沉默信息调节因子2相关酶类3(silent mating type information regulation 2 homolog 3,SIRT3)是位于线粒体基质中的一种去乙酰化酶,在线粒体能量代谢、氧化应激调节、延缓衰老等方面具有重要的作用。本研究通过采用SIRT3-EGFP报告基因系统及实时荧光定量聚合酶链式反应技术,从白藜芦醇、山柰酚、石榴皮鞣素、安石榴苷、漆树黄酮、咖啡醇、矢车菊素、飞燕草素葡萄糖苷这8种多酚类化合物中筛选SIRT3激活剂,并评价筛选得到的SIRT3多酚激活剂对于中波紫外线(ultraviolet radiation B,UVB)诱导的人正常皮肤永生化角质形成细胞(human immortalized keratinocytes,HaCaT)衰老的抑制作用。结果表明,白藜芦醇、山柰酚、安石榴苷、漆树黄酮和咖啡醇具有显著增强SIRT3基因表达水平的作用(P<0.05);白藜芦醇、安石榴苷、漆树黄酮以及咖啡醇均能有效降低UVB诱导的细胞内活性氧水平;这5种多酚均显著提高了谷胱甘肽(reduced glutathione,GSH)/氧化型谷胱甘肽(oxidized glutathione,GSSG)比值(P<0.05),白藜芦醇与咖啡醇显著增强了超氧化物歧化酶2活性(P<0.05、P<0.01);此外,安石榴苷不仅可以激活SIRT3,修复细胞的抗氧化系统,且显著降低了细胞衰老标志物SA-β-gal的相对活力(P<0.05)。结论:白藜芦醇、山柰酚、安石榴苷、漆树黄酮和咖啡醇均能激活SIRT3基因的表达,其中安石榴苷能够显著修复UVB诱导的HaCaT细胞衰老,其抗衰老机制可能与SIRT3-烟酰胺腺嘌呤二核苷酸磷酸-GSH/GSSG抗氧化途径有关。

The biodegradation behavior of an ultrafne-grained(UFG) magnesium alloy ZE41A containing rare-earth(RE),obtained through multi-pass of equal channel angular pressing(EACP),was investigated by electrochemical measurements in Hank’s *** highest value of charge transfer resistance was obtained in the electrochemical impedance spectroscopy plots of the ECAPed samples with the largest pressing pass,while a movement of corrosion potential toward noble direction and a decrement of corrosion current density were observed in the potentiodynamic polarization curves of the ECAPed samples with further pressing *** results indicated that ECAP could be an effcient way to reduce the biodegradation rate of the ZE41A alloy.

Effects of age on survival and morbidity in gastric cancer patients undergoing gastrectomy

AIM To evaluate clinicopathological features and surgical outcomes of gastric cancer in elderly and non-elderly patients after inverse probability of treatment weighting(IPTW) method using propensity *** We enrolled a total of 448 patients with histologically confirmed primary gastric carcinoma who received gastrectomies. Of these, 115 patients were aged > 80 years old(Group A), and 333 patients were aged < 79 years old(Group B). We compared the surgical outcomes and survival of the two groups after *** Postoperative complications, especially respiratory complications and hospital deaths, were significantly more common in Group A than in Group B(P < 0.05). Overall survival(OS) was significantly lower in Group A patients than in Group B patients. Among the subset of patients who had pathological Stage Ⅰ?disease, OS was significantly lower in Group A(P < 0.05) than Group B, whereas cause-specific survival was almost equal in the two groups. In multivariate analysis, pathological stage, histology, and extent of lymph node dissection wereindependent prognostic values for *** When the gastrectomy was performed in gastric cancer patients, we should recognized high mortality and comorbidities in that of elderly. More extensive lymph node dissection might improve prognoses of elderly gastric cancer patients.

Corneal topographic changes and surgically induced astigmatism following combined phacoemulsification and 25-gauge vitrectomy

AIM： To evaluate corneal topographic changes and surgically induced astigmatism （SIA） after combined phacoemulsification and 25-gauge transconjunctival sutureless vitrectomy （25-G TSV）.METHODS： A retrospective study on 96 eyes of 87 patients who underwent combined phacoemulsification and 25-G TSV. The different topographic parameters and SIA were analyzed pre- and ***： There was no significant changes in corneal topographic parameters at different follow up periods. Only surface regularity index changed significantly in the 2nd postoperative week and then returned to baseline values thereafter. Mean SIA gradually decreased to reach 0.12 D by the 6th postoperative ***： Corneal surface and astigmatic changes are insignificant in either early or late postoperative periods following combined phacoemulsification and 25-G TSV. The SIA was the minimum among previous reports on sutureless vitrectomy alone or combined with phacoemulsification. Improvement of SIA did not stop at the 3rd postoperative month but it continued till the 6th month postoperatively.

Improving corrosion resistance of RE-containing magnesium alloy ZE41A through ECAP

Significant grain refinement was achieved in rare earth (RE) containing aeronautic magnesium alloy ZE41A through equal-channel angular pressing (ECAP) using rotary die at 603 K. Influence of ECAP pass number on its microstructure change and corrosion behavior was investigated by optical microscope (OM)/scanning electron microscope (SEM) observation and potentiostatic polarization tests in aqueous solution of NaCl, respectively. The results showed that ultrafine equiaxial grains (about 2.5 μm) were obtained over 16 passes due to plastic-induced grain refinement accommodated by dynamic recrystallization. The lower corrosion current density and nobler corrosion potential correlated with large number of pressing passes were attributed to the low tendency toward localized corrosion with broken secondary phase after homogenization on ultrafine-grained Mg matrix. The multi-pass ECAP method made the ZE41A aeronautic magnesium alloy more attractive since severe plastic deformation may significantly improve its corrosion resistance besides superior mechanical properties.
