Cationic liposome-mediated transfection of CD40 ligand gene inhibits hepatic tumor growth of hepatocellular carcinoma in mice

Objective: To evaluate the efficacy of cationic liposome-mediated CD40 ligand (CD40L) gene therapy for hepato- cellular carcinoma. Methods: 1×106 of parental H22 cells or H22 cells transfected with the expression vector containing murine CD40L cDNA encoding the entire coding region (pcDNA3.1+-mCD40L) were inoculated subcutaneously into the left flanks of syngenic BALB/C mice. The tumor-bearing mice (tumor nodules 10 mm in maximal diameter) received the treatment of the intratumoral injection of pcDNA3.1+-mCD40L/Transfectam, pcDNA3.1+, or phosphate-buffered saline (PBS), or no treatment. The mice were monitored for tumor growth weekly. We examined mCD40L messenger ribonucleic acid (mRNA) expression by reverse transcription polymerase chain reaction (RT-PCR) and the histologic changes in tumors at two weeks after intratumoral injection using immunohistochemical staining of tumor tissues. Results: All mice inoculated with parental H22 cells developed a tumor subcutaneously, and the tumor size increased progressively within three weeks. However, the mice receiving H22-CD40L cells exhibited complete regression of the tumor two weeks after tumor cell inoculation. The tumor-bearing animals with the treatment of pcDNA3.1+ or PBS, or without treatment had progressive tumor growth, while those mice treated with pcDNA3.1+-mCD40L exhibited a significant inhibition of tumor growth. RT-PCR analysis showed that 783-bp fragments cor- responding to the mCD40L mRNA were amplified only from pcDNA3.1+-mCD40L treated tumors. The tumor samples from pcDNA3.1+-mCD40L-treated mice showed significant lymphocyte infiltration, apoptotic bodies, and confluent necrosis in the tumor tissues. Conclusion: The tumorigenicity of CD40L-expressing cells was abrogated when the cells were implanted subcu- taneously. In vivo gene therapy of established liver tumor nodules in mice by the intratumoral injection of pcDNA3.1+-mCD40L led to significant tumor inhibition. There was mCD40L mRNA expression in the

Dear Editor,This letter is concerned with stability analysis and stabilization design for sampled-data based load frequency control(LFC) systems via a data-driven method. By describing the dynamic behavior of LFC systems based on a data-based representation, a stability criterion is derived to obtain the admissible maximum sampling interval(MSI) for a given controller and a design condition of the PI-type controller is further developed to meet the required MSI. Finally, the effectiveness of the proposed methods is verified by a case study.

Experimental and numerical simulation of the attenuation effect of blast shock waves in tunnels at different altitudes

Traffic engineering such as tunnels in various altitudinal gradient zone are at risk of accidental explosion,which can damage personnel and *** prediction of the distribution pattern of explosive loads and shock wave propagation process in semi-enclosed structures at various altitude environment is key research focus in the fields of explosion shock and fluid *** effect of altitude on the propagation of shock waves in tunnels was investigated by conducting explosion test and numerical *** on the experimental and numerical simulation results,a prediction model for the attenuation of the peak overpressure of tunnel shock waves at different altitudes was *** results showed that the peak overpressure decreased at the same measurement points in the tunnel entrance under the high altitude *** contrast,an increase in altitude accelerated the propagation speed of the shock wave in the *** average error between the peak shock wave overpressure obtained using the overpressure prediction formula and the measured test data was less than15%,the average error between the propagation velocity of shock waves predicted values and the test data is less than 10%.The method can effectively predict the overpressure attenuation of blast wave in tunnel at various altitudes.

Increased Orexin Expression Promotes Sleep/Wake Disturbances in the SOD1-G93A Mouse Model of Amyotrophic Lateral Sclerosis

Background：Sleep/wake disturbances in patients with amyotrophic lateral sclerosis （ALS） are well-documented,however,no animal or mechanistic studies on these disturbances *** is a crucial neurotransmitter in promoting wakefulness in sleep/wake regulation,and may play an important role in sleep disturbances in *** this study,we used SOD1-G93A transgenic mice as an ALS mouse model to investigate the sleep/wake disturbances and their possible mechanisms in ***：Electroencephalogram/electromyogram recordings were performed in SOD1-G93A transgenic mice and their littermate control mice at the ages of 90 and 120 days,and the samples obtained from these groups were subjected to quantitative reverse transcriptase-polymerase chain reaction,western blotting,and enzyme-linked immunosorbent ***：For the first time in SOD1-G93A transgenic mice,we observed significantly increased wakefulness,reduced sleep time,and up-regulated orexins （prepro-orexin,orexin A and B） at both 90 and 120 *** analysis confirmed moderate to high correlations between sleep/wake time （total sleep time,wakefulness time,rapid eye movement [REM] sleep time,non-REM sleep time,and deep sleep time） and increase in orexins （prepro-orexin,orexin A and B）.Conclusion：Sleep/wake disturbances occur before disease onset in this ALS mouse *** orexins may promote wakefulness and result in these disturbances before and after disease onset,thus making them potential therapeutic targets for amelioration of sleep disturbances in *** studies are required to elucidate the underlying mechanisms in the future.

Bending behaviors of large diameter thin-walled CP-Ti tube in rotary draw bending

The rotary draw bending(RDB) of large diameter thin-walled(LDTW) tube needs the strict cooperation of multiple processing parameters to avoid possible multiple *** to the specific properties,the bending behaviors of LDTW commercial pure titanium(CP-Ti) tube are much more complex to achieve precise *** the CP-Ti tube of 50.8 mm(out diameter, D)×0.508 mm(wall thickness,t)×101.6 mm(bending radius,R) as a representative component,the bending behaviors of LDTW CP-Ti tube under different processing conditions were *** experiments and analytical analysis,the bending characteristics of the CP-Ti tube were ***,based on the orthogonal experimental design,a series of three-dimensional FE models of RDB for LDTW CP-Ti tube were established,and the effects of processing parameters on the bending behaviors were numerically *** results show that:1) Wrinkling is the primary behavior for the LDTW CP-Ti tube in RDB,and the larger the difference between the maximum wall thickening and the maximum wall thinning degrees of the bent tube,the larger the wrinkling tendency;2) The bending behaviors of the LDTW CP-Ti tube are very sensitive to the some processing parameters,and the wrinkling is significantly affected by the mandrel shank diameter,while the wall thinning is remarkably affected by the clearance between wiper die and tube,mandrel shank *** qualified bent tube with the wall thinning of 11.43%,the cross-section distortion of 2.69%and the wrinkling height less than 2%is then obtained.

Effect of gradual increase of salt on performance and microbial community during granulation process

Salinity was considered to have effects on the characteristics,performance microbial communities of aerobic granular *** study investigated granulation process with gradual increase of salt under different *** identical sequencing batch reactors were operated,while the influent of Ra and Rb was subjected to stepwise increments of NaCl concentrations(0-4 g/L and 0-10 g/L).The presence of filamentous bacteria may contribute to granules formed under lower salinity conditions,potentially leading to granules *** removal efficiency achieved in both reactors although therewas a small accumulation of nitrite in Rb at later *** removal efficiencies of chemical oxygen demand(COD),total nitrogen(TN),and total phosphorus(TP)in Ra were 95.31%,93.70%and 88.66%,while the corresponding removal efficiencies in Rb were 94.19%,89.79%and 80.74%.Salinity stimulated extracellular polymeric substances(EPS)secretion and enriched EPS producing bacteria to help maintain the integrity and stability of the aerobic *** nitrifying bacteria were responsible for NH_(4)^(+)-N and NO_(2)^(-)-N oxidation of salinity systems and large number of denitrifying bacteria were detected,which ensure the high removal efficiency of TN in the systems.

以水厂坝鱼道2015年8月至2016年7月监测资料为基础,进行统计分析,并通过与同类鱼道对比,对该鱼道的过鱼效果进行评价。分析了鱼道的过鱼种类组成,季节、上游水位变化对鱼道过鱼效果的影响。监测期内,水厂坝鱼道过鱼数量共1 043尾,39种,分属3目、10科37属,鱼道的主要过鱼季节是4—10月,当鱼道水温在20~35℃之间变化,上游水位32.6~33.0 m之间变化时,对应的鱼道水流条件可使鱼道达到较好的过鱼效果。与国内外其他鱼道相比,有39种鱼类能够找到水厂坝鱼道进口,并顺利通过鱼道,高于其他几个鱼道。但该鱼道过鱼数量较其他鱼道偏少。研究成果有助于流溪河上游水库开展生态调度及鱼类资源保护等工作,为推动流溪河其他梯级闸坝及珠三角平原区低水头闸坝工程增设鱼道提供参考。

Indole derivatives,especially bisindolylesters,have attracted intense attention due to their important applications in medicinal chemistry and organic *** we develop a Lewis acid-catalyzed efficient and regioselective strategy to prepare a series of symmetric and unsymmetric bisindolylesters in high to excellent yields under mild *** systematic investigations,which include stoichiometric nuclear magnetic resonance(NMR)experiments and structural characterization of intermediates,have provided insights into the possible reaction mechanism for this B(C_(6)F_(5))_(3)-catalyzed addition ***,the sequential employment of Al(C_(6)F_(5))_(3)and B(C_(6)F_(5))_(3)as catalysts enabled us to successfully prepare the unsymmetric bisindolyl-compounds in one-pot two-step manner without the separation step.
