Haploidentical hematopoietic cell transplantation with or without an unrelated cord blood unit for adult acute myeloid leukemia:a multicenter,randomized,open-label,phase 3 trial

Coinfusion of unrelated cord blood(UCB)units in haploidentical hematopoietic cell transplantation(haplo-HCT)(haplo-cord HCT)for hematopoietic malignancies showed promising results in previous reports,but the efficiency of haplo-cord HCT in acute myeloid leukemia(AML)still lacks sufficient *** multicenter,randomized,phase 3 trial(*** NCT03719534)aimed to assess the efficacy and safety of haplo-cord HCT in AML patients.A total of 268 eligible patients aged 18-60 years,diagnosed with measurable residual disease in AML(excluding acute promyelocytic leukemia),with available haploidentical donors and suitable for allotransplantation,were randomly allocated(1:1)to receive haplo-cord HCT(n=134)or haplo-HCT(n=134).The 3-year overall survival(OS)was the primary endpoint in this *** median follow-up was 36.50 months(IQR 24.75-46.50).The 3-year OS of Haplo-cord HCT group was better than haplo-HCT group(80.5%,95%confidence interval[CI]:73.7-87.9 vs.67.8%95%CI 60.0-76.5,p=0.013).Favorable progression-free survival(70.3%,95%CI 62.6-78.8 vs.57.6%,95%CI 49.6-67.0,p=0.012)and cumulative incidence of relapse(12.1%,95%CI 12.0-12.2 vs.30.3%,95%CI 30.1-30.4,p=0.024)were observed in haplo-cord HCT *** 3-4 adverse events(AEs)within two years posttransplantation in the two groups were ***-cord HCT patients exhibited a faster cumulative incidence of neutrophil recovery(p=0.026)and increased T-cell reconstitution in the early period ***-cord HCT can improve OS in AML patients without excessive AEs,which may exert additional benefits for recipients of haplo-HCT.

Molecular profiling of cell-surface proteins is a powerful strategy for precise cancer *** mass cytometry(MC)enables synchronous detection of over 40 cellular parameters,its full potential in disease classification is challenged by the limited types of recognition probes currently *** this work,we synthesize a panel of heavy isotopeconjugated aptamers to profile cancer-associated signatures on the surface of hematological malignancy(HM)*** on 15 molecular signatures,we performed cell-surface profiling that allowed the precise classification of 8 HM cell *** with machine-learning technology,this aptamer-based MC platform also achieved multiclass identification of HM subtypes in clinical sampleswith 100%accuracy in the training cohort and 80%accuracy in the test ***,we report an effective and practical strategy for precise cancer classification at the singlecell level,paving the way for its clinical use in the near future.

行业神崇拜虽然指向一种“守旧”的精神状态和行为实践,但经过一些作家的逆写,得到了现代的“再利用”。酬神与渎神场面的变奏,隐含着解放个性的可能,是中国向现代进发的精神投影。大工业生产时代,作为历史资源的行业神崇拜,获得了现代性的实践方式,但现代作家拒绝征用其作为团结革命队伍的手段,这透露出革命知识分子对行业神崇拜可能引发力量的忧惧。性别立场昭然的当代故事中,女革命者的进步性没有因为敬神遭到批评,她们顺着“畏神—造神—成神”的脉络,跃向了主体的自由。而文学中行业神崇拜的归来,则展示出当代作家对文化传统的充足自信,预示着中国“信仰”的文艺表达进入了深度模式。

民国时期的上海理发店是脱胎于传统空间的生成性消费场所。作家对理发店的空间书写,以对物我互动、劳动技术、两性关系、身体权力等命题的观察和分析,取代对殖民话语和消费观念的统摄性评判。眼光的调整引发了对传统消费空间意蕴的倒转与叛逆,从而显示出与主体欲望自由释放相对应的被动体验。由此可见,对半殖民地空间书写的研究,不仅应拓展研究对象的范围,还应关注不同空间的成形历程与非通约的特质,发掘其背后潜藏的多重意蕴。

尤为密集的“!”是《我们夫妇之间》突出的文本现象,其生成语境与书写意图皆有其特殊性,是透视个性创作与集体话语的一面镜子。频频出现的感叹号帮助文本构成了潜在的双重视角,较为全面地呈现了进城干部的城市体验、革命生活和精神动向,导向了对乡村与城市、干部与群众、革命与婚姻等问题的思考。感叹号的高频使用是建国初期文学的整体症候,它承接了五四和抗战时期文学的写作策略和模式,亦是“文革”中文艺统制的先声。这些高频使用感叹号的创作既是对集体话语的个人回应,也是对世界现代、后现代思潮本土化的开掘,孕育着中国现当代文化和文学的无限可能。

欧美现代主义文学的研究,不能只聚焦现代性的经典角度,还需要从私人空间、家庭关系等与之相对的维度切入。以《阿斯彭文稿》为例,家庭是小说空间构设的重点,也是考察作品观念表达的有效视角。小说通过大宅、花园、威尼斯城三重空间的并置,探索了家庭空间及其权力边界,瓦解了私人空间和公共空间、内部和外部的简单对立。在道德观念上,《阿斯彭文稿》的故事徘徊在高雅保守和世俗开放之间,透露出19世纪旅欧文人重塑价值观的繁难。小说中的家庭元素值得剖析,它与风姿多彩的都市空间、性别意识一起,绘制出欧美文学走向现代的丰富性。沿着这一方向,我们有望打开欧美现代主义文学研究的新可能。

Arsenic trioxide(As2O3)is recently found to have therapeutic potential in systemic sclerosis(SSc),a life-threatening multi-system fibrosing autoimmune disease with type I interferon(IFN-I)*** activated plasmacytoid dendritic cells(pDCs)are responsible for IFN-I secretion and are closely related with fibrosis establishment in *** this study,we showed that high concentrations of As2O3 induced apoptosis of pDCs via mitochondrial pathway with increased BAX/BCL-2 ratio,while independent of reactive oxygen species ***,at clinical relevant concentrations,As2O3 preferentially inhibited IFN-αsecretion as compared to other cytokines such as TNF-α,probably due to potent down-regulation of the total protein and mRNA expression,as well as phosphorylation of the interferon regulatory factor7(IRF7).In addition,As2O3 induced a suppressive phenotype,and in combination with cytokine inhibition,it down-regulated pDCs’capacity to induce CD4+T cell proliferation,Thl/Th22 polarization,and B cell differentiation towards ***,chronically activated pDCs from SSc patients were not resistant to the selective IFN-αinhibition,and regulatory phenotype induced by ***,our data suggest that As2O3 could target pDCs and exert its treatment efficacy in SSc,and more autoimmune disorders with IFN-I signature.

牺牲是南京大屠杀题材小说的核心话题。中外作家叙述牺牲的方式,折射出各自国家的思想统制和生死观念的演化,包含建构式牺牲、效死型牺牲、圣母型牺牲等情形。中国作家的大屠杀叙事侧重对民族认同和战胜信念的强化,构成对牺牲亡国论的回击。在外籍作家的视阈中,中华民族对劫难的携手承担,往往被拆解贬抑成军民对峙、性别对立的内部冲突,继而服务于日军的美化和外侨荣光的撒播。立足跨国、跨文化的视野,深入分析南京大屠杀题材小说中的牺牲观念,能为今后的文学创作提供价值引导,为铭记历史和塑造记忆提供中正的精神指向。
