Inhibitory Effects of Simvastatin on Oxidized Low-Density Lipoprotein-lnduced Endoplasmic Reticulum Stress and Apoptosis in Vascular Endothelial Cells

Background：Oxidized low-density lipoprotein （ox-LDL）-induced oxidative stress and endothelial apoptosis are essential for atherosclerosis. Our previous study has shown that ox-LDL-induced apoptosis is mediated by the protein kinase RNA-like endoplasmic reticulum kinase （PERK）/eukaryotic translation initiation factor 2α-subunit （eIF2α）/CCAAT/enhancer-binding protein homologous protein （CHOP） endoplasmic reticulum （ER） stress pathway in endothelial cells. Statins are cholesterol-lowering drugs that exert pleiotropic effects including suppression of oxidative stress. This study aimed to explore the roles of simvastatin on ox-LDL-induced ER stress and apoptosis in endothelial cells.Methods：Human umbilical vein endothelial cells （HUVECs） were treated with simvastatin （0.1, 0.5, or 2.5 μmol/L） or DEVD-CHO （selective inhibitor of caspase-3, 100 μmol/L） for 1 h before the addition of ox-LDL （100 μg/ml） and then incubated for 24 h, and untreated cells were used as a control group. Apoptosis, expression of PERK, phosphorylation of eIF2α, CHOP mRNA level, and caspase-3 activity were measured. Comparisons among multiple groups were performed with one-way analysis of variance （ANOVA） followed by post hoc pairwise comparisons using Tukey’s tests. A value of P 〈 0.05 was considered statistically significant.Results：Exposure of HUVECs to ox-LDL resulted in a significant increase in apoptosis （31.9% vs. 4.9%, P 〈 0.05）. Simvastatin （0.1, 0.5, and 2.5 μmol/L） led to a suppression of ox-LDL-induced apoptosis （28.0%, 24.7%, and 13.8%, F = 15.039, all P 〈 0.05, compared with control group）. Ox-LDL significantly increased the expression of PERK （499.5%, P 〈 0.05） and phosphorylation of eIF2α （451.6%, P 〈 0.05）, if both of which in the control groups were considered as 100%. Simvastatin treatment （0.1, 0.5, and 2.5 μmol/L） blunted ox-LDL-induced expression of PERK （407.8%, 339.1%, and 187.5%, F = 10.121, all P 〈 0.05, compared with control group） and phosphorylation of eIF2α （407.8%, 339.1%, 187.5%, F = 11.430, all P 〈 0.05, compared w

Clinical utility of cytokine analysis in the diagnosis and efficacy monitoring of vitreoretinal lymphoma

AIM:To investigate the value of cytokine analysis in aqueous humor(AH)for discriminating vitreoretinal lymphoma(VRL)from uveitis and for evaluating the efficacy of intravitreal methotrexate(MTX)injections.METHODS:This retrospective study was done on 28 VRL patients between 2013 and 2019.AH interleukin(IL)-10,IL-6,IL-8,vascular endothelial growth factor(VEGF),and vascular cell adhesion molecules(VCAM)were measured in 28 VRL patients and 38 uveitis patients.As to the respective examinations for distinguishing VRL from uveitis,the diagnostic accuracy was evaluated by receiver operating characteristic(ROC)curve analysis.The response to treatment was monitored by observing changes in bestcorrected visual acuity(BCVA),ocular manifestation,and AH cytokine levels in 21 patients with VRL who had undergo multiple intravitreal injections of MTX.RESULTS:Compared with uveitis patients,VRL patients had higher IL-10 level(P1.55 and 88.2%and 81.1%,respectively,for IL-10>76.7 pg/m L.In 21 patients who had undergo repeated injections,improvements in BCVA,clinical remission of VRL and continuous decrease in cytokine levels over time were observed.In those patients,the BCVA correlated with the aqueous levels of IL-10 and IL-6 during the course of disease treatment.CONCLUSION:The combination of the aqueous cytokine profiles can be instrumental for conventional diagnostic methods and for progression monitoring and treatment response.

Effects of internals on phase holdup and backmixing in a slightly-expanded-bed reactor with gas–liquid concurrent upflow

Five different internals were designed,and their effects on phase holdup and backmixing were investigated in a gas–liquid concurrent upflow reactor where the spherical alumina packing particles of three diameters(3.0,4.5 and6.0 mm)were slightly expanded under the conditions of varied superficial gas velocities(6.77×10-2-3.61×10-1 m·s-1)and superficial liquid velocities(9.47×10-4-2.17×10-3 m·s-1).The experimental results show that the gas holdup increases with the superficial gas velocity and particle size,opposite to the variational trend of liquid holdup.When an internal component is installed amid the upflow reactor,a higher gas holdup,a less liquid holdup and a larger Peclet number characterizing the weaker backmixing are obtained compared to those in the bed without internals under the same operating conditions.Additionally,the minimal backmixing is observed in the reactor equipped with the internals with a novel multi-step design.Finally,empirical correlations were proposed for estimating gas holdup,liquid holdup and Peclet number with the relative deviations within 11%,12%and 25%,respectively.

Vascular endothelium refers to a single layer of endothelial cells that line the inner surface of blood vessels,serving as barriers and transducers between the circulating blood in the lumen and the rest of the vessel wall.Endothelial cells play essential roles in many aspects of vascular biology,such as barrier functions,thrombosis/fibrinolysis,inflammation,angiogenesis,vasoconstriction and vasodilation.

Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis

Objective PERK/elF2/CHOP is a major signaling pathway mediating endoplasmic reticulum （ER） stress related with atherosclerosis. Oxidized LDL （ox-LDL） also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis. The present study was conducted to explore the regulatory effect of ox-LDL on PERK/elF2a/CHOP signaling pathway in vascular endothelial cells. Methods The effects of ox-LDL on PERK and p-elF2a protein expression of primary human umbilical vein endothelial cells （HUVECs） were investigated by Western blot analysis. PERK gene silencing and selective elF2a phosphatase inhibitor, salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis, caspase-3 activity, and CHOP mRNA level. Results Ox-LDL treatment significantly increased the expression of PERK, PERK-mediated inactivation of elF2a phosphorylation, and the expression of CHOP, as well as the caspase-3 activity and apoptosis. The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral shRNA or by selective elF2a phosphatase inhibitor, salubrinal. Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/elF2a/CHOP ER-stress pathway. It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis.

A review of application of base editing for the treatment of inner ear disorders

The development of applications for the CRISPR/Cas9(clustered regularly interspaced short palindromic repeats/CRISPR-associated nuclease 9)system has increased greatly in recent years,especially in the area of gene therapy by efficient in vivo genome editing.Although great success has been achieved in repairing and rewriting genomes through homology-directed repair coupled with Cas9 nuclease cleavage,its in vivo efficiency is insufficient for gene therapy.Base editing is a next-generation genome-editing tool that does not involve double-stranded DNA breaks and uses components of the CRISPR system together with other enzymes to make point mutations directly in cellular DNA or RNA.Base editors,composed of an engineered deaminase and a catalytically impaired CRISPR/Cas9 variant,are powerful tools for targeted base editing in cells and organisms.In non-dividing cells,base editors can directly transform one base or base pair into another,efficiently installing a point mutation.Undesired by-products of editing are seldom generated during this procedure.Herein we review the different base-editing platforms,including their deaminase recruitment strategies and editing outcomes,and the in vivo delivery of base editors.Additionally,we summarize therapeutic applications of base editing in disorders of the inner ear.

Erosion behavior of a ship propeller material of QAl9-4 alloy

The erosion behavior of a QA19-4 alloy as a ship propeller material was investigated.The effects of the solution,the impact angles and the sand content were considered.The test results demonstrate that the mass loss of the alloy in 3.5 wt%NaCl solution is 1.35 times that in tap water,due to the corrosive effect of Cl^(-).The mass loss of the alloy increases as the impact angle increases up to~30°and consequently decreases as the impact angle increases.Also,this feature is typical for the ductile metal behavior.At the impact angle of 0°,this feature is associated with the predominant erosion mechanisms,such as shallow plowing and surface fatigue;at 30°,this feature is micro-cutting,deep plowing and surface fatigue;and at 45°,it becomes indentation accompanied by extruded lips.The mass loss and surface roughness of the alloy increase as the sand content increases under the testing conditions.

通过拉伸性能测试对比研究了Cl-加速模拟混凝土孔隙液盐雾腐蚀前后三种钢筋的抗震性能变化特点。结果表明三种锈蚀钢筋的各项抗震性能均随着其平均质量锈蚀率的增加而下降。三种钢筋锈后其屈屈比、最大力下总延伸率和能量吸收功都体现出了较好的线性退化规律(线性拟合度R 2多在0.90以上)。对比三种锈蚀钢筋的抗震性能各项性能指标的性能退化因子λ,发现各项性能指标的性能退化因子λ均表现为:HPB300>HRB400E>HRB500E。V含量越多,钢筋抗震性能越好。对同一种锈蚀钢筋的各项性能退化因子λ进行比较,发现三种钢筋性能退化因子λ均表现为:能量吸收功>最大力下总延伸率(A gt)>屈屈比>强屈比。

Living gymnosperms comprise four major groups:cycads,Ginkgo,conifers,and gnetophytes.Relationships among/within these lineages have not been fully resolved.Next generation sequencing has made available a large number of sequences,including both plastomes and single-copy nuclear genes,for reconstruction of solid phylogenetic trees.Recent advances in gymnosperm phylogenomic studies have updated our knowledge of gymnosperm systematics.Here,we review major advances of gymnosperm phylogeny over the past 10 years and propose an updated classification of extant gymnosperms.This new classification includes three classes(Cycadopsida,Ginkgoopsida,and Pinopsida),five subclasses(Cycadidae,Ginkgoidae,Cupressidae,Pinidae,and Gnetidae),eight orders(Cycadales,Ginkgoales,Araucariales,Cupressales,Pinales,Ephedrales,Gnetales,and Welwitschiales),13 families,and 86 genera.We also described six new tribes including Acmopyleae Y.Yang,Austrocedreae Y.Yang,Chamaecyparideae Y.Yang,Microcachrydeae Y.Yang,Papuacedreae Y.Yang,and Prumnopityeae Y.Yang,and made 27 new combinations in the genus Sabina.

Clinical characteristics of ocular toxocariasis in adults in north China

AIM: To investigate the clinical features of adult patients with ocular toxocariasis(OT) in north China and to diagnose adults OT patients in early stage.METHODS: Clinical data of 24 adults with OT were retrospectively analyzed.Slit lamp photographs and fundus photographs and other imaging examinations of all the patients were reviewed.A questionnaire concerning the pet ownership and place of residence was completed to investigate the possible infection origin.Descriptive statistical analyses were performed on the demographic data, clinical features, funduscopic findings and ELISA results.RESULTS: Among the 24 patients diagnosed with OT by Toxocara IgG antibody in intraocular fluid, 16(66.7%) eyes were right eye.The onset age of 12 eyes(50.0%) was between 30 and 40 years old, and 21(87.5%) eyes were of peripheral granuloma type.The most common sign was vitreous opacity.Granulomas were detected in all the eyes, and 20(83.3%) patients resided in rural area.In 4 patients, the concentration of anti-Toxocara antibody both in anterior humor and in vitreous humor were detected, and the results showed the concentration in vitreous humor was much higher than aqueous humor.CONCLUSION: Our study analyzes the clinical manifestation of OT in adults, which may have been underrecognized before.Eye side, residence, and detection of granuloma may help us in diagnosis of OT in patients with monocular vitreous opacity.For adult patients with presumed OT, negative results of anti-Toxocara antibody in anterior humor cannot rule out the possibility of OT, further detection of vitreous humor is suggested for final diagnosis.
