The Moon provides a unique environment for investigating nearby astrophysical events such as *** samples retain valuable information from these events,via detectable long-lived“fingerprint”radionuclides such as^(60)*** this work,we stepped up the development of an accelerator mass spectrometry(AMS)method for detecting^(60)Fe using the HI-13tandem accelerator at the China Institute of Atomic Energy(CIAE).Since interferences could not be sufficiently removed solely with the existing magnetic systems of the tandem accelerator and the following Q3D magnetic spectrograph,a Wien filter with a maximum voltage of±60 kV and a maximum magnetic field of 0.3 T was installed after the accelerator magnetic systems to lower the detection background for the low abundance nuclide^(60)Fe.A 1μm thick Si_(3)N_(4) foil was installed in front of the Q3D as an energy *** particle detection,a multi-anode gas ionization chamber was mounted at the center of the focal plane of the ***,an^(60)Fe sample with an abundance of 1.125×10^(-10)was used to test the new AMS *** results indicate that^(60)Fe can be clearly distinguished from the isobar^(60)*** sensitivity was assessed to be better than 4.3×10^(-14)based on blank sample measurements lasting 5.8 h,and the sensitivity could,in principle,be expected to be approximately 2.5×10^(-15)when the data were accumulated for 100 h,which is feasible for future lunar sample measurements because the main contaminants were sufficiently separated.

Mammalian cells express two isoforms of Cu- and Zn-containing superoxide dismutases（SODs）, CuZn-SOD and extracellular SOD（EC-SOD）, involved in the defense system against reactive oxygen species（ROS）. The two SODs have structurally homologous centre domain with distinct N- and C-terminuses, resulting in the different characteristics of the structure and function of the two molecules. We generated a hybrid SOD molecule（namely hy- SOD） via replacing the N- and C-terminuses of hCuZnSOD with the counterparts of hEC-SOD. The hySOD was expressed in host Pichia pastoris and the purified protein was a dimer with a molecular weight of about 34000. A series of activity analyses indicates that the hySOD is similar to hEC-SOD in heat-stability, and has the activity of protecting the host cell against heat shock and oxidative stress. Our results show evidence for the study on the compound activity of multiple SOD molecules, and may be important for understanding the relationship between structure and function of hEC-SOD and hCuZnSOD.

Ficin is an important component of plants in Ficus family such as fig latex. It is of special significance in medicine and industry because it exhibits activity throughout a wide range of temperature and pH values. In this work, we purified a component of ficin from the latex homogeneity of shandong fig trees, and the properties of the purified ficin were studied. The current findings revealed that heavy metal ions were able to inhibit ficin, while DTT, L-cysteine, and β-ME were found to promote ficin activity. It was also observed that the half life of ficin at 65 °C was longer than 1 h and the Michaelis constant（Km） for casein hydrolyzation was determined to be 1.56 mg/mL. Our study shows that this purified ficin is a cysteine protease.

研究旨在探究饲料中添加酵母培养物“水产益康”对大口黑鲈(Micropterus salmoides)生长性能、肠道健康、免疫力和病菌攻击后存活率等方面的影响,在基础对照饲料中添加质量分数0.1%和0.3%的酵母培养物“水产益康”(分别为0.1%添加组和0.3%添加组),配置3种等氮等脂的实验饲料。将360尾初始质量为(32.37±0.15)g的大口黑鲈分为3个处理,每个处理设3个重复,每个网箱放养40尾鱼,生长实验周期为11周。结果表明:两个添加组的生长性能与对照组相比没有显著差异(P>0.05);在肠道结构方面,0.3%添加组的绒毛长度较对照组显著增加,隐窝深度较对照组显著下降(P0.05);嗜水气单胞菌(Aeromonas hydrophila)攻毒结果显示,攻毒后两实验组的累计存活率较对照组升高,在252h以后有显著区别(P<0.05)。以上结果表明,饲料中添加0.1%—0.3%培养物对大口黑鲈肠道健康、抗氧化能力、免疫和抗病力有积极作用。

Setosphaeria turcica,an essential phytopathogenic fungus,is the primary cause of serious yield losses in corn; however,its pathogenic mechanism is poorly *** cloned STK2,a newly discovered mitogen-activated protein kinase gene with a deduced amino acid sequence that is 96% identical to MAK2 from Phaeosphaeria nodorum,56% identical to KSS1 and 57% identical to FUS3 from Saccharomyces *** deduce Stk2 function in *** and to identify the genetic relationship between STK2 and KSS1/FUS3 from ***,a restructured vector containing the open reading frame of STK2 was transformed into a fus3/kss1 double deletion mutant of *** results show that the STK2 complementary strain clearly formed pseudohyphae and ascospores,and the strain grew on the surface of the medium after rinsing with sterile water and the characteristics of the complementary strain was the same as the wild-type ***,STK2 complemented the function of KSS1 in filamentation and invasive growth,as well as the mating behavior of FUS3 in ***,however,its exact functions in *** will be studied in the future research.

The mitogen-activated protein kinase （MAPK）, a key signal transduction component in the MAPK cascade pathway, regulates a variety of physiological activities in eukaryotes. However, little is known of the role MAPK plays in phytopathogenic fungi. In this research, we cloned the MAPK gene STK1 from the northern corn leaf blight pathogen Setosphaeria turcica and found that the gene shared high homology with the high osmolality glycerol （HOG） MAPK gene HOG1 of Saccharomy- ces cerevisiae. In addition, gene knockout technology was employed to investigate the function of STKI. Gene knockout mutants （KOs） were found to have altered hyphae morphology and no conidiogenesis, though they did show similar radial growth rate compared to the wild-type strain （WT）. Furthermore, microscope observations indicated that STK1 KOs did not form normal appressoria at 48 h post-inoculation on a hydrophobic surface. STK1 KOs had reduced virulence, a significantly altered Helminthosporium turcicum （HT）-toxin composition, and diminished pathogenicity on the leaves of susceptible inbred corn OH43. Mycelium morphology appeared to be significantly swollen and the radial growth rates of STK1 KOs declined in comparison with WT under high osmotic stress. These results suggested that STK1 affects the hyphae development, conidiogenesis, and pathogenicity of S. turcica by regulating appressorium development and HT-toxin biosynthesis. Moreover, the gene appears to be involved in the hypertonic stress response in S. turcica.

Abundant and diverse domestic mammals living on the Tibetan Plateau provide useful materials for investigating adaptive evolution and genetic convergence. Here, we used 327 genomes from horses, sheep,goats, cattle, pigs and dogs living at both high and low altitudes, including 73 genomes generated for this study, to disentangle the genetic mechanisms underlying local adaptation of domestic mammals. Although molecular convergence is comparatively rare at the DNA sequence level, we found convergent signature of positive selection at the gene level, particularly the EPAS1 gene in these Tibetan domestic mammals. We also reported a potential function in response to hypoxia for the gene C10 orf67, which underwent positive selection in three of the domestic mammals. Our data provide an insight into adaptive evolution of high-altitude domestic mammals, and should facilitate the search for additional novel genes involved in the hypoxia response pathway.
