Enhanced therapeutic effects for human pancreatic cancer by application K-ras and IGF-IR antisense oligodeoxynucleotides

AIM: To investigate the combined effects of K-ras antisense oligodeoxynucleotide (K-ras ASODN) specif ic to GTT point mutation at codon 12 and type Ⅰ insulin-like growth factor receptor (IGF-IR) antisense oligodeoxynucleotide (IGF-IR ASODN) on proliferation and apoptosis of human pancreatic cancer Patu8988 cells in vitro and in vivo. METHODS: K-ras gene point mutation and its style at codon 12 of human pancreatic cancer cell line Patu8988 were detected by using polymerase chain reaction with special sequence primers (PCR-SSP) and sequence analysis. According to the mutation style, K-ras mutation ASODN specifi c to K-ras point mutation at codon 12 was designed and composed. After K-ras ASODN and IGF-IR ASODN treated on Patu8988 cells respectively or cooperatively, the proliferation and morphological change of Patu8988 cells were analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, colony forming assay andtransmission electron microscopy; the expression of K-ras and IGF-IR mRNA and protein in the treated cells was measured by reverse-transcript polymerase chain reaction (RT-PCR) and flow cytometry respectively; apoptosis was determined by flow cytometry. The combined antitumor activity of K-ras ASODN and IGF-IR ASODN was evaluated in BALB/c nude mice bearing human pancreatic cancer inoculated with Patu8988 cells. RESULTS: The results of PCR-SSP and sequence analysis showed that the human pancreatic cancer cell line Patu8988 had point mutation at codon 12, and the mutation style was GGT→GTT. 2-32 μg/mL K-ras ASODN and 2-32 μg/mL IGF-IR ASODN could inhibit Patu8988 cells' growth, induce apoptosis and decrease the expression of K-ras and IGF-IR mRNA and protein alone. However, there was much more effective inhibition of growth and induction of apoptosis by their combination than by each one alone. Dear Editor,This letter presents an improved repetitive controller(IRC) that uses a complex-coefficient filter to enhance the tracking performance of a system for periodic signals. Compared with the low-pass filter used in the conventional repetitive controller(CRC), the complex-coefficient filter causes less change in the phase and amplitude of a signal at the frequencies of the periodic signal, especially at the fundamental frequency, when the two filters have the same cutofffrequency.

Flotation separation of chalcopyrite from pyrite using mineral fulvic acid as selective depressant under weakly alkaline conditions

Mineral fulvic acid(MFA)was used as an eco-friendly pyrite depressant to recover chalcopyrite by flotation with the use of the butyl xanthate as a *** experiments showed that MFA produced a stronger inhibition effect on pyrite than on *** separation of chalcopyrite from pyrite was realized by introducing 150 mg/L MFA at a pulp pH of approximately *** copper grade,copper recovery,and separation efficiency were 28.03%,84.79%,and 71.66%,*** adsorption tests,zeta potential determinations,and localized electrochemical impedance spectroscopy tests showed that more MFA adsorbed on pyrite than on chalcopyrite,which weakened the subsequent interactions between pyrite and the *** force microscope imaging further confirmed the adsorption of MFA on pyrite,and X-ray photoelectron spectroscopy results indicated that hydrophilic Fe-based species on the pyrite surfaces increased after exposure of pyrite to MFA,thereby decreasing the floatability of pyrite.

Pyridyl-based ketones and 1,6-diketones are both attractive and invaluable scaffolds which play pivotal roles in the construction and structural modification of a plethora of synthetically paramount natural products, pharmaceuticals, organic materials and fine chemicals. In this context, we herein demonstrate an unprecedented, robust and generally applicable synthetically strategy to deliver these two crucial ketone frameworks via visible-light-induced ring-opening coupling reactions of cycloalcohols with vinylazaarenes and enones, respectively. A plausible mechanism involves the selective β-C-C bond cleavage of cycloalcohols enabled by proton-coupled electron transfer and ensuing Giese-type addition followed by single electron reduction and protonation. The synthetic methodology exhibits broad substrate scope,excellent functional group compatibility as well as operational simplicity and environmental friendliness.

Environmental Inequality in China’s Urban Expansion:A Case Study of Guangzhou,China

Environmental inequality is a prevalent issue in developing countries undergoing urban *** expansion induces the formation and evolution of environmental inequality by creating environmental and structural conditions that lead to the spatial relocation of environmental hazards and the socio-spatial segregation of different groups in developing *** study investigated the spatial patterns and temporal trends of environmental inequality under urban expansion in Guangzhou,a megacity in *** considered how environmental disparities and socio-demographic attributes interact in terms of industrial pollution exposure using additive semiparametric quantile regression,combined with spatial visualisation,on the basis of the economic and population census data from 1990 to *** study revealed that urban expansion sparked the spatial displacement of environmental risks and the social-spatial differentiation,exposing the peripheral regions and disadvantaged groups to higher environmental risks.A reciprocal transformation occurred between central and peripheral regions,as well as a process of redistributing environmental risks across social *** the context of urban expansion in developing countries,the causes of environmental inequality shifted from individual socio-economic differences to structural factors,such as industrial layout and social division of labour in cities,leading to the spatial displacement and concealment of environmental *** study provides insights and guidance for policymakers to address the issue of environmental inequality in the context of urban expansion.

Evaluating the accuracy of earth rotation parameters based on the BDS observations

The Bei Dou satellite system(BDS) has progressed with the full operationalization of the secondgeneration regional system(BDS-2) and the third-generation global system(BDS-3). This technology plays a crucial role in determining Earth Rotation Parameters(ERPs). In this study, we determine the ERPs based on the observations of BDS-2, BDS-3 and BDS-2+BDS-3, with the time spanning from August18, 2022, to August 18, 2023. The IERS EOP 20C04 series is used as a reference to evaluate the accuracy of the ERP estimates. We analyze the impact of different numbers of reference stations, polyhedron volumes, observation arc lengths, satellite types, and satellite systems on solving ERPs using BDS-2 and BDS-3 observation data provided by the International GNSS Service(IGS) stations. When selecting a specific satellite type, it is necessary to choose an appropriate observation arc length based on different numbers of reference stations while maximizing the volume of the formed polyhedron to achieve optimal efficiency and accuracy in parameter estimation. When both the number of reference stations and observation arc length are fixed, higher precision of the ERPs can be achieved using observations from MEO than MEO + IGSO and MEO + IGSO + GEO. Moreover, when considering only IGSO and MEO satellites as options for analysis purposes, BDS-3 provides higher accuracy compared to BDS-2. In summary, when using BDS for ERP estimation and MEO satellite observations with the same observation arc length,selecting stations from reference stations with larger polyhedral volumes can significantly improve the efficiency and accuracy of parameter estimation.

Axial emission characteristics of an ionic liquid electrospray thruster with a circular emitter

Ionic Liquid Electrospray Thrusters(ILETs)are well suited for micro-nano satellite applications due to their small size,low power consumption,and high specific ***,the limited thrust of a single-emitter ILET restricts its use in space *** optimize the performance of ILETs and make them suitable for a wider range of space missions,we designed a Circular-emitter ILET(CILET)to convert a one-dimensional(point)emission into a twodimensional(line)*** CILET can self-organize multiple Taylor cones *** cones were photographed and the axial emission currents were measured under different voltage and pressure difference conditions with a CILET experimental *** emission can be divided into two stable states and one unstable state based on the flow and current *** current in Stable state Ⅰ increases non-linearly with the voltage,while that in Stable state Ⅱ is nearly linear with respect to the *** number of cones increases with the voltage in stable states,while the cones become short and crowded under high-voltage *** variation law of the number of cones can be explained with the self-organization *** variation in the current exhibits a good correlation with the number of *** study demonstrates the feasibility of circular emitters and experimentally indicates that the emission current is improved by approximately two orders of magnitude compared to that of a single capillary.
