Micro RNA-320 family is downregulated in colorectal adenoma and affects tumor proliferation by targeting CDK6

AIM: To investigate the microR NA(miR NA) expression during histological progression from colorectal normal mucosa through adenoma to carcinoma within a lesion. METHODS: Using microarray, the sequential changes in miR NA expression profiles were compared in colonic lesions from matched samples; histologically, nonneoplastic mucosa, adenoma, and submucosal invasive carcinoma were microdissected from a tissue sample. Cell proliferation assay was performed to observe the effect of miR NA, and its target genes were predicted using bioinformatics approaches and the expression profile of SW480 transfected with the miR NA mimics. mR NA and protein levels of the target gene in colon cancer cell lines with a mimic control or miR NA mimics were measured using qR TPCR and Western blotting. The expression levels of miR NA and target gene in colorectal tissue samples were also ***: Microarray analysis identified that the miR-320 family, including miR-320 a, miR-320 b, miR-320 c, miR-320 d and miR-320 e, were differentially expressed in adenomaand submucosal invasive carcinoma. The mi R-320 family, which inhibits cell proliferation, is frequently downregulated in colorectal adenoma and submucosal invasive carcinoma tissues. Seven genes including CDK6 were identified to be common in the results of gene expression array and bioinformatics analyses performed to find the target gene of the miR-320 family. We confirmed that mR NA and protein levels of CDK6 were significantly suppressed in colon cancer cell lines with miR-320 family mimics. CDK6 expression was found to increase from non-neoplastic mucosa through adenoma to submucosal invasive carcinoma tissues and showed an inverse correlation with miR-320 family ***: MiR-320 family affects colorectal tumor proliferation by targeting CDK6, plays important role in its growth, and is considered to be a biomarker for its early detection.

Reliability of urinary tests for antibody to Helicobacterpylori in patients with pulmonary tuberculosis

AIM: Although the quality of currently available urinary tests for detecting antibody to Helicobacter pylori(H pylori) have been proved in some populations, the accuracy has not been studied regarding patients who suffer from pulmonary tuberculosis with multi-drug treatments. The present study was conducted to evaluate the accuracy of these urinary tests for antibody to H pylori in these patients. METHODS: Serum samples from 61 inpatients with pulmonary tuberculosis were tested using enzyme immunoassay, and urine samples were assayed by enzyme-linked immunosorbent assay method (URINELISA) and immunochromatography method (RAPIRAN). Medicines prescribed to the patients were recorded for medical charts, to evaluate the influences on the results of urinary tests. RESULTS: The sensitivity, specificity, and consistency of URINELJSA against the serum test were 93.1%, 65.6%, and 78.6% respectively, and those of RAPIRAN were 86.2%, 93.7%, and 90.1% respectively, which were almost equal to the data previously reported. Prescribed medicines had little influence on the results. CONCLUSION: The two urinary tests for detecting H pylori antibody have a diagnostic accuracy in patients with pulmonary tuberculosis given multiple anti-tuberculosis drugs.

Exploring the Lives of Mothers Raising Children with Autism Spectrum Disorder in Japan

Background: Examining the lives that mothers experience and build will allow us to deepen our understanding of children with ASD and their mothers and facilitate developing support methods. The study aimed to examine the lives of mothers raising children with autism spectrum disorder (ASD) and investigate their sources of support. Method: We conducted a qualitative inductive study using semi-structured interviews to identify characteristics of the lives that mothers have created. Results: Semi-structured interviews were conducted with 11 mothers having children with ASD. The analysis comprised three stages of coding and yielded eight categories. The lives of these mothers contained three themes: preoccupation with parenting children with ASD and their siblings;evolving mother;and using social resources. Mothers engaged in “assessing the characteristics, growth, and changes in the child with ASD”, had a “preoccupation with parenting children with ASD”, and were “thinking about the future of the child with ASD”, and “having goals and plans for parenting” while having “consideration toward the child’s siblings”. During this process, mothers experienced “changes in perspective or approach” and created lifestyles while “receiving help from people around them” and engaged in the “use of social resources”. Conclusions: To avoid becoming preoccupied with parenting and being burdened by their lifestyle, mothers require social support to monitor their perceptions. Furthermore, the utilization of social resources requires the supporting individuals to understand the characteristics of children with ASD, provide appropriate information, and assist in decision-making.

Subcutaneous Emphysema Caused by Sodium Hypochlorite plus RC-Prep

Introduction: “RC-Prep”, a chemo-mechanical preparation for root canals, includes EDTA and urea peroxide as ingredients. For lubrication and its chelating effect, RC-Prep is useful as an adjunct in root canal enlargement. Furthermore, its cleaning action, due to its foaming properties in combi-nation with sodium hypochlorite, can be useful in root canal treatment in many situations. Methods: In a case of upper gingiva swelling, we used sodium hypochlorite with RC-Prep in the treatment of an infected root canal of the left maxillary premolar at the first visit. Within 2 min after use, the patient’s face became swollen. We kept the root canal open, and conducted a CT scan to investigate. Results: We diagnosed subcutaneous emphysema and prescribed 1 week of pain killers and antibiotics. The pain and swelling in the face finally resolved at 11 days after the incident. Conclusions: The cause of the emphysema was considered to have been the use of a sodium hypo-chlorite solution after using RC-Prep for an opened root apex.

Molecular processes in iron and zinc homeostasis and their modulation for biofortification in rice

More than a billion people suffer from iron or zinc deficiencies globally. Rice(Oryza sativa L.) iron and zinc biofortification; i.e., intrinsic iron and zinc enrichment of rice grains, is considered the most effective way to tackle these deficiencies. However, rice iron biofortification, by means of conventional breeding, proves difficult due to lack of sufficient genetic variation. Meanwhile,genetic engineering has led to a significant increase in the iron concentration along with zinc concentration in rice grains. The design of impactful genetic engineering biofortification strategies relies upon vast scientific knowledge of precise functions of different genes involved in iron and zinc uptake, translocation and storage. In this review, we present an overview of molecular processes controlling iron and zinc homeostasis in rice. Further,the genetic engineering approaches adopted so far to increase the iron and zinc concentrations in polished rice grains are discussed in detail, highlighting the limitations and/or success of individual strategies. Recent insight suggests that a few genetic engineering strategies are commonly utilized for elevating iron and zinc concentrations in different genetic backgrounds, and thus, it is of great importance to accumulate scientific evidence for diverse genetic engineering strategies to expand the pool of options for biofortifying farmer-preferred cultivars.

Gas Permeation Properties of Organic-inorganic Ultrathin Membranes

Recently, a great interest has been noted in the synthesis of inorganic membranes for gas separation. One of the candidates is a carbon molecular sieve (CMS) membrane, which is synthesized by the pyrolysis of a polymer. However, CMS membranes are very brittle and fragile. Additionally, it requires more careful handling and it is very difficult to prepare a thin CMS membrane because of its poor mechanical property. Therefore, the gas permeances of CMS membranes were not very high.

Silicon-containing polymers have been intensively studied in the past half-century, and a great number of papers, patents and books have been published on the synthesis, properties and applications. Silicon-containing polymers, especially polymers containing silicon atoms in the backbone, including some major species as polysiloxane, polysilane, polysilazane, etc. have chains constructed of alternately arranged silicon and oxygen atoms with organic groups attached to the silicon atoms, which are the most important silicon-containing polymers so far.
