Recent advances and remaining challenges in lung cancer therapy

Lung cancer remains the most common cause of cancer death.Given the continued research into new drugs and combination therapies,outcomes in lung cancer have been improved,and clinical benefits have been expanded to a broader patient population.However,the overall cure and survival rates for lung cancer patients remain low,especially in metastatic cases.Among the available lung cancer treatment options,such as surgery,radiation therapy,chemotherapy,targeted therapies,and alternative therapies,immunotherapy has shown to be the most promising.The exponential progress in immuno-oncology research and recent advancements made in the field of immunotherapy will further increase the survival and quality of life for lung cancer patients.Substantial progress has been made in targeted therapies using tyrosine kinase inhibitors and monoclonal antibody immune checkpoint inhibitors with many US Food And Drug Administration(FDA)-approved drugs targeting the programmed cell death ligand-1 protein(e.g.,durvalumab,atezolizumab),the programmed cell death-1 receptor(e.g.,nivolumab,pembrolizumab),and cytotoxic T-lymphocyte-associated antigen 4(e.g.,tremelimumab,ipilimumab).Cytokines,cancer vaccines,adoptive T cell therapies,and Natural killer cell mono-and combinational therapies are rapidly being studied,yet to date,there are currently none that are FDA-approved for the treatment of lung cancer.In this review,we discuss the current lung cancer therapies with an emphasis on immunotherapy,including the challenges for future research and clinical applications.

Prox1 Suppresses Proliferation and Drug Resistance of Retinoblastoma Cells via Targeting Notch1

Objective Retinoblastoma(RB)is a prevalent type of eye cancer in youngsters.Prospero homeobox 1(Prox1)is a homeobox transcriptional repressor and downstream target of the proneural gene that is relevant in lymphatic,hepatocyte,pancreatic,heart,lens,retinal,and cancer cells.The goal of this study was to investigate the role of Prox1 in RB cell proliferation and drug resistance,as well as to explore the underlying Notch1 mechanism.Methods Human RB cell lines(SO-RB50 and Y79)and a primary human retinal microvascular endothelial cell line(ACBRI-181)were used in this study.The expression of Prox1 and Notch1 mRNA and protein in RB cells was detected using quantitative real time-polymerase chain reaction(RT-qPCR)and Western blotting.Cell proliferation was assessed after Prox1 overexpression using the Cell Counting Kit-8 and the MTS assay.Drug-resistant cell lines(SO-RB50/vincristine)were generated and treated with Prox1 to investigate the role of Prox1 in drug resistance.We employed pcDNA-Notch1 to overexpress Notch1 to confirm the role of Notch1 in the protective function of Prox1.Finally,a xenograft model was constructed to assess the effect of Prox1 on RB in vivo.Results Prox1 was significantly downregulated in RB cells.Overexpression of Prox1 effectively decreased RB cell growth while increasing the sensitivity of drug-resistant cells to vincristine.Notch1 was involved in Prox1’s regulatory effects.Notch1 was identified as a target gene of Prox1,which was found to be upregulated in RB cells and repressed by increased Prox1 expression.When pcDNA-Notch1 was transfected,the effect of Prox1 overexpression on RB was removed.Furthermore,by downregulating Notch1,Prox1 overexpression slowed tumor development and increased vincristine sensitivity in vivo.Conclusion These data show that Prox1 decreased RB cell proliferation and drug resistance by targeting Notch1,implying that Prox1 could be a potential therapeutic target for RB.

Impact of Polymer matrix on Polymer Mechanochromism from Excited State Intramolecular Proton Transfer

Mechanochromic polymers based on non-covalent changes have attracted much attention recently.Herein,we report the impact of inter/intramolecular hydrogen bonds on polymer mechanochromism from the excited state intramolecular proton transfer (ESIPT) process.PhMz-NH2-OH and PhMz=2A are designed and obtained by simple and high-yield synthesis,and are connected into polyurethane and poly(methyl acrylate-co-2-ethylhexyl acrylate),respectively.In the initial state,the PhMz-NH2-OH@PU sample shows blue fluorescence from the excited enol form (E*) excitons,owing to intermolecular hydrogen bonds that interrupt the ESIPT reactions but the PhMz=2A@Pma-2-EA sample expresses cyan fluorescence belonging to the excited keto form (K*) emission,implying that the intramolecular hydrogen bonds matter.Furthermore,under stretching,external force can tune the emission of the PhMz=2A@Pma-2-EA sample from K* to E* state.Though external force can putatively still promote a bond rotation,ESIPT reactions remain equivalently interrupted in both the relaxed and stressed states in a hydrogen-bond donating environment.DFT calculation confirms the force-induced increase in dihedral angle for the transition of ESIPT-on/off.Thus,PhMz-NH2-OH@PU and PhMz=2A@Pma-2-EA showed disparate initial ESIPT states and further different responses/sensitivity to force.This study reports a novel and efficient strategy for enriching mechanochromic investigation and extending the applications of ESIPT reactions.

Photoinduced superlubricity on TiO_(2) surfaces

Superlubricity control is of great interest in both industry and scientific research,and several methods have been proposed to achieve this goal.In this work,ultraviolet(UV)light was introduced into titanium dioxide(TiO_(2))and silicon nitride(Si_(3)N_(4))tribosystems to accomplish photoinduced superlubricity.The friction coefficients(COFs)between Si_(3)N_(4) balls and TiO_(2) plates in the mixtures of sulfuric acid(H^(2)SO_(4))solution and glycerol solution were obviously reduced,and the system entered the superlubricity region(COFmation of silica(SiO_(2))layers on the surfaces of Si_(3)N_(4) balls and the elastohydrodynamic effects were determined to be fundamental to the low friction in this experiment,and the enhancement of the combination between the TiO_(2) surface and the hydroxy group of glycerol by UV illumination was the key to the photoinduced superlubricity in this system.These findings showed one method for achieving superlubricity by introducing a light field that could be further applied to special working conditions.

Quantifying Contribution of Recycled Moisture to Precipitation in Temperate Glacier Region,Southeastern Tibetan Plateau,China

Recycled moisture is an important indicator of the renewal capacity of regional water resources.Due to the existence of yulong Snow Mountain,Lijiang in yunnan Province,southeast of the Qinghai-Tibet Plateau,China,is the closest ocean glacier area to the equator in Eurasia.Daily precipitation samples were collected from 2017 to 2018 in Lijiang to quantify the effect of sub-cloud evaporation and recycled moisture on precipitation combined with the d-excess model during monsoon and non-monsoon periods.The results indicated that the d-excess values of precipitation fluctuated between–35.6‰and 16.0‰,with an arithmetic mean of 3.5‰.The local meteoric water line(LMWL)wasδD=7.91δ~(18)O+2.50,with a slope slightly lower than the global meteoric water line(GMWL).Subcloud evaporation was higher during the non-monsoon season than during the monsoon season.It tended to peak in march and was primarily influenced by the relative humidity.The source of the water vapour affected the proportion of recycled moisture.According to the results of the Hybrid Single-Particle Lagrangian Integrated Trajectory(HYSPLIT)model,the main sources of water vapour in Lijiang area during the monsoon period were the southwest and southeast monsoons.During the non-monsoon period,water vapour was transported by a southwesterly flow.The recycled moisture in Lijiang area between march and October 2017 was 10.62%.Large variations were observed between the monsoon and non-monsoon seasons,with values of 5.48%and 25.65%,respectively.These differences were primarily attributed to variations in the advection of water vapour.The recycled moisture has played a supplementary role in the precipitation of Lijiang area.

Epitranscriptomics focuses on the RNA-modification-mediated post-transcriptional regulation of gene expression. The past decade has witnessed tremendous progress in our understanding of the landscapes and biological functions of RNA modifications, as prompted by the emergence of potent analytical approaches. The hematopoietic system provides a lifelong supply of blood cells, and gene expression is tightly controlled during the differentiation of hematopoietic stem cells (HSCs). The dysregulation of gene expression during hematopoiesis may lead to severe disorders, including acute myeloid leukemia (AML). Emerging evidence supports the involvement of the mRNA modification system in normal hematopoiesis and AML pathogenesis, which has led to the development of small-molecule inhibitors that target N6-methyladenosine (m6A) modification machinery as treatments. Here, we summarize the latest findings and our most up-to-date information on the roles of m6A and N7-methylguanine in both physiological and pathological conditions in the hematopoietic system. Furthermore, we will discuss the therapeutic potential and limitations of cancer treatments targeting m6A.

IndRT-GCNets: Knowledge Reasoning with Independent Recurrent Temporal Graph Convolutional Representations

Due to the structural dependencies among concurrent events in the knowledge graph and the substantial amount of sequential correlation information carried by temporally adjacent events,we propose an Independent Recurrent Temporal Graph Convolution Networks(IndRT-GCNets)framework to efficiently and accurately capture event attribute information.The framework models the knowledge graph sequences to learn the evolutionary represen-tations of entities and relations within each period.Firstly,by utilizing the temporal graph convolution module in the evolutionary representation unit,the framework captures the structural dependency relationships within the knowledge graph in each period.Meanwhile,to achieve better event representation and establish effective correlations,an independent recurrent neural network is employed to implement auto-regressive modeling.Furthermore,static attributes of entities in the entity-relation events are constrained andmerged using a static graph constraint to obtain optimal entity representations.Finally,the evolution of entity and relation representations is utilized to predict events in the next subsequent step.On multiple real-world datasets such as Freebase13(FB13),Freebase 15k(FB15K),WordNet11(WN11),WordNet18(WN18),FB15K-237,WN18RR,YAGO3-10,and Nell-995,the results of multiple evaluation indicators show that our proposed IndRT-GCNets framework outperforms most existing models on knowledge reasoning tasks,which validates the effectiveness and robustness.

Spatial distribution of supra-permafrost groundwater in the Qinghai-Tibet Engineering Corridor using inversion models

Supra-permafrost groundwater(SPG)is a key factor that causes damage to highways and railways in the Qinghai-Tibet Engineering Corridor(QTEC).It is difficult to monitor SPG in the field due to their complex formation mechanisms and movement characteristics.Traditional single-site field monitoring studies limit the spatial and temporal precision of SPG spatial distribution.To determine the moisture content of shallow soils and the SPG distribution along the QTEC,this work employed the temperature vegetation dryness index and remote sensing models for groundwater table distribution models.The accuracies of the models were validated using mea-surements obtained from different sites in the corridor.In the permafrost zones of the QTEC,72%,22%and 6%of the SPG were located at depths of 0.5-1,1 m,respectively.Meanwhile,79.4%of the area along the Qinghai-Tibet Highway(QTH)(Xidatan-Tanggula)section contained SPG.In these sections with SPG,37.9%have an SPG table at depths of 0.5-0.8 m.This study preliminarily explored the SPG distribution in the QTEC with a 30 m resolution.The findings can help improve the spatial scale of SPG research,provide a basis for the analysis of the hydrothermal mechanisms,and serve as a guide in the assessment of operational risks and road structure designs.

Double-confined nanoheterostructure Sb/Sb2S3@Ti3C2Tx@C toward ultra-stable Li-/Na-ion batteries

Antimony-based materials with high capacities and moderate potentials are promising anodes for lithium-/-sodium-ion batteries.However,their tremendous volume expansion and inferior conductivity lead to poor structural stability and sluggish reaction kinetics.Herein,a doubleconfined nanoheterostructure Sb/Sb2S3@Ti3C2Tx@C has been fabricated through a solvothermal method followed by low-temperature heat treatment.The dual protection of‘‘MXene’’and‘‘carbon’’can better accommodate the volume expansion of Sb/Sb2S3.The strong covalent bond （Ti–S,Ti–O–Sb,C–O–Sb） can firmly integrate Sb-based material with Ti3C2Txand carbon,which significantly improves the structure stability.In addition,the carbon layer can restrain the oxidation of MXenes,and the nano-Sb/Sb2S3can facilitate electron/ion transport and suppress the restacking of MXenes.The heterogeneous interface between Sb and Sb2S3can further promote interfacial charge transfer.The MXene-Sb/Sb2S3@C-1 with the optimal Sb content shows high specific capacities,comparable rate properties and ultra-stable cycling performances (250 m Ah·g-1after 2500 cycles at 1 A·g-1for sodium-ion batteries).Ex situ X-ray diffractometer （XRD）test reveals the storage mechanism including the conversion and alloying process of MXene-Sb/Sb2S3@C-1.Cyclic voltammetry （CV） test results demonstrate that the pseudocapacitance behavior is dominant in MXene-Sb/Sb2S3@C-1,especially at large current.This design paves the way for exploring high-performance alloy-based/conversion-type anode for energy storage devices.

Causal role of immune cells in obstructive sleep apnea hypopnea syndrome:Mendelian randomization study

BACKGROUND Despite being one of the most prevalent sleep disorders,obstructive sleep apnea hypoventilation syndrome(OSAHS)has limited information on its immunologic foundation.The immunological underpinnings of certain major psychiatric diseases have been uncovered in recent years thanks to the extensive use of genome-wide association studies(GWAS)and genotyping techniques using highdensity genetic markers(e.g.,SNP or CNVs).But this tactic hasn't yet been applied to OSAHS.Using a Mendelian randomization analysis,we analyzed the causal link between immune cells and the illness in order to comprehend the immunological bases of OSAHS.AIM To investigate the immune cells'association with OSAHS via genetic methods,guiding future clinical research.METHODS A comprehensive two-sample mendelian randomization study was conducted to investigate the causal relationship between immune cell characteristics and OSAHS.Summary statistics for each immune cell feature were obtained from the GWAS catalog.Information on 731 immune cell properties,such as morphologic parameters,median fluorescence intensity,absolute cellular,and relative cellular,was compiled using publicly available genetic databases.The results'robustness,heterogeneity,and horizontal pleiotropy were confirmed using extensive sensitivity examination.RESULTS Following false discovery rate(FDR)correction,no statistically significant effect of OSAHS on immunophenotypes was observed.However,two lymphocyte subsets were found to have a significant association with the risk of OSAHS:Basophil%CD33dim HLA DR-CD66b-(OR=1.03,95%CI=1.01-1.03,P<0.001);CD38 on IgD+CD24-B cell(OR=1.04,95%CI=1.02-1.04,P=0.019).CONCLUSION This study shows a strong link between immune cells and OSAHS through a gene approach,thus offering direction for potential future medical research.
