Inhibitory Effects of Simvastatin on Oxidized Low-Density Lipoprotein-lnduced Endoplasmic Reticulum Stress and Apoptosis in Vascular Endothelial Cells

Background：Oxidized low-density lipoprotein （ox-LDL）-induced oxidative stress and endothelial apoptosis are essential for atherosclerosis. Our previous study has shown that ox-LDL-induced apoptosis is mediated by the protein kinase RNA-like endoplasmic reticulum kinase （PERK）/eukaryotic translation initiation factor 2α-subunit （eIF2α）/CCAAT/enhancer-binding protein homologous protein （CHOP） endoplasmic reticulum （ER） stress pathway in endothelial cells. Statins are cholesterol-lowering drugs that exert pleiotropic effects including suppression of oxidative stress. This study aimed to explore the roles of simvastatin on ox-LDL-induced ER stress and apoptosis in endothelial cells.Methods：Human umbilical vein endothelial cells （HUVECs） were treated with simvastatin （0.1, 0.5, or 2.5 μmol/L） or DEVD-CHO （selective inhibitor of caspase-3, 100 μmol/L） for 1 h before the addition of ox-LDL （100 μg/ml） and then incubated for 24 h, and untreated cells were used as a control group. Apoptosis, expression of PERK, phosphorylation of eIF2α, CHOP mRNA level, and caspase-3 activity were measured. Comparisons among multiple groups were performed with one-way analysis of variance （ANOVA） followed by post hoc pairwise comparisons using Tukey’s tests. A value of P 〈 0.05 was considered statistically significant.Results：Exposure of HUVECs to ox-LDL resulted in a significant increase in apoptosis （31.9% vs. 4.9%, P 〈 0.05）. Simvastatin （0.1, 0.5, and 2.5 μmol/L） led to a suppression of ox-LDL-induced apoptosis （28.0%, 24.7%, and 13.8%, F = 15.039, all P 〈 0.05, compared with control group）. Ox-LDL significantly increased the expression of PERK （499.5%, P 〈 0.05） and phosphorylation of eIF2α （451.6%, P 〈 0.05）, if both of which in the control groups were considered as 100%. Simvastatin treatment （0.1, 0.5, and 2.5 μmol/L） blunted ox-LDL-induced expression of PERK （407.8%, 339.1%, and 187.5%, F = 10.121, all P 〈 0.05, compared with control group） and phosphorylation of eIF2α （407.8%, 339.1%, 187.5%, F = 11.430, all P 〈 0.05, compared w

Hypertrophic cardiomyopathy(HCM)is genetic cardiomyopathy with the risk of sudden death characterized by abnormal sarcomere protein.^([1])Although some pathogenic genes related to HCM have been reported,^([2])the disease phenotype and pathogenesis cannot be fully elucidated,which brings great difficulties to the diagnosis and treatment of HCM,especially the exploration of HCM treatment strategies.

Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis

Objective PERK/elF2/CHOP is a major signaling pathway mediating endoplasmic reticulum （ER） stress related with atherosclerosis. Oxidized LDL （ox-LDL） also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis. The present study was conducted to explore the regulatory effect of ox-LDL on PERK/elF2a/CHOP signaling pathway in vascular endothelial cells. Methods The effects of ox-LDL on PERK and p-elF2a protein expression of primary human umbilical vein endothelial cells （HUVECs） were investigated by Western blot analysis. PERK gene silencing and selective elF2a phosphatase inhibitor, salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis, caspase-3 activity, and CHOP mRNA level. Results Ox-LDL treatment significantly increased the expression of PERK, PERK-mediated inactivation of elF2a phosphorylation, and the expression of CHOP, as well as the caspase-3 activity and apoptosis. The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral shRNA or by selective elF2a phosphatase inhibitor, salubrinal. Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/elF2a/CHOP ER-stress pathway. It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis.

Miniature Boat Fabrication with Striking Loading Capacity in Seawater from Hydrophobic Steel Mesh

A superhydrophobic steel mesh film was prepared by combination of macro-scale rough surface and low surface energy material treatment through a facile coating method. The contact angle for seawater is measured to be as high as 130.16°. A reformed Cassie-Baxter equation was applied for the theoretical predictions of this novel material for the first time. Good agreement between the predictions and experiments was obtained. The loading capacities of these boats, fabricated from the resulting hydrophobic steel meshes, were also characterized. The highest loading weight about 17.50 g was obtained by the steel mesh treated by 2.0wt% (heptadecafluoro-1,1,2,2,-tetradecyl)trimethoxysilane solution. The striking loading capacity of this miniature boat may be attributed to the air film trapped around the mesh surface. This novel superhydrophobic steel mesh material have wide applications on medical materials, marine materials and smart materials.

Background:Triple-negative breast cancer(TNBC)is the most aggressive subtype and occurs in approximately 15%–20%of diagnosed breast cancers.TNBC is characterized by its highly metastatic and recurrent features,as well as a lack of specific targets and targeted therapeutics.Epidermal growth factor receptor(EGFR)is highly expressed in a variety of tumors,especially in TNBC.LR004-VC-MMAE is a new EGFR-targeting antibody–drug conjugate produced by our laboratory.This study aimed to evaluate its antitumor activities against EGFR-positive TNBC and further studied its possible mechanism of antitumor action.Methods:LR004-VC-MMAE was prepared by coupling a cytotoxic payload(MMAE)to an anti-EGFR antibody(LR004)via a linker,and the drug-to-antibody ratio(DAR)was analyzed by HIC-HPLC.The gene expression of EGFR in a series of breast cancer cell lines was assessed using a publicly available microarray dataset(GSE41313)and Western blotting.MDA-MB-468 and MDA-MB-231 cells were treated with LR004-VC-MMAE(0,0.0066,0.066,0.66,6.6 nmol/L),and the inhibitory effects of LR004-VC-MMAE on cell proliferation were examined by CCK-8 and colony formation.The migration and invasion capacity of MDA-MB-468 and MDA-MB-231 cells were tested at different LR004-VCMMAE concentrations(2.5 and 5 nmol/L)with wound healing and Transwell invasion assays.Flow cytometric analysis and tumorsphere-forming assays were used to detect the killing effects of LR004-VC-MMAE on cancer stem cells(MDA-MB-468 and MDA-MB-231 cells).The mouse xenograft models were also used to evaluate the antitumor efficacy of LR004-VC-MMAE in vivo.Briefly,BALB/c nude mice were subcutaneously inoculated with MDA-MB-468 or MDAMB-231 cells.Then they were randomly divided into 4 groups(n=6 per group)and treated with PBS,naked LR004(10 mg/kg),LR004-VC-MMAE(10 mg/kg),or doxorubicin,respectively.Tumor sizes and the body weights of mice were measured every 4 d.The effects of LR004-VC-MMAE on apoptosis and cell cycle distribution were analyzed by flow cytometry.Western blotting was used to d

Some issues in diagnostics of solar chromospheric magnetic fields with the Mg b_2 line

Up to now, exact measurements of chromospheric magnetic fields have not been as successful as those done in the photosphere. We are currently engaging in diagnostics of chromospheric magnetic fields with the Mg b2 line by employing the Multi-Channel Solar Telescope at Huairou Solar Observing Station. Therefore, how to improve accuracy in the measurement is the main issue of our present study. To this end, we first study linear calibration coefficients for longitudinal and transverse components of chromospheric fields, which vary with wavelength, in the case of a weak field assumption. Then the polarization crosstalk introduced by instruments is analyzed in detail with two numerical simulation methods. Comparisons of the po- larization signals between cases with and without correction are presented. The result indicates that polarization accuracy is greatly improved after crosstalk correction.

Chromosomal and Genetic Analysis of a Human Lung Adenocarcinoma Cell Line OM

Background： Lung cancer has become the leading cause of death in many regions.Carcinogenesis is caused by the stepwise accumulation of genetic and chromosomal changes.The aim of this study was to investigate the chromosome and gene alterations in the human lung adenocarcinoma cell line OM.Methods： We used Giemsa banding and multiplex fluorescence in situ hybridization focusing on the human lung adenocarcinoma cell line OM to analyze its chromosome alterations.In addition, the gains and losses in the specific chromosome regions were identified by comparative genomic hybridization （CGH） and the amplifications of cancer-related genes were also detected by polymerase chain reaction （PCR）.Results： We identified a large number of chromosomal numerical alterations on all chromosomes except chromosome X and 19.Chromosome 10 is the most frequently involved in translocations with six different interchromosomal translocations.CGH revealed the gains on chromosome regions of 3q25.3-28, 5p13, 12q22-23.24, and the losses on 3p25-26, 6p25, 6q26-27, 7q34-36, 8p22-23, 9p21-24, 10q25-26.3, 12p 13.31-13.33 and 17p 13.1-13.3.And PCR showed the amplification of genes： Membrane metalloendopeptidase （MME）, sucrase-isomaltase （SI）, butyrylcholinesterase （BCHE）, and kininogen （KNG）.Conclusions： The lung adenocarcinoma cell line OM exhibited multiple complex karyotypes, and chromosome 10 was frequently involved in chromosomal translocation, which may play key roles in tumorigenesis.We speculated that the oncogenes may be located at 3q25.3-28, 5p13, 12q22-23.24, while tumor suppressor genes may exist in 3p25-26, 6p25, 6q26-27, 7q34-36, 8p22-23, 9p21-24, 10q25-26.3, 12p 13.31-13.33, and 17p 13.1-13.3.Moreover, at least four genes （MME, SI, BCHE, and KNG） may be involved in the human lung adenocarcinoma cell line OM.

Synthesis and Characterization of Fluorinated Polyurethane Containing Carborane in the Main Chain: Thermal, Mechanical and Chemical Resistance Properties

In this study, two fluorinated polyurethanes（FPU） containing carborane groups in the main chains were firstly designed and synthesized via the reaction of hexamethylene diisocyanate trimer（HDI trimer） with fluorinated polyesters（CFPETs） having hydroxyl-terminated carborane groups at room temperature. The structures of carborane fluorinated polyesters（CFPETs） and polyurethanes（CFPUs） were characterized by gel permeation chromatography（GPC）, Fourier transform infrared（FTIR） spectroscopy and nuclear magnetic resonance（NMR） measurements. The thermal stability, mechanical properties, Shore A hardness, solvent resistance and acid-alkali resistance of the carborane fluorinated polyurethane films were also studied. Thermogravimetric analysis（TGA） tests manifested that the introduction of carborane groups into the main chain of fluorinated polyurethane endowed the obtained fluorinated polyurethane with excellent thermal stability. The thermal decomposition temperature of carborane fluorinated polyurethane（CFPU） increased by 190 °C compared with that of the carborane-free fluorinated polyurethane（FPU）. Even at 800 °C, CFPU showed the char yield of 66.5%, which was higher than that of FPU（34.3%）. The carborane-containing fluorinated polyurethanes also showed excellent chemical resistance and prominent mechanical property even after the cured films being immersed into Jet aircraft oil or 37% HCl for 168 h or at high temperature（700 °C）. It is found that the structural characteristics of carborane group and the compacted structure of CFPU effectively improve the thermal stability, mechanical property, solvent resistance and acid-alkali resistance of the carborane-free fluorinated polyurethane. These excellent properties make CFPU as the useful raw materials to prepare the high temperature resistant coatings or adhesives for automotive engines, engine or fuel tank of aircraft and other equipment working in high-temperature or high concentrations of acid-alkali environments.

Electrochemical dissolution behavior of gold and its main coexistent sulfide minerals in acid thiocyanate solutions

Electrochemical tests were developed to investigate the electrochemical dissolution behavior of gold and its main coexistent sulfide minerals in acid thiocyanate solutions.The optimal leaching conditions for gold in acidic thiocyanate system were pH 2,0.15 mol·L^(-1) thiocyanate and 0.2 g·L^(-1)Fe^(3+).Fe^(3+) addition to the acidic thiocyanate system promoted gold dissolution significantly,arsenopyrite dissolution was inhibited,chalcopyrite dissolution was increased,and the dissolution behavior of other associated minerals remained mostly unchanged.Thiocyanate made gold and associated mineral leaching easier.The galvanic corrosion effect of gold and its main coexistent sulfide minerals in an acidic thiocyanate-free system was that the chalcocite,arsenopyrite and pyrite acted as a cathode to reduce anodic gold dissolution;galena as an anode undergoes oxidation to inhibit anodic gold dissolution.There was almost no galvanic corrosion behavior between stibnite,yellow sphalerite and black sphalerite and gold.Thiocyanate addition changed the galvanic corrosion behavior of stibnite and yellow sphalerite in the thiocyanate system,which inhibited anodic gold dissolution.In the acidic thiocyanate system in the presence of ferric iron,the arsenopyrite promoted anodic gold dissolution,the chalcocite and gold were mostly free of galvanic corrosion,and the remaining minerals inhibited anodic gold dissolution.

Black hairy tongue associated with allo peripheral blood hematopoietic stem cell transplantation

Tongue lesions resulting from mucositis are a frequent complication of high-dose chemotherapy and irradiation. They are very common in patients with hematopoiefic stem cell transplantation, and tongue lesions due to other causes have also been reported. Black hairy tongue （BHT） is a special tongue lesion, not rare in the population with tobacco abuse, but so far it has not been reported after allo peripheral blood hematopoietic stern cell transplantation （allo-PBHST）. Here we presented a patient who developed BHT after allo-PBHST and discussed the factors that may cause this condition.
