The treatment of PML/RARA+acute promyelocytic leukemia(APL)with all-trans-retinoic acid and arsenic trioxide(ATRA/ATO)has been recognized as a model for translational medicine *** an altered microenvironment is a general cancer hallmark,how APL blasts shape their plasma composition is poorly ***,we reported a cross-sectional correlation network to interpret multilayered datasets on clinical parameters,proteomes,and metabolomes of paired plasma samples from patients with APL before or after ATRA/ATO induction *** study revealed the two prominent features of the APL plasma,suggesting a possible involvement of APL blasts in modulating plasma *** was characterized by altered secretory protein and metabolite profiles correlating with heightened proliferation and energy consumption in APL blasts,and the other featured APL plasma-enriched proteins or enzymes catalyzing plasma-altered metabolites that were potential trans-regulatory targets of PML/***,results indicated heightened interferon-gamma signaling characterizing a tumor-suppressing function of the immune system at the first hematological complete remission stage,which likely resulted from therapy-induced cell death or senescence and ensuing supraphysiological levels of intracellular ***,our work sheds new light on the pathophysiology and treatment of APL and provides an information-rich reference data cohort for the exploratory and translational study of leukemia microenvironment.

自身免疫性脑炎急性症状性痫性发作指发生在免疫介导性脑炎活动期的癫痫发作。自身免疫相关性癫痫是指存在免疫病因,并有长期无诱因癫痫发作倾向的疾病。两者通常都对抗癫痫发作药物反应差,早期识别免疫病因并开始免疫治疗或许能帮助患者达到良好的预后。但目前尚无针对不同类型脑炎痫性发作的药物使用准则,自身免疫相关性癫痫也缺乏诊断标准和治疗原则。本文综述并讨论了两者可能的发病机制、临床表现和推荐的治疗方法,旨在为两者的诊治提供理论依据,并对未来的研究方向有所帮助。

The Ly-6 and uPAR(LU)domain-containing proteins represent a large family of cell-surface *** particular,mouse Ly-6A/Sca-1 is a widely used marker for various stem cells;however,its human ortholog is *** this study,based on a systematic survey and comparative genomic study of mouse and human LU domain-containing proteins,we identified a previously unannotated human gene encoding the candidate ortholog of mouse Ly-6A/*** gene,hereby named LY6A,reversely overlaps with a lncRNA gene in the majority of exonic *** found that LY6A is aberrantly expressed in pituitary tumors,but not in normal pituitary tissues,and may contribute to *** to mouse Ly-6A/Sca-1,human LY6A is also upregulated by interferon,suggesting a conserved transcriptional regulatory mechanism between humans and *** cloned the full-length LY6A cDNA,whose encoded protein sequence,domain architecture,and exon‒intron structures are all well conserved with mouse Ly-6A/*** expression of the LY6A protein in cells demonstrates that it acts the same as mouse Ly-6A/Sca-1 in their processing and glycosylphosphatidylinositol anchoring to the cell ***,these studies unveil a novel human gene encoding a candidate biomarker and provide an interesting model gene for studying gene regulatory and evolutionary mechanisms.

As muscle activity during growth is considerably important for mandible quality and morphology, reducing dietary loading directly influences the development and metabolic activity of mandibular condylar cartilage （MCC）. However, an overall investigation of changes in the protein composition of MCC has not been fully described in literature. To study the protein expression and putative signaling in vivo, we evaluated the structural changes of MCC and differentially expressed proteins induced by reducing functional loading in rat MCC at developmental stages. Isobaric tag for relative and absolute quantitation-based 2D nano-high performance liquid chromatography （HPLC） and matrix-assisted laser desorption/ionization time-of-flight/ time-of-flight （MALDI-TOF/TOF） technologies were used. Global protein profiling, KEGG and PANTHER pathways, and functional categories were analyzed. Consequently, histological and tartrate-resistant acid phosphatase staining indicated the altered histological structure of condylar cartilage and increased bone remodeling activity in hard-diet group. A total of 805 differentially expressed proteins were then identified. GO analysis revealed a significant number of proteins involved in the metabolic process, cellular process, biological regulation, localization, developmental process, and response to stimulus. KEGG pathway analysis also suggested that these proteins participated in various signaling pathways, including calcium signaling pathway, gap junction, ErbB signaling pathway, and mitogen-activated protein kinase signaling pathway. Collagen types I and II were further validated by immunohistochemical staining and Western blot analysis. Taken together, the present study provides an insight into the molecular mechanism of regulating condylar growth and remodeling induced by reducing dietary loading at the protein level.

Disabled homolog 2 （DAB2） is frequently deleted or epigenetically silenced in many human cancer cells＇ Therefore, DAB2 has always been regarded as a tumor suppressor gene. However, the role of DAB2 in tumor progression and metastasis remains unclear. In this study, DAB2 expression was upregulated along with human prostate cancer （PCa） progression. DAB2 overexpression or knockdown effects in LNCaP and PC3 cell lines were verified to address the biological functions of DAB2 in PCa progression and metastasis. LNCaP and PC3 cell lines were generated from human PCa cells with low and high metastatic potentials, respectively. The results showed that DAB2 shRNA knockdown can inhibit the migratory and invasive abilities of PC3 cells, as well as the tumorigenicity, whereas DAB2 overexpression enhanced LNCaP cell migration and invasion. Further investigation showed that DAB2 regulated the cell migration associated genes in PC3 cells, and the differential DAB2 expression between LNCaP and PC3 cells was partly regulated by histone 4 acetylation. Therefore, DAB2 may play an important role in PCa progression and metastasis.

[目的/意义]文章对现有研究中各影响因素的差异性进行研究和证实,为开展社交环境下的信息服务以及信息规避行为领域的研究提供参考借鉴。[方法/过程]文章运用元分析方法,从30篇实证研究中,提取53个效应值进行异质性检验、发表偏倚检验、整体效应检验和调节效应分析等相关分析。[结果/结论]心理因素和信息因素中包含的8个影响因素与用户信息规避行为有相关关系,其中信息杂乱因素的相关性最强,情绪因素的相关性最弱。同时,感知目标障碍受性别、隐私担忧、信息质量和学科差异的调节作用比较明显;隐私担忧受情绪的调节作用比较明显。

【目的/意义】文章对现有研究中各影响因素的差异性进行研究和证实,一方面能够为不持续使用行为后续相关研究提供参考,另一方面可为社交媒体运营商优化服务策略、减少消极影响提供理论指导。【方法/过程】使用元分析方法,从77篇相关实证研究中,提取87个效应值进行效应值计算、发表偏倚检验、异质性检验、整体效应检验和调节效应分析等综合定量分析。【结果/结论】研究发现,情感因素中的社交媒体倦怠和态度因素中的不满意与不持续使用行为呈强相关关系,信息过载和隐私关注同样是不持续使用行为的影响因素。同时,社交媒体用户不持续使用行为的影响因素会受性别、年龄、隐私关注和信息过载的调节。【创新/局限】使用元分析方法分析了各影响因素对用户不持续使用行为的影响强度,对已有研究中存在的结论差异进行了证实。但部分高质量文献由于没有相关可用数据被排除在外,影响了分析结果的全面性。

Turbulence Statistics of Thermo-Buoyancy Supercritical Fuel Flow in a Regenerative Cooling Channel

Active regenerative cooling with supercritical hydrocarbon fuel is considered as the most promising thermal protection *** existence of buoyancy force would lead to strongly anisotropic flow and thermal transport *** is closely related to the cooling performance of the *** elucidate the mechanisms of turbulent transport,the large eddy simulation(LES) was performed to assess turbulence statistics within different turbulence *** results indicated that the buoyancy and inertial force together dominated the change of turbulent ***,the spatial thermal buoyancy effect significantly suppressed the vertical velocity *** is due to the laminar motion caused by the buoyancy force,thereby weakening the thermal *** the statistics of velocity fluctuation,it was found that the buoyancy force and inertial force greatly weaken the vertical and streamwise velocity fluctuation,*** the statistics of thermal transport,the results pointed out that the near-wall heat transport characteristics need to be paid more *** thickness of the temperature mixing boundary layer led to the attenuation of vertical heat flux,which inhibited vertical temperature diffusion and predisposed to extreme conditions of heat transfer *** results can enhance the academic understanding of the heat transfer mechanism of supercritical fluids,and give guidance for further applications of thermal protection.

Output feedback stabilization of stochastic high-order planar nonlinear systems with stochastic inverse dynamics and output-constraint

In this paper, we solve the output feedback control problem of stochastic high-order planar nonlinear systems with output constraint and stochastic integral input-to-state stability(SiISS) inverse dynamics. By employing a key coordinate transformation, a stochastic nonlinear system with output constraint and SiISS inverse dynamics is converted into an unconstrained system. By skillfully constructing an observer and adopting SiISS small-gain conditions, we develop a new output feedback control design and analysis method, and prove that all the closed-system signals are bounded almost surely, the output constraint is not violated almost surely, and the equilibrium point of the closed-loop system is stochastically asymptotically stable.
