Thioredoxin and thioredoxin-interacting protein as prognostic markers for gastric cancer recurrence

AIM:To evaluate the potential of thioredoxin (TXN) and thioredoxin-interacting protein (TXNIP) expression as biomarkers for predicting gastric cancer recurrence. METHODS:TXN and TXNIP expression levels were acquired from gene expression microarray data for 65 human gastric cancer tissues. We determined whether each gene expression level was associated with cancer recurrence and investigated the relationship between the two genes. For validation, the expression levels of TXN and TXNIP were measured by quantitative real- time reverse transcription polymerase chain reaction in 68 independent stage Ⅲ gastric cancer patients. The correlation between gene expression and cancer prognosis was evaluated. Immunohistochemical staining was performed to investigate the protein expression levels of TXN and TXNIP and to characterize the expression patterns of each protein. RESULTS:TXN was a prognosis-related gene (P = 0.009), whereas TXNIP, a TXN inhibitor, demonstrated a negative correlation with TXN in the gene expression microarray data. In the 68 stage Ⅲ patients, the expression levels of both TXN and TXNIP had a statistically significant effect on recurrence-free survival (RFS, P = 0.008 and P = 0.036, respectively). The low TXN and high TXNIP expression group exhibited a better prognosis than the other groups, and the high TXN and low TXNIP expression group exhibited a poorer prognosis (P < 0.001 for RFS and P = 0.001 for overall survival). More than half of the patients in the simulta-neously high TXN and low TXNIP expression group ex- perienced a recurrence within 1 year after curative surgery, and the 5-year survival rate of the patients in this group was 29%, compared with 89% in the low TXN and high TXNIP expression group. The TXN protein was overexpressed in 65% of the gastric cancer tissues, whereas the TXNIP protein was underexpressed in 85% of the cancer cells. AIM： To evaluate the effect of ranitidine on gastric mucosal changes and on GI bleeding in long distance runhers. METHODS： Twenty-four long distance runners （M： 16, F： 8, age： 18.2＋ 1.5 years） participated in this study. A symptom questionnaire, stool hemoccult test, and upper gastrointestinal （GI） endoscopy were performed on the subjects prior to the study. The subjects took oral ranitidine （150 mg, b.i.d.） for two weeks. The upper GI endoscopy and stool Hemoccult tests were repeated after the treatment. RESULTS： Twenty-two of the 24 runners had at least one upper G1 mucosal lesion before the medication. The Endoscopic improvements were seen in eleven of the 14 cases of erosive gastritis and four of the 5 cases of esophagitis. Six subjects were Heine occult positive prior to the study, but only one was positive after the medication. CONCLUSION： Gastric mucosal lesions and GI bleeding in long distance runners seem to be associated to acidrelated factors mediated by the high level of regular running. Ranitidine seems to be and effective prophylaxis to prevent gastric mucosal lesions and GI bleeding.

AIM To evaluate the efficacy and safety of HL tablet extracted from magnolia officinalis for treating patients with nonalcoholic fatty liver disease(NAFLD).METHODS Seventy-four patients with NAFLD diagnosed by ultrasonography were randomly assigned to 3 groups given high dose(400 mg) HL tablet, low dose(133.4 mg) HL tablet and placebo, respectively, daily for 12 wk. The primary endpoint was post-treatment change of hepatic fat content(HFC) measured by magnetic resonance spectroscopy. Secondary endpoints included changes of serum aspartate aminotransferase, alanine aminotransferase(ALT), cholesterol, triglyceride, free fatty acid, homeostasis model assessment-estimated insulin resistance, and body mass index(BMI).RESULTS The mean HFC of the high dose HL group, but not of the low dose group, declined significantly after 12 wk of treatment(high dose vs placebo, P = 0.033; low dose vs placebo, P = 0.386). The mean changes of HFC from baseline at week 12 were-1.7% ± 3.1% in the high dose group(P = 0.018),-1.21% ± 4.97% in the low dose group(P = 0.254) and 0.61% ± 3.87% in the placebo group(relative changes compared to baseline, high dose were:-12.1% ± 23.5%, low dose:-3.2% ± 32.0%, and placebo: 7.6% ± 44.0%). Serum ALT levels also tended to decrease in the groups receiving HL tablet while other factors were unaffected. There were no moderate or severe treatment-related safety issues during the *** HL tablet is effective in reducing HFC without any negative lipid profiles, BMI changes and adverse effects.

AIM:To compare outcomes from radiofrequency ablation(RFA) and hepatectomy for treatment of colorectal liver metastasis(CRLM).METHODS:From January 2000 to December 2009,408 patients underwent curative intent treatment for *** excluded patients using the criteria:size of CRLM > 3 cm,number of CRLM ≥ 5,percutaneous RFA,follow-up period 2 cm) CRLM(P = 0.034).However,survival curves were similar for single or small(≤ 2 cm) CRLM(P = 0.714,P = 0.740).CONCLUSION:Hepatectomy is better than RFA for the treatment of ***,RFA might be suitable for selected patients with single,small(≤ 2 cm) CRLM.

Chinese cabbage was cultivated in upland soil with the addition of biochar in order to investigate the potential for reduction of greenhouse gas emissions. Barley straw biochar(BSB) was introduced in a Wagner pot(1/5000a) in amounts of 0(BSB0, control), 100(BSB100), 300(BSB300), and 500(BSB500) kg 10a^(-1). After the addition of BSB into the upland soil, carbon dioxide(CO_2) emission increased while methane(CH_4) and nitrous oxide(N_2O) emissions decreased. The highest CO2 flux was measured for the BSB500 sample,(84.6 g m^(-2)) followed by BSB300, BSB100, and BSB0 in decreasing order. Relative to those of control, the total CH_4 flux and N_2O flux for the BSB500 treatment were lower by 31.6% and 26.1%, respectively. The global warming potential(GWP) of the treatment without biochar was 281.4 g CO_2 m-2 and those for treatments with biochar were in the range from 194.1 to 224.9 g CO_2 m^(-2). Therefore, introducing BSB into upland soil to cultivate Chinese cabbages can reduce the global warming potential.

A new putative transposon was identified in the tobacco budworm, Helio- this virescens. This transposon was characterized as a full length CORE-SINE （65 bp of ＂CORE＂ core specific nucleotide short interspersed elements） that resembled sequences from three other lepidopterans and humans. In particular, the A-box and B-box regions of this sequence most closely conformed to the signature of CORE-SINEs from widely divergent species. This CORE-SINE was present as a polymorphism in a hypervariable region of the gene hscp, which is the target of pyrethroid insecticides and other xenobiotics in the nerve axon. We described this new putative transposon as Noct-1 due to its presence in a noctuid moth. This is the first description of a full-length CORE-SINE with the A-box, B-box, target site duplication, and candidate core domain from an insect.

Antiproliferative Effects of Zinc-Citrate Compound on Hormone Refractory Prostate Cancer

Objective： To investigate the antiproliferative effects of zinc-citrate compound on hormone refractory prostate cancer （HRPC）. Methods： HRPC cell line （DU145） and normal prostate cell line （RWPE-1） were treated with zinc, citrate and zinc-citrate compound at different time intervals and concentrations to investigate the effect of zinc-citrate compound. Mitochondrial （m）-aconitase activity was determined using aconitase assay. DNA laddering analysis was performed to investigate apoptosis of DU145 cells. Molecular mechanism of apoptosis was investigated by Western blot analysis of P53, P21w^fl, Bcl-2, Bcl-xL and Bax, and also caspase-3 activity analysis. Results： Treatment with zinc-citrate compound resulted in a time- and dose-dependent decrease in cell number of DU145 cells in comparison with RWPE-1. M-aconitase activity was significantly decreased. DNA laddering analysis indicated apoptosis of DU145 cells. Zinc-citrate compound increased the expression of P21wall and P53, and reduced the expression of Bcl-2 and Bcl-xL proteins but induced the expression of Bax protein. Zinc-citrate compound induced apoptosis of DU145 cells by activation of the caspase-3 pathway. Conclusion： Zinc-citrate compound can induce apoptotic cell death in DU145, by caspase-3 activation through up-regulation of apoptotic proteins and down-regulation of antiapoptotic proteins.

Biochar has a large specific surface area,well-developed pore structure,abundant surface functional groups,and superior nutrient supply capacity,which is widely available and environmentally friendly with its advantages in waste resource utilization,heavy metal(loid)remediation,and carbon *** review focuses on the interactions between biochar(including raw biochar,functional biochar(modified/engineered/designer biochar),and composite biochar)and rhizosphere during the remediation of soil contaminated with heavy metal(loid)s(Pb,As,Cd,Hg,Co,Cu,Ni,Zn,Cr,etc.)and the effects of these interactions on the microbial communities and root exudates(enzymes and low-molecular-weight organic acids(LMWOAs)).In terms of microorganisms,biochar affects the composition,diversity,and structure of microbial communities through the supply of nutrients,provision of microbial colonization sites,immobilization of heavy metal(loid)s,and introduction of exogenous *** regard to root exudates,biochar provides electron transfer support between the microorganisms and exudates,regulates the secretion of enzymes to resist the oxidative stress stimulated by heavy metal(loid)s,ameliorates rhizosphere acidification caused by LMWOAs,and promotes the activity of soil *** roles and mechanisms of biochar on rhizosphere soils are discussed,as well as the challenges of biochar in the remediation of heavy metal(loid)-contaminated soils,and the issues that need to be addressed in future research are foreseen.
