Mesenchymal stem cells(MSCs)derived from human embryonic stem cells(hESCs)have significant potential for cell-mediated bone regeneration.Our recent study revealed that inhibiting the epigenetic regulator EZH2 plays a key role in promoting the mesodermal differentiation of hESCs.In this study,an epigenome-wide analysis of hESCs and MSCs revealed that growth differentiation factor 6(GDF6),which is involved in bone formation,was the most upregulated gene associated with MSCs compared to hESCs.Furthermore,we identified GDF6 as a repressive target of EZH2 and found that ectopic GDF6 selectively promoted hESC differentiation towards the mesodermal lineage and enriched the MSC population.Our results provide molecular insights governing the mesenchymal commitment of hESCs and identify an inducing factor that offers strong promise for the future of regenerative medicine.

Phenolic compounds,as well as other aromatic compounds,have been reported to be abundant in hadal trenches.Although high-throughput sequencing studies have hinted at the potential of hadal microbes to degrade these compounds,direct microbiological,genetic and biochemical evidence under in situ pressures remain absent.Here,a microbial consortium and a pure culture of Pseudomonas,newly isolated from Mariana Trench sediments,efficiently degraded phenol under pressures up to 70 and 60 MPa,respectively,with concomitant increase in biomass.By analyzing a high-pressure(70 MPa)culture metatranscriptome,not only was the entire range of metabolic processes under high pressure generated,but also genes encod-ing complete phenol degradation via ortho-and meta-cleavage pathways were revealed.The isolate of Pseudomonas also contained genes encoding the complete degradation pathway.Six transcribed genes(dmpKLMNOP_(sed))were functionally identified to encode a multicomponent hydroxylase catalyzing the hydroxylation of phenol and its methylated derivatives by heterogeneous expression.In addition,key catabolic genes identified in the metatranscriptome of the high-pressure cultures and genomes of bacterial isolates were found to be all widely distributed in 22 published hadal microbial metagenomes.At microbiological,genetic,bioinformatics,and biochemical levels,this study found that microorganisms widely found in hadal trenches were able to effectively drive phenolic compound degradation under high hydrostatic pressures.This information will bridge a knowledge gap concerning the microbial aromatics degradation within hadal trenches.

For decades, the search for potential signs of Martian life has attracted strong international interest and has led to significant planning and scientific implementation. Clearly, in order to detect potential life signals beyond Earth, fundamental questions, such as how to define such terms as “life” and “biosignature”, have been given considerable attention. Due to the high costs of direct exploration of Mars, Mars-like regions on Earth have been invaluable targets for astrobiological research, places where scientists could practice the search for “biosignatures” and refine ways to detect them. This review summarizes scientific instrumental techniques that have resulted from this work. Instruments must necessarily be our “eyes” and “hands” as we attempt to identify and quantify biosignatures on Mars.Scientific devices that can be applied in astrobiology include mass spectrometers and electromagnetic-spectrum-based spectrometers,redox potential indicators, circular dichroism polarimeters, in situ nucleic acid sequencers, life isolation/cultivation systems, and imagers.These devices and how to interpret the data they collect have been tested in Mars-analog extreme environments on Earth to validate their practicality on Mars. To anticipate the challenges of instrumental detection of biosignatures through the full evolutionary history of Mars, Terrestrial Mars analogs are divided into four major categories according to their similarities to different Martian geological periods(the Early-Middle Noachian Period, the Late Noachian-Early Hesperian Period, the Late Hesperian-Early Amazonian Period, and the Middle-Late Amazonian Period). Future missions are suggested that would focus more intensively on Mars’ Southern Hemisphere, once landing issues there are solved by advances in spacecraft engineering, since exploration of these early terrains will permit investigations covering a wider continuum of the shifting habitability of Mars through its geological history. Study on Flow Field Simulation at Transmission Towers in Loess Hilly Regions Based on Circular Boundary Constraints

When using high-voltage transmission lines for energy transmission in loess hilly regions,local extreme wind fields such as turbulence and high-speed cyclones occur from time to time,which can cause many kinds of mechanical and electrical failures,seriously affecting the reliable and stable energy transmission of the power grid.The existing research focuses on the wind field simulation of ideal micro-terrain and actual terrain with mostly single microterrain characteristics.Model boundary constraints and the influence of constrained boundaries are the main problems that need to be solved to accurately model and simulate complex flow fields.In this paper,a flow field simulation method based on circular boundary constraints is carried out.During the study,the influence of the model boundary and the selection conditions of themodeling range are systematically analyzed.It is more suitable to make sure that the air domain is 4 times higher than the height of the hill undulations,in addition to ensuring that there should be a minimum of 400 m between the study region and the boundary.Then,an actual terrain model of a power grid line in Shanxi is established,through the method proposed in this paper,the wind speed at the location of the transmission tower line under different wind directions is analyzed,and it is found that when the incidence direction is 45 degrees north by east the wind speed is the highest.The findings demonstrate that the circular boundary model has the advantage of more easily adjusting the wind incidence direction,in addition to theoretically reducing the errors caused by traditional models in boundary processing.It can accurately obtain the distribution characteristics of the flow field affected by the terrain,and quickly screen out the extreme working conditions that are most harmful to the transmission lines in the actual transmission line for energy transmission in complex loess hilly regions.

HCV genotypes have been documented in clinical practice.The aim of this study was to determine the replication priority of different HCV genotypes in a Chinese HCV positive cohort.Serum samples from 491 apparently healthy Chinese blood donors testing positive for HCV antibodies and naive to antiviral drug therapy were tested.Genotyping analysis showed that genotypes 1b and 2a were predominant and accounted for 77.6%of the HCV infections.Among the genotype groups,individuals infected with genotype 2a had an HCV RNA viral load(108 copies/mL)about 200-fold(lg,2.3)greater than those infected with other genotypes(10^(4)–10^(5)copies/mL)indicating a replication priority of genotype 2a.However,there was no correlation between HCV genotype and antibody response suggesting that the amplification advantage of genotype 2a results from a favorable interaction with the host cellular environment.In conclusion,HCV genotypes 1b and 2a are the predominant genotypes in China and genotype 2a possesses a significant replication priority compared with the other genotypes.This suggests the existence of host cellular factors that may act as drug-targets for entirely clearing HCV infection in the future.

1949年以来中国学者在自然界分离到数百株乙型脑炎（乙脑）病毒株，并对这些病毒株进行神经毒力的研究，发现毒株问存在明显的毒力差异，特别是神经外毒力，差异的原因与自然界环境如媒介、宿主和气候等有关。乙脑的显性和隐性感染则与病毒株的毒力强弱密切相关。对乙脑病毒的变异进行研究，发现病毒可以通过小鼠脑内或不同的细胞传代而毒力逐步减弱，特别是神经外毒力降低更快，并且可以通过空斑把强弱病毒分离从而挑选到低毒力的弱毒株。通过不同减毒试验，获得多株不同毒力的弱毒株用于人体、猪和马的免疫。特别是其中发现病毒通过实验动物体内的非神经组织传代，能够进一步降低病毒的残余毒力并提高其免疫性，应用该创新性技术成功筛选到1株高度减毒而稳定并保持高免疫性的SA14-14-2株，用该株病毒生产的活疫苗，自1989年取得生产许可证以来已在国内外广泛应用，证实其安全高效达到国际领先水平，并于2013年取得WHO的预认证。

This paper proposes a homogeneous vehicle platoon longitudinal controller considering the effect of electronic throttle opening angle.In particular,a nonlinear longitudinal controller is proposed by integrating the consensus between connected autonomous vehicles（CAVs）,the influence of interactions between vehicles and the effect of electronic throttle（ET） opening angle.Then,the convergence condition is deduced analytically by using the Routh–Hurwitz stability criterion.The controller can not only ensure the consensus of CAVs,but also avoid rear-end collision and negative velocity.Finally,simulations are carried out in the presence of external interference to verify the robustness and effectiveness of the proposed controller with respect to position,velocity and acceleration/deceleration profiles.

Rapid and accurate detection of immunoglobulin E(Ig E) in serum and reduction of serum dosage are of great significance for clinical detection. Herein, we described a rapid magnetic separation of Ig E from patient serum based on Fe3 O4@Si O2-NTA@026 sdab as the capture probe and multiple horseradish peroxidase(HRP)-labeled antibodies linked gold nanoparticles(Au NPs) as chemiluminescence(CL) signal amplifier for ultrasensitive detection of total Ig E. Results showed that the limit of detection of our immunosensor system in serum samples was 0.03 k U/L, which is lowest in comparison with current methods, and far lower than that of Immuno CAP for Ig E detection(0.1 k U/L). Furthermore, our immunosensor possessed satisfied repeatability and accuracy, as well as good stability. In comparison with the Immuno CAP for the quantitative detection of Ig E, highly consistent results were achieved in 20 serum samples. Specially, this method was also successfully utilized for assessing the Ig E traces in breast cancer patients,which provides a new idea for the diagnosis of early cancer. Therefore, we believe that such versatile immunosensor will offer an alternative method for the on-site monitoring and determination of various Ig E-related diseases.
