Sensitivity and resistance risk analysis of Didymella bryoniae populations to fluopyram

Fluopyram is an succinate dehydrogenase inhibitors(SDHI)fungicide that has been registered in China to control gummy stem blight(GSB)in watermelons for many ***,whether the field pathogens of GSB are still sensitive to fluopyram or not is ***,we collected 69 Didymella bryoniae isolates from the fields that usually use fluopyram to control GSB to determine the sensitivity *** EC_(50)(50%inhibition effect)values of fluopyram against *** ranged from 0.0691 to 0.3503μg mL^(–1) and the variation factor was *** mean EC_(50) value was(0.1579±0.0669)μg mL^(–1) and the curve of sensitivity was *** resistant strains were found in the isolates,which means that the pathogens were still sensitive to *** minimal inhibition concentration(MIC)of fluopyram against *** was 3μg mL^(–1).Four low-resistant mutants and two medium-resistant mutants were obtained using fungicide taming and the resistance of mutants could be inherited *** growth rate of mutants decreased significantly compared with that of wild-type strains while the biomass of most mutants was similar to that of wild-type *** sensitivity of most resistant mutants to various stresses was increased compared with that of wild-type *** virulence of mutants receded except for low-resistant mutant XN51FR-1,which had the same lesion area as XN51 on the watermelon *** results indicated that the fitness of resistant mutants was decreased compared with that of wild-type *** cross-resistance assay indicated that fluopyram-resistant mutants were positive cross-resistant to all six SDHI fungicides in this test but were still sensitive to fluazinam and *** the resistance risk of *** to fluopyram was *** addition,we found that the SdhB gene of low-resistant mutant XN30FR-1 had three new point mutations at positions K258N,A259P,and ***-resistant mutant XN52FR-1 showed a mutation at position H277N an

质膜H+-三磷酸腺苷酶(plasma membrane H+-ATPase,PMA),简称质膜ATP酶,属于质子泵家族蛋白,广泛存在于植物和真菌的质膜上;它们的主要作用是维持细胞营养摄取所需的跨膜电化学质子梯度和调控pH值。质膜ATP酶是真菌生命活动所必须的,该酶缺失后,突变体的生长出现明显缺陷甚至无法生长,这使其具有作为杀真菌剂靶标的潜力;另外,由于真菌和植物的质膜ATP酶同源性较低,故以质膜ATP酶为靶标开发的杀真菌剂具有生物安全性。最近明确的酿酒酵母Saccharomyces cerevisiae和粗糙脉孢霉Neurospora crassa质膜ATP酶的冷冻电镜结构揭示了其六聚体的状态,阐明了质膜ATP酶的作用机制,为基于结构的药物设计提供了理论基础。本文主要对真菌中质膜ATP酶的结构和功能进行系统阐述,并对质膜ATP酶作为新型杀真菌剂靶标的研究现状进行综述,旨在为新型杀真菌剂的发现和应用提供理论依据。

Progressive paradoxical sleep deprivation impairs partial memory following learning tasks in rats

BACKGROUND： Complex learning tasks result in a greater number of paradoxical sleep phases, which can improve memory. The effect of paradoxical sleep deprivation, induced by ＂flower pot＂ technique, on spatial reference memory and working memory require further research. OBJECTIVE： To observe the effect of progressive paradoxical sleep deprivation in rats, subsequent to learning, on memory using the Morris Water Maze. DESIGN, TIME AND SETTING： Controlled observation experiment. The experiment was performed at the Laboratory of Neurobiology, Department of Anatomy, Histology and Embryology, School of Basic Medical Sciences, Lanzhou University from December 2006 to October 2007. MATERIALS： Twenty-eight, male, Wistar rats, 3-4 months old, were provided by the Experimental Animal Center of Lanzhou University. The Morris Water Maze and behavioral analyses system was purchased from Genheart Company, Beijing, China. METHODS： All animals, according to a random digits table, were randomly divided into paradoxical sleep deprivation, tank control, and home cage control groups. Paradoxical sleep deprivation was induced by the ＂flower pot＂ technique for 72 hours, housing the rats on small platforms over water. Rats in the ＂tank control＂ and ＂home cage control＂ groups were housed either in a tank with large platforms over the water or in normal cages without paradoxical sleep deprivation. MAIN OUTCOME MEASURES： Morris Water Maze was employed for task learning and spatial memory testing. Rats in all groups were placed at six random starting points each day for four consecutive days. Each placement was repeated for two trials; the first trial represented reference memory and the second working memory. Rats in the first trial were allowed to locate the submerged platform within 120 seconds. Data, including swimming distance, escape latency, swimming velocity, percentage of time in correct quarter, and memory scores were recorded and analyzed automatically by behavioral analyses systems

Effects of gamma-aminobutyric acid A-receptor antagonist on sleep-wakefulness cycles following lesion to the ventrolateral preoptic area in rats

BACKGROUND： Neurons expressing gamma-aminobutyric acid （GABA） play an important role in the regulation of wakefulness to sleep, as well as the maintenance of sleep. However, the role of GABAergic neurons in the tuberomammillary nucleus （TMn）, with regard to the sleep-wakefulness cycle, is poorly understood. OBJECTIVE： To investigate the effects of GABAergic neurons in the TMn on the sleep-wakefulness cycle. DESIGN, TIME AND SETTING： Randomized controlled study, performed at the Laboratory of Neurobiology, Department of Anatomy, Histology and Embryology, School of Basic Medical Sciences, Lanzhou University from July 2007 to February 2008. MATERIALS： Fifteen healthy, adult, male, Sprague Dawley rats were randomly divided into three groups（n = 5）： control, ventrolateral preoptic area （VLPO） lesion, and VLPO lesion plus GABAA receptor antagonist-treated. Ibotenic acid and bicuculline were provided by Sigma （St. Louis, USA）. METHODS： Four electroencephalogram screw electrodes were implanted into the skull at a frontal region （two） and parietal bones （two） on each side. Three flexible electromyogram wire electrodes were placed into the nuchal muscles. On day 8, a fine glass micropipette （10–20 mm tip diameter） containing ibotenic acid solution （10 nmol/L） was injected into the VLPO in both hemispheres following bone wax removal under anesthesia. One week after the second surgery, sleep-wakefulness states were recorded in rats from the VLPO lesion group. On day 10 after VLPO lesion, bicuculline （10 nmol/L）, a GABAA-receptor antagonist, was microinjected into the TMn and sleep-wakefulness states were recorded for 24 hours. MAIN OUTCOME MEASURES： Duration of the sleep-wakefulness cycle in each group using a Data acquisition unit （Micro1 401 mk2） and Data collection software （Spike Ⅱ）. RESULTS： VLPO lesion induced an increased duration of wakefulness （W, 13.17%） and light slow-wave sleep （SWS1, 28.9%）, respectively. Deep slow-wave sleep （SWS2, 43.74%） and paradoxical

Genetic diversity and phylogenetic analysis of Chinese Han and Li ethnic populations from Hainan Island by 30 autosomal insertion/deletion polymorphisms

With the characteristics of low mutation rate,length variation and short amplicon size,insertion/deletion polymorphisms(InDels)have the advantages of both short tandem repeats(STRs)and single nucleotide polymorphisms(SNPs).Herein,people of two ethnicities from Hainan Island were genotyped for the first time using the Investigator DIPplex *** investigated the forensic parameters of the 30 InDels and the phylogenetic relationships among different *** accumulated powers of discrimination and powers of exclusion were 0.9999999999646 and 0.9897 in the Hainan Han population and 0.9999999999292 and 0.9861 in the Hainan Li population,***,population comparisons among geographically,ethnically and linguistically diverse populations via cluster heatmap,multidimensional scaling,principal component analysis,phylogenetic tree and STRUCTURE analyses demonstrated that the Hainan Han population had genetic similarities to the other Han,She and Tujia populations,while the Hainan Li population had close genetic relationships to the Zhuang and Miao groups;both populations had a high degree of genetic differentiation from most Turkic-speaking *** results suggested that the 30 autosomal InDels are highly polymorphic and informative,which are suitable for human identification and population genetics.

Massively parallel sequencing of 231 autosomal SNPs with a custom panel:a SNP typing assay developed for human identification with Ion Torrent PGM

The custom-designed single nucleotide polymorphism(SNP)panel amplified 231 autosomal SNPs in one PCR reaction and subsequently sequenced with massively parallel sequencing(MPS)technology and Ion Torrent personal genome machine(PGM).SNPs were chosen from SNPforID,IISNP,HapMap,dbSNP,and related published *** concordance was obtained between available MPS calling and Sanger sequencing with 9947A and 9948 *** SNPs(rs4606077,rs334355,rs430046,rs2920816,rs4530059,rs1478829,rs1498553,rs7141285,rs12714757 and rs2189011)with low coverage or heterozygote imbalance should be optimized or excluded from the *** data had sufficiently high coverage and gave reliable SNP calling for the remaining 221 loci with the custom MPS-SNP panel.A default DNA input amount of 10 ng per reaction was recommended by Ampliseq technology but sensitivity testing revealed positive results from as little as 1 ng input *** testing with this panel is possible through analysis of the F MAR(frequency of major allele reads)values at most loci with enough high coverage depth and low level of sequencing *** results indicate the potential advantage of the custom MPS-SNP assays and Ion Torrent PGM platform for forensic study.

Identification of the Mislabeled Breast Cancer Samples by Mitochondrial DNA Haplotyping

The task to identify whether an archival malignant tumor specimen had been mislabeled or interchanged is a challenging one for forensic genetic&The nuclear DNA(nDNA)markers were affected by the aberration of tumor cells,so they were not suitable for personal identification when the tumor tissues were *** this study,we focused on a new solution-mitochondrial single nucleotide polymorphism(mtSNP)haplotyping by a multiplex SNaPshot *** validate our strategy of haplotyping with 25 mtSNPs,we analyzed 15 pairs of cancerous/healthy tissues taken from patients with ductal breast *** haplotypes of all the fifteen breast cancer tissues were matched with their paired breast *** heteroplasmy at 2 sites,14783A/G and 16519C/T was observed in one breast tissue,which indicated a mixture of related mitochondrial ***,only one haplotype was retained in the paired breast cancer tissue,which could be considered the result of proliferation of tumor *** allele drop-out and allele drop-in were observed when 39 STRs and 20 tri-allelic SNPs of nDNA were *** to nDNA markers applied,25 mtSNPs were more stable without interference from aberrance of breast ***,two cases were presented where the investigation of haplotype with 25 mtSNPs was used to prove the origin of biopsy specimen with breast *** mislabeling of biopsy specimen with breast cancer could be certified in one case but could not be supported in the other *** highlight the importance of stability of mtSNP haplotype in breast *** was implied that our multiplex SNaPshot assay with 25 mtSNPs was a useful strategy to identify mislabeled breast cancer specimen.

Population genetics and forensic efficiency of 30 InDel markers in four Chinese ethnic groups residing in Sichuan

Sichuan Province is located at the transitional junction regions of the Qinghai-Tibet Plateau and the low-altitude *** also serves as the corridor of Sino Tibetan-speaking population migration and expansion since neolithic expansion of Proto-Tibeto-Burman populations from Middle/Upper Yellow River during Majiayao period(3300-2000 BC).However,the population structure and the corresponding genetic diversity of forensic-related markers in this region remain ***,we genotyped 30 insertion-deletion(InDel)markers in 444 samples from four ethnic groups(Han,Tibetan,Hui and Yi)from Sichuan Province using the Investigator■ DIPplex kit to explore the characteristics of population genetics and forensic genetic *** the loci were found to be in Hardy-Weinberg Equlibrium(HWE)after applying a Bonferroni correction and no pairwise loci showed prominent linkage *** combined matching probability(CMP)and the combined power of discrimination(CPD)are larger than 1.8089×10^(11)and 0.9999999995,*** component analysis,multi-dimen-sional scaling plots and Neighbour-Joining tree among 65 worldwide populations indicated that Sichuan Hui and Han are genetically close to Hmong-Mien and Tai-Kadai-speaking popula-tions,and Sichuan Tibetan and Yi bear a strong genetic affinity with Tibeto-Burman-speaking *** model-based genetic structure further supports the genetic affinity between the studied populations and linguistically close populations.

焦虑症常与睡眠障碍共存且相互作用,一方面睡眠障碍可引起焦虑,另一方面焦虑患者常出现失眠、睡眠片段化等焦虑样睡眠改变。基底外侧杏仁核和中央杏仁核与脑内多个睡眠-觉醒调控核团存在纤维联系,是调控焦虑样行为与睡眠的关键核团。本研究总结杏仁核解剖结构、纤维联系,以及杏仁核调节焦虑样行为与睡眠障碍的作用机制与神经环路,对进一步理解睡眠与焦虑相互作用的潜在机制具有重要意义,为临床治疗焦虑症和睡眠障碍提供理论基础。
