Low-Rank Optimal Transport for Robust Domain Adaptation

When encountering the distribution shift between the source(training) and target(test) domains, domain adaptation attempts to adjust the classifiers to be capable of dealing with different domains. Previous domain adaptation research has achieved a lot of success both in theory and practice under the assumption that all the examples in the source domain are welllabeled and of high quality. However, the methods consistently lose robustness in noisy settings where data from the source domain have corrupted labels or features which is common in reality. Therefore, robust domain adaptation has been introduced to deal with such problems. In this paper, we attempt to solve two interrelated problems with robust domain adaptation:distribution shift across domains and sample noises of the source domain. To disentangle these challenges, an optimal transport approach with low-rank constraints is applied to guide the domain adaptation model training process to avoid noisy information influence. For the domain shift problem, the optimal transport mechanism can learn the joint data representations between the source and target domains using a measurement of discrepancy and preserve the discriminative information. The rank constraint on the transport matrix can help recover the corrupted subspace structures and eliminate the noise to some extent when dealing with corrupted source data. The solution to this relaxed and regularized optimal transport framework is a convex optimization problem that can be solved using the Augmented Lagrange Multiplier method, whose convergence can be mathematically proved. The effectiveness of the proposed method is evaluated through extensive experiments on both synthetic and real-world datasets.

Reactive oxidative species(ROos)production-driven ferroptosis plays a role in acute kidney injury(Akl).However,its exact molecular mechanism is poorly understood.Scavenger receptor CD36 has important roles in oxidizing lipids,lipid accumulation,metabolic syndrome,and insulin resistance in chronic kidney disease,but its roles remain unexplored in AKl.The present study investigated the role and mechanism of CD36 in regulating proximal tubular cell ferroptosis and AKl.The expression of CD36 was found to be significantly up-regulated in AKI renal tissues and correlated with renal function,which might serve as an independent biomarker for AKl patients.Moreover,in adult mice subjected to AKl,deletion of CD36(CD36-/-)induced tubular cell Ros accumulation,ferroptosis activation,and renal injury.Mechanistically,combining LC-MS/MS,co-IP,and ubiquitination analyses revealed that CD36 could specifically bind to ferroptosis suppressor protein 1(FSP1)and regulate its ubiquitination at sites K16 and K24,leading to FSP1 degradation and progression of ferroptosis in AKl.The present results emphasize a novel mechanism of CD36 in cisplatin-induced AKl.The discovery of the special CD36 roles in promoting ferroptosis and AKI development by regulating the ubiquitination of FSP1 in proximal tubular cells may be potential therapeutic targets for AKl.Moreover,CD36 may play a key role in the progression of AKl.Therefore,targeting CD36 may provide a promising treatment option for AKI.

Various molecular analytical techniques have provided more and more information which is necessary for the accurate classification of diseases.Such information has enabled researchers to shift from morphology-based classification to molecular characteristics-based classification,also known as molecular classification.Molecular classifications of diseases can be carried out at the DNA,RNA,and protein levels.At the DNA level,for example,molecular classification can be done based on genetic mutation,polymorphisms,cytogenetic changes in genomes or DNA methylation differences.

颗粒增强粘接层结构参数对连接强度的影响及工艺优化

基于Box-Behnken design(BBD)响应面法,开展颗粒增强粘接试验研究,构建粘接层结构参数(粘接层厚度、颗粒粒径和颗粒体积分数)与失效载荷和能量吸收值之间的多元回归模型,辨析粘接层结构参数对接头力学性能的影响规律;同时,结合多目标遗传算法,开展粘接工艺的多目标优化,并进行试验验证。结果表明:单因素分析中,颗粒体积分数对接头强度的影响最大;交互作用分析中,粘接层厚度和颗粒体积分数对接头连接强度的影响最为显著。通过Pareto解集确定的最优粘接层结构参数组合为粘接层厚度0.531 mm、颗粒粒径61.765μm、颗粒体积分数4.099%,遗传算法的预测值与试验值之间的误差分别为5.7%(失效载荷)和0.9%(能量吸收值)。

Laboratory research and pharmacoepidemiology provide support for metformin as a potential antitumor agent.However,the lack of a clear understanding of the indications of metformin limits its efficacy.Here,we performed a genome-wide CRISPR knockout negative screen to identify potential targets that might synergize with metformin.Next-generation sequencing of pooled genomic DNAs isolated from surviving cells after 18 days of metformin treatment(T18)compared to those of the untreated cells at day 0(T0)yielded candidate genes.Knockdown of a group of cyclin-dependent kinases(CDKs),including CDK1,CDK4,and CDK6,confirmed the results of the screen.Combination treatment of the CDKs inhibitor abemaciclib with metformin profoundly inhibited tumor viability in vitro and in vivo.Although cell cycle parameters were not further altered under the combination treatment,investigation of the metabolome revealed significant changes in cell metabolism,especially with regard to fatty acid oxidation,the tricarboxylic acid cycle and aspartate metabolism.Such changes appeared to be mediated through inhibition of the mTOR pathway.Collectively,our study suggests that the combination of CDKs inhibitor with metformin could be recognized as a potential therapy in future clinical applications.

基于火焰稳定在回流区剪切层中的假设,建立了预混火焰近贫燃熄火极限特性的理论分析模型.一旦火焰稳定在剪切层中,由于剪切层同时从两侧卷吸流体,所以进入火焰的流体将是自由流与回流区流体的混合物.如果是贫燃火焰,回流区流体由燃烧产物和多余的氧化剂组成而没有燃料,因此进入剪切层火焰的混合物当量比将低于自由流的当量比.如此一来,即使自由来流是可燃的,剪切层中混合物的当量比也可能超出可燃极限.分析表明,对于回流区稳定的贫燃火焰,进入剪切层火焰的混合物有效当量比将低于自由来流的当量比.根据理论分析建立了近贫燃熄火极限的回流区稳定燃烧模式图,其中涉及4个参数:自由来流当量比、剪切层卷吸比、吹熄极限和再点火极限.分析揭示出4种可能的燃烧模式:超稳定火焰、亚稳定火焰、振荡火焰和熄火.特别地,在参数空间发现了一个振荡区,它可能为近吹熄火焰引入一种新的固有不稳定性机制.在这种机制中,剪切层中的当量比振荡实际上是由燃烧过程本身驱动的,因为火焰的位置/振荡影响着进入回流区的流体成分进而影响剪切层中的当量比.因此,当量比振荡与燃烧过程之间的反馈循环由于周期性的火焰吹熄和闪回/再稳定而得以封闭.
