency 7.045 × 10^(-5) Hz is found in the narrow-line Seyfert 1galaxy Mrk 142 in the data of XMM-Newton collected on 2020 April *** find that the QPO signal is statistically significantly larger than the 5σ level and highly coherent with quality factor Q > 5 at the 0.3–10 keV band by using the method of the Lomb–Scargle Periodogram,the Weighted Wavelet Z-transform and the *** analyze the data in 0.3–0.6 keV,0.6–1 keV,1–3 keV and 3–10 keV energy bands,and find obvious QPO signals at 0.3–0.6 keV and 1–3 keV *** then analyze the time-average spectra and time variability at the QPO frequency of 7.045 × 10^(-5) Hz,and use a model to fit *** find that the QPO signal mainly comes from the X-ray hot corona.

The role of cAMP-dependent protein kinase A in the formation of long-term memory in Bactrocera dorsalis

The cAMP-dependent protein kinase A(PKA)signaling pathway has long been considered critical for long-term memory(LTM)*** studies have mostly focused on the role of PKA signaling in LTM induction by multiple spaced conditioning with less attention to LTM induction by a single ***,we conducted behavioral-pharmacology,enzyme immunoassay and RNA interference experiments to study the role of the PKA signaling pathway in LTM formation in the agricultural pest Bactrocera dorsalis,which has a strong memory capacity allowing it to form a two-day memory even from a single conditioning *** found that either blocking or activating PKA prior to conditioning pretreatment affected multiple spaced LTM,and conversely,they did not affect LTM formed by single *** was further confirmed by enzyme-linked immunosorbent assay(ELISA)and silencing of the protein kinase regulatory subunit 2 and catalytic subunit *** together,these results suggest that activating PKA during memory acquisition helps to induce the LTM formed by multiple spaced conditioning but not by a single *** findings challenge the conserved role of PKA signaling in LTM,which provides a basis for the greater diversity of molecular mechanisms underlying LTM formation across species,as well as possible functional and evolutionary implications.
