Prospects in the application of ultrasensitive chromosomal aneuploidy detection in precancerous lesions of gastric cancer

Gastric cancer(GC)is a prevalent malignant tumor within the digestive system,with over 40%of new cases and deaths related to GC globally occurring in *** advancements in treatment modalities,such as surgery supplemented by adjuvant radiotherapy or chemotherapeutic agents,the prognosis for GC remains *** targeted therapies and immunotherapies are currently under invest-igation,but no significant breakthroughs have been *** have indicated that GC is a heterogeneous disease,encompassing multiple subtypes with distinct biological characteristics and ***,personalized treatment based on clinical features,pathologic typing,and molecular typing is crucial for the diagnosis and management of precancerous lesions of gastric cancer(PLGC).Current research has categorized GC into four subtypes:Epstein-Barr virus-positive,microsatellite instability,genome stability,and chromosome instability(CIN).Technologies such as multi-omics analysis and gene sequencing are being employed to identify more suitable novel testing methods in these *** these,ultrasensitive chromosomal aneuploidy detection(UCAD)can detect CIN at a genome-wide level in subjects using low-depth whole genome sequencing technology,in conjunction with bioinformatics analysis,to achieve qualitative and quantitative detection of chromosomal *** editorial reviews recent research advancements in UCAD technology for the diagnosis and management of PLGC.

The Lamiaceae family is renowned for its terpenoid-based medicinal components,but Leonurus,which has traditional medicinal uses,stands out for its alkaloid-rich ***,the principal active compound found in Leonurus,has demonstrated promising effects in reducing blood lipids and treating ***,the biosynthetic pathway of leonurine remains largely ***,we present the chromosome-level genome sequence assemblies of Leonurus japonicus,known for its high leonurine production,and Leonurus sibiricus,characterized by very limited leonurine *** integrating genomics,RNA sequencing,metabolomics,and enzyme activity assay data,we constructed the leonurine biosynthesis pathway and identified the arginine decarboxylase(ADC),uridine diphosphate glucosyltransferase(UGT),and serine carboxypeptidase-like(SCPL)acyltransferase enzymes that catalyze key reactions in this *** analyses revealed that the UGT–SCPL gene cluster evolved by gene duplication in the ancestor of Leonurus and neofunctionalization of SCPL in ***,which contributed to the accumulation of leonurine specifically in ***,our comprehensive study illuminates leonurine biosynthesis and its evolution in Leonurus.

Examining the pollination biology of plant species is not only crucial for enhancing our understanding of their reproductive biology,but also essential for elucidating their adaptation and evolutionary ***,we investigated the breeding system and pollination biology of two closely related species in *** flower of *** is unique in the genus with purple and chartaceous(paper-like)sepals,fewer stamens with yellow anthers and purple *** three seasons of field observation and exclusion experiments,we determined that *** was primarily pollinated by a hornet species,Vespa bicolor,which also served as the most efficient *** contrast,*** was primarily pollinated by large *** purpurea exhibited a significantly higher cumulative nectar volume than ***,which could be a crucial factor in attracting ***.A control experiment further demonstrated that removing the nectar leaf(petal)significantly decreased the visiting frequency of *** system studies revealed that both species were self-compatible,yet they primarily underwent outcrossing in natural *** study presents a compelling case of possible pollinator shift in *** accompanied by morphological divergence.A more in-depth investigation of this system would offer crucial insights into the extent to which pollinators are involved in the plant speciation process and whether they contribute to reproductive isolation between closely related species.

Evolution of mechanical properties,localized corrosion resistance and microstructure of a high purity Al-Zn-Mg-Cu alloy during non-isothermal aging

The evolution of mechanical properties,localized corrosion resistance of a high purity Al-Zn-Mg-Cu alloy during non-isothermal aging(NIA)was investigated by hardness test,electrical conductivity test,tensile test,intergranular corrosion test,exfoliation corrosion test,slow strain rate tensile test and electrochemical test,and the mechanism has been discussed based on microstructure examination by optical microscopy,electron back scattered diffraction,scanning electron microscopy and scanning transmission electron *** NIA treatment includes a heating stage from 40℃to 180℃with a rate of 20℃/h and a cooling stage from 180℃to 40℃with a rate of 10℃/*** results show that the hardness and strength increase rapidly during the heating stage of NIA since the increasing temperature favors the nucleation and the growth of strengthening precipitates and promotes the transformation of Guinier-Preston(GPI)zones toη'*** the cooling stage,the sizes ofη'phase increase with a little change in the number density,leading to a further slight increase of the hardness and *** NIA proceeds,the corroded morphology in the alloy changes from a layering feature to a wavy feature,the maximum corrosion depth decreases,and the reason has been analyzed based on the microstructural and microchemical feature of precipitates at grain boundaries and subgrain boundaries.

Acoustically actuated magnetoelectric(ME)antenna based on the efficient oscillation of magnetic dipoles has recently been considered as a promising solution for portable very-low-frequency ***,the severe nonlinear dynamic behavior in the case of strong-field excitation results in insufficient radiation capability and poor communication performance for a conventional ME *** this work,we propose to suppress the nonlinearity of an ME antenna by neutralizing the spring-hardening effect in amorphous Metglas and the spring-softening effect in piezoelectric ceramics through an ME multilayered transmitter(ME-MLTx)*** a driving voltage of 50 V_(pp)at the resonance frequency of 21.2 kHz,a magnetic flux density as high as 108 fT at a distance of 100 m is produced from a single *** addition,ME-MLTx performs a decreased mechanical quality factor(Q_(m))less than 40.65,and,thus,a broadened bandwidth of 500 Hz is ***,a communication link transmitting binary American Standard Code for Information Interchange-coded message is built,which allows for an error-free communication with a distance of 18 m and a data rate of 300 bit/s in the presence of heavy environment *** communication distance can be further estimated over 100 m when using a femtotesla-class-inductive magnetic field *** obtained results are believed to bring ME antennas one step closer to being applicable in very-low-frequency communications.

Migration pattern of a population of Barn Swallows(Hirundo rustica) breeding in East Asian tropical region

Birds exhibit a high degree of migratory diversity, which is influenced by various ecological factors and life history strategies. Conducting studies on tropical bird migration, of which research is scarce, and comparing it with temperate birds can enhance our understanding of bird migration behaviour and its underlying mechanisms. In this study, we explored the migration behaviour of a breeding population of the Barn Swallow(Hirundo rustica) in Zhanjiang, southern China, a region located in the northern tropics, using light-level geolocators. From 2021 to 2023, we deployed geolocators on 92 breeding swallows and retrieved geolocators successfully from 23 individuals. These swallows all exhibited migratory behaviour, and wintering on various islands in Southeast Asia. They displayed sex differences in their wintering locations. All males concentrated in Borneo, while females primarily chose Borneo but also dispersed to the Philippines, South China Sea, and Vietnam for wintering. The studied swallow population adopted a seasonal migration pattern of “indirect in autumn, direct in spring”, bypassing the ecological barrier of the South China Sea in autumn and tending to directly cross it in spring migration. Moreover, the distance and duration of autumn migration was significantly longer than those of the spring migration. Compared to temperate Barn Swallows, the Barn Swallow population breeding in Zhanjiang adopts a pattern of “intra-tropical migration” and initiates autumn migration earlier. The formation of their migration pattern may be limited by ecological and physiological factors.

Provenance of Upper Jurassic in North Yellow Sea Basin， Eastern North China Craton

Base on the detrital zircon U-Pb dating data， this paper discusses the provenance and constructs source-to-sink system of the Upper Jurassic in the North Yellow Sea Basin (NYSB)， Eastern North China Craton (ENCC). In addition， to avoid the bias of detrital zircon ages caused by variations in zircon fertility， we gather 1709 whole rock zirconium content values (ppm) from granitic rocks with different ages in the North China Craton (NCC). Base on mean Zr content of granitic rocks in each age， these granitic rocks can be divided into four groups: Group A consists of Jurassic， Group B consists of Paleozoic， Neo-Mesoproterozoic and Neoarchean， Group C consists of Early Cretaceous and Triassic， and Group D consists of Paleoproterozoic. This research assigns these groups zircon fertility factors (ZFF) of 1， 1.4， 1.8 and 2.2. The U-Pb age of detrital zircons from Late Jurassic sediments corrected by ZFF are statistically analyzed (K-S test， Similarity， Cross-correlation， Linkeness， Multidimensional scaling). And combining with palaeocurrent and palaeogeography， we construct two source-to-sink systems: 1) Jiaodong Uplift and Sulu Orogenic belt as the source area and the North Yellow Sea Basin as the sink area; 2) Liaodong Peninsula as the source area and the North Yellow Sea Basin as the sink area.

DNA-delivered monoclonal antibodies targeting the p53 R175H mutant epitope inhibit tumor development in mice

tThe tumor suppressor p53 is the most common mutated gene in cancer,with the R175H as the most frequent p53 missense ***,there are currently no approved targeted therapies or immunotherapies against mutant ***,we characterized and inves-tigated a monoclonal antibody(mAb)that recognizes the mutant p53-R175H for its affinity,specificity,and activity against tumor cells in *** then delivered DNA plasmids expres-sing the anti-R175H mAb or a bispecific antibody(BsAb)into mice to evaluate their therapeutic *** results showed that the anti-R175H mAb specifically bound to the p53-R175H an-tigen with a high affinity and recognized the human mutant p53-R175H antigen expressed on HEK293T or MC38 cells,with no cross-reactivity with wild-type *** cultured cells,the anti-R175H mAb showed higher cytotoxicity than the control but did not induce antibody-dependent cellular *** made a recombinant MC38 mouse cell line(MC38-p53-R175H)that overexpressed the human p53-R175H after knocking out the endogenous mutant p53 *** vivo,administration of the anti-R175H mAb plasmid elicited a robust anti-tumor effect against MC38-p53-R175H in *** administration of the anti-R175H BsAb plasmid showed no therapeutic effects,yet potent anti-tumor activity was observed in combination with the anti-PD-1 *** results indicate that targeting specific mutant epitopes using DNA-delivered mAbs or BsAbs presents a form of improved natural immunity derived from tumor-infiltrating B cells and plasma cells against intracellular tumor antigens.

Monitoring the prevalence of antimicrobial resistance genes(ARGs)is vital for addressing the global crisis of antibiotic-resistant bacterial *** its importance,the characterization of ARGs and microbiome structures,as well as the identification of indicators for routine ARG monitoring in pig farms,are still lacking,particularly concerning variations in antimicrobial exposure in different countries or ***,metagenomics and random forest machine learning were used to elucidate the ARG profiles,microbiome structures,and ARG contamination indicators in pig manure under different antimicrobial pressures between China and *** showed that Chinese pigs exposed to high-level antimicrobials exhibited higher total and plasmid-mediated ARG abundances compared to those in European pigs(P<0.05).ANT(6)-Ib,APH(3')-IIIa,and tet(40)were identified as shared core ARGs between the two pig ***,the core ARGs identified in pig populations were correlated with those found in human populations within the same geographical *** and Prevotella were identified as the dominant genera in the core microbiomes of Chinese and European pigs,*** ARG markers and 43 biomarkers were able to differentiate between the Chinese and European pig manure samples with accuracies of 100%and 98.7%,*** for assessing ARG contamination in Chinese and European pigs also achieved high accuracy(r=0.72-0.88).Escherichia flexneri in both Chinese and European pig populations carried between 21 and 37 *** results of this study emphasize the importance of global collaboration in reducing antimicrobial resistance risk and provide validated indicators for evaluating the risk of ARG contamination in pig farms.
