Demethylating agent decitabine induces autologous cancer testis antigen specific cytotoxic T lymphocytes in vivo

Background Cancer testis antigens （CTAs） are a novel group of tumor associated *** agent decitabine was reported to be able to up-regulate CTAs through its hypomethylation mechanism,thus enhance the immunogenicity of leukemia ***,few researches have ever focused on the questions that whether this immunostimulatory effect of decitabine could induce autologous CTA specific cytotoxic T lymphocytes （CTLs） in vivo,and if so,whether this effect contributes to disease *** this study,we aimed to show that decitabine could induce specific autologous CTLs against some mouse CTAs in leukemia cells in vitro and in *** Several mouse CTAs were screened by *** specific to one of the CTAs named P1A was detected and sorted by P1A specific dimer by flow *** activity of specific CTLs was measured by real time *** We firstly screened expression of some CTAs in mouse leukemia cells before and after decitabine treatment and found that decitabine treatment did up-regulate expression of many *** we measured the CTLs＇ activity specific to a mouse CTA P1A in vivo and showed that this activity increased after decitabine ***,we sorted these in vivo induced P1A specific CTLs by flow cytometry and demonstrated their cytotoxicity against decitabine treated leukemia *** Our study showed the autologous immune response induced by decitabine in *** more importantly,we firstly proved that this response may contribute to disease *** believe that this immunostimulatory effect is another anti-cancer mechanism of decitabine,and this special effect would inspire new applications of decitabine in the field of leukemia treatment in the future.

Phosphorus(P) leaching is a major problem in greenhouse vegetable production with excessive P fertilizer application. Substitution of inorganic P fertilizer with organic fertilizer is considered a potential strategy to reduce leaching, but the effect of organic material addition on soil P transformation and leaching loss remains unclear. The X-ray absorption nearedge structure(XANES) spectroscopy technique can determine P speciation at the molecular level. Here, we integrated XANES and chemical methods to explore P speciation and transformation in a 10-year field experiment with four treatments: 100% chemical fertilizer(4 CN), 50% chemical N and 50% manure N(2CN+2MN), 50% chemical N and 50% straw N(2CN+2SN), and 50% chemical N and 25% manure N plus 25% straw N(2CN+2 MSN). Compared with the 4 CN treatment, the organic substitution treatments increased the content of labile P by 13.7–54.2% in the 0–40 cm soil layers, with newberyite and brushite being the main constituents of the labile P. Organic substitution treatments decreased the stable P content;hydroxyapatite was the main species and showed an increasing trend with increasing soil depth. Straw addition(2CN+2SN and 2CN+2 MSN) resulted in a higher moderately labile P content and a lower labile P content in the subsoil(60–100 cm). Moreover, straw addition significantly reduced the concentrations and amounts of total P, dissolved inorganic P(DIP), and particulate P in leachate. DIP was the main form transferred by leaching and co-migrated with dissolved organic carbon. Partial least squares path modeling revealed that straw addition decreased P leaching by decreasing labile P and increasing moderately labile P in the subsoil. Overall, straw addition is beneficial for developing sustainable P management strategies due to increasing labile P in the upper soil layer for the utilization of plants, and decreasing P migration and leaching.

Expression of PIN Genes in Rice （Oryza sativa L.）： Tissue Specificity and Regulation by Hormones

Twelve genes of the PIN family in rice were analyzed for gene and protein structures and an evolutionary relationship with reported AtPINs in Arabidopsis. Four members of PIN1 （designated as OsPINla-d）, one gene paired with AtPIN2 （OsPIN2）, three members of PIN5 （OsPIN5a-c）, one gene paired with AtPIN8 （OsPIN8）, and three monocot-specific PINs （OsPIN9, OsPINIOa, and b） were identified from the phylogenetic analysis. Tissue-specific expression patterns of nine PIN genes among them were investigated using RT-PCR and GUS reporter. The wide variations in the expression domain in different tissues of the PIN genes were observed. In general, PIN genes are up-regulated by exogenous auxin, while different responses of different PIN genes to other hormones were found.

Implicit Trust Based Context-Aware Matrix Factorization for Collaborative Filtering

Matrix factorization （MF） has been proved to be a very effective technique for collaborative filtering （ CF）, and hence has been widely adopted in today＇s recommender systems, Yet due to its lack of consideration of the users＇ and items＇ local structures, the recommendation accuracy is not fully satisfied. By taking the trusts among users＇ and between items＇ effect on rating information into consideration, trust-aware recommendation systems （TARS） made a relatively good performance. In this paper, a method of incorporating trust into MF was proposed by building user-based and item-based implicit trust network under different contexts and implementing two implicit trust-based context-aware MF （]TMF） models. Experimental results proved the effectiveness of the methods.

Effects of Chinese medicine Tongxinluo on hyperglycemia and beta-cell damage in streptozotocin-induced diabetic rats

Background Oxidative stress has been implicated in the onset and progression of diabetes. Tongxinluo is a traditional Chinese medicine with potent antioxidant properties. The aim of this study was to test the hypothesis that pretreatment with Tongxinluo has similar effects as melatonin on preventing hyperglycemia and beta-cell damage in a rat model of streptozotocin （STZ）-induced diabetes. Methods Forty male Sprague Dawley rats were randomly assigned to four groups （n=10 each）： normal control （NC） group; STZ group （70 mg/kg, i.p.）; Tongxinluo （1.0 g-kg-^-d-1） pretreated （TXL＋STZ） group and melatonin （200 iJg-kg-~.d-1） pretreated （MLT＋STZ） group. Tongxinluo and melatonin were administered by gavage beginning 8 days before STZ injection and continuing until the end of the study （15 days after STZ administration）. Blood glucose levels and body weights, malondialdehyde （MDA）, and reduced glutathione （GSH） levels were measured, and immunofluorescence studies were performed in all of the groups. Results Pretreatment with Tongxinluo, as with melatonin, attenuated severe hyperglycemia and weight loss induced by STZ.. In pancreatic homogenates, MDA levels were significantly lower and GSH levels were significantly higher in Tongxinluo pretreated group and in melatonin pretreated group than those in STZ group. Values of insulin staining were significantly improved in Tongxinluo pretreated group and in melatonin pretreated group as compared with those in STZ group. Conclusions Tongxinluo, as melatonin, prevented hyperglycemia and beta-cell destruction induced by STZ in rats through reducing oxidative stress in pancreatic tissues. Tongxinluo may provide an alternative therapy for the prevention and treatment of diabetes.

The Rabi oscillations in two-component Bose-Einstein condensates with a coupling drive are studiedby means of a pair of bosonic operators. The coupling drive and initial phase difference will affect the amplitudeand the period of the Rabi oscillations. The Rabi oscillations will vanish in the evolution of the condensate densityfor some special initial phase differences (ψ = 0 or π). Our theory provides not only an analytical framework forquantitative predictions for two-component condensates, but also gives an intuitive understanding of some mysteriousfeatures observed in experiments and numerical. simulations.

Risk factors of infected pancreatic necrosis secondary to severe acute pancreatitis

BACKGROUND： Severe acute pancreatitis（SAP） remains a clinical challenge with considerable morbidity and *** early identification of infected pancreatic necrosis（IPN）, a life-threatening evolution secondary to SAP, is obliged for a more preferable prognosis. Thus, the present study was conducted to identify the risk factors of IPN secondary to SAP. METHODS： The clinical data of patients with SAP were retrospectively analyzed. Univariate and multivariate logistic regression analyses were sequentially performed to assess the associations between the variables and the development of IPN secondary to SAP. A receiver operating characteristic（ROC） curve was created for each of the qualified independent risk factors. RESULTS： Of the 115 eligible patients, 39（33.9%） progressed to IPN, and the overall in-hospital mortality was 11.3%（13/115）.The early enteral nutrition（EEN）（P=0.0092, OR=0.264）, maximum intra-abdominal pressure（IAP）（P=0.0398, OR=1.131）and maximum D-dimer level（P=0.0001, OR=1.006） in the first three consecutive days were independent risk factors associated with IPN secondary to SAP. The area under ROC curve（AUC） was 0.774 for the maximum D-dimer level in the first three consecutive days and the sensitivity was 90% and the specificity was 58% at a cut-off value of 933.5 μg/L; the AUC was 0.831 for the maximum IAP in the first three consecutive days and the sensitivity was 95% and specificity was 58%at a cut-off value of 13.5 mm Hg. CONCLUSIONS： The present study suggested that the maximum D-dimer level and/or maximum IAP in the first three consecutive days after admission were risk factors of IPN secondary to SAP; an EEN might be helpful to prevent the progression of IPN secondary to SAP.

Non-perturbative Aspects of QCD and Parameterized Quark Propagator

Based on the Global Color Symmetry Model, the non-perturbative Q, CD vacuum is investigated in the parameterized fully dressed quark propagator. Our theoretical predictions for various quantities characterized the QCD vacuum are in agreement with those predicted by many other phenomenologieal QCD inspired models. The successful predictions clearly indicate the extensive validity of our parameterized quark propagator used here. A detailed discussion on the arbitrariness in determining the integration cut-off parameter ofμ in calculating QCD vacuum condensates and a good method, which avoided the dependence of calculating results on the cut-off parameter is also strongly recommended to readers.
