To prepare the PrP specific monoclonal antibodies （mAbs） that can be used for the detection of mammalian prions and study of pathogenesis of prion diseases. Methods Several BALB/c mice were immunized with recombinant hamster prion protein （HaPrP）. Three hybridoma cell lines designated as B7, B9, and B10, secreting monoclonal antibodies against HaPrP, were established by hybridoma technique. The mAbs reactivities were evaluated with EliSA, Western blot, and immunohistochemistry. Results The mAbs produced by these cell lines reacted well with different recombinant hamster PrP proteins. Western blot analyses showed that mAbs B7 and B9 reacted with PrP^Sc from the scrapie-infected animals after proteinase K digestion with three glycosylated forms. The mAbs exhibited cross-reactivity with various PrPc from several other mammalian species, including humans and cattles, lmmunohistochemistry assays confirmed that mAbs B7 and B9 could recognize not only extracellular but also intracellular PrPso. Conclusion The mAbs of prion protein are successfully generated by hybridoma technique and can be applied for the diagnosis of prion associated diseases.

In this work, we provide 2189 photometrically- and kinematically-selected candidate members of 24 star clusters from the LAMOST DR2 catalog. We perform two-step membership identification： selection along the stellar track in the colormagnitude diagram, i.e., photometric identification, and selection from the distribution of radial velocities, i.e. the kinematic identification. We find that the radial velocities from the LAMOST data are very helpful in the membership identification. The mean probability of membership is 40% for the sample selected with radial velocity. With these 24 star clusters, we investigate the performance of the radial velocity and metallicity estimated with the LAMOST pipeline. We find that the systematic offsets in radial velocity and metallicity are 0.85 ± 1.26 km s-1and-0.08 ± 0.04 dex, with dispersions of 5.47＋1.16-0.71 km s-1and 0.13＋0.04-0.02 dex, respectively. Finally, we propose that the photometrically-selected candidate members of the clusters covered by the LAMOST footprint should be assigned higher priority so that more candidate stars can be observed.

To treat technical problems of data acquisition,alarm management, and historical data archiving of radiation protection control system in Thorium Molten Salt Reactor, a network prototype system was designed based on experimental physics and industrial control *** level of many locations in Jiading campus of Shanghai Institute of Applied Physics is being continuously monitored, and boundaries of accelerator facilities and radiochemical areas are defined by the access control system. In this paper, we introduce the control system design,including human–machine interfaces, alarm system, historical data archiving system, and the software for access control. The software development followed the standard of Capability Maturity Model Integration(CMMIò) Level3, and the software had passed a third-party test, which indicated that the functionality and the reliability could fulfill the requirements of the radiation protection system.

AIM: To investigate the protective effect and mechanisms of ghrelin postconditioning against hypoxia/reoxygenation (H/R)-induced injury in human gastric epithelial cells. METHODS: The model of H/R injury was established in gastric epithelial cell line (GES-1) human gastric epithelial cells. Cells were divided into seven groups: normal control group (N); H/R postconditioning group; DMSO postconditioning group (DM); ghrelin postconditioning group (GH); D-Lys3-GHRP-6 + ghrelin postconditioning group (D + GH); capsazepine + ghrelin postconditioning group (C + GH); and LY294002 + ghrelin postconditioning group (L + GH). 3-(4,5-dimethylthazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay was used to detect GES-1 cell viability. Hoechst 33258 fluorochrome staining and flow cytometry were conducted to determine apoptosis of GES-1cells. Spectrophotometry was performed to determine release of lactate dehydrogenate (LDH). Protein expression of Bcl-2, Bax, Akt, and glycogen synthase kinase (GSK)-3β was determined by western blotting. Expression of vanilloid receptor subtype 1 (VR1), Akt and GSK-3β was observed by immunocytochemistry. RESULTS: Compared with the H/R group, cell viability of the GH group was significantly increased in a dosedependent manner (55.9% ± 10.0% vs 69.6% ± 9.6%, 71.9% ± 17.4%, and 76.3% ± 13.3%). Compared with the H/R group, the percentage of apoptotic cells in the GH group significantly decreased (12.38% ± 1.51% vs 6.88% ± 0.87%). Compared with the GH group, the percentage of apoptotic cells in the D + GH group, C + GH group and L + GH groups significantly increased (11.70% ± 0.88%, 11.93% ± 0.96%, 10.20% ± 1.05% vs 6.88% ± 0.87%). There were no significant differences in the percentage of apoptotic cells between the H/R and DM groups (12.38% ± 1.51% vs 13.00% ± 1.13%). The phenomenon of multiphase flow in porous media is confronted in various fields of science and industrial applications. Owing to the complicated porous structure, the flow mechanisms are still not completely resolved. A critical and fundamental question is the variation of pore structure and REV sizes among different types of porous media. In this study, a total of 22 porous samples were employed to systematically evaluate the pore-based architecture and REV sizes based on X-ray CT image analysis and pore network modelling. It is found that the irregular grain shapes give rise to large specific areas,narrow and thin throats in identical sand packs. The packs with more types of sands, or composited by the sands with larger difference in diameter, have larger specific area, smaller tortuosity and pore *** on the REV measurement through porosity solely, the REV sizes of sand packs are generally in the order of magnitude of 10^(-2) m L, while it is at least one order of magnitude smaller in rock cores. Our result indicates that the combination of porosity and Euler number is an effective indicator to get the REV sizes of porous samples.
