ADP-dependent glucokinase controls metabolic fitness in prostate cancer progression

Background Cell metabolism plays a pivotal role in tumor progression,and targeting cancer metabolism might effectively kill cancer *** aimed to investigate the role of hexokinases in prostate cancer(PCa)and identify a crucial target for PCa *** The Cancer Genome Atlas(TCGA)database,online tools and clinical samples were used to assess the expression and prognostic role of ADP-dependent glucokinase(ADPGK)in *** effect of ADPGK expression on PCa cell malignant phenotypes was validated in vitro and in *** proteomics,metabolomics,and extracellular acidification rate(ECAR)and oxygen consumption rate(OCR)tests were performed to evaluate the impact of ADPGK on PCa *** underlying mechanisms were explored through ADPGK overexpression and knockdown,co-immunoprecipitation(Co-IP),ECAR analysis and cell counting kit-8(CCK-8)*** ADPGK was the only glucokinase that was both upregulated and predicted worse overall survival(OS)in prostate adenocarcinoma(PRAD).Clinical sample analysis demonstrated that ADPGK was markedly upregulated in PCa tissues ***-PCa *** ADPGK expression indicates worse survival outcomes,and ADPGK serves as an independent factor of biochemical *** vitro and in vivo experiments showed that ADPGK overexpression promoted PCa cell proliferation and migration,and ADPGK inhibition suppressed malignant ***,proteomics,and ECAR and OCR tests revealed that ADPGK significantly accelerated glycolysis in ***,ADPGK binds aldolase C(ALDOC)to promote glycolysis via AMP-activated protein kinase(AMPK)*** was positively correlated with ADPGK,and high ALDOC expression was associated with worse survival outcomes in *** In summary,ADPGK is a driving factor in PCa progression,and its high expression contributes to a poor prognosis in PCa *** accelerates PCa glycolysis and progression by activating ALDOC-AMPK signaling,suggesting

Reactive oxygen species（ROS） plays a key role in human heart diseases. Glutathione peroxidase（GPX） functions as an antioxidant as it catalyzes the reduction of hydroperoxide. In order to investigate the antioxidant effect of human selenium-containing single-chain Fv（Se-scFv-B3）, a new mimic of GPX, a model system of hydrogen peroxide（H202）-induced rat cardiac myocyte damage was established. The cardiac myocyte damage was characte- rized in terms of cell viability, lipid peroxidation, cell membrane integrity, and intracellular H202 level. The Se-scFv-B3 significantly reduced H2O2-induced cell damage as shown by the increase of cell viability, the decline of malondialdehyde（MDA） production, lactate dehydrogenase（LDH） release, and intracellular H2O2 level. So Se-scFv-B3 may have a great potential in the treatment of human heart diseases induced by ROS.

Prognostic value of primary tumor surgery in seminoma patients with distant metastasis at diagnosis:a population-based study

The aims of this study were to determine the prog no stic value of primary tumor surgery and identify optimal can didates for such surgery among patients with semi noma and dista nt metastasis at diag no *** ide ntified 521 patients with seminoma and distant metastasis at diagnosis between 2004 and 2014 from the Surveillanee,Epidemiology,and End Results *** these patients,434 had undergone surgery,whereas 87 had *** prognostic value of primary tumor surgery was assessed by Kaplan-Meier methods,log-rank analyses,and multivariate Cox's proportional hazards *** curves and forest plots were also *** analysis indicated that patients who underwent surgery had a better 5-year overall survival and cancer-specific survival tha n those who did *** analyses dem on strated that primary tumor surgery is an in depende nt prog no stic factor for overall survival and cancer-specific survival,along with age at diagnosis,Mstage,and marital *** addition,primary tumor surgery still had con siderable prog no stic value in the subgroup of patie nts with lymph node ***,forest plots demonstrated that patie nts with Mia stage,N1 or N2-3 stage,and a youn ger age at diagnosis(dings indicate that primary tumor surgery is correlated with improved survival in patients with seminoma and distant ***,primary tumor surgery is an independent prognostic indicator for patients with seminoma and distant metastasis.

Risk factors associated with early and late HAT after adult liver transplantation

AIM: To identify risk factors that might contribute to hepatic artery thrombosis (HAT) after liver transplantation (LT). METHODS: The perioperative and follow-up data of a total of 744 liver transplants, performed from February 1999 to July 2010, were retrospectively reviewed. HAT developed in 20 patients (2.7%). HAT was classified as early (occurring in fewer than 30 d post LT) or late (occurring more than 30 d post LT). Early HAT devel-oped in 14 patients (1.9%). Late HAT developed in 6 patients (0.8%). Risk factors associated with HAT were analysed using the chi(2) test for univariate analysis and logistic regression for multivariate analysis. RESULTS: Lack of ABO compatibility, recipient/donor weight ratio >= 1.15, complex arterial reconstruction, duration time of hepatic artery anastomosis > 80 min, duration time of operation > 10 h, dual grafts, number of units of blood received intraoperatively >= 7, number of units of fresh frozen plasma (FFP) received intraoperatively >= 6, postoperative blood transfusion and postoperative FFP use were significantly associated with early HAT in the univariate analysis (P = 1.15 (OR = 4.499), duration of hepatic artery anastomosis > 80 min (OR = 5.429), number of units of blood received intraoperatively >= 7 (OR = 4.059) and postoperative blood transfusion (OR = 6.898). Graft type (whole/living-donor/split), duration of operation > 10 h, retransplantation, rejection reaction, recipients with diabetes preoperatively and recipients with a high level of blood glucose or diabetes postoperatively were significantly associated with late HAT in the univariate analysis (P 10 h (OR = 6.394), retransplantation (OR = 21.793) and rejection reactions (OR = 16.936). The bedrock weathered crust in front of the Altun Mountains in the Qaidam Basin,western China,is different from others because this is a salt-lake basin,where saline water fluid infiltrates and is deposited in the overlying strata.A large amount of gypsum infills the bedrock weathered crust,and this has changed the pore *** core observation,polarized light microscopy,electron probe,physical property analysis and field emission scanning electron microscopy experiments,the characteristics of the weathered bedrock have been *** are cracks and a small number of dissolved pores in the interior of the weathered *** micropores are widely developed,especially the various matrix cracks formed by tectonics and weathering,as well as the stress characteristics of small dissolved pores,and physical properties such as porosity and ***‘dual structure’developed in the bedrock is important for guiding the exploration of the lake basin bedrock for natural gas.

Background and Aims:Nonalcoholic fatty liver disease(NAFLD)is associated with metabolic *** study aimed to explore the role of metabolic disorders in screening advanced fibrosis in NAFLD ***:A total of 246 histologically-proven NAFLD patients were enrolled across 14 *** compared the severity of fibrosis in patients with different components of metabolic *** on standard noninvasive tests and metabolic disorders,we developed new algorithms to identify advanced ***:Metabolic syndrome(MetS)was frequent in NAFLD patients(133/246,54%).Patients with MetS had a higher proportion of significant fibrosis(p=0.014)and higher LSM values(9.2 kPa,vs.7.4 kPa,p=0.002)than those without *** with more metabolic disorders had higher fibrosis stages(p=0.017).Reduced highdensity lipoprotein cholesterol(odds ratio[OR]:2.241,95%confidence interval[CI]:1.004–5.002,p=0.049)and raised fasting glucose(OR:4.500,95%CI:2.083–9.725,p<0.001)were significantly associated with advanced *** these two metabolic disorders as a screening tool,a sensitivity,specificity and accuracy of 92%,81%and 83%was achieved,*** the new algorithms combining metabolic disorders with noninvasive measurements,the number of patients requiring liver biopsy was reduced,especially in combination with the Fibrosis-4 score and metabolic disorders(36%to 17%,p<0.001).In addition,this stepwise algorithm could achieve a high accuracy(85%)and high negative predictive value(93%).Conclusions:Metabolic disorders should be taken into consideration in the diagnosis of advanced *** further validation and investigation,new algorithms could be recommended in primary care units to spare patients from unnecessary referral and liver biopsies.
