Aralia elata inhibits neurodegeneration by downregulating O-GlcNAcylation of NF-κB in diabetic mice

AIM： To investigate the role of O-GIcNAcylation of nuclear factor-kappa B （NF-KB） in retinal ganglion cell （RGC） death and analysedthe effect of Aralia elata （AE） on neurodegen- eration in diabetic mice. METHODS： C57BL/6mice with streptozotocin-induced diabetes were fed daily with AE extract or control （CTL） diet at the onset of diabetes mellitus （DM）. Two months af- tar injection of streptozotocin or saline, the degree of cell death and the expression of O-GIcNAc transferase （OGT）, N-acetyl-b-D-glucosaminidase （OGA）, O-GIcNAcylated pro- teins, and O-GIcNAcylation of NF-KB were examined. RESULTS： AE did not affect the metabolic status of diabetic mice. The decrease in the inner retinal thickness （P〈0.001 vs CTL, P〈0.01 vs DM） and increases in RGCs with terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling （P〈0.001 vs CTL, P〈0.0001 vs DM）, glial activation, and active caspase-3 （P〈0.0001 vs CTL, P〈0.0001 vs DM） were blocked in diabetic retinas of AE extract-fed mice. Expression levels of protein O-GIcNAcylation and OGT were increased in diabetic retinas （P〈0.0001 vs CTL）, and the level of O-GIcNAcylation of the NF-KB p65 subunit was higher in diabetic retinas than in controls （P〈0.0001 vs CTL）. AE extract downregulated O-GIcNAcylation of NF-KB and prevented neurodegeneration induced by hyperglycemia （P〈0.0001 vs DM）. CONCLUSION： O-GIcNAcylation of NF-KB is concerned in neuronal degeneration and that AE prevents diabetes-in- duced RGC apoptosis via downregulation of NF-KB O-GI- cNAcylation. Hence, O-GIcNAcylation may be a new object for the treatment of DR, and AE may have therapeutic pos- sibility to prevent diabetes-induced neurodegeneration.

Kale Juice Improves Coronary Artery Disease Risk Factors in Hypercholesterolemic Men

Objective To evaluate the effect of 3-month kale （Brassica oleracea acephala） juice supplementation on coronary artery disease risk factors among hypercholesterolemic men. Methods Thirty-two men with hypercholesterolemia （〉 200 mg/dL） were recruited after annual health examinations among the faculty and staff at university. The subjects consumed 150 mL of kale juice per day for a 12-week intervention period. Dietary and anthropometric assessments were performed and blood samples were collected to evaluate biochemical profiles before and after supplementation. Results Serum concentrations of HDL-cholesterol, and HDL- to LDL-cholesterol ratio were significantly increased by 27% （P〈0.0001） and 52% （P〈0.0001）, respectively. The LDL-cholesterol concentration and the atherogenic index were significantly reduced by 10% （P=0.0007） and 24.2% （P〈0.0001）, respectively without affecting body mass index, waist and hip circumferences, or nutrient intakes after three months of supplementation. While there was no difference in the concentration of malondialdehyde, significant increase in glutathione peroxidase activity （P=0.0005） were accompanied by a significant increase in the serum selenium level （P=0.0132）. It was also found that the responses of these risk factors to kale juice administration were dependent on smoking status. Conclusion Regular meals supplementation with kale juice can favorably influence serum lipid profiles and antioxidant systems, and hence contribute to reduce the risks of coronary artery disease in male subjects with hyperlipidemia.

AIM： To investigate the anti-cancer mechanisms of Korean mistletoe lectin （Viscum album coloratura agglutinin, VCA） using a human colon cancer cell line （COLO）. METHODS： Cytotoxic effects of VCA on COLO cells were determined by 3-（4,5-dimethylthiazol-2-yl）-2,5- diphenyltetrazolium bromide （MTI-） assay in vitro and tumor-killing effects in vivo. To study the mechanisms involved, the expression of various pro-caspases, anti- apoptotic proteins, and death receptors was determined by western blot. To determine which death receptor is involved in VCA-induced apoptosis of COLO cells, cytotoxicity was examined by MTT assay after treatment with agonists or antagonists of death receptors. RESULTS： VCA killed COLO cells in a time- and dosedependent manner and induced complete regression of tumors in nude mice transplanted with COLO cells. Treatment of COLO cells with VCA activated caspase-2, -3, -8, and -9 and decreased expression of anti-apoptotic molecules including receptor interacting protein, nuclear factor-κB, X-linked inhibitor of apoptosis protein, and Akt/protein kinase B. We then examined the involvement of death receptors in VCA-induced apoptosis. Only tumor necrosis factor receptor 1, among the death receptors examined, was involved in apoptosis of COLO cells, evidenced by inhibition of VCA-induced apoptosis and decreased activation of caspases, particularly caspase-8, by tumor necrosis factor receptor 1 antagonizing ***： VCA-induced apoptotic COLO cell death is due to the activation of caspases and inhibition of antiapoptotic proteins, in part through the tumor necrosis factor receptor 1 signaling pathway.

A case study of environmental characteristics on urban road-surface and air temperatures during heat-wave days in Seoul

High road-surface temperature due to heat waves can lead to dangerous driving conditions such as tire blowouts and deformation induced by thermal stress on the roads. In this study, a Mobile Observation Vehicle dataset, with high spatial and temporal resolutions for the heat-wave episode that occurred on 16–17 August 2018, is used to understand environmental characteristics on urban road-surface and air temperatures in Seoul. This study demonstrates that the magnitude of urban road-surface temperature is dependent on the differences in incoming solar radiation due to screening of high-rise buildings in the Gangnam area, and is associated with the topographical features in the Gangbuk area. The road-surface temperature in the section of darker-colored asphalts was higher than that of lighter-colored asphalts, with a mean difference of 6.8°C, and both surface and air temperatures on the iron plate were highest, with means of 51.7°C and 35.1°C,respectively. In addition, during the water-sprinkling period, road-surface temperature was cooled by about 8.7°C(19%) compared with that in the period without water-sprinkling, but there was no significant change in air temperature. The current results could be practically used to improve roadsurface temperature prediction models for civil engineers or road managers.

Species delimitation in the green algal genus Codium (Bryopsidales) from Korea using DNA barcoding

Codium, one of the largest marine green algal genera, is difficult to delimit species boundary accurately based on morphological identification only. DNA barcoding is a powerful tool for discriminating species of seaweeds. The plastid elongation factor TU （tufA） is considered as maker to perform DNA barcoding of green algal species than rbcL gene due to universality and rapid evolution rate. We conducted DNA barcoding application to Codium specimens from the Jeju Island, Korea to overcome the limit of morphological identification and to confirm the species diversity. As a result of applying tufA marker, we newly generated fifty-five tufA barcodes to resolve eight species. TufA marker exhibited 6.1%–21.8% interspecific divergences, wider than the gap of rbcL exon 1, 3.5%–11.5%. Molecular analysis of rbcL exon 1 sequences of Codium revealed eight distinct species like tufA analysis separated in five phylogenetic groups. DNA barcoding of the genus Codium using tufA marker is more helpful to overcome the limit of morphological identification, and this is more potential to reveal cryptic species and to resolve the relationships among subspecies than rbcL analysis alone. The complement of tufA barcoding and rbcL analyses including morphology for the genus Codium in the northwestern Pacific will give much more reliable achievement for discovering species diversity and resolving the phylogenetic relationships.

BACKGROUND Carotid blowout syndrome(CBS)is a rupture of the carotid artery and is mainly caused by radiation and resection of head and neck cancers or direct tumor invasion of the carotid artery *** is a life-threatening clinical *** is no established and effective mode of management of ***,there is no established preceding sign or symptom;therefore,preventive efforts are not clinically *** SUMMARY We described two cases of CBS that occurred in patients with head and neck cancer after definitive chemoradiotherapy(CRT)using three-dimensional conformal intensity-modulated radiation *** men aged 61 and 56 years with locally advanced head and neck cancer were treated with definitive *** completing CRT,both of them achieved complete ***,they had persistent severe pain in the oropharyngeal mucosal region and the irradiated neck despite the use of opioid analgesics and rehabilitation for relief of contracted ***,continuous follow-up imaging studies showed no evidence of cancer *** to twelve months after completing CRT,the patients visited the emergency room complaining about massive oronasal *** showed rupture of carotid artery pseudoaneurysms on the irradiated *** attempting to secure hemostasis with carotid arterial stent insertion and coil embolization,both patients died because of repeated bleeding from the *** In patients with persistent pain in irradiated sites,clinicians should be suspicious of progressing or impending CBS,even in the three-dimensional conformal intensity-modulated radiation therapy era.

In this paper,we consider the numerical stability of gravity-capillary waves generated by a localized pressure in water of finite depth based on the forced Korteweg-de Vries(FKdV)framework and the polynomial *** stability studies are focused on the symmetric solitary wave for the subcritical flow with the Bond number greater than one *** its steady symmetric solitarywave-like solutions are randomly perturbed,the evolutions of some waves show stability in time regardless of the randomness while other waves produce unstable *** representing the perturbation with a random variable,the governing FKdV equation is interpreted as a stochastic *** polynomial chaos expansion of the random solution has been used for the study of stability in two *** it allows us to identify the stable solution of the stochastic governing *** it is used to construct upper and lower bounding surfaces for unstable solutions,which encompass the fluctuations of waves.

A novel isoxazole compound CM2-II-173 inhibits the invasivephenotype of triple-negative breast cancer cells

Invasion and metastasis are important hallmarks of breast cancer and are the leading cause of patient ***-negative breast cancer (TNBC) is an aggressive type of breast cancer characterized by a poor prognosis and a lackof effective targeted therapies. The present study investigated the inhibitory effect of a novel FTY720 derivative on theinvasive phenotype of TNBC cells. Here, we showed that a novel compound with an isoxazole ring, 4-(3-Decylisoxazol-5-yl)-1-hydroxy-2-(hydroxymethyl)butan-2-aminium chloride (CM2-II-173), significantly inhibitedinvasiveness of MDA-MB-231 TNBC cells. Expression of matrix metalloproteinase (MMP)-9 and invasiveness ofMCF10A normal breast cells induced by sphingosine-1-phosphate (S1P) were reduced by CM2-II-173 *** of pMEK1, pAkt, pERK, and p38 MAPK by S1P were inhibited by treatment with *** and anchorage-independent growth of MDA-MB-231 TNBC cells were significantly decreased by CM2-II-173. CM2-II-173 efficiently induced apoptosis in MDA-MB-231 TNBC cells. CM2-II-173 significantly inhibitedinvasive phenotypes of breast, liver, prostate, and ovarian cancer cells. CM2-II-173 exhibited a more potent effect onthe invasiveness of MDA-MB-231 TNBC cells compared to FTY720. Taken together, this study demonstrated thatCM2-II-173 has the potential to be a lead compound that can inhibit cancer progression of not only TNBC cells, butalso of liver, prostate, and ovarian cancer cells.

Effect of change of sand properties on travel distance of ricocheted debris
