An Electrical Stimulation Culture System for Daily Maintenance-Free Muscle Tissue Production

Low-labor production of tissue-engineered muscles(TEMs)is one of the key technologies to realize the practical use of muscleactuated *** study developed and then demonstrated the daily maintenance-free culture system equipped with both electrical stimulation and medium replacement *** avoid ethical issues,immortal myoblast cells C2C12 were *** system consisting of gel culture molds,a medium replacement unit,and an electrical stimulation unit could produce 12 TEMs at one *** contractile forces of the TEMs were measured with a newly developed microforce measurement *** the TEMs cultured without electrical stimulation generated forces of almost 2mN and were shortened by 10%in tetanic *** the contractile forces,electrical stimulation by a single pulse at 1 Hz was most effective,and the contractile forces in tetanus were over 2.5 *** the other hand,continuous pulses decreased the contractile forces of ***-stained cross-sections showed that myoblast cells proliferated and fused into myotubes mainly in the peripheral regions,and fewer cells existed in the internal *** must be due to insufficient supplies of oxygen and nutrients inside the *** increasing the supplies,one TEM might be able to generate a force up to around 10 *** tetanic forces of the TEMs produced by the system were strong enough to actuate microstructures like previously reported crawling *** daily maintenance-free culture system which could stably produce TEMs strong enough to be utilized for microrobots should contribute to the advancement of biohybrid devices.

The genus Macridiscus Dall,1902(Bivalvia:Veneridae)contains a few species which are important elements from warm temperate to tropical fauna of the Western *** firstly proposed by Dall(1902,Proceedings of the United States National Museum 26:335-412), Macridiscus was widely accepted as a subgenus of the genus Gomphina until 2006,Mikkelsen et al.(Zoological Journal of the Linnean Society 148:439-521.)suggested Macridiscus should be separated from genus Gomphina and elevated to an independent genus based on morphology and molecular *** taxonomy of the genus Macridiscus has in the past been based only on shell characters,and there has been no agreement about the number of valid species in this *** this study,we explore the taxonomy and phylogeny of Macridiscus species in order to resolve the systematics of the genus of Macridiscus,based not only on shell characters but also on mitochondrial and nuclear DNA *** morphological characters,the sequences of the cytochrome c oxidase subunit I(COI)of mitochondrial DNA and the first internal transcribed spacer region(ITS1)between the 18S and 5.8S ribosomal DNA were highly concordant and clearly suggested that three species should be recognized in the genus Macridiscus:*** (R(o|¨)mer,1860),***.("aequilatera"auctt.)(Lutaenko,2001)and ***(Koch,in Philippi,1843).Our finding supports Lutaenko's(2001,Phuket Marine Biological Center Special Publication 25:465-486.)classification of the genus Macridiscus(which he put under the genus Gomphina as a subgenus),and the morphological characters and geographical distribution of the three species were redescribed based on the molecular data.

Variations and mutations in the hepatitis B virus genome and their associations with clinical characteristics

Hepatitis B virus(HBV) infection is major global issue, because chronic HBV infection is strongly associated with liver cancer. HBV spread worldwide with variousmutations and variations. This variability, called quasispecies, is derived from no proof-reading capacity of viral reverse transcriptase. So far, thousands of studies reported that the variety of genome is closely related to the geographic distribution and clinical characteristics. Recent technological advances including capillary sequencer and next generation sequencer have made in easier to analyze mutations. The variety of HBV genome is related to not only antigenicity of HBs-antigen but also resistance to antiviral therapies. Understanding of these variations is important for the development of diagnostic tools and the appropriate therapy for chronic hepatitis B. In this review, recent publications in relation to HBV mutations and variations are updated and summarized.

Hepatocellular carcinoma metastasizing to the skull base involving multiple cranial nerves

We describe a rare case of HCV-related recurrent multiple hepatocellular carcinoma (HCC) metastasizing to the skull base involving multiple cranial nerves in a 50-year- old woman. The patient presented with symptoms of ptosis, fixation of the right eyeball, and left abducens palsy, indicating disturbances of the right oculomotor and trochlear nerves and bilateral abducens nerves. Brain contrast-enhanced computed tomography (CT) revealed an ill-defined mass with abnormal enhancement around the sella turcica. Brain magnetic resonance imaging (MRI) disclosed that the mass involved the clivus, cavernous sinus, and petrous apex. On contrast-enhanced MRI with gadolinium-chelated contrast medium, the mass showed inhomogeneous intermediate enhancement. The diagnosis of metastatic HCC to the skull base was made on the basis of neurological findings and imaging studies including CT and MRI, without histological examinations. Further studies may provide insights into various methods for diagnosing HCC metastasizing to the craniospinal area.

Genetic Variation in the Testis-Specific HASPIN Gene Encoding a Serine/Threonine Protein Kinase in Infertile Japanese Males

HASPIN is a serine/threonine protein kinase predominantly expressed during spermatogenesis and localized in the nucleus. The HASPIN gene is conserved from yeast to mammals and plants. To investigate any possible associations between HASPIN polymorphisms and impaired spermatogenesis in Japanese males, we screened for mutations in the HASPIN coding sequence (CDS) using DNA from 282 sterile male patients and 262 fertile male volunteers. Polymorphisms were found at 10 positions within the HASPIN CDS. Among these 10 polymorphisms, 5 were found only in the infertile group, 3 of which were nonsynonymous. These polymorphisms found only in the infertile patients may be a cause of male infertility and thus valuable candidates for further study of this condition.

Preliminary Clinical Study Using a Novel Wound Dressing Composed of Hyaluronic Acid and Collagen Containing EGF

This clinical trial aimed to evaluate the efficacy and safety of a novel wound dressing composed of hyaluronic acid (HA) and collagen (Col) containing epidermal growth factor (EGF), referred to as EGF-wound dressing. EGF-wound dressing was prepared by freeze-drying a mixed aqueous solution of high-molecular-weight HA, low-molecular-weight HA and heat-denatured Col containing EGF. EGF-wound dressing was applied to skin defects, such as intractable skin ulcers, burn ulcers, traumatic skin defects and skin donor-site wounds. The dressing was changed twice a week for a period of 6 weeks or longer, if necessary. The primary endpoints were size of wound area, formation of granulation tissue, extent of epithelialization, infection control and macroscopic appearance. Effectiveness, safety and overall clinical evaluation were scored by plastic surgeons, as authorized by the Japanese Society of Plastic and Reconstructive Surgery. This study was registered with the University Hospital Medical Information Network (UMIN0000005264). Healthy granulation tissue and rapid epithelialization were observed for a given period after application of EGF-wound dressing onto the wounds. Most cases were assessed as having achieved good or excellent results. This clinical study demonstrated that EGF-wound dressing was beneficial in the treatment of various skin defects.

Background: The ability to climb stairs (ascending and descending stair without using a handrail) and rise from a chair (rising from chairs without using an elbow rest) are among the most important measures of physical function for ADL evaluation for the independent living, and assessed by the questioners on many epidemiological studies in elderly. But little is known about the relationship between the self-reported performance level of the tasks and lower leg strength and power in very elderly people. The purpose of this study was to ascertain the relationship between the self-reported performance level of two tasks and the lower leg function in community-dwelling 80-year-old population. Methods: Out of 994 persons who were 80 years old living in Morioka City, Iwate Prefecture, Japan, 607 individuals (236 men and 371 women) underwent a physical fitness test that included measurements of leg extensor power and knee extensor strength. The ability to climb up stair and to rise from a chair was assessed by self- reported questionnaire which was ranked in three levels. The area under the receiver-operating characteristic (ROC) curve (AUC) was used to evaluate the measurements. Results: The cut-off points for the leg extensor power of subjects who could completely perform the stair-climbing and chair-rising functions were determined to be approximately 8.6 watt/kg body mass for men and 5.6 watt/kg body mass for women. In addition, the cut-off points for the knee extensor strength of subjects who could completely perform the stair-climbing and chair-rising functions were determined to be approximately 0.97 kg/kg body mass for men and 0.84 kg/kg body mass for women. Conclusions: From a practical viewpoint, the present study suggested that the cut-off points of leg extensor power and knee extensor strength can be used as targets in simple self-reported questionnaires to help in screening for mobility in 80-year-old
