Flowering time is a crucial rice trait that influences its adaptation to various environments, cropping schedules, and agronomic characteristics. Rice breeders have exploited spontaneous mutations in heading date genes to regulate the flowering time. In the present study, we investigated how breeders in Fukui Prefecture regulated days to heading while developing promising rice varieties. Genome-wide association studies (GWAS) identified Hd1, Hd16, and Hd18 as the major genes controlling days to heading in the population. However, we suspected that this highly bred population might exhibit genomic stratification, which could lead to spurious or false correlations in the GWAS. Thus, we also conducted correlation and partial correlation analyses, which uncovered another key heading date gene, Hd17, that GWAS failed to detect because of its linkage disequilibrium with the major effect gene Hd16. Examination of haplotype frequencies across different breeding periods revealed that the early-heading Hd16 (Hd16(E)) and late-heading Hd17 (Hd17(L)) were increasingly co-selected in the Hd1 functional population. Varieties carrying this Hd16(E)/Hd17(L) combination exhibited days to heading in the range of 70-80, which corresponds to the peak temperature and sunshine period and is also optimal for grain quality and yield components in the Fukui environment. The present study highlights that it is imperative to remain vigilant for Type I (false positives) and Type II (false negatives) errors when performing GWAS on highly bred populations and to implement appropriate countermeasures by accounting for gene-by-gene interactions established through the breeding process. We also discuss the effectiveness of Hd16(E), which is not used outside Japan for subtle days to heading control but is widely used in Japan at certain latitudes.

In this study, we tried to improve the performance by giving a pre-swirling flow to the radial inflow that occurred in the semi-opened axial fan. In addition, the flow fields of rotor outlet were clarified experimentally, and the effect of pre-swirling flow was considered. The experiment was carried out using a performance test wind tunnel with a square cross section of 880 mm. Three types of casings were prepared, in which the blade tip protruded 0%, 20%, and 40% of the meridional chord length. They were called R25, R15, and R05, respectively, in the casing bellmouth model code. Guide blades for generating a pre-swirling flow were installed on the vertical wall surface of the casing. In addition, a vertical wall was installed 60% upstream of the meridional chord length as an obstacle to prevent axial inflow. The velocity fields of the rotor outlet were measured using a hot-wire anemometer. From the results, the pre-swirling flow did not significantly affect the fan performance. When there was no obstacles wall upstream, there was a partial increase in efficiency, but the difference was not so large. When there was an obstacle wall upstream, the efficiency increased overall in the case of R15, but in the case of R05, the efficiency increased only in the low flow rate region, and conversely decreased in the high flow rate region. By observing the blade outlet flow fields when the performance was improved, it was confirmed that the influence of the tip leakage vortex was weakened.

WISE Infrared Search for Young Stellar Objects Associated with Starless Cores

This study presents the results of an infrared search of young stellar objects (YSOs) associated with cores with high optical extinction and no associated infrared IRAS source. Four hundred YSO candidates were identified in the WISE photometric catalog based on the infrared excess attributed to the circumstellar materials and proto-planetary disks. One-hundred and forty-nine cores do not have YSO candidates. Whereas, 32 cores harbor only Class I candidates and 107 cores have Class II candidates. Ninety-one cores that were previously identified as starless cores, were found to contain YSOs. The ratio of the number of starless cores to the number of star-forming cores suggests that the typical timescale from molecular cloud core formation to the birth of a star is in the range of 0.5 - 1.4 Myr.

AIM To investigate the possibility of diagnosing gastric cancer from an unstained pathological tissue using Raman spectroscopy, and to compare the findings to those obtained with conventional histopathology. METHODS We produced two consecutive tissue specimens from areas with and without cancer lesions in the surgically resected stomach of a patient with gastric cancer. One of the two tissue specimens was stained with hematoxylin and eosin and used as a reference for laser irradiation positioning by the spectroscopic method. The other specimen was left unstained and used for Raman spectroscopy analysis. RESULTS A significant Raman scattering spectrum could be obtained at all measurement points. Raman scattering spectrum intensities of 725 cm^(-1) and 782 cm^(-1), are associated with the nucleotides adenine and cytosine, respectively. The Raman scattering spectrum intensity ratios of 782 cm^(-1)/620 cm^(-1), 782 cm^(-1)/756 cm^(-1), 782 cm^(-1)/1250 cm^(-1), and 782 cm^(-1)/1263 cm^(-1) in the gastric adenocarcinoma tissue were significantly higher than those in the normal stomach *** The results of this preliminary experiment suggest the feasibility of our spectroscopic method as a diagnostic tool for gastric cancer using unstained pathological specimens.

AIM: To evaluate the neuroprotective effect of a dietary supplement(ClearVision EX~?;CV) against glutamateinduced excitotoxicity in ***: We evaluated the protective effects CV on glutamate-induced cell toxicity of an immortalized mouse hippocampal cell line(HT-22) in vitro and N-methyl-Daspartate(NMDA) induced retinal injury in vivo. Once-daily oral administration of CV or vehicle(5% Arabic gum) was started the day before the NMDA injection and continued until the end of the study. Electroretinograms(ERGs) were recorded to evaluate the retinal function at 2 d after NMDA injection. Furthermore, a histological evaluation, Western blot analysis, and immunohistochemistry were performed for assessing the signal transduction pathway. RESULTS: HT-22 cell death was induced by the addition of glutamate and co-incubation with CV protected against it. Oral administration of CV inhibited the decrease in scotopic threshold response amplitudes induced by the intravitreal injection of NMDA and those of the thickness of the inner retinal layer in the histological evaluation. The increased phosphorylated levels of extracellular signalregulated kinase(ERK) but not cAMP response element binding protein(CREB) or Akt were observed 1 h after NMDA injection in both the vehicle-and CV-treated rats;however, p ERK activation was no more upregulated at 3 h after NMDA injection. pERK upregulation was observed in Müller ***: CV shows a protective effect against both glutamate-induced HT-22 cell death and NMDAinduced retinal damage. pERK upregulation in the Müller cells plays a key role in the protective effect of CV against glutamate-induced retinal toxicity.

In this review, we focused on the relationship between central blood pressure and chronic kidney diseases(CKD). Wave reflection is a major mechanism that determines central blood pressure in patients with CKD. Recent medical technology advances have enabled non-invasive central blood pressure measurements. Clinical trials have demonstrated that compared with brachial blood pressure, central blood pressure is a stronger risk factor for cardiovascular(CV) and renal diseases. CKD is characterized by a diminished renal autoregulatory ability, an augmented direct transmission of systemic blood pressure to glomeruli, and an increase in proteinuria. Any elevation in central blood pressure accelerates CKD progression. In the kidney, interstitial inflammation induces oxidative stress to handle proteinuria. Oxidative stress facilitates atherogenesis, increases arterial stiffness and central blood pressure, and worsens the CV prognosis in patients with CKD. A vicious cycle exists between CKD and central blood pressure. To stop this cycle, vasodilator antihypertensive drugs and statins can reduce central blood pressure and oxidative stress. Even in early-stage CKD, mineral and bone disorders(MBD) may develop. MBD promotes oxidative stress, arteriosclerosis, and elevated central blood pressure in patients with CKD. Early intervention or prevention seems necessary to maintain vascular health in patients with CKD.

Evaluation of Blood Pressure Control Medicines Using Health and Medical Checkup Data in Japan: Alternative Methods for Randomized Controlled Trials

Background: Many people take medicines to control high blood pressure (BP), or hypertension. Randomized clinical trials (RCT) are usually used for the evaluation of effects of medicines. However, RCT have some serious problems. Data and Methods: We evaluated the effects of BP medicines in Japan using a dataset containing 113,979 cases. We employed four statistical methods in the analysis. First, we simply compared the systolic blood pressure (SBP) of individuals with and without BP medicines. We then used a regression model with a dummy variable, representing taking medicines or not. We replaced the dummy variable by its expected value, and estimated the regression model again. Finally, we selected individuals who had both taken and not taken medicines at different times. The effect of sample selection was also considered in the estimation. Results: For the simple comparison, SBP with BP medicines was 11 mmHg higher than without medicines. In the next regression analysis, SBP with BP medicines was still 5 mmHg higher. When the dummy variable was replaced by its expected value, SBP with medicines decreased by 7 mmHg. For individuals taking medicines at some times and not at others, SBP decreased by 9 and 8 mmHg in models with and without a sample bias correction, respectively. Conclusion: The methods eliminated some problems of RCT and might be attractive. However, we obtained contradictory conclusions depending on the statistical methods employed, despite using the identical dataset. Statistical methods must be selected carefully to obtain a reliable evaluation. Limitations: The dataset was observatory, and the sample period was only 3 years.

Prorenin receptor （PRR） is a multi-functioning protein possessing at least four different roles：（1） working as a receptor for renin and prorenin producing angiotensin I from angiotensinogen thus enhancing the tissue renin-angiotensin system; （2） inducing intracellular signals when a ligand binds to PRR; （3） participating in the functions of vacuolar proton ATPase; and （4） constitut-ing the Wnt signaling receptor complex. Here, the roles of PRR in kidney physiology and diabetic conditions as well as recent fndings regarding a soluble form of PRR are discussed. We also propose the possible mechanism concerning diabetic nephropathy as “trade-off hypothe-sis” from a PRR point of view. In brief, under hypergly-cemic conditions, injured podocytes degrade degener-ated proteins and intracellular organelles which require V-ATPase and PRR for vesicle internal acidification. Sustained hyperglycemia overproduces PRR molecules, which are transported to the transmembrane and bind to increased serum prorenin in the diabetic condition. This enhances tissue renin-angiotensin system and PRR-mediated mitogen-activated protein kinase signals, resulting in increased injurious molecules such as transforming growth factor-β, cyclooxygenase2, interleukin1β, and tumor necrosis factor-α ending in diabetic ne-phropathy progression. Although many fndings led us to better PRR understanding, future works should elu-cidate which PRR functions, of the four discussed here, are dominant in each cell and kidney disease context.
