The roles of RACK1 in the pathogenesis of Alzheimer's disease

The receptor for activated C kinase 1(RACK1)is a protein that plays a crucial role in various signaling pathways and is involved in the pathogenesis of Alzheimer's disease(AD),a prevalent neurodegenerative disease.RACK1 is highly expressed in neuronal cells of the central nervous system and regulates the pathogenesis of AD.Specifically,RACK1 is involved in regulation of the amyloid-β precursor protein processing through α-or β-secretase by binding to different protein kinase C isoforms.Additionally,RACK1 promotes synaptogenesis and synaptic plasticity by inhibiting N-methyl-D-aspartate receptors and activating gamma-aminobutyric acid A receptors,thereby preventing neuronal excitotoxicity.RACK1 also assembles inflammasomes that are involved in various neuroinflammatory pathways,such as nuclear factor-kappa B,tumor necrosis factor-alpha,and NOD-like receptor family pyrin domain-containing 3 pathways.The potential to design therapeutics that block amyloid-β accumulation and inflammation or precisely regulate synaptic plasticity represents an attractive therapeutic strategy,in which RACK1 is a potential target.In this review,we summarize the contribution of RACK1 to the pathogenesis of AD and its potential as a therapeutic target.

We report the magnetostrictive performance of the Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites.Measurement of M?ssbauer spectra and the XRD results indicate that the easy magnetic direction of the Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)compound is alongdirection with spontaneous magnetostrictionλ111 of1900 ppm.The-oriented Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites with particle volume fractions of20 vol%-55 vol%were prepared.Magnetization measurements demonstrate that the composite is highly anisotropic.The raw material cost of Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)is about 85%of that of the Terfenol-D(Tb_(0.3)Dy_(0.7)Fe_(2))composition.Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composites possess comparable magnetostriction with those composites made of Tb_(0.3)Dy_(0.7)Fe_(2)particles.At room temperature,the magnetostrictionλ_Ⅱof the Nd_(0.2)Tb_(0.3)Dy_(0.5)Fe_(1.93)/epoxy composite reaches 420 ppm at a low field of 80 kA/m and about1000 ppm at 239 kA/m.In addition,the composite exhibits a large saturation magnetostrictionλ_Ⅱover1200 ppm from 300 to 100 K,suggesting its potential applications as a low-cost magnetostrictive material in a wide temperature range.Therefore,the current study offers an opportunity to widen the operating temperature range of the magnetostrictive composite.

Cellular structures are commonly used to design energy-absorbing structures,and origami structures are becominga prevalent method of cellular structure design.This paper proposes a foldable cellular structure based on theWaterbomb origami pattern.The geometrical configuration of this structure is described.Quasi-static compressiontests of the origami tube cell of this cellular structure are conducted,and load-displacement relationship curvesare obtained.Numerical simulations are carried out to analyze the effects of aspect ratio,folding angle,thicknessand number of layers of origami tubes on initial peak force and specific energy absorption(SEA).Calculationformulas for initial peak force and SEA are obtained by the multiple linear regression method.The degree ofinfluence of each parameter on the mechanical properties of the single-layer tube cell is compared.The resultsshow that the cellular structure exhibits negative stiffness and periodic load-bearing capacity,as well as foldingangle has the most significant effect on the load-bearing and energy-absorbing capacity.By adjusting the designparameters,the stiffness,load-bearing capacity and energy absorption capacity of this cellular structure can beadjusted,which shows the programmable mechanical properties of this cellular structure.The foldability andthe smooth periodic load-bearing capacity give the structure potential for application as an energy-absorbingstructure.

Antarctic krill （Euphausia superba Dana） was collected using a High Speed Collector and an Isaac-Kidd midwater trawl （IKMT） net during the austral summer of 2007/2008 and 2008/2009 in the circumpolar and Prydz Bay regions of the Southern Ocean, respectively. Combined with the simultaneous recording of environmental factors, spatial distribution, population structure and growth condition of E. superba were studied. The abundance of E. superba in the Weddell Sea was higher than in Prydz Bay. However, the abundance of E. superba in both the Weddell Sea and Prydz Bay was lower than figures reported in previous krill surveys for the same time period. With respect to the total study area, E. superba displayed a normal growing state during the two expeditions. E. superba grew relatively poorly in some stations, which may be due to the late retreat of sea ice or lower chlorophyll a concentrations. The number of juvenile E. superba collected using the High Speed Collector was proportionally greater in stations located at the edge of the sea ice, while adults dominated in long-term non-ice regions. This phenomenon reflects the different distribution pattern between juvenile and adult krill. The population structure of E. superba differed between sea regions, which may affect recruitment.

Protein post-translational modifications(PTMs)are at the heart status of cellular signaling events and broadly involved in tumor progression.CD147 is a tumor biomarker with various PTMs,promoting tumor metastasis and metabolism reprogramming.Nevertheless,the relationship between the PTMs of CD147 and apoptosis has not been reported.In our study,we produced a specific anti-CD147-K71 di-methylation(CD147-K71me2)antibody by immunizing with a di-methylated peptide and observed that the level of CD147-K71me2 in non-small cell lung cancer(NSCLC)tissues were lower than that in NSCLC adjacent tissues.SETDB1 was identified as the methyltransferase catalyzing CD147 to generate CD147-K71me2.RNA-seq showed that FOSB was the most significant differentially expressed gene(DEG)between wild-type CD147(CD147-WT)and K71-mutant CD147(CD147-K71R)groups.Subsequently,we found that CD147-K71me2 promoted the expression of FOSB by enhancing the phosphorylation of p38,leading to tumor cell apoptosis.In vivo experiments showed that CD147-K71me2 significantly inhibited tumor progression by promoting cell apoptosis.Taken together,our findings indicate the inhibitory role of CD147-K71me2 in tumor progression from the perspective of post-translational modification,which is distinct from the pro-cancer function of CD147 itself,broadening our perspective on tumor-associated antigen CD147.

Comprehensive analysis of clinical and biological value of ING family genes in liver cancer

BACKGROUND Liver cancer(LIHC)is a malignant tumor that occurs in the liver and has a high mortality in cancer.The ING family genes were identified as tumor suppressor genes.Dysregulated expression of these genes can lead to cell cycle arrest,senescence and/or apoptosis.ING family genes are promising targets for anticancer therapy.However,their role in LIHC is still not well understood.AIM To have a better understanding of the important roles of ING family members in LIHC.METHODS A series of bioinformatics approaches(including gene expression analysis,genetic alteration analysis,survival analysis,immune infiltration analysis,prediction of upstream microRNAs(miRNAs)and long noncoding RNAs(lncRNAs)of ING1,and ING1-related gene functional enrichment analysis)was applied to study the expression profile,clinical relationship,prognostic significance and immune infiltration of ING in LIHC.The relationship between ING family genes expression and tumor associated immune checkpoints was investigated in LIHC.The molecular mechanism of ING1 mediated hepatocarcinogenesis was preliminarily discussed.RESULTS mRNA/protein expression of different ING family genes in LIHC was analyzed in different databases,showing that ING family genes were highly expressed in LIHC.In 47 samples from 366 LIHC patients,the ING family genes were altered at a rate of 13%.By comprehensively analyzing the expression,clinical pathological parameters and prognostic value of ING family genes,ING1/5 was identified.ING1/5 was related to poor prognosis of LIHC,suggesting that they may play key roles in LIHC tumorigenesis and progression.One of the target miRNAs of ING1 was identified as hsa-miR-214-3p.Two upstream lncRNAs of hsa-miR-214-3p,U91328.1,and HCG17,were identified.At the same time,we found that the expression of ING family genes was correlated with immune cell infiltration and immune checkpoint genes.CONCLUSION This study lays a foundation for further research on the potential mechanism and clinical value of ING family genes in the treatment and pr

Microstructural characterization and mechanical properties of(TiC+TiB)/TA15 composites prepared by an in-situ synthesis method

Titanium matrix composites reinforced with ceramic particles are considered a promising engineering material due to their combination of high specific strength,low density,and high modulus.In this study,the TA15-based composites reinforced with a volume fraction of 10% to 25%(TiB+TiC)were prepared using powder metallurgy and casting technique.Microstructural characterization and phase constitution were examined using optical microscopy(OM),scanning electron microscopy(SEM),and X-ray diffraction(XRD).In addition,the microhardness,room temperature(RT)and high temperature(HT)tensile properties of the composites were evaluated.Results revealed that the reinforcements are distributed uniformly even in the composites with a high volume of TiB and TiC.However,as the volume fraction exceeds 15%,TiB and TiC particles become coarsening and exhibit rod-like and dendritic-like morphology.Microhardness increases gradually from 321.2 HV for the base alloy to a maximum of 473.3 HV as the reinforcement increases to 25vol.%.Tensile test results indicate that a reinforcement volume fraction above 20% is beneficial for enhancing tensile strength and yield strength at high temperatures,but it has an adverse effect on room temperature elongation.Conversely,if the reinforcement volume fraction is below 20%,it can improve high-temperature elongation when the temperature exceeds 600℃.

Carbonaceous materials in seismic fault zones may considerably influence seismic fault slip;however,the formation mechanism of carbonaceous materials remains unclear.In this study,we proposed a novel hypothesis for the formation of carbonaceous materials in fault gouge.Thus,we conducted a CO_(2) hydrogenation experiment in a high-temperature reactor at a co-seismic temperature,with fault gouge formed during the Wenchuan earthquake as the catalyst.Our experimental results demonstrate that carbonaceous materials in fault zones are formed on the fault gouge during the chemical reaction process,suggesting that the carbonaceous materials are possibly generated from the catalytic hydrogenation of CO_(2),followed by thermal cracking of its products.The results of this study provide a theoretical basis for understanding fault behavior and earthquake physics.

BACKGROUND Glucose and lipid metabolic disorder in patients with type 2 diabetes mellitus(T2DM)is associated with the levels of serum tumor markers of the digestive tract,such as cancer antigen(CA)199.Therefore,tumor markers in T2DM are important.AIM To evaluate the expression of serum tumor markers[CA199,CA242,and carcinoembryonic antigen(CEA)]and the clinical implications of the expression in T2DM.METHODS For this observational study conducted at Hefei BOE Hospital,China,we enrolled 82 patients with first-onset T2DM and 51 controls between April 2019 and December 2020.Levels of fasting blood glucose(FBG),tumor markers(CA199,CEA,and CA242),glycosylated hemoglobin(HbA1c),etc.were measured and group index levels were compared.Moreover,FBG and HbA1c levels were correlated with tumor marker levels.Tumor markers were tested for diagnostic accuracy in patients with>9%HbA1c using the receiver operating curve(ROC)curve.RESULTS The T2DM group had high serum FBG,HbA1c,CA199,and CEA levels(P9%)revealed significant differences in CEA and CA199 levels(P<0.05).The areas under the ROC curve for CEA and CA199 were 0.853 and 0.809,respectively.CA199,CEA,and CA242 levels positively correlated with HbA1c(r=0.308,0.426,and 0.551,respectively)and FBG levels(r=0.236,0.231,and 0.298,respectively).CONCLUSION As compared to controls,serum CEA and CA199 levels were higher in patients with T2DM.HbA1c and FBG levels correlated with CA199,CEA,and CA242 levels.Patients with poorly controlled blood sugar must be screened for tumor markers.
