Dear editor,Henoch-Schönlein purpura (HSP) is the commonestvasculitis in children, typically affecting children agedthree to ten years.[1] It is a multi-systemic vasculitismediated by type III hypersensitivity with deposition ofimmunoglobulin. An immune complex-mediated vasculitisaffects small vessels of the skin, joints, kidneys, andgastrointestinal (GI) tracts. HSP is usually a self-limitingcondition and resolves within six to eight weeks.[1]

Focus on the gut-brain axis: multiple sclerosis, the intestinal barrier and the microbiome

The brain-gut axis serves as the bidirectional connection between the gut microbiome, the intestinal barrier and the immune system that might be relevant for the pathophysiology of inflammatory demyelinating diseases. People with multiple sclerosis have been shown to have an altered microbiome, increased intestinal permeability and changes in bile acid metabolism. Experimental evidence suggests that these changes can lead to profound alterations of peripheral and central nervous system immune regulation. Besides being of pathophysiological interest, the brain-gut axis could also open new avenues of therapeutic targets. Modification of the microbiome, the use of probiotics, fecal microbiota transplantation, supplementation with bile acids and intestinal barrier enhancers are all promising candidates. Hopefully, pre-clinical studies and clinical trials will soon yield significant results.

The benefits of neuroinflammation for the repair of the injured central nervous system

Inflammation of the nervous system(neuroinflammation)is now recognized as a hallmark of virtually all neurological *** neuroinflammatory conditions such as multiple sclerosis,there is prominent infiltration and a long-lasting representation of various leukocyte subsets in the central nervous system(CNS)*** in classic neurodegenerative disorders,where such immense inflammatory infiltrates are absent,there is still evidence of activated CNS-intrinsic *** consequences of excessive and uncontrolled neuroinflammation are injury and death to neural elements,which manifest as a heterogeneous set of neurological ***,it is now readily acknowledged,due to instructive studies from the peripheral nervous system and a large body of CNS literature,that aspects of the neuroinflammatory response can be beneficial for CNS *** recognized benefits of inflammation to the CNS include the preservation of CNS constituents(neuroprotection),the proliferation and maturation of various neural precursor populations,axonal regeneration,and the reformation of myelin on denuded ***,we highlight the benefits of neuroinflammation in fostering CNS recovery after neural injury using examples from multiple sclerosis,traumatic spinal cord injury,stroke,and Alzheimer’s *** focus on CNS regenerative responses,such as neurogenesis,axonal regeneration,and remyelination,and discuss the mechanisms by which neuroinflammation is pro-regenerative for the ***,we highlight treatment strategies that harness the benefits of neuroinflammation for CNS regenerative responses.

Temperature-induced Evolution of Micro-structure and Chain Dynamics in Thermoplastic Polyurethane with Low Hard Segment Content as Studied by In Situ Synchrotron SAXS and Time-Domain NMR Experiments

The microstructural evolution of a thermoplastic polyurethane(TPU)with low hard segment content has been monitored utilizing in situ real-time synchrotron small angle X-ray scattering(SAXS)and time-domain nuclear magnetic resonance(NMR)*** TPU is composed of 23 wt% of[4,4-methylenediphenyl diisocyanate(MDI)]-[1,4-butanediol(BD)]chain segments,which form hard domains,as[polytetrahydrofuran(PTHF)]forming soft *** number and distribution of monomer units in hard blocks is determined by the successive self-nucleation and annealing thermal fractionation *** situ SAXS method reveals heating-induced increase in the spacing of hard and soft domains,while time-domain ^(1)H-NMR characterizes the changes in the phase composition and chain dynamics in these domains.A glassy fraction of short MDI-BD chain segments in hard domains passes through T_(g) above ambient *** higher temperatures,MDI-BD nanocrystals start to *** length distribution of MDI-BD chain segments causes a distribution in crystal sizes and wide melting temperature *** melting is accompanied by the mixing of MDI-BD with PTHF segments in soft domains,and by increase in segmental mobility in these *** 180℃,the TPU melt is homogeneous on the scale above nanometers according to SAXS data.

Self-assembly of nitrogen-doped TiO2 with exposed {001} facets on a graphene scaffold as photo-active hybrid nanostructures for reduction of carbon dioxide to methane

Tailored synthesis of well-defined anatase TiO2-based crystals with exposed {001} facets has stimulated incessant research interest worldwide due to their scientific and technological importance. Herein, anatase nitrogen-doped TiO2 （N-TiO2） nanoparticles with exposed {001} facets deposited on the graphene （GR） sheets （N-TiO2-001/GR） were synthesized for the first time via a one-step solvothermal synthetic route using NH4F as the morphology-controlling agent. The experimental results exemplified that GR was uniformly covered with anatase N-TiO2 nanoparticles （10-17 nm）, exposing the {001} facets. The percentage of exposed {001} facets in the N-TiO2-001/GR nanocomposites was calculated to be ca. 35%. Also, a red shift in the absorption edge and a strong absorption in the visible light range were observed due to the formation of Ti-O-C bonds, resulting in the successful narrowing of the band gap from 3.23 to 2.9 eV. The photocatalytic activities of the as-prepared photocatalysts were evaluated for CO2 reduction to produce CH, in the presence of water vapor under ambient temperature and atmospheric pressure using a low-power 15 W energy-saving daylight lamp as the visible light source--in contrast to the most commonly employed high-power xenon lamps--which rendered the process economically and practically feasible. Among all the studied photocatalysts, the N-TiO2-001/GR nanocomposites exhibited the greatest CH4 yield of 3.70 p-mol＇gcatalyst 1, approxi- mately 11-fold higher activity than the TiO2-001. The enhancement of photocatalyfic performance was ascribed to the effective charge anti-recombination of graphene, high absorption of visible light region relative to the {101} facets. and high catalytic activity of {001} facets

目的探讨金雀异黄素(Genistein)在去卵巢(OVX)小鼠非酒精性脂肪性肝病(NAFLD)中的保护作用及机制。方法取40只6周龄雌性C57BL/6小鼠建立OVX小鼠模型,随机分为5组,每组8只,分别为对照组、模型4周组、模型6周组、模型8周组、模型10周组。相同环境下,采用高脂饮食对5组OVX小鼠进行饮食造模,病理学检查显示,经10周高脂饮食诱导NAFLD成功。另取40只6周龄雌性C57BL/6小鼠随机分为5组:空白组、假手术(Sham)组、OVX组、OVX+L-Genistein(4 mg/kg体质量)组、OVX+H-Genistein(8 mg/kg体质量)组。Sham组进行相同的OVX手术过程,但不结扎卵巢动脉和切除卵巢。空白组小鼠正常饮食喂养,其余各组供给高脂饮食。将Genistein溶解在DMSO中,Sham组和OVX组的动物仅用溶媒溶液处理,所有小鼠每天灌胃给药1次,持续10周。记录小鼠体质量及脏器指数,处死小鼠并收集血清及肝组织。试剂盒检测血清ALT、AST活性及TG、TC水平。HE和油红O染色观察肝组织病理。通过Western Blot分析肝组织中脂质代谢相关的固醇调节元件结合蛋白1(SREBP-1c)、过氧化物酶体增殖物激活受体α(PPARα)的蛋白表达。计量资料多组间比较采用单因素方差分析,进一步两组间比较采用Dunnett-t检验。结果高脂饮食10周后,Genistein低、高剂量组的体质量、肝指数和肝质量均低于OVX组(P值均<0.05)。此外,Genistein显著下调了血清TC、TG水平(P值均<0.05),降低了血清中AST和ALT活性(P值均<0.05)。HE和油红O染色结果表明,Genistein低、高剂量组较OVX组脂滴累积明显减少。Western Blot分析结果显示,给予Genistein干预后,SREBP-1c的蛋白表达量明显下降,PPARα的蛋白表达量明显升高(P值均<0.05)。结论Genistein对OVX小鼠NAFLD具有保护作用,其部分作用机制可能与调节SREBP-1c、PPARα的表达有关,从而促进脂肪酸氧化和抑制肝脏脂质合成。
