RFC2 may contribute to the pathogenicity of Williams syndrome revealed in a zebrafish model

Williams syndrome(WS)is a rare multisystemic disorder caused by recurrent microdeletions on 7q11.23,characterized by intellectual disability,distinctive craniofacial and dental features,and cardiovascular *** studies have explored the roles of individual genes within these microdeletions in contributing to WS ***,we report five patients with WS with 1.4 Mb-1.5 Mb microdeletions that include RFC2,as well as one patient with a 167-kb microdeletion involving RFC2 and six patients with intragenic variants within *** investigate the potential involvement of RFC2 in WS pathogenicity,we generate a rfc2 knockout(KO)zebrafish using CRISPR-Cas9 ***,we generate a KO zebrafish of its paralog gene,rfc5,to better understand the functions of these RFC genes in development and *** rfc2 and rfc5 KO zebrafish exhibit similar phenotypes reminiscent of WS,including small head and brain,jaw and dental defects,and vascular ***-seq analysis reveals that genes associated with neural cell survival and differentiation are specifically affected in rfc2 KO *** addition,heterozygous rfc2 KO adult zebrafish demonstrate an anxiety-like behavior with increased social *** results suggest that RFC2 may contribute to the pathogenicity of WS,as evidenced by the zebrafish model.

To the Editor： A lumbar epidural block is used to treat patients with a herniated intervertebral disc, spinal stenosis, or lumbosacral radiculopathy. This approach reduces radicular pain by decreasing inflammation of nerve roots through epidural corticosteroid injection. Patients with moderate-to-severe spinal stenosis who taking various anticoagulant agents have more risk for epidural hematoma.

Macrophage migration inhibitory factor(miF)is a chemokine that plays an essential role in immune system *** studies suggested that miF protects neurons in ischemic ***,few studies are reported on the role of miF in neurological recovery after ischemic *** purpose of this study is to identify the molecular mechanism of neuroprotection mediated by *** neuroblastoma cells were incubated in Dulbecco’s modified Eagle’s medium under oxygen-glucose deprivation(OGD)for 4 hours and then returned to normal aerobic environment for reperfusion(OGD/R).30 ng/mL miF recombinant(30 ng/mL)or ISO-1(miF antagonist;50μM)was administered to human neuroblastoma *** cell cultures were assigned to one of four groups:control,OGD/R,OGD/R with miF,OGD/R with *** viability was analyzed using WST-1 *** levels of brain-derived neurotrophic factor(BDNF),microtubule-associated protein 2(MAP2),Caspase-3,Bcl2,and Bax were detected by western blot assay and immunocytochemistry in each group to measure apoptotic ***-1 assay results revealed that compared to the OGD/R group,cell survival rate was significantly higher in the OGD/R with miF group and lower in the OGD/R with ISO-1 *** blot assay and immunocytochemistry results revealed that expression levels of BDNF,Bcl2,and MAP2 were significantly higher,and expression levels of Caspase-3 and Bax were significantly lower in the miF group than in the OGD/R *** levels of BDNF,Bcl2,and MAP2 were significantly lower,and expression levels of Caspase-3 and Bax were significantly higher in the ISO-1 group than in the OGD/R *** administration promoted neuronal cell survival and induced high expression levels of BDNF,MAP2,and Bcl2(anti-apoptosis)and low expression levels of Caspase-3 and Bax(pro-apoptosis)in an OGD/R *** results suggest that miF administration is effective for inducing expression of BDNF and leads to neuroprotection of neuronal cells

Comparison of the efficacy and safety of sedation between dexmedetomidine-remifentanil and propofol-remifentanil during endoscopic submucosal dissection

AIM: To compare the efficacy and safety of sedation protocols for endoscopic submucosal dissection(ESD) between dexmedetomidine-remifentanil and ***: Fifty-nine patients scheduled for ESD were randomly allocated into a dexmedetomidineremifentanil(DR) group or a propofol-remifentanil(PR) group. To control patient anxiety, dexmedetomidine or propofol was infused to maintain a score of 4-5 on the Modified Observer's Assessment of Alertness/Sedation scale. Remifentanil was infused continuously at a rate of 6 μg/kg per hour in both groups. The ease of advancing the scope into the throat, gastric motility grading, and satisfaction of the endoscopist and patient were assessed. Hemodynamic variables and hypoxemic events were compared to evaluate patient ***: Demographic data were comparable between the groups. The hemodynamic variables and pulse oximetry values were stable during the procedure in both groups despite a lower heart rate in the DR group. No oxygen desaturation events occurred in either group. Although advancing the scope into the throat was easier in the PR group("very easy" 24.1% vs 56.7%, P = 0.010), gastric motility was moresuppressed in the DR group("no + mild" 96.6% vs 73.3%, P = 0.013). The endoscopists felt that the procedure was more favorable in the DR group("very good + good" 100% vs 86.7%, P = 0.042), whereas patient satisfaction scores were comparable between the groups. En bloc resection was performed 100% of the time in both groups, and the complete resection rate was 94.4% in the DR group and 100% in the PR group(P = 0.477). CONCLUSION: The efficacy and safety of dexmedetomidine and remifentanil were comparable to propofol and remifentanil during ESD. However, the endoscopists favored dexmedetomidine perhaps due to lower gastric motility.

BACKGROUND： Mesenchymal stem cells （MSCs） appear to be a good alternative to Schwann cells in the treatment of peripheral nerve injury. Fetal stem cells, like umbilical cord blood （UCB） and umbilical cord （UC） stem cells, have several advantages over adult stem cells. OBJECTIVE： To assess the effects of UC-derived MSCs （UCMSCs） and UCB-derived MSCs （UCBMSCs） in repair of sciatic nerve defects. DESIGN, TIME AND SETTING： A randomized controlled animal experiment was performed at the laboratory of Department of Oral and Maxillofacial Surgery, Seoul National University Dental Hospital, from July to December 2009. MATERIALS： UCMSCs were provided by the Research Institute of Biotechnology, Dongguk University. UCBMSCs were provided by the Laboratory of Stem Cells and Tumor Biology, College of Veterinary Medicine, Seoul National University. Dulbecco＇s modified Eagle＇s medium （DMEM） was purchased from Gibco-BRL, USA. METHODS： Seven-week-old Sprague-Dawley rats were randomly and evenly divided into three groups： DMEM, UCBMSCs, and UCMSCs. A 10-mm defect in the left sciatic nerve was constructed in all rats. DMEM （15 μL） containing 1×10^6 UCBMSCs or UCMSCs was injected into the gap between nerve stumps, with the surrounding epineurium as a natural conduit. For the DMEM group, simple DMEM was injected. MAIN OUTCOME MEASURES： At 7 weeks after sciatic nerve dissection, dorsal root ganglia neurons were labeled by fluorogold retrograde labeling. At 8 weeks, electrophysiology and histomorphometry were performed. At 2, 4, 6, and 8 weeks after surgery, sciatic nerve function was evaluated using gait analysis. RESULTS： The UCBMSCs group and the UCMSCs group exhibited similar sciatic nerve function and electrophysiological indices, which were better than the DMEM group, as measured by gait analysis （P 〈 0.05）. High performance of TiO_(2)/Cu_(x)O photoelectrodes for regenerative solar energy storage in a vanadium photoelectrochemical cell

Photocatalysts for harvesting solar energy to either electricity or chemical fuels have attracted much attention recently, but they have big obstacles such as wide bandgaps and rapid charge recombinations to overcome for final applications. In this study, we investigates a useful method to utilize vanadium redox pairs, which are commonly applied for vanadium redox flow batteries, to diminish charge recombinations and thus to enhance photocurrent response in regenerative solar energy storage. The results reveal significant improvements in photocurrent density by forming cuprous and cupric oxides in TiO_(2)/Cu_(x)O electrodes under solar AM 1.5 illuminations using the vanadium photoelectrochemical storage cell at 0.025 mol L^(-1) of vanadium redox species in the acid electrolytes. In addition, the stabilized photocurrent density of the copper content optimized TiO_(2)/Cu_(x)O electrodes is almost tripled from the TiO_(2) only electrode because the charge recombinations can be mitigated with the content optimized TiO_(2)/Cu_(x)O electrodes. Therefore, the optimized TiO_(2)/Cu_(x)O electrode results in the highest charge storing performance in the catholyte chamber, and the roles of vanadium redox species are also clearly demonstrated.

Direct synthesis of highly conductive poly（3,4- ethylenedioxythiophene）：poly（4-styrenesulfonate）（PEDOT：PSS）/graphene composites and their applications in energy harvesting systems

We report for the first time highly conductive poly（3,4-ethylenedioxythiophene）： poly（4-styrenesulfonate）（PEDOT：PSS）/graphene composites fabricated by in situ polymerization and their applications in a thermoelectric device and a platinum （Pt）-free dye-sensitized solar cell （DSSC） as energy harvesting systems. Graphene was dispersed in a solution of poly（4-styrenesulfonate）（PSS） and polymerization was directly carried out by addition of 3,4-ethylenedioxythiophene （EDOT） monomer to the dispersion. The content of the graphene was varied and optimized to give the highest electrical conductivity. The composite solution was ready to use without any reduction process because reduced graphene oxide was used. The fabricated film had a conductivity of 637 ***-1, corresponding to an enhancement of 41%, after the introduction of 3 wt.% graphene without any further complicated reduction processes of graphene being required. The highly conductive composite films were employed in an organic thermoelectric device, and the device showed a power factor of 45.7 μW·m^-1K^-2 which is 93% higher than a device based on pristine PEDOT：PSS. In addition, the highly conductive composite films were used in Pt-free DSSCs, showing an energy conversion efficiency of 5.4%, which is 21% higher than that of a DSSC based on PEDOT：PSS.

Slip-enhanced Reverse Electrodialytic Power Generation in Ion-selective Nanochannels

Power generation by reverse electrodialysis in ion-selective nanochannels is numerically investigated. Especially,in the present study, the influence of hydrodynamic slip at the surface of nanochannels is investigated. The current-potential characteristics of the nanochannels are calculated by solving several governing equations:Nernst-Planck equation for the ionic concentrations, the Poisson equation for the electric potential, and the Navier-Stokes equation for the diffusioosmotic flow. Hydrodynamic slip is applied as the boundary condition at the surface of nanochannels. As the slip length increases, the diffusioosmotic flow velocity and electrical conductance of ions increase because the friction at the surface of nanochannels decreases. It is shown that the power generation is enhanced by 44% with a moderate 100nm slip length by using a nanochannel with 10nm height.
