Tendinopathy is a prevalent musculoskeletal disorder,accounting for over 30%of musculoskeletal ***,current therapeutic strategies for tendinopathy are limited and often yield unsatisfactory clinical ***,there is a critical need to explore novel therapeutic approaches with minimal side effects for tendinopathy and its early *** this study,we successfully designed and synthesized a copper(I)-based nanocluster,named Cu_(11),which possesses remarkable enzyme-like and ROS-scavenging *** unique combination of properties qualifies Cu_(11)as an attractive anti-inflammatory and anti-ROS *** Cu_(11)clusterzymes specifically target mitochondria,effectively scavenging excessive reactive oxygen species(ROS)and reducing oxidative ***,Cu_(11)clusterzymes inhibit the activation of key signaling pathways involved in inflammation,namely tumor necrosis factor(TNF),mitogen-activated protein kinase(MAPK),and nuclear factor(NF)-κB,leading to a decrease in the release of proinflammatory ***,our in vivo experiments using a collagenase-induced acute Achilles tendinopathy model demonstrated that Cu_(11)clusterzymes effectively alleviate inflammation and oxidative stress,without causing systemic *** study highlights the potential of copper-based clusterzymes as therapeutic agents for the treatment of Achilles tendinopathy and other inflammatory *** unique enzyme-like and ROS-scavenging activities of Cu_(11)make it a promising candidate for further development and clinical translation.

Cell-free expression systems have emerged as a versatile and powerful platform for metabolic engineering,biosynthesis and synthetic biology ***,successful examples of the synthesis of complex natural products using this system are still ***,a structurally unique and complex diketopiperazine alkaloid,is a clinically promising antibiotic that selectively inhibits the transcription termination factor ***,we established a modular cell-free expression system with cascade catalysis for the biosynthesis of bicyclomycin from a chemically synthesized *** six cell-free expressed biosynthetic enzymes,including five iron-andα-ketoglutarate-dependent dioxygenases and one cytochrome P450 monooxygenase,were active in converting their substrates to the corresponding *** co-expressed enzymes in the cell-free module were able to complete the related partial *** vitro biosynthesis of bicyclomycin was also achieved by reconstituting the entire biosynthetic pathways(i.e.,six enzymes)using the modular cell-free expression *** study demonstrates that the modular cell-free expression system can be used as a robust and promising platformforthe biosynthesis of complex antibiotics.

Costunolide alleviates HKSA-induced acute lung injury via inhibition of macrophage activation

Staphylococcus aureus(***)infection leads to a severe inflammatory response and causes acute lung injury(ALI),eventually threatening human ***,it is of importance to find an agent to inhibit inflammation and reduce ***,we found that costunolide,a sesquiterpene lactone,displays anti-inflammatory effects and ameliorates heat-killed ***(HKSA)-induced *** treatment attenuated HKSA-induced murine ALI in which pulmonary neutrophil infiltration was inhibited,lung edema was decreased,and the production of pro-inflammatory cytokines was significantly *** addition,costunolide dose-dependently inhibited the generation of IL-6,TNF-α,IL-1β,and keratinocyte-derived cytokine(KC),as well as the expression of iNOS,in HKSA-induced ***,costunolide attenuated the phosphorylation of p38 MAPK and cAMP response element-binding protein(CREB).Collectively,our findings suggested that costunolide is a promising agent for alleviating bacterialinduced ALI via the inhibition of the MAPK signaling pathways.

The long-term safety and effectiveness of once-daily tadalafil is crucial,but limited data are available in Chinese patients witherectile dysfunction(ED).In this post-marketing,multicenter,randomized,open-label trial with 2-year follow-up,635 ED caseswere randomized to receive daily oral tadalafil 2.5 mg or 5 mg for 3 months,of whom 580 continued once-daily tadalafil 5 mg for21 ***-emergent adverse events in the 12-month and 24-month period were similar,with the most common beingviral upper respiratory tract infection,upper respiratory tract infection,and *** improvement from baseline inthe International Index of Erectile Function-Erectile Function(IIEF-EF)score was detected at month 12(least squares mean[LSM]change:7.9,95%confidence interval[CI]:7.5–8.4,P1)and was maintained to month 24(LSM change:8.6,95%CI:8.1–9.0,P1).The proportions of patients regaining normal erectile function(IIEF-EF score≥26)were 43.7%and 48.0%at months 12 and 24,*** Assessment Questionnaire results showed improved erection function in 97.5%ofpatients and improved ability to engage in sexual activity in 95.9%of patients at month 12;these values were 96.1%and 95.0%at month 24,*** quality of sexual life score based on the Sexual Life Quality Questionnaire(SLQQ)was increased by52.2%at month 12 and by 55.3%at month 24(both P1).The treatment satisfaction score determined by SLQQ(mean±standard deviation)was 62.4±21.0 at month 12 versus 65.9±20.2 at month ***-year daily application of tadalafil 5mg in Chinese men with ED showed a favorable safety profile and durable improvement in sexual performance and satisfaction.

Despite the extensive application of advanced oxidation processes(AOPs)in water treatment,the efficiency of AOPs in eliminating various emerging contaminants such as halogenated antibiotics is constrained by a number of *** moieties exhibit strong resistance to oxidative radicals,affecting the dehalogenation and detoxification *** address these limitations of AOPs,advanced reduction processes(ARPs)have been ***,a novel nucleophilic reductant—namely,the carbon dioxide radical anion(CO_(2)^(·-))—is introduced for the simultaneous degradation,dehalogenation,and detoxification of florfenicol(FF),a typical halogenated *** results demonstrate that FF is completely eliminated by CO_(2)^(·-),with approximately 100%of Cland 46%of Freleased after 120 min of *** detoxification is observed,which exhibits a linear response to the release of free inorganic halogen ions(R^(2)=0.97,p1).The formation of halogen-free products is the primary reason for the superior detoxification performance of this method,in comparison with conventional hydroxyl-radical-based *** identification and density functional theory(DFT)calculations reveal the underlying dehalogenation mechanism,in which the chlorine moiety of FF is more susceptible than other moieties to nucleophilic attack by CO_(2)^(·-).Moreover,CO_(2)^(·-)-based ARPs exhibit superior dehalogenation efficiencies(>75%)in degrading a series of halogenated antibiotics,including chloramphenicol(CAP),thiamphenicol(THA),diclofenac(DLF),triclosan(TCS),and ciprofloxacin(CIP).The system shows high tolerance to the pH of the solution and the presence of natural water constituents,and demonstrates an excellent degradation performance in actual groundwater,indicating the strong application potential of CO_(2)^(·-)-based ARPs in real ***,this study elucidates the feasibility of CO_(2)^(·-)for the simultaneous degradation,dehalogenation,and detoxification of halog

＜正＞Pain is an unpleasant sensory and emotional experience associated with actual or potential tissue damage, including a sensory-discriminative and an emotional-affective *** evidence from morphological, electrophysiological,neuroimaging,and behavioral studies as well as early clinical observation indicates that the anterior cingulate cortex（ACC） is involved in pain-related ***,little is known about the molecular and cellular mechanisms underlying pain-related aversion in *** previous studies showed that NMDAR-dependent ERK signaling cascade in rat ACC contributes to pain *** the present study we observed the role of p38,another member of MAP（mitogen-activated protein） kinase family,in ACC in pain-related negative emotion using Western Blot and behavioral ***-plantar injection of 5%formalin produced a robust increase in phosphorylated MKK3/6（pMKK3/6）,upstream of p38/MAPK,and phosphorylated p38（p-p38） in the bilateral rostral ACC（rACC）.Using formalin-induced conditioned place avoidance（F-CPA） in rats,which is believed to reflect the pain-related negative emotion,we found that blockade of p38/MAPK activation in the rACC with p38 inhibitor SB203580（0.1μg,1μg per side） but not low dose（0.01μg per side）,or another kind of p38 inhibitor SB239063（0.1μg,1μg per side） dose-dependently prevented the induction of ***,these blockades affected neither formalin-induced two-phase spontaneous pain nor low-intensity electric foot shock-induced CPA *** together,these results indicate that activation of p38/MAPK contributes to pain-related negative emotion,which will provide new molecule mechanisms underlying pain affect and bring new treatments for relieving emotional pain-related disturbance.

针对复杂曲面多轴数控加工过程的复杂性,提出了一种面向多轴加工的分层递阶构造模型,即利用铣削过程的几何、运动学与力学模型综合分析多轴加工规律.切削工件几何模型采用深度元素法,通过分析刀具与工件接触区域,得到刀具在不同瞬时的轴向范围及沿轴向各微元的切入角和切出角,转换为铣削力模型所需的几何信息,同时运动学模型又为几何与力学模型提供计算依据.采用序列二次规划法优化进给速度,使机床工作在多个约束条件的最大范围值内.五轴螺旋桨加工实验表明,在满足加工精度条件下,优化的进给速度使零件加工效率提高约20%.

Many economically important crops and vegetables belonging to the cruciferous family are heavily endangered by clubroot disease caused by Plasmodiophora brassicae *** of clubroot resistant cultivars based on mapping and cloning of resistant genes is commonly regarded as the most cost-effective and efficient way to fight against this *** traditional way of R gene functional validation requires stable transformation that is both time-and *** this study,a rapid and efficient hairy-root transgenic protocol mediated by Agrobacterium rhizogenes was *** transformation positive rate was over 80%in Brassica napus showed by GUS reporter gene and this transformation only took 1/6 of the time compared with stable *** system was applicable to different *** varieties and other cruciferous crops including Brassica rapa and Brassica *** particular,two known CR genes,CRA3.7.1 and CRA8.2.4 were used respectively,as example to show that the system works well for CR gene study combined with subsequent *** infection in *** importantly,it works both in over-expression that led to disease resistance,as well as in RNAi which led to disease susceptible ***,this system can be used in batch-wise identification of CR genes,and also offered the possibility of manipulating key genes within the *** genome that could improve our knowledge on host-pathogen interaction.
