The integrated repair of bone and cartilage boasts advantages for osteochondral restoration such as a long-term repair effect and less deterioration compared to repairing cartilage alone.Constructing multifactorial,spatially oriented scaffolds to stimulate osteochondral regeneration,has immense significance.Herein,targeted drugs,namely kartogenin@polydopamine(KGN@PDA)nanoparticles for cartilage repair and miRNA@calcium phosphate(miRNA@CaP)NPs for bone regeneration,were in situ deposited on a patterned supramolecular-assembled 2-ureido-4[lH]-pyrimidinone(UPy)modified gelation hydrogel film,facilitated by the dynamic and responsive coordination and complexation of metal ions and their ligands.This hydrogel film can be rolled into a cylindrical plug,mimicking the Haversian canal structure of natural bone.The resultant hydrogel demonstrates stable mechanical properties,a self-healing ability,a high capability for reactive oxygen species capture,and controlled release of KGN and miR-26a.In vitro,KGN@PDA and miRNA@CaP promote chondrogenic and osteogenic differentiation of mesenchymal stem cells via the JNK/RUNX1 and GSK-3β/β-catenin pathways,respectively.In vivo,the osteochondral plug exhibits optimal subchondral bone and cartilage regeneration,evidenced by a significant increase in glycosaminoglycan and collagen accumulation in specific zones,along with the successful integration of neocartilage with subchondral bone.This biomaterial delivery approach represents a significant toward improved osteochondral repair.

An Emergency Shelter Location Model Based on the Sense of Security and the Reliability Level

Based on the value function of the prospect theory,this paper constructs a security function,which is used to describe the victims’feelings about the distance in emergency evacuation.Since different paths between the demand points and the emergency shelters are generally of different importance degrees,they are divided into main paths and auxiliary paths.The security function values and the reliability levels of main paths and auxiliary paths are given different weights.The weighted sum of the security function values and the weighted sum of the reliability level function values of all demand points are maximized to determine the location and the number of the emergency shelters,the transfer paths,the reinforced edges and the incremental reliability level of the selected edge.In order to solve the model,a two-stage simulated annealing-particle swarm optimization algorithm is proposed.In this algorithm,the particle swarm optimization(PSO)algorithm is embedded into the simulated annealing(SA)algorithm.The cumulative probability operator and the cost probability operator are formed to determine the evolution of the particles.Considering the budget constraint,the algorithm eliminates the shelter combinations that do not meet the constraint,which greatly saves the calculation time and improves the efficiency.The proposed algorithm is applied to a case,which verifies its feasibility and stability.The model and the algorithm of this paper provide a basis for emergency management departments to make the earthquake emergency planning.

Bioinspired Multifunctional Self-Sensing Actuated Gradient Hydrogel for Soft-Hard Robot Remote Interaction

The development of bioinspired gradient hydrogels with self-sensing actuated capabilities for remote interaction with soft-hard robots remains a challenging endeavor. Here, we propose a novel multifunctional self-sensing actuated gradient hydrogel that combines ultrafast actuation and high sensitivity for remote interaction with robotic hand. The gradient network structure, achieved through a wettability difference method involving the rapid precipitation of MoO_(2) nanosheets, introduces hydrophilic disparities between two sides within hydrogel. This distinctive approach bestows the hydrogel with ultrafast thermo-responsive actuation(21° s^(-1)) and enhanced photothermal efficiency(increase by 3.7 ℃ s^(-1) under 808 nm near-infrared). Moreover, the local cross-linking of sodium alginate with Ca^(2+) endows the hydrogel with programmable deformability and information display capabilities. Additionally, the hydrogel exhibits high sensitivity(gauge factor 3.94 within a wide strain range of 600%), fast response times(140 ms) and good cycling stability. Leveraging these exceptional properties, we incorporate the hydrogel into various soft actuators, including soft gripper, artificial iris, and bioinspired jellyfish, as well as wearable electronics capable of precise human motion and physiological signal detection. Furthermore, through the synergistic combination of remarkable actuation and sensitivity, we realize a self-sensing touch bioinspired tongue. Notably, by employing quantitative analysis of actuation-sensing, we realize remote interaction between soft-hard robot via the Internet of Things. The multifunctional self-sensing actuated gradient hydrogel presented in this study provides a new insight for advanced somatosensory materials, self-feedback intelligent soft robots and human–machine interactions.

La_(2)O_(3)-modified BiYbO_(3)–Pb(Zr,Ti)O_(3) ternary piezoelectric ceramics with enhanced electrical properties and thermal depolarization temperature

High-performance Pb(Zr_(1−x)Ti_(x))O_(3)(PZT)piezoceramics are urgently desired by the market in view of their expanded operating temperature range,reduced property temperature dependence,and enhanced sensitivity and acoustic power.In this work,we reported a kind of low-cost and high-performance 0.06BiYbO_(3)–0.94Pb(Zr_(0.48)Ti_(0.52))O_(3) ternary piezoceramics;the modifying effects of La_(2)O_(3) on this perovskite system were investigated in terms of the structures,electrical properties,and thermal depolarization behaviors of ceramics.The field-dependent dielectric and conduction properties indicated that there are close correlations among oxygen vacancies(VO),conducting electrons,and intrinsic conduction process.The degradation in ferroelectric properties observed in those samples doped with more than 0.15 wt%of La_(2)O_(3) indicated that the occupying mechanisms of La^(3+)changed from the donor substitution for Pb^(2+)to the isovalent substitution for Bi^(3+).The thermally depoling micromechanisms of ceramics were revealed from the thermodynamic processes of defect dipoles and intrinsic dipoles within ferroelectric domains.The sample doped with 0.15 wt%of La_(2)O_(3) shows excellent electrical properties with TC=387℃,d33=332 pC/N,TKε=5.81×10^(−3)℃−1,Pr=20.66μC/cm^(2),Td=356℃.The significantly enhanced electrical properties and thermal depolarization temperature benefited from the donor substitution of La3+,decreasing the oxygen vacancy concentration in the lattice and possibly optimizing the ferroelectric domain structure of ceramics.

miR-125b reverses cisplatin resistance by regulating autophagy via targeting RORA/BNIP3L axis in lung adenocarcinoma

The platinum-based chemotherapy is one of the most frequently used treatment protocols for lung adenocarcinoma(LUAD),and chemoresistance,however,usually results in treatment failure and limits its application in the clinic.It has been shown that microRNAs(miRNAs)play a significant role in tumor chemoresistance.In this study,miR-125b was identified as a specific cisplatin(DDP)-resistant gene in LUAD,as indicated by the bioinformatics analysis and the real-time quantitative PCR assay.The decreased serum level of miR-125b in LUAD patients was correlated with the poor treatment response rate and short survival time.MiR-125b decreased the A549/DDP proliferation,and the multiple drug resistance-and autophagy-related protein expression levels,which were all reversed by the inhibition of miR-125b.In addition,xenografts of human tumors in nude mice were suppressed by miR-125b,demonstrating that through autophagy regulation,miR-125b could reverse the DDP resistance in LUAD cells,both in vitro and in vivo.Further mechanistic studies indicated that miR-125b directly repressed the expression levels of RORA and its downstream BNIP3L,which in turn inhibited autophagy and reversed chemoresistance.Based on these findings,miR-125b in combination with DDP might be an effective treatment option to overcome DDP resistance in LUAD.

单细胞水平解析早期糖尿病肾病免疫细胞间的相互作用

为探究影响小鼠糖尿病肾病(diabetic kidney disease,DKD)进展的免疫细胞及细胞间相互作用,本研究应用CellChat软件对单细胞RNA测序(single-cell RNA-sequencing,scRNA-Seq)数据进行分析。预测在早期DKD肾脏免疫细胞中,作为CCL、MIF和CXCL三个主要的信息流之一的MIF信号显著降低,主要表现在Mif及其受体基因Cd44、Cxcr4和Cxcr2的表达降低;同时,F13a1^(+)巨噬细胞不再响应MIF信号。早期DKD肾脏中,F13a1^(+)巨噬细胞是细胞间相互作用数量和强度变化最大的免疫细胞。F13a1^(+)巨噬细胞信号配体表达的改变导致细胞间信号ANGPTL、IL2和IL16消失以及KIT和PROS信号出现。值得注意的是,在早期DKD肾脏中,F13a1^(+)巨噬细胞显著降低了与免疫细胞活化相关的转录因子基因Etv1(ETS variant transcription factor 1)的表达。采用基因集富集功能分析(gene set enrichment analysis,GSEA)法对差异表达基因(differentially expressed genes,DEGs)的功能研究发现,早期DKD肾脏中F13a1^(+)巨噬细胞的趋化因子媒介的信号通路、免疫反应和髓系白细胞媒介的免疫等生物过程下调;而免疫反应的激活、吞噬作用和造血祖细胞分化等生物过程上调。应用饮食诱导的糖尿病小鼠肾脏验证发现,除KIT信号外,以上结果均有良好复现性。本研究揭示F13a1^(+)巨噬细胞可能通过降低Etv1的表达调节其免疫相关活性,并通过改变MIF信号以及TSLP-(IL7R^(+)CRLF2)、IL16-CD4、ANGPTL4-SDC3/4和PROS1-AXL配体-受体的相互作用调控F13a1^(+)巨噬细胞与免疫细胞间的通信,从而在小鼠DKD的发生发展中发挥关键作用。

Low-fat microwaved peanut snacks production: Effect of defatting treatment on structural characteristics, texture, color, and nutrition

This study develops low-fat microwaved peanut snacks(LMPS) using partially defatted peanuts(PDP) with different defatting ratios, catering to people’s pursuit of healthy, low-fat cuisine. The effects of defatting treatment on the structural characteristics, texture, color, and nutrient composition of LMPS were comprehensively explored. The structural characteristics of LMPS were characterized using X-ray micro-computed tomography(Micro-CT) and scanning electron microscope(SEM). The results demonstrated that the porosity, pore number, pore volume,brightness, brittleness, protein content, and total sugar content of LMPS all significantly increased(P<0.05) with the increase in the defatting ratio. At the micro level, porous structure, cell wall rupture, and loss of intracellular material could be observed in LMPS after defatting treatments. LMPS made from PDP with a defatting ratio of 64.44% had the highest internal pore structural parameters(porosity 59%, pore number 85.3×105, pore volume 68.23 mm3), the brightest color(L* 78.39±0.39), the best brittleness(3.64±0.21) mm–1), and the best nutrition(high protein content,(34.02±0.38)%; high total sugar content,(17.45±0.59)%; low-fat content,(27.58±0.85)%). The study provides a theoretical basis for the quality improvement of LMPS.

A knowledge graph(KG)is a knowledge base that integrates and represents data based on a graph-structured data model or topology.Geoscientists have made efforts to construct geosciencerelated KGs to overcome semantic heterogeneity and facilitate knowledge representation,data integration,and text analysis.However,there is currently no comprehensive paleontology KG or data-driven discovery based on it.In this study,we constructed a two-layer model to represent the ordinal hierarchical structure of the paleontology KG following a top-down construction process.An ontology containing 19365 concepts has been defined up to 2023.On this basis,we derived the synonymy list based on the paleontology KG and designed corresponding online functions in the OneStratigraphy database to showcase the use of the KG in paleontological research.

人口老龄化背景下体育消费的健康效应——基于全国消费大数据的实证证据

基于健康资本需求理论将体育消费视作一种健康投资,提出个体体育消费的增长可降低其医疗消费支出的假设;采用2018—2021年全国消费大数据,实证检验个体体育消费的健康效应。结果显示,体育消费占个体年度总消费的比例每提高1个单位,医疗消费占个体年度总消费的比例降低0.203个单位。新冠病毒感染疫情对体育消费的健康效应有所抑制但依然显著。进一步分析表明,体育消费的健康效应对35岁及以上人群的影响会随着年龄的增长明显增强,对65岁及以上老龄人口的影响效果最为突出。认为:在应对人口老龄化和医疗负担增加等重大挑战时,需要加快普及对体育消费的健康效应认知,大力促进全人群体育消费。

Effect of the electro-acupuncture on senile plaques and its formation in APPD/PS1D double transgenic mice

Alzheimer’s disease(AD)is a progressive neurodegenerative disease and its incidence will increase with age and is aggravating.The senile plaques(SPs)are one of three main pathological features in AD patients,which are formed by amyloid b-protein(Ab)overaccumulation.b-amyloid precursor protein(APP),b-site APP cleavage(BACE1),and insulin degrading enzyme(IDE)proteins participate in the process of Ab production and degradation.At present,the pathogenesis of AD is not yet clear and the current treatment methods can only relief the related symptoms of AD.The electro-acupuncture(EA)is a traditional Chinese medicine treatment combined the acupuncture and electrical stimulation and the treatment effect can also be controlled by transform the electrical frequency.Thus,in this experiment,we carried out behavioral test,immunohistochemistry(IHC),and Western Blot(WB)after different period treatments to the model mice by electro-acupuncturing“Baihui”and“Shenshu”acupoints in APPt/PS1t double transgenic mice.It was found that the EA therapy can improve the ability of learning,memory and spatial exploration,and reduce the deposition of SPs in brain of AD model mice,and reduce the expressions of APP and BACE1,increase the expression of IDE protein.These results prompt that EA can effectively alleviate the pathological process of AD.We speculate that EA may play a comprehensive role in preventing the development of AD,considering the previous data.
