Osteoarthritis(OA)is characterized by cartilage degradation,pain,and synovitis.1 Joint inflammation driven by cytokines has been demonstrated to cause cartilage degradation and pain.2 However,approaches to neutralize cytokines,such as IL-1 and TNF-α,known to be involved in OA have shown poor clinical efficacy.3 There is an unmet clinical need to find better anti-inflammatory and pain targets for OA therapy and to elucidate the role of other cytokines in OA *** studies have shown that IL-22 and its receptor IL-22R play central roles in inflammation and diseases such as psoriasis,ulcerative colitis,graft-versus-host disease,certain infections and tumors,as well as in liver and pancreas damage.4,5 The role of IL-22/IL-22R and the potential for therapeutic targeting of both proteins in OA remain largely unknown,which we sought to investigate.

Dear Editor,Influenza affects humans of all ages;however,elderly people have increased susceptibility to infections and are especially predisposed to complications.1 A number of global efforts have increased influenza vaccination administration in elderly people to reduce complications,particularly when seasonal influenza infections peak.2 To date,much is known about how influenza infection and host immunity interact during pathogenesis,which has led to a number of vaccines.3,4 However,the impact of influenza vaccines in older people is modest.2 Hence,the underlying pathophysiological mechanisms responsible for the worsening of influenza infection in old age are still elusive.

The main methods of coalbed methane(CBM)development are drainage and depressurization,and a precise prediction of coal reservoir pressure is thus crucial for the evaluation of reservoir potentials and the formulation of reasonable development *** work established a new reservoir pressure prediction model based on the material balance equation(MBE)of coal reservoir,which considers the self-regulating effects of coal reservoirs and the dynamic change of equivalent drainage area(EDA).According to the proposed model,the reservoir pressure can be predicted based on reservoir condition data and the actual production data of a single *** with traditional reservoir pressure prediction models which regard EDA as a fixed value,the proposed model can better predict the average pressure of ***,orthogonal experiments were designed to evaluate the sensitivity of reservoir parameters on the reservoir pressure prediction results of this proposed *** results show that the saturation of irreducible water is the most sensitive parameter,followed by Langmuir volume and reservoir porosity,and Langmuir pressure is the least sensitive *** addition,the pressure drop of reservoirs is negatively correlated with the saturation of irreducible water and the Langmuir volume,while it is positively correlated with *** work analyzed the reservoir pressure drop characteristics of the CBM wells in the Shizhuangnan Block of the Qinshui Basin,and the results show that the CBM reservoir depressurization can be divided into three types,i.e.,rapidly drop type,medium-term stability type,and slowly drop *** drainage features of wells were reasonably interpreted based on the comprehensive analysis of the reservoir depressurization type;the latter was coupled to the corresponding permeability dynamic change characteristics,eventually proving the applicability of the proposed model.

The ongoing coronavirus disease 2019(COVID-19)pandemic is a global public health crisis,causing social and economic disasters in many *** China,two-consecutive negative results of nucleic acid tests for SARS-CoV-2 from the respiratory samples are required to end the quarantine of COVID-19 ***,clinicians face a dilemma in case of patients with long-term viral *** report described an unusual COVID-19 case who had persistent viral RNA positivity for more than 4 months after initial illness in the presence of low neutralizing antibodies,but without prolonged clinical *** anti-viral drug treatments had no impact and there was no evidence of *** the patient was self-quarantined at home,no infection occurred to the three family members living with her for 15 to 19 *** viral culture in BSL-3 laboratory on the 102nd day after symptom onset was *** the 129th day on,8 continuous nucleic acid tests of sputum samples showed negative *** patient was discharged on 137th days since symptom *** conclusion,viral RNA shedding in the sputum of the COVID-19 patient may last over 4 *** no evidence shows the existence of infectious virus,two-consecutive negative nucleic acid tests may not be the prerequisite for ending quarantine of COVID-19 patients with prolonged viral shedding.

Immune responses of dendritic cells combined with tumor-derived autophagosome vaccine on hepatocellular carcinoma

Background： To induce and collect tumor-derived autophagosomes （DRibbles） from tumor cells as an antitumor vaccine by inhibiting the functions of proteasomes and lysosomes. Methods： Dendritic cells （DCs） generated from peripheral blood mononuclear cell （PBMC） of hepatocellular carcinoma （HCC） patients were cocultured with DRibbles, and then surface molecules of DCs, as well as surface molecules on DCs, were determined by flow cytometry. Meanwhile, immune responses of the DCs-DRibbles were examined by mixed lymphocyte reactions. Results： DRibbles significantly induced the expression of CD80, CD83, CD86 and HLA-DR on DCs. The enzyme-linked immunosorbnent assay （ELISA） showed that IFN-γ, levels after vaccination increased than before in most patients, but CDS＋ proportion of PBMC increased only in nine patients. Higher levels of IFN-γ, were detected in the CD8＋ cells than CD4＋ T cells. These results suggested that DCs-DRibbles vaccine could induce antigen-specific cellular immune response on HCC and could prime strong CD8＋ T cell responses, supporting it as a tumor vaccine candidate. Conclusions： Our results demonstrate that HCC/DRibbles-pulsed DCs immunotherapy might be deployed as an effective antitumor vaccine for HCC immunotherapy in clinical trials.
