Black rice， as a dietary supplement， has received increasing attention because of its beneficial health properties. Although black rice and its major ingredients have been considered a potential therapy for ulcerative colitis (UC)， its effect and underlying mechanism of action remain obscure. In this study， black rice was demonstrated to improve Dextran Sulfate Sodium Salt (DSS)-induced UC and restore intestinal barrier dysfunction. Cyanidin 3-O-glucoside (C3G) is the most abundant ingredient in black rice. Similarly， in vivo and in vitro studies have demonstrated that C3G alleviates colitis and intestinal barrier dysfunction. Proteomic analysis showed that C3G significantly reduced abnormally elevated swiprosin-1 levels in mice with colitis. Furthermore， the effect of C3G on mitigating colitis was inhibited after swiprosin-1 was overexpressed in intestinal tissue in vivo and Caco-2 cells in vitro. Acyl-CoA synthetase long-chain family member 1 (ACSL1) was identified as a potential protein that interacts with swiprosin-1 by co-immunoprecipitation and liquid chromatography–tandem mass spectrometry. By integrating metabolomics experiments and bioinformatics analysis of single-cell sequencing， ACSL1 was found to selectively bind to swiprosin-1 and regulate downstream linoleic acid metabolism in colitis. Moreover， C3G was observed to prevent the localization of ACSL1 within mitochondria via swiprosin-1， thereby inhibiting the oxidation of linoleic acid. This study demonstrated that C3G targeted swiprosin-1 to ameliorate UC via ASCL1-mediated linoleic acid metabolism， thereby providing a novel potential insight into the mechanism of C3G protection against colitis and establishing the groundwork for its clinical application.

Design and synthesis of novel antifungal triazole derivatives with good activity and water solubility

In order to find novel antifungal agents with good activity and aqueous solubility, a series of SYN-2869 analogues containing a pyridine ring were synthesized and evaluated for their in vitro antifungal activity and water solubility. The results indicated that some compounds showed potent activity against six pathogenic fungi. In particular, the analogue 17a having an isobutyl substitution on the triazolone exhibited significant broad spectrum antifungal activity. In addition, the water solubility of compound l？a was sufficiently improved over SYN-2869.

Design,synthesis of novel antifungal triazole derivatives with high activities against Aspergillus fumigatus

Based on the active site of Aspergillus fumigatus lanosterol 14α-demethylase （AF-CYP51）, novel triazole compounds were designed. Their chemical synthesis and the antifungal activities were reported. The results showed that all the target compounds exhibited excellent activities with broad spectrum; in which compounds 4, 12 and 15 showed comparable activities against A. fumigatus to the control drug itraconazole.

To improve the aqueous solubility of an itraconazole analogue, compound 1 （YL-24）, a series of novel prodrugs were synthesized. Among these prodrugs, the phosphate disodium salt compound 7 exhibited excellent aqueous solubility （9.8 mg/mL） at near-neutral pH and sufficient stability in buffer solutions, along with favorable pharmacokinetic profiles. In particular, compounds 1 and 7 provided moderate survival efficacy in murine systemic Candida albicans SC5314 infection model, but their efficacy was weaker than that of fluconazole.

A series of novel triazole derivatives containing y-lactam were designed and synthesized, and their structures were confirmed by IH NMR, 13C NMR and HRMS, The in vitro antifungal activities of the target compounds were evaluated, The results showed that all of the compounds exhibited stronger activity against the six clinically important fungi tested than fluconazole. 3D and 3E showed comparative activity against the fungi tested except for Candida glabrata and Aspergillus fumigatus as voriconazole. In addition, the docking model for 2A and CYP51 was investigated.

Based on the active site of Candida albicans lanosterol 14α-demethylase (CACYP51), novel triazole compounds structurally different from the current triazole drugs were designed and synthesized. In vitro antifungal activities showed that compounds 10, 11, 16 and 20 exhibited strong activities. In addition, compounds 10, 11 and 16 also displayed certain activities against fluconazole-resistant fungi.

An Online Q-Learning Method for Linear-Quadratic Nonzero-Sum Stochastic Differential Games with Completely Unknown Dynamics

In this paper, the authors design a reinforcement learning algorithm to solve the adaptive linear-quadratic stochastic n-players non-zero sum differential game with completely unknown dynamics. For each player, a critic network is used to estimate the Q-function, and an actor network is used to estimate the control input. A model-free online Q-learning algorithm is obtained for solving this kind of problems. It is proved that under some mild conditions the system state and weight estimation errors can be uniformly ultimately bounded. A simulation with five players is given to verify the effectiveness of the algorithm.
