Endogenous prostaglandin D<sub>2</sub>synthesis inhibits e-selectin generation in human umbilical vein endothelial cells

We examined the role of prostaglandin D2 (PGD2) in the formation of E-selectin following inter-leukin-1 (IL-1) stimulation in human umbilical vein endothelial cells (HUVEC) transfected with lipocaline-type PGD2 synthase (L-PGDS) genes. HUVEC were isolated from human umbilical vein and incubated with 20 U/mL IL-1 and various concentrations of authentic PGD2. The isolated HUVEC were also transfected with L-PGDS genes by electroporation. The L-PGDS-transfected HUVEC were used to investigate the role of endogenous PGD2 in IL-1-stimulated E-selectin biosynthesis. We also used an anti-PGD2 antibody to examine whether an intracrine mechanism was involved in E-selectin production. PGD2 and E-selectin levels were determined by radio-immunoassay and enzyme- immunoassay, respectively. E-selectin mRNA was assessed by real-time RT-PCR. IL-1-stimulated E-selectin production by HUVEC was dose-dependently inhibited by authentic PGD2 at concentrations greater than 10–6 mol/L. L-PGDS gene-transfected HUVEC produced more PGD2 than HUVEC transfected with the reporter gene alone. IL-1 induced increases in E-selectin production in HUVEC transfected with the reporter genes alone. However, this effect was significantly attenuated in the case of IL-1 stimulation of HUVEC trans-fected with L-PGDS genes, and accompanied by an apparent suppression of E-selectin mRNA expression. Neutralization of extracellular PGD2 by anti-PGD2- specific antibody influenced neither E-selectin mRNA expression nor E-selectin biosynthesis. HUVEC transfected with L-PGDS genes showed increased PGD2 synthesis. This increase was associated with attenuation of both E-selectin generation and E-selectin mRNA expression. The results suggest that endogenous PGD2 decreases E-selectin synthesis and E-selectin mRNA expression, probably through an intracrine mechanism.

Low Salt Diet in Pregnant Mothers Is Associated with Enhanced Salt Appetite in Their Offspring of Dahl Salt-Sensitive Rats

Objectives: We investigated an influence of salt intake during gestation or lactation in salt preference of weaning Dahl salt sensitive (Dahl S) strain. Material and Methods: Nine-week-old female Dahl S rats, after mated with the male, were divided into 1) high-salt group fed a 4% NaCl diet (high-salt mother) or 2) low-salt group fed a 0.3% NaCl diet (low-salt mother) during gestation or lactation periods. Using 0.4%, 0.6% and 0.8% (w/v) saline solutions, we assessed salt preference in their offspring after weaning. Systolic blood pressure (SBP) was determined by tail cuff method. Results: Both the male and female offspring from low-salt mothers during gestation consumed equal amounts of any saline solution. However, the amount of each saline solution was higher in the offspring of low-salt mothers than those of high-salt mothers. This resulted in a significant increase of salt intake in both the male and female offspring of low-salt mothers compared with those of high-salt mothers. In contrast, both the male and female offspring from low-mothers during lactation rather preferred the lower concentration of saline solution, and this resulted in less salt intake in the offspring of low-salt mothers than those of high-salt mothers. Conclusions: We demonstrated that low salt diet in pregnant mothers increased salt intake of their offspring. The offspring of mothers fed a low salt diet during lactation had less salt than those of high-salt mothers. The influence of salt intake of mothers in their offspring varies along with the growth stage of their babies.

Fructose-Induced Cognitive Dysfunction Is Associated with Increased Oxidative Stress in the Rat Brains

High fructose consumption is a risk factor for diabetes. Type 2 diabetes is associated with cognitive and memory impairment. To determine if fructose intake might affect memory function independently of diabetes development, we investigated the effects of monosaccharides on memory in prediabetic state. We examined whether 2.5% apple juice solution could influence cognitive function in salt-sensitive Dahl rats using passive avoidance methods. Furthermore, we examined the role of monosaccharides intake in cognitive function and oxidative stress in the brain. Four-week consumption of either apple juice or 2.5% fructose did not affect the blood glucose concentrations;however, apple juice, but not fructose, significantly decreased cognitive function compared with that of control rats given water. Second, Wistar rats aged 4 weeks were assigned to four groups given water, or 1.25% glucose, or fructose, or galactose solutions for 11 weeks. The fructose group had only slightly higher blood glucose concentrations than the control group. However, memory function engraved at age of 7 weeks and evaluated for 8 weeks in a passive avoidance test, was significantly decreased in the fructose and galactose groups compared with those in the glucose group. Oxidative stress in the brain, assessed by the tissue malondialdehyde (MDA) content, was significantly increased in the fructose group compared with that in the control group, and the decrease in cognitive function significantly correlated with the MDA content. These findings suggest that long-term apple juice consumption decreases memory function, possibly through an increase in oxidative stress in the brain, in turn induced by fructose overloading. Fructose-induced cognitive dysfunction is likely mediated by mechanisms other than insulin resistance.

Continuous Subcutaneous Administration of Miso Extracts Attenuates Salt-Induced Hypertension in Dahl Salt-Sensitive Rats

Objective: Long-term intake of Miso attenuates hypertension in Dahl salt-sensitive (Dahl S) rats through an increased urinary sodium excretion. We examined whether a bolus injection into the peritoneal cavity (i.p.) or a continuous subcutaneous infusion of a Miso extract attenuates hypertension in Dahl S rats. Materials and Methods: We investigated the effects of a bolus, i.p. injection of 50 mg Miso extract in 0.5 mL on hypertension in Dahl S rats, and examined whether a long-term subcutaneous infusion of the Miso extract (50 mg Miso/day), using an osmotic mini-pump working for 14 days, attenuates hypertension in Dahl S rats. Results: A bolus, i.p. injection of 50 mg Miso extract decreased SBP in 2 hrs. The reduction was significant at 4 hrs, and SBP returned to the baseline at 24 hrs. The SBP reduction at 4 hrs after the injection was increasingly greater each day during the 4 days. The SBP reduction by the Miso extract was dose-dependent and the antihy-pertensive activity occurs in a <5 kDa fraction of the extract. The subcutaneous infusion of 50 mg Miso extract/day for 14 days significantly attenuated hypertension in Dahl S rats. The SBP reduction was associated with significant decreases in the heart and kidney weights. Urinary protein excretion significantly decreased in the Miso group. The SBP reduction was not associated with increases in either urinary sodium excretion or fractional excretion of sodium. Conclusions: SBP reduction by very low-dose of the Miso extract may be mediated partly by mechanisms other than renal sodium handling.

An Observational Study on the Clinical and Nutritional Factors Determining the Kidney Function at One Year after Transplantation

Objective: The aim of the present study was to investigate the clinical and nutritional factors influencing the renal function of the transplanted kidney during a one-year follow-up period after transplantation. Patients: The present prospective observational study included 52 patients who underwent kidney transplantation at Jichi Medical University Hospital from 2014 to 2016. Results: The serum creatinine (sCr) concentration at one month after transplantation was closely related to the concentration at 12 months. The recipients were divided into two groups based on the sCr concentration at one month after transplantation. Recipients with a sCr concentration greater than or equal to the median were classified into Group H, while those with concentrations that were less than the median were classified into Group L. A stepwise multiple regression analysis demonstrated that the salt intake in male recipients was an independent predictor of the renal function at 12 months (β = 0.663, p β = 0.618, p < 0.01). Moreover, in female recipients, the BMI and serum uric acid values in Group H were significantly higher than those in Group L. The BMI values of the female recipients in Group H were significantly higher than those in Group L at all times during the follow-up period (p < 0.01). Conclusion: The daily salt intake was independently associated with the renal function at one year after kidney transplantation in male recipients. In female recipients, the BMI was independently associated with the sCr concentration during the one-year follow-up period.

Who needs further evaluations to diagnose upper urinary tract urothelial cancers among patients with abnormal findings by enhanced CT?

Objective:We evaluated who would need further evaluations such as retrograde pyelography(RP)and/or ureteroscopy to diagnose upper urinary tract urothelial cancers(UUTUCs)when abnormal findings for the upper urinary tract(UUT)were detected by enhanced computed tomography(CT).Methods:We retrospectively analyzed 125 patients who underwent enhanced CT for various reasons and had abnormal findings for the *** whose tumors were suspected to be of extraureteral origin were *** patients received RP and/or ureteroscopy to evaluate the ***:The median age of the 125 patients was 70 years and gross hematuria(26.4%)was the most frequently observed ***,ureteroscopy and both were performed for 121,59 and 55 patients,*** revealed tumor-like lesions in 58 patients and the other patients had non-tumor-like *** were found in 43(34.4%)of the 125 *** of them had tumor-like lesions on *** 58 patients who had tumor-like lesions on CT,univariate and multivariate analyses revealed that tumor diameter and tumor enhancement were significant predictive factors for *** curve analysis of enhanced CT to diagnose UUTUCs revealed that a tumor diameter of 18 mm was the best cutoff *** sensitivity,specificity and accuracy were 90.0%,98.8% and 92.7% for RP and 95.5%,100% and 97.1%for ureteroscopy,*** of them had high sensitivity,specificity and ***:We should decide to evaluate the UUT according to the tumor diameter on enhanced *** we evaluate the UUT in patients with tumor diameters of less than 20 mm,ureteroscopy is recommended.
