Degradation of warm permafrost and talik formation on the Qinghai-Tibet Plateau in 2006-2021

Permafrost is degrading globally,particularly those with low thermal stability on the Qinghai-Tibet Plateau,owing to climate change.However,the inadequacy of direct research on permafrost degradation based on in-situ monitoring limits the prediction of permafrost degradation and engineering practices.This study explored the processes and modes of permafrost degradation into talik by analyzing ground temperature data from five points in the hinterland of the Qinghai-Tibet Plateau from 2006 to 2021.The results showed that the degradation of the warm permafrost layer with a geothermal gradient of zero occurred simultaneously in the top and bottom directions.The rate of permafrost degradation from the top down and bottom up increase during the degradation process,but the increase of the former is more drastic after the formation of thawed interlayer.Additionally,the construction of the Qinghai-Tibet Railway changed the degradation modes of the permafrost in adjacent natural sites through horizontal heat transfer,particularly after through talik formation under the embankment.The findings suggest that taking countermeasures before or immediately after forming thawed interlayer is more effective.When evaluating the thermal impact of projects in warm permafrost regions,special attention should be given to the horizontal heat transfer process that may result from the formation of a through talik.

Field observations of the thermal stability of permafrost under buildings with an underfloor open ventilation space and pile foundations in warm permafrost at high altitudes

Pile foundations combined with ventilation spaces under floors are the most common method in buildings over permafrost.The safety and stability of buildings are closely related to the temperature of permafrost.However,there are limitations of understanding on this method in the high-altitude,warm(>−1℃)permafrost areas on the Qinghai–Tibet Plateau.In this study,the thermal stability of permafrost foundation soils under buildings with an underfloor open ventilation space and pile foundations in warm permafrost at high altitudes was studied through field observations of ground and air temperatures,wind speed,net radiation from 2017 to 2021.The results indicated that the open ventilation space exerted an effective cooling effect on the underlying permafrost and pile foundations from March to October,while a thermal insulation effect was observed from November to February of the following year,but overall,the cooling effect dominated;the cooling effect of open ventilation spaces differed spatially.The permafrost temperature on the south-facing side was higher than that on the north-facing side,and those on the east and west sides were higher than that directly under the open ventilation space of the building.This study also demonstrated that radiation shielded by the building was a main factor of the cooling effect of open ventilation spaces,and the cooling effect of open ventilation spaces could accelerate the back-freezing of the cast-in-place(CIP)pile foundations.This structure could effectively maintain the frozen state of the underlying warm permafrost at high elevations on the interior Qinghai–Tibet Plateau.

Current situation and prospect for the diagnosis and treatment of pediatric critical rare diseases in China

The onset of critical rare diseases (RDs) in children is rapid and dangerous, accompanied by a high mortality rate, which brings a heavy burden to both families and society. Multiple malformations, neuromuscular diseases, metabolic diseases, and heart diseases are the most common types of RDs in children of China, often manifesting with multiple organ dysfunction. At present, the diagnosis and treatment of critical RDs in children face challenges such as prolonged diagnosis time, a high misdiagnosis rate, limited treatment modalities, and a significant disease burden. However, with the progress in genetic testing technology, the establishment of multidisciplinary diagnosis and treatment platforms, and the implementation of relevant RD policies in China, children with critical RDs will received enhanced medical services, experience improved prognoses, and reintegrate into social life.

STRAP binds to and promotes the repair of N1-methyldeoxyadenosine in DNA

N1-methyladenosine(m1A)is an important RNA modification that functions in various biological processes by interacting with cellular proteins.However,the binding proteins of N1-methyldeoxyadenosine(1mdA)in DNA remain largely unknown.Herein,we employed a quantitative proteomics strategy to identify the potential binding proteins of 1mdA in human cells.Our results revealed that serine–threonine kinase receptor-associated protein(STRAP)can bind to 1mdA-carrying DNA.We further demonstrated that STRAP participates in alkylating agent-induced DNA damage response and can promote the repair of 1mdA embedded in DNA.Moreover,we investigated the effects of STRAP on 1mdA-induced perturbation in transcription using a shuttle vector-and next-generation sequencing-based assay,and found that STRAP is involved in the transcriptional bypass of 1mdA in human cells.Together,our study revealed STRAP as a novel 1mdA-binding protein in human cells and provided new insight into the biological implications of STRAP and 1mdA modification in human diseases.

Lanthanide-based microlasers:Synthesis,structures,and biomedical applications

The large size of lasers limits their applications in confined spaces,such as in biosensing and in vivo brain tissue imaging.In this regard,micron-sized lasers have been developed.They exhibit great potential for biological detecting,remote sensing,and depth tracking due to their small sizes,sensitive properties of their spectral fingerprints,and flexible positional modulation in the microenvironment.Lanthanide-based luminescent materials that possess long excited-state lifetime,narrow emission bandwidth,and upconversion behaviors are promising as gain mediums for novel microlasers.In addition,lanthanide-based microlasers could be generated under natural ambient conditions with pumped or continuous light sources,which significantly promotes the practical applications of microlasers.Recent progress in the design,synthesis,and biomedical applications of lanthanide-based microlasers has been outlined in this review.Lanthanide ions doped and upconverted lanthanide-based microlasers are highlighted,which exhibit advantageous structures,miniaturized dimensions,and high lasing performance.The applications of lanthanide-based microlasers are further discussed,the upconverted microlasers show great advantages for biological applications owing to their tunable excitation and emission characteristics and excellent environmental stability.Moreover,perspectives and challenges in the field of lanthanide-based microlasers are presented.

Transplantation of fibrin-thrombin encapsulated human induced neural stem cells promotes functional recovery of spinal cord injury rats through modulation of the microenvironment

Recent studies have mostly focused on engraftment of cells at the lesioned spinal cord,with the expectation that differentiated neurons facilitate recovery.Only a few studies have attempted to use transplanted cells and/or biomaterials as major modulators of the spinal cord injury microenvironment.Here,we aimed to investigate the role of microenvironment modulation by cell graft on functional recovery after spinal cord injury.Induced neural stem cells reprogrammed from human peripheral blood mononuclear cells,and/or thrombin plus fibrinogen,were transplanted into the lesion site of an immunosuppressed rat spinal cord injury model.Basso,Beattie and Bresnahan score,electrophysiological function,and immunofluorescence/histological analyses showed that transplantation facilitates motor and electrophysiological function,reduces lesion volume,and promotes axonal neurofilament expression at the lesion core.Examination of the graft and niche components revealed that although the graft only survived for a relatively short period(up to 15 days),it still had a crucial impact on the microenvironment.Altogether,induced neural stem cells and human fibrin reduced the number of infiltrated immune cells,biased microglia towards a regenerative M2 phenotype,and changed the cytokine expression profile at the lesion site.Graft-induced changes of the microenvironment during the acute and subacute stages might have disrupted the inflammatory cascade chain reactions,which may have exerted a long-term impact on the functional recovery of spinal cord injury rats.

Th17/Treg balance and macrophage polarization ratio in lower extremity arteriosclerosis obliterans

Objective:To explore the balance of peripheral blood T helper 17 cells/regulatory T cell(Th17/Treg)ratio and the polarization ratio of M1 and M2 macrophages in lower extremity arteriosclerosis obliterans(ASO).Methods:A rat model of lower extremity ASO was established,and blood samples from patients with lower extremity ASO before and after surgery were obtained.ELISA was used to detect interleukin 6(IL-6),IL-10,and IL-17.Real-time RCR and Western blot analyses were used to detect Foxp3,IL-6,IL-10,and IL-17 expression.Moreover,flow cytometry was applied to detect the Th17/Treg ratio and M1/M2 ratio.Results:Compared with the control group,the iliac artery wall of ASO rats showed significant hyperplasia,and the concentrations of cholesterol and triglyceride were significantly increased(P<0.01),indicating the successful establishment of ASO.Moreover,the levels of IL-6 and IL-17 in ASO rats were pronouncedly increased(P<0.05),while the IL-10 level was significantly decreased(P<0.05).In addition to increased IL-6 and IL-17 levels,the mRNA and protein levels of Foxp3 and IL-10 in ASO rats were significantly decreased compared with the control group.The Th17/Treg and M1/M2 ratios in the ASO group were markedly increased(P<0.05).These alternations were also observed in ASO patients.After endovascular surgery(such as percutaneous transluminal angioplasty and arterial stenting),all these changes were significantly improved(P<0.05).Conclusions:The Th17/Treg and M1/M2 ratios were significantly increased in ASO,and surgery can effectively improve the balance of Th17/Treg,and reduce the ratio of M1/M2,and the expression of inflammatory factors.

Monitoring and evaluation of the water quality of the Lower Neches River, Texas, USA

Increasing bacteria levels in the Lower Neches River caused by Hurricane Harvey has been of a serious concern.This study is to analyze the historical water sampling measurements and real-time water quality data collected with wireless sensors to monitor and evaluate water quality under different hydrological and hydraulic conditions.The statistical and Pearson correlation analysis on historical water samples determines that alkalinity,chloride,hardness,conductivity,and pH are highly correlated,and they decrease with increasing flow rate due to dilution.The flow rate has positive correlations with Escherichia coli,total suspended solids,and turbidity,which demonstrates that runoff is one of the causes of the elevated bacteria and sediment loadings in the river.The correlation between E.coli and turbidity indicates that turbidity greater than 45 nephelometric turbidity units in the Neches River can serve as a proxy for E.coli to indicate the bacterial outbreak.A series of statistical tools and an innovative two-layer data smoothing filter are developed to detect outliers,fill missing values,and filter spikes of the sensor measurements.The correlation analysis on the sensor data illustrates that the elevated sediment/bacteria/algae in the river is either caused by the first flush rain and heavy rain events in December to March or practices of land use and land cover.Therefore,utilizing sensor measurements along with rainfall and discharge data is recommended to monitor and evaluate water quality,then in turn to provide early alerts on water resources management decisions.

Geobacter sulfurreducens promoted the biosynthesis of reduced graphene oxide and coupled it for nitrobenzene reduction

In order to explore an efficient and green method to deal with nitrobenzene(NB)pollutant,reduced graphene oxide(r GO)as an electron shuttle was applied to enhance the extracellular electron transfer(EET)process of Geobacter sulfurreducens,which was a typical electrochemically active bacteria(EAB).In this study,r GO biosynthesis was achieved via the reduction of graphene oxide(GO)by G.sulfurreducens PCA within 3 days.Also,the r GOPCA combining system completely reduced 50-200μmol/L of NB to aniline as end product within one day.SEM characterization revealed that PCA cells were partly wrapped by rGO,and therefore the distance of electron transfer between strain PCA and r GO material was reduced.Beside,the ID/IGof GO,r GO,and r GO-PCA combining system were 0.990,1.293 and 1.31,respectively.Moreover,highest currents were observed in r GO-PCA-NB as 12.950μA/-12.560μA at -408 m V/156 m V,attributing to the faster electron transfer efficiency in EET process.Therefore,the NB reduction was mainly due to:(I)direct EET process from G.sulfurreducens PCA to NB;(II)r GO served as electron shuttle and accelerated electron transfer to NB,which was the main degradation pathway.Overall,the biosynthesis of r GO via GO reduction by Geobacter promoted the NB removal process,which provided a facile strategy to alleviate the problematic nitroaromatic pollution in the environment.
