The effect of spatial temperature gradient on the microstructural evolution of a 308L stainless steel during the directed energy deposition(DED)process was experimentally investigated.A novel cooling system was designed and incorporated to a DED system in order to control the temperature gradient along the deposition direction during *** deposition,the workpiece was placed on a lifting platform,and as the deposition process proceeded,the platform and workpiece were gradually lowered into cooling water so that the temperature gradient along the deposition direction could be controlled and maintained stable during the deposition *** microstructure characterization results indicated that a deposition strategy with higher G and G/R values(where G is temperature gradient and R is solidification rate)produced finer cellular grains that were better aligned with the deposition direction,while a deposition strategy with lower G and G/R values produced columnar grains with larger primary arm spacing and less aligned with the deposition direction.

Two eukaryotic vectors expressing 9 tandem repeats of human MUCI（VNTR）, VR1012-VNTR, and pEGFP-VNTR, were constructed by cloning VNTR gene into VR1012 and pEGFP, respectively. VNTR stably expressing murine Lewis lung carcinoma（LLC） cell line（VNTR^＋ LLC） was established by Lipofectamine-mediated transfection of pEGFP-VNTR into LLC cells. The EGFP expression was observed under a fluorescent microscope and VNTR expression in VNTR^＋ LLC cells was confirmed by means of Western blotting. A syngenic graft tumor model was generated by subcutaneous injection of VNTR^＋ LLC cells into C57/BL6 mice and tumor size increased rapidly with time and in a cell qumber dependent manner. VNTR mRNA expression in the tumor formed was confirmed by RT-PCR. After the third immunization mice were challenged subcutaneously with 5×10^5 VNTR^＋ LLC cells, a significant reduction of subcutaneous tumor growth was observed in the groups immunized with VNTR plasmid DNA compared with that in the groups immunized with the vector DNA alone. Thus, the suppression of subcutaneous tumor was antigen-specific. This model is useful for the development of tumor vaccines targeting MUCI VNTRs.

NH_(3)-SCR was one of the most promising de NO_x technologies and it has been widely applied in industrial NO_x ***,with the further development of energy transformation in power generation sector,the development of NH_(3)-SCR catalysts is facing some new *** is becoming an urgent problem to solve low catalytic activity and stability of NH_(3)-SCR catalysts at the working condition of ultra-low temperature(≤200℃)and high concentrations of H_(2)O+SO_(2) due to the gradual deployment of new energy power ***,the traditional coal-fired power plants would need flexible operation with the increasing share of renewable energy *** NH_(3)-SCR catalysts which were applied in coal-fired power industry would be requested to work in a wide temperature window from 200℃ to 500℃ in the near ***,in this review,we summarized the progress of NH_(3)-SCR catalysts in solving these different industrial problems in recent *** the research directions which were deserved to be focused on the development of NH_(3)-SCR catalysts for the energy transition of power generation sector are proposed.

Survivin, a novel member of inhibitor of apoptosis（IAP） protein family, is aberrantly expressed in cancer but undetectable in normal, differentiated adult tissues. The cancer-specific expression of survivin, coupled with its importance in inhibiting cell death and in regulating cell division makes it a useful diagnostic marker of cancer and a potential target for cancer treatment. Survivin cDNA amplified from the total RNA of 293 cells through RT-PCR was cloned into prokaryotic expression vector pRSET-B. The recombinant plasmid pRSET-B-Surv was expressed in *** BL21, and the relative molecule mass（Mr） of expressed fusion protein was approximately 21000. The recombinant protein was purified through Ni^2＋ affinity chromatography column and characterized by SDS-PAGE and Western blot. The purified recombinant protein was then injected into rabbits, and antisurvivin polyclonal antibody with a high titer was obtained.

Relationships between Terrain Features and Forecasting Errors of Surface Wind Speeds in a Mesoscale Numerical Weather Prediction Model

Numerical weather prediction(NWP)models have always presented large forecasting errors of surface wind speeds over regions with complex *** this study,surface wind forecasts from an operational NWP model,the SMS-WARR(Shanghai Meteorological Service-WRF ADAS Rapid Refresh System),are analyzed to quantitatively reveal the relationships between the forecasted surface wind speed errors and terrain features,with the intent of providing clues to better apply the NWP model to complex terrain *** terrain features are described by three parameters:the standard deviation of the model grid-scale orography,terrain height error of the model,and slope *** results show that the forecast bias has a unimodal distribution with a change in the standard deviation of *** minimum ME(the mean value of bias)is 1.2 m s^(-1) when the standard deviation is between 60 and 70 m.A positive correlation exists between bias and terrain height error,with the ME increasing by 10%−30%for every 200 m increase in terrain height *** ME decreases by 65.6%when slope angle increases from(0.5°−1.5°)to larger than 3.5°for uphill winds but increases by 35.4%when the absolute value of slope angle increases from(0.5°−1.5°)to(2.5°−3.5°)for downhill *** sensitivity experiments are carried out with a model output statistical(MOS)calibration model for surface wind speeds and ME(RMSE)has been reduced by 90%(30%)by introducing terrain parameters,demonstrating the value of this study.

The induction of human immunodeficiency virus（HIV）-specific T-cell response is generally considered as critical to the development of effective immunity to HIV type 1 （ HIV-1 ）. Recombinant Avipoxvirus vectors are used widely for vaccination against HIV-1, where the induction of a cytotoxic CD8 ＋ T-cell（CTL） response seems to be an important component of protective immunity. A recombinant fowlpox virus（rFPV/Gag-pol） expressing the Gag-pol protein of HIV was constructed and characterized. The specific expression protein in CEF cells infected by recombinant fowlpox and the specific antibody in the sera of mice immunized with rFPV were analyzed via Western-blot.

Polymer density-functional theories(PDFTs)have distinct advantages in the study of polyelectrolyte(PE)systems over experiments and molecular *** we give an introductory review of some PDFTs recently developed for PE *** start with a general formalism of PDFTs and its relation to the widely used polymer self-consistent field theory(SCFT),then explain the various correlations that are neglected in SCFT but can be accounted for in PDFTs,including those due to the excluded-volume interaction and chain connectivity of uncharged polymers,the electrostatic correlations of small ions,and the chain correlations in *** also list some applications of PDFTs for PE systems,and finally give some perspectives on future *** hope that our review can attract more researchers to apply and further develop PDFTs as a promising class of theoretical and computational tools.

Objective To investigate a potential relationship between Solute carrier family 30 （zinc transporter） member 8 （SLC3OAS） rs13266634 variant and efficacy of rosiglitazone or repaglinide in treating newly diagnosed Chinese type 2 diabetes patients. Methods A total of 209 diabetic patients without any antihyperglycemic history were recruited and treated with repaglinide or rosiglitazone randomly for 48 weeks （104 and 105 patients, respectively）. Anthropometric measurements and clinical laboratory tests were carried out before and after the treatment. An non-synonymous variant rs13266634 was genotyped by matrix-assisted laser desorption ionization-time of flight mass spectroscopy. Results Ninety-one patients in repaglinide group and ninety-three patients in rosiglitazone group completed the study. 6 value of homeostasis model assessment of beta cell function （HOMA-B） and 6 value of fasting proinsulin levels were statistically significant between three genotype groups （P=0.0149 and 0.0246, respectively） after rosiglitazone treatment. However, no genotype association was observed in the repaglinide or rosiglitazone group with other parameters. Conclusion The SLC3OA8 variant was associated with the efficacy of insulin sensitizer monotherapy on insulin secretion in patients with newly diagnosed type 2 diabetes mellitus in Shanghai, China.

The evolution of microstructure,texture,and magnetic properties with random texture,near-copper texture,weak near-cube texture,and strong λ fiber(/ND(normal direction))before rolling of non-oriented electrical steel was *** recrystallized hot bands with different textures but similar grain sizes were prepared by pre-annealing at low-temperature and high-temperature normalization *** was observed that the final annealed products exhibited similar recrystallized *** contrast,the final annealed product with more 2 fiber before rolling exhibited a stronger cube *** the fiber before rolling becoming stronger,the proportion of(111)deformed matrices became larger,which could be observed in the early recrystallization *** overwhelmingly dominant 2 orientation nuclei are formed in the(111)deformed matrix and become the dominant ***,the best magnetic properties are obtained in the products with strong 2 fiber before rolling,corresponding to the strong cube texture and low anisotropyparameter.
