Bio-inspired macrostructure array(MAA,size:submillimeter to millimeter scale)materials with special wettability(MAAMs-SW)have attracted significant research attention due to their outstanding performance in many applications,including oil repellency,liquid/droplet manipulation,anti-icing,heat transfer,water collection,and oil–water separation.In this review,we focus on recent developments in the theory,design,fabrication,and application of bio-inspired MAAMs-SW.We first review the history of the basic theory of special wettability and discuss representative structures and corresponding functions of some biological surfaces,thus setting the stage for the design and fabrication of bio-inspired MAAMs-SW.We then summarize the fabrication methods of special wetting MAAs in terms of three categories:additive manufacturing,subtractive manufacturing,and formative manufacturing,as well as their diverse functional applications,providing insights into the development of these MAAMs-SW.Finally,the challenges and directions of future research on bio-inspired MAAMs-SW are briefy addressed.Worldwide efforts,progress,and breakthroughs from surface engineering to functional applications elaborated herein will promote the practical application of bio-inspired MAAMs-SW.

Lipocalins exhibit functional diversity,including roles in retinol transport,invertebrate cryptic coloration,and stress response.However,genome-wide identification and characterization of lipocalin in the insect lineage have not been thoroughly explored.Here,we found that a lineage-specific expansion of the lipocalin genes in Lepidoptera occurred in large part due to tandem duplication events and several lipocalin genes involving insect coloration were expanded more via tandem duplication in butterflies.A comparative analysis of conserved motifs showed both conservation and divergence of lepidopteran lipocalin family protein structures during evolution.We observe dynamic changes in tissue expression preference of paralogs in Bombyx mori,suggesting differential contribution of paralogs to specific organ functions during evolution.Subcellular localization experiments revealed that lipocalins localize to the cytoplasm,nuclear membrane,or nucleus in BmN cells.Moreover,several lipocalin genes exhibited divergent responses to abiotic and biotic stresses,and 1 lipocalin gene was upregulated by 30o fold in B.mori.These results suggest that lipocalins act as signaling components in defense responses by mediating crosstalk between abiotic and biotic stress responses.This study deepens our understanding of the comprehensive characteristics of lipocalins in insects.

Despite the mounting attention being paid to vegetation growth and their driving forces for water-limited ecosystems, the relative contributions of atmospheric and soil moisture dryness stress on vegetation growth are an ongoing debate. Here we comprehensively compare the impacts of high vapor pressure deficit(VPD) and low soil water content(SWC) on vegetation growth in Eurasian drylands during 1982–2014.The analysis indicates a gradual decoupling between atmospheric dryness and soil dryness over this period,as the former has expanded faster than the latter. Moreover, the VPD–SWC relation and VPD–greenness relation are both non-linear, while the SWC–greenness relation is near-linear. The loosened coupling between VPD and SWC, the non-linear correlations among VPD–SWC-greenness and the expanded area extent in which SWC acts as the dominant stress factor all provide compelling evidence that SWC is a more influential stressor than VPD on vegetation growth in Eurasian drylands. In addition, a set of 11 Earth system models projected a continuously growing constraint of SWC stress on vegetation growth towards 2100. Our results are vital to dryland ecosystems management and drought mitigation in Eurasia.

Background:High proliferative rate of cancer cells requires autophagy to maintain nutrient supply and intracellular homeostasis.As a result,impairing autophagic flux could be a novel strategy of cancer therapy.Aims and Objectives:In this study,the mechanism of a xanthone derivative isolated from Garcinia mangostana,garcinone E(GE),was investigated.Materials and Methods:Fluorescence assay was used to observe the accumulation and location of autophagosome and lysosome.Flow cytometry with Lyso-tracker red,MDC,and AO staining were applied to evaluate the lysosome accumulation and cellular acidity.Western blot and RT-qPCR were performed to evaluate the protein and mRNA levels,respectively.Results:GE could cause enhancement of LC3 II and p62 and the accumulation of autophagosome and lysosome.Meanwhile,it limited the protein level of Rab7,increased lysosomal pH,and inhibited the maturation of lysosomal hydrolases such as Cathepsin L,therefore blockaded the fusion of autophagosome and lysosome.Moreover,GE acted as a TFEB modulator by downregulating its protein level,which might contribute to autophagy dysfunction in ovarian cancer cells.Conclusions:GE interfered autophagosome–lysosome fusion in cancer cells,which demonstrated its application as an autophagy regulator and a potential therapeutic agent.

How to improve machine learning models for lithofacies identification by practical and novel ensemble strategy and principles

Typically, relationship between well logs and lithofacies is complex, which leads to low accuracy of lithofacies identification. Machine learning (ML) methods are often applied to identify lithofacies using logs labelled by rock cores. However, these methods have accuracy limits to some extent. To further improve their accuracies, practical and novel ensemble learning strategy and principles are proposed in this work, which allows geologists not familiar with ML to establish a good ML lithofacies identification model and help geologists familiar with ML further improve accuracy of lithofacies identification. The ensemble learning strategy combines ML methods as sub-classifiers to generate a comprehensive lithofacies identification model, which aims to reduce the variance errors in prediction. Each sub-classifier is trained by randomly sampled labelled data with random features. The novelty of this work lies in the ensemble principles making sub-classifiers just overfitting by algorithm parameter setting and sub-dataset sampling. The principles can help reduce the bias errors in the prediction. Two issues are discussed, videlicet (1) whether only a relatively simple single-classifier method can be as sub-classifiers and how to select proper ML methods as sub-classifiers;(2) whether different kinds of ML methods can be combined as sub-classifiers. If yes, how to determine a proper combination. In order to test the effectiveness of the ensemble strategy and principles for lithofacies identification, different kinds of machine learning algorithms are selected as sub-classifiers, including regular classifiers (LDA, NB, KNN, ID3 tree and CART), kernel method (SVM), and ensemble learning algorithms (RF, AdaBoost, XGBoost and LightGBM). In this work, the experiments used a published dataset of lithofacies from Daniudi gas field (DGF) in Ordes Basin, China. Correlation between serum lipid profiles and cognitive impairment in old age:a cross-sectional study

Background The correlation between cognitive function and lipid profiles,including total cholesterol,low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)and triglycerides,is inconsistent.Aims This cross-sectional study investigated the association between serum lipid levels and the prevalence of cognitive impairment among community-dwelling older adults and explored this difference in association by gender and urban-rural residency.Methods Participants aged 65 and above in urban and rural areas were recruited between 2018 and 2020,selected from the Hubei Memory and Aging Cohort Study.Detailed neuropsychological evaluations,clinical examinations and laboratory tests were conducted in community health service centres.Multivariate logistic regression was used to analyse the correlation between serum lipid profiles and the prevalence of cognitive impairment.Results We identified 1336 cognitively impaired adults(≥65 years)-1066 with mild cognitive impairment and 270 with dementia-from 4746 participants.Triglycerides level was correlated with cognitive impairment in the total sample(χ2=6.420,p=0.011).In gender-stratified multivariate analysis,high triglycerides in males reduced the risk of cognitive impairment(OR:0.785,95%CI:0.623 to 0.989,p=0.040),and high LDL-C in females increased the risk of cognitive impairment(OR:1.282,95%CI:1.040 to 1.581,p=0.020).In both gender-stratified and urban-rural stratified multivariate analyses,high triglycerides reduced the risk of cognitive impairment in older urban men(OR:0.734,95%CI:0.551 to 0.977,p=0.034),and high LDL-C increased the risk of cognitive impairment in older rural women(OR:1.830,95%CI:1.119 to 2.991,p=0.016).Conclusions There are gender and urban-rural differences in the correlation of serum lipids with cognitive impairment.High triglycerides levels may be a protective factor for cognitive function in older urban men,while high LDL-C levels may be a risk factor for cognitive function in older rural women.

The deformation behavior and the microstruc-ture evolution of as-cast AZ91D magnesium alloy without pre-homogenization treatment were systematically inves-tigated. The flow stress behavior was studied by com-pression tests in strain rate range of 0.001-1.000 s^-1 and deformation temperature range of 220-380 ℃ with a maximum deformation strain of 60 %. The dependence of flow stress on deformation temperature and strain rate was described by hyperbolic sine constitutive equation. Through regression analysis, the average apparent activa- tion energy and coefficient of strain rate sensitivity were estimated to be 181.98 kJ.mol^-1 and 0.14, respectively. The results also reveal that the variation of peak stress depends on strain rate and deformation temperature. Microstructure observation shows that, at temperatures higher than 300 ℃ and strain rates lower than 0.01 s^-1, DRX developed extensively at the grain boundaries and in the core of coarse grains, resulting in a more homogeneous microstructure. Furthermore, the effects of strain, defor-mation temperature, strain rate, and eutectic β phase on the microstructure evolution of as-cast AZ91D magnesium alloy were discussed.

Understanding the relative roles of local environmental effects and spatial effects on phytoplankton community is of essential importance to study the biogeography of them at regional scale. However, the determinants that driving the biogeography of phytoplankton communities in the coastal area of northern Zhejiang still remained unclear. We surveyed phytoplankton community compositions in water columns associated with environmental and spatial influences across five subzones that geographically covering this region over four seasons. Diatoms and dinoflagellates were recorded as the main dominant groups and Coscinodiscus oculs-iridis, Coscinodiscus jonesianus, and Skeletonema costatum, were identified as the major abundant species existing in all seasons.Spatially structured environmental conditions, rather than pure spatial or environmental factors, substantially shaped the biogeography of phytoplankton community, with the former mainly comprised of water temperature,dissolved oxygen, phosphate, pH, and salinity, and the latter referring to a non-negligible factor. This study was the first integrated research that combining environmental filtering with spatial factors in structuring phytoplankton communities at a complete tempo-spatial scale. Our results may facilitate to the further study of harmful algal blooms early-warning in this region.

Systematic screening reveals synergistic interactions that overcome MAPK inhibitor resistance in cancer cells

Objective:Effective adjuvant therapeutic strategies are urgently needed to overcome MAPK inhibitor(MAPKi)resistance,which is one of the most common forms of resistance that has emerged in many types of cancers.Here,we aimed to systematically identify the genetic interactions underlying MAPKi resistance,and to further investigate the mechanisms that produce the genetic interactions that generate synergistic MAPKi resistance.Methods:We conducted a comprehensive pair-wise sgRNA-based high-throughput screening assay to identify synergistic interactions that sensitized cancer cells to MAPKi,and validated 3 genetic combinations through competitive growth,cell viability,and spheroid formation assays.We next conducted Kaplan-Meier survival analysis based on The Cancer Genome Atlas database and conducted immunohistochemistry to determine the clinical relevance of these synergistic combinations.We also investigated the MAPKi resistance mechanisms of these validated synergistic combinations by using co-immunoprecipitation,Western blot,qRTPCR,and immunofluorescence assays.Results:We constructed a systematic interaction network of MAPKi resistance and identified 3 novel synergistic combinations that effectively targeted MAPKi resistance(ITGB3+IGF1R,ITGB3+JNK,and HDGF+LGR5).We next analyzed their clinical relevance and the mechanisms by which they sensitized cancer cells to MAPKi exposure.Specifically,we discovered a novel protein complex,HDGF-LGR5,that adaptively responded to MAPKi to enhance cancer cell stemness,which was up-or downregulated by the inhibitors of ITGB3+JNK or ITGB3+IGF1R.Conclusions:Pair-wise sgRNA library screening provided systematic insights into elucidating MAPKi resistance in cancer cells.ITGB3-+IGF1R-targeting drugs(cilengitide+linsitinib)could be used as an effective therapy for suppressing the adaptive formation of the HDGF-LGR5 protein complex,which enhanced cancer stemness during MAPKi stress.
