CD200Rhigghh neutrophils with dysfunctional autophagy establish systemic immunosuppression by increasing regulatory T cells

Distinct neutrophil populations arise during certain pathological conditions.The generation of dysfunctional neutrophils during sepsis and their contribution to septicemia-related systemic immune suppression remain unclear.In this study,using an experimental sepsis model that features immunosuppression,we identified a novel population of pathogenic CD200R^(high) neutrophils that are generated during the initial stages of sepsis and contribute to systemic immune suppression by enhancing regulatory T(T_(reg))cells.Compared to their CD200R^(low)counterparts,sepsis-generated CD200Rhigh neutrophils exhibit impaired autophagy and dysfunction,with reduced chemotactic migration,superoxide anion production,and TNF-αproduction.Increased soluble CD200 blocks autophagy and neutrophil maturation in the bone marrow during experimental sepsis,and recombinant CD200 treatment in vitro can induce neutrophil dysfunction similar to that observed in CD200R^(high) neutrophils.The administration of anα-CD200R antibody effectively reversed neutrophil dysfunction by enhancing autophagy and protecting against a secondary infection challenge,leading to increased survival.Transcriptome analysis revealed that CD200R^(high) neutrophils expressed high levels of Igf1,which elicits the generation of Treg cells,while the administration of anα-CD200R antibody inhibited Treg cell generation in a secondary infection model.Taken together,our findings revealed a novel CD200R^(high) neutrophil population that mediates the pathogenesis of sepsis-induced systemic immunosuppression by generating Treg cells.

Aim:Serum amyloid A(SAA)is an important mammalian acute reactant.Here,weaim to investigate the effect of SAA on apoptosis and its mechanism of action inhuman amniotic WISH cells.Methods:The expression of formyl peptide receptor(FPRL1),which is reported as a SAA receptor,was tested using RT-PCR andligand binding assay with radio-labeled FPRL1 ligand.The effect of SAA onproliferating cell population was evaluated by thymidine incorporation assay.The protein phosphorylation levels and caspase-3 activity were detected by West-ern blot assay.Results:SAA inhibits thymidine incorporation in human amnioticWISH cells.A SAA-induced decrease of proliferating cell population was accom-panied with nuclear condensation and caspase-3 activation in WISH cells,sug-gesting that SAA induces WISH cell apoptosis.Since FPRL1 has been reportedas a SAA receptor,we investigated the effects of several FRPL1 agonists on aproliferating cell population in WISH cells.Among the tested FPRL1 agonists,only SAA induced a decrease of proliferating cell population in WISH cells.Onthe downstream signaling of SAA,we found that SAA stimulated extracellularsignal-regulated kinase and p38 kinase,which were not inhibited by pertussistoxin(PTX),ruling out the role of PTX-sensitive G-proteins.Furthermore a SAA-induced decrease of proliferating cell population was not affected by PTX,sug-gesting that SAA inhibits WISH cell apoptosis in a PTX-sensitive G-protein-independent manner.A SAA-induced decrease of a proliferating cell populationwas completely blocked by PD98059 and SB203580,suggesting that mitogen-activated protein kinase activities are essentially required for the process.Conclusion:SAA is a novel inducer for WISH cell apoptosis,and the PTX-insen-sitive pathway is involved in the process.

Aim:To investigate the effects of sphingosylphosphorylcholine(SPC)on humanmonocyte-derived dendritic cell(DC)chemotaxis.Methods:Human DC were gen-erated from peripheral blood monocytes by culturing them with granulocyte mac-rophage-colony stimulating factor and interleukin-4.The effect of SPC on the DCchemotactic migration was measured by chemotaxis assay.Intracellular signalingevent involved in the SPC-induced DC chemotaxis was investigated with severalinhibitors for specific kinase.The expression of the SPC receptors was examinedby reverse transcription polymerase chain reaction.Results:We found that SPCinduced chemotactic migration in immature DC(iDC)and mature DC(mDC).Interms of SPC-induced signaling events,mitogen activated protein kinase activa-tion and Akt activation in iDC and mDC were stimulated.SPC-induced chemotaxiswas mediated by extracellular signal-regulated protein kinase and phosphoino-sitide-3-kinase,but not by calcium in both iDC and mDC.Although mDC expressovarian cancer G protein-coupled receptor 1,but not G protein-coupled receptor 4,iDC do not express any of these receptors.To examine the involvement of sphin-gosine-1-phosphate(S1P)receptors,we checked the effect of an S1P receptorantagonist(VPC23019)on SPC-induced DC chemotaxis.VPC23019 did not affectSPC-induced DC chemotaxis.Conclusion:The results suggest that SPC may playa role in regulating DC trafficking during phagocytosis and the T cell-stimulatingphase,and the unique SPC receptor,which is different from SIP receptors,isinvolved in SPC-induced chemotaxis.

Measurement of alumina film induced ablation of internal insulator in solid rocket environment

This study investigates the ablation of internal insulation induced by the deposition of an alumina film with different lateral film speeds.A sub-scale test solid rocket motor(SRM)was designed in an impinging jet configuration to form an alumina film on the sample and to encourage the lateral movement of the film by a high-speed wall jet.Fifteen static fire tests of the test SRM were conducted with six different jet velocities(V_(jet)=100 m/s,150 m/s,200 m/s,268 m/s,330 m/s,and 450 m/s)that indirectly affected the velocity of the wall jet and the deposition rate of alumina droplets.The ablation velocity was deduced from the difference in the sample thickness after a test using a coordinate measuring machine.The droplet deposition mass flux and wall jet velocity were obtained via two-phase flow simulation with the same jet velocity and effective pressure.As a result,the characteristics of alumina-induced ablation and the changes in ablation with jet velocities were obtained.The area within0.8×jet diameter was focused upon,where the ratio of ablation velocity to incoming alumina mass was constant for each jet velocity,and showed a similarity in jet structure.When the ablation velocity was increased from 2.05 to 9.98 mm/s with increasing jet velocity,the ratio of the ablation velocity and alumina mass flux decreased from 1.07×10^(-4)to 0.49×10^(-4)m^(3)/kg as Al_(2)O_(3)-C reactions became less efficient with a reduced residence time of the film.Because the decrease in residence time by the wall jet is more pronounced for slow reactions involved in Al_(2)O_(3)-C reactions,fast reactions in Al_(2)O_(3)-C reactions are less affected and result in a convergence of the volumetric rate of ablation per unit mass of alumina.

Quantitative analysis of mechanical properties associated with aging treatment and microstructure in Mg-Al-Zn alloys through machine learning

The present study proposes a methodology for predicting the mechanical properties of AZ61 and AZ91alloys associated with microstructure,texture and aging parameters and estimating predictor importance.For this,we investigate quantitative correlations between microstructure,texture and mechanical properties of aged AZ61 and AZ91 rods through machine learning.This regression analysis focuses on the precipitation behavior of Mg17Al12as the main second phase of Mg-Al-Zn alloys with respect to aging conditions.To simplify data generation,only SEM images were used to quantify the features of discontinuous and continuous precipitates.To overcome the lack of data and make the most of the measured data,we devised a method to extend the existing dataset by a factor of 9 using the mean and standard deviation of the measured data.Artificial neural networks predicted tensile and compressive yield strengths and resultant yield asymmetry with a high accuracy of over 98%using 11 predictors for a total of 288datasets.Decision tree learning quantitatively assessed the importance of predictors in determining the mechanical properties of aged AZ61 and AZ91 rods.

Paleo-shoreline changes in moraine dammed lake Khagiin Khar, Khentey Mountains, Central Mongolia

The formation and evolution of glacier moraine-dammed lakes are closely related to past glacier expansion and retreat. Geomorphic markers such as lacustrine terraces and beach ridges observed in these lakes provide important evidence for regional paleoenvironment reconstruction. We document the magnitude of paleo-shoreline fluctuations and timings of highstands of lake water by using cosmogenic 10Be surface exposure dating and optically stimulated luminescence(OSL) dating on samples collected from lacustrine sediment and bedrock strath in Lake Khagiin Khar. The lake was initially impounded by glacier moraine at the Global Last Glacial maximum(gLGM;21–19 ka), and the lake reached its maximum paleo-shoreline level of 1840 m at sea level(a.s.l.). At that time, the stored lake water amount was up to seven times greater and the surface area was three times larger than the present values. The paleolake experienced higher shoreline levels at 1832, 1822, and 1817 m a.s.l. and reached the present lake level after 0.4 ka. We interpret that decrease in the paleolake level was caused by spillover. The increase in melt water after the gLGM and the Late Glacial exceeded the storage threshold of the lake, and the paleolake water overflowed across the lowest drainage divides. The lake spilled over across the lowest bedrock ridge at 15.9 ± 0.6 ka, and the outlet was incised since that time at a rate of 3.72 ± 0.15 mm/yr. The initial stream of the Khiidiin Pass River was disturbed by LGM moraine damming and was rerouted into the present course running through moraine after the spillover at 15.9 ± 0.6 ka.

Synthesis and characterization of Ba(Co_xNb_(1-x))O_3 nanoparticles by hydrothermal processing

The nanosized Ba(CoxNb1-x)O3(BCN) particles were prepared under high temperature and pressure conditions by precipitation from metal nitrates with aqueous potassium hydroxide. Ba(CoxNb1-x)O3 powders were obtained in the temperature range of 170-210 ℃ for 6 h. The results show that the average size of the synthesized particles increases with increasing reaction temperature. The average size of the synthesized particles is about 10 nm. The crystalline phase of the synthesized particles is found to be Ba(CoxNb1-x)O3. Ceramics derived from the nano BCN powders could achieve high sintering density at a relatively low sintering temperature.

Synthesis and characterization of metallic Pd embedded TiO_2 nanoparticles by reverse micelle and sol-gel processing

Metallic Pd clusters were embedded into TiO2 nanoparticles that were synthesized within reverse micelle via a solution reduction of Pd(NO3)2 by hydrazine hydrate. The size of the particles can be controlled by manipulating the relative rates of the hydrolysis and condensation reactions of titanium tetra-isopropoxide within the micro-emulsion. The spherical equivalent size for the Pd clusters formed in TiO2 matrix was estimated to be around (3±1) nm. The presence of Pd in the matrix was demonstrated by EDS spectroscopy. The effects of synthesis parameters, such as the concentration of Pd solution, the molar ratio of water to TIP, and the molar ratio of water to surfactant, were discussed.
