Evaluation and clinical application of a new method for detecting ADAMTS13 activity

Background A severe deficiency of ADAMTS13 activity contributes to the pathogenesis of thrombotic thrombocytopenic purpura （TTP）. Measuring the activity of ADAMTS13 is helpful for the diagnosis of TTP and the prognostic monitor in TTP patients. Most available assays are cumbersome and costly, so not easily adapted to routine laboratories. ADAMTS13 cleaves von Willebrand factor （VVVF） within the domain A2, located between domains A1 and A3. Therefore, specific assays for ADAMTS13 activity could be based on the different structures of VWF before and after the cleavage. Using this hypothesis we try to establish a new and simple method to determine ADAMTS13 activity. Methods First, plasma samples were exposed in denaturing condition to allow cleavage of VWF by ADAMTS13. Then, the ADAMTS13 activity was measured with two novel monoclonal antibodies, SZ-129 and SZ-125, which specifically recognize the VWF A1 and A3 domains by using a two-site sandwich ELISA. Compared with a residual-collagen binding assay （R-CBA）, plasma ADAMTS13 activities in 161 samples were assessed, and the inhibitory activities of ADAMTS13 autoantibody in 24 TTP patients were determined. The relationship of these two assays was analyzed by linear correlation, and the sensitivity and specificity of the new assay was also evaluated. Results Plasma ADAMTS13 activities in normal people and TTP, acute myocardial infarction （AMI）, and idiopathic thrombocytopenic purpura （ITP） patients determined by the new assay were （89.75±7.93）%, （17.63±18.71）%, （68.55±18.08）%, （85.83±9.84）%, respectively. Results were consistent with those of R-CBA, the squared correlation factor was 0.9183 of the two assays. The new assay can easily discriminate a ＂TTP plasma sample from a non-TTP plasma sample （P 〈0.01）, and the coefficient of variation for the new assay was 6.17%. Improved Approximation Schemes for Early Work Scheduling on Identical Parallel Machines with a Common Due Date

We study the early work scheduling problem on identical parallel machines in order to maximize the total early work,i.e.,the parts of non-preemptive jobs that are executed before a common due *** preprocessing and constructing an auxiliary instance which has several good properties,for any desired accuracy,we propose an efficient polynomial time approximation scheme with running time O(f(1/ε)n),where n is the number of jobs and f(1/ε)is exponential in 1/ε,and a fully polynomial time approximation scheme with running time O(1/ε^(2m+1)+n)when the number of machines is fixed.

Evolutionary analysis of allotetraploid hybrids of red crucian carp×common carp,based on ISSR,AFLP molecular markers and cloning of cyclins genes

The allotetraploid hybrids of red crucian carp × common carp are the first reported artificially cultured polyploid fish with bisexual fertility and stable inheritance in vertebrate. Using ISSR and AFLP markers and the cyclins genes, the genomes and cyclin gene sequence changes were analyzed between the allotetraploid hybrids and their parents. The results indicated that the allotetraploids inherited many genetic characteristics from their parents and the genetic characteristics were stable after 15 generations. However, the allotetraploids had a closer genetic relationship with their original female parents and represented a bias toward the maternal progenitor. DNA fingerprinting analysis showed that the allotetraploids had undergone sequences deletion from their original parents and that the deleted sequences were mostly from the male parent's genome. Some non-parental bands were found in the allotetraploid hybrids. Sequences analysis of the cyclin A1 and B1 genes showed nonsynonymous substitutions of single nucleotides in codons that were different from their original parents, leading to non-parental amino acid loci. We speculate that the non-additivity in the allotetraploids, compared with their progenitors, could be an adjustment to the genomic shock from heterozygosity and polyploidy, allowing maintenance of genetic stability.

This paper studies a robust fault compensation and vibration suppression problem of flexible hypersonic *** controlled plant is represented by a cascade system composed of a nonlinear Ordinary Differential Equation(ODE)and an Euler-Bernoulli Beam Equation(EBBE),in which the vibration dynamics is coupled with the rigid dynamics and suffers from distributed faults.A state differential transformation is introduced to transfer distributed faults to an EBBE boundary and a longitudinal dynamics is refined by utilizing T-S fuzzy IF-THEN rules.A novel T-S fuzzy based fault-tolerant control algorithm is developed and related stability conditions are *** robust exponential stability and well-posedness are proved by using the modified l_(0)-semigroup based Lyapunov direct approach.A simulation study on the longitudinal dynamics of flexible hypersonic vehicles effectively verifies the validity of the developed theoretical results.

A Long-Time-Step-Permitting Tracer Transport Model on the Regular Latitude–Longitude Grid

If an explicit time scheme is used in a numerical model, the size of the integration time step is typically limited by the spatial resolution. This study develops a regular latitude–longitude grid-based global three-dimensional tracer transport model that is computationally stable at large time-step sizes. The tracer model employs a finite-volume flux-form semiLagrangian transport scheme in the horizontal and an adaptively implicit algorithm in the vertical. The horizontal and vertical solvers are coupled via a straightforward operator-splitting technique. Both the finite-volume scheme's onedimensional slope-limiter and the adaptively implicit vertical solver's first-order upwind scheme enforce monotonicity. The tracer model permits a large time-step size and is inherently conservative and monotonic. Idealized advection test cases demonstrate that the three-dimensional transport model performs very well in terms of accuracy, stability, and efficiency. It is possible to use this robust transport model in a global atmospheric dynamical core.

With the development of intelligent technology,the problem of Li DAR-based localization has played an increasingly important role in the localization of robots in unmanned *** the dense point clouds from the environment requires a large amount of memory,specific and stable structural features,such as pole-like objects in the environment,can serve as the ideal landmarks for localization in unmanned systems,which can effectively reduce the memory usage and mitigate the effects of dynamic environment *** this paper,the authors propose a pole-like objects extraction approach and then,it is applied into the localization system of mobile *** the same experimental conditions,the proposed method can extract more pole-like objects and achieve better long-term localization accuracy than some existing methods in different urban environmental datasets.
