Herb induced liver injury by Xianling Gubao Tablets: A case assessed for causality using updated RUCAM and integrated evidence chain

Objective:A typical case of Xianling Gubao(XLGB)Tablets-induced liver injury was systematically studied in the clinic and the laboratory.Methods:A patient with herb-induced liver injury(HIli)and a history of taking XLGB Tablets before disease onset was engaged as the study subject,and the case was diagnosed according to the updated Roussel Uclaf Causality Assessment Method(RUCAM)and the integrated evidence chain(iEC)method recommended by the Guidelines for Diagnosis and Treatment of Herb-induced liver Injury(HIli Guidelines).Results:Clinical history,biochemical indexes and imaging tests were used to exclude the influence of fundamental diseases and confusing liver diseases such as viral,alcoholic and autoimmune liver diseases on the diagnosis.Based on an investigation of the patient’s medication history,she was suspected to have HIli caused by XLGB Tablets,as the patient was only taking an oral preparation of XLGB Tablets,and the influence of other drugs on the diagnosis was excluded.This patient with alanine aminotransferase(ALT)3 upper limit of normal(ULN)and a calculated R of 6 was diagnosed with possible acute drug-induced hepatocellular injury.The relationship was considered“highly probable”(score of 9)using the updated RUCAM of 2016.Moreover,the fingerprint similarity between the preparation taken by the patient and a commercially available preparation was 0.99,suggesting that the patient was consuming XLGB Tablets rather than another drug.LC-MS technology and the Agilent Fake TCM-Drugs database were used to investigate the drug,and no chemical additions were found.Examination of the drug for pesticide residues,heavy metals,aflatoxins and other exogenous substances indicated compliance with the content limits of the Chinese Pharmacopoeia.Conclusion:In summary,the final diagnosis of XLGB-induced liver injury reached the clinical diagnosis of HIli and was acute severe hepatocellular injury type by the updated RUCAM and iEC.Therefore,this study provides scientific evidence regarding the causality evaluation o

With its more sustainable production mode and higher healthy food supplying,cultivated meat is gradually regarded as the promising alternative meat food in future by human beings.Most of the countries in the planet had participated in the cut-throat competition for sustainable protein searching,especially for the regions with low food self-sufficiency rate.In this paper,by reviewing the commercial market participation,product development stage,and technical research progress related to cultivated meat in detail,we demonstrated the industrialization progress of cultivated meat and explored the challenges of cultivated meat in the near future.1)Seeded cell and serum free medium were the basic raw materials,which were directly related to the food safety regulation,production cost control and productivity improvement;2)Bioreactor and regulation were the cornerstone of industrialization,which were closely related to the capacity expansion,product marketing;3)Technological improvements in taste,texture,safety,scale,cost were the foundation of realizing product iterative upgrade and market acceptance.

Chiral transfer amidst one-dimensional linear polymers and twodimensional network covalent organic frameworks:Striking a fine balance between helicity and crystallinity

The current landscape of chiral covalent organic frameworks(COFs)predominantly centered on constructing asymmetric molecular-scale chirality,often introducing an inherent contradiction to the COF symmetry and limiting diversity.Herein,we overcome these challenges by achieving chiral transfer between one-dimensional(1D)imine linear polymers(LPs)and two-dimensional(2D)networkβ-ketoenamine COFs composed of achiral monomers.We successfully synthesize several 1D imine LPs with mesoscopic helical chirality,comprising achiral C2-symmetric terephthalaldehyde and diamine linkers in a chiral supramolecular transcription system.Leveraging the irreversible tautomerism mechanism within the linker replacement approach,terephthalaldehyde(TPA)units in these helical 1D LPs are substituted with C3-symmetric 1,3,5-triformylphloroglucinol(TP),yielding the corresponding 2D networkβ-ketoenamine COFs.Crystallinity and helicity of the resultantβ-ketoenamine COFs intimately hinge on reaction conditions,including the aldehyde stoichiometry of Tp and TPA,as well as the quantity and concentration of the catalyst employed.Under optimized conditions,the nucleation and growth were precisely governed,achieving a harmonious equilibrium of crystallinity and helicity within the generated 2D networkβ-ketoenamine COFs,even with covalent bond rupture,recombination,and topological transition(from[C2+C2]to[C3+C2]).Impressively,the ground state chirality inherent to helical 1D LPs seamlessly transfers to helical 2D networkβ-ketoenamine COFs.This study not only offers new perspectives on the development of chiral functional COFs,but also provides fresh insights into the precise control of COFs'microscopic morphology.

Research on organized method and regulatory mechanism of Cell cultured meat based on modified silk

Cell cultured meat has been extensively studied as an environmentally friendly， energy-saving and more effective technology. However， there are many technical bottlenecks， especially the regulatory mechanism and manufacturing method of in vitro myogenesis. Based on an edible modified silk protein scaffold， with 3D culturing， in situ differentiated and transcriptome analysis， this study describes novel scaffolds and fabrication methods for cell cultured meat. The results showed that the effective space and utilization efficiency for cell culture of the scaffold is 26-1000 that of the traditional culture dish; it could form a tissue-like structure. Transcriptomics revealed the regulatory pathways and key factors of different cycles. It clarifies that the multi-cycle process of myoblast myogenesis in vitro is different from the single feedback regulation in vivo. More importantly， a novel scaffold-based cell cultured meat manufacturing method was developed， further develop a new tissue culture solution that is different from existing cell culture meat production.For manufacturing processes， it provides a new cell culture meat technology system， provides a theoretical basis for the regulation of cell proliferation and muscle growth， and lays the technical foundation for in situ tissue culture of cell cultured meat in vitro.

Sequentially bridged MXene platelets for strong high‐temperature EM‐IR bi‐stealth sheets

Combination of flexible multifunctional stealth technology properties such as electromagnetic(EM)and infrared(IR)stealth is crucial to the development of aerospace,military,and electronic fields,but the synthesis technology still has a significant challenge.Herein,we have successfully designed and synthesized highly flexible MXene@cellulose lamellae/borate ion(MXCB)sheets with strong high‐temperature EM‐IR bi‐stealth through sequential bridging of hydrogen and covalent bonds.The resultant MXCB sheets display high conductivity and good mechanical features such as flexibility,stretchability,fatigue resistance,and ultrasonic damage.MXCB sheets have a high tensile strength of 795 MPa.Furthermore,MXCB sheets with different thicknesses indicate exceptional high‐temperature thermal‐camouflage characteristics.This reduces the radiation temperature of the target object(>300°C)to 100°C.The conductivity of MXCB sheet with 3μm thickness is 6108 S/cm and the EM interference(EMI)shielding value is 39.74 dB.The normalized surface‐specific EMI SE absolute shielding effectiveness(SSE/t)is as high as 39312.78 dB·cm2/g,which remained 99.39%even after 10,000 times repeated folding.These multifunctional ultrathin MXCB sheets can be arranged by vacuum‐assisted induction to develop EM‐IR bi‐stealth sheet.

GCAGA: A Gini Coefficient-Based Optimization Strategy for Computation Offloading in Multi-User-Multi-Edge MEC System

To support the explosive growth of Information and Communications Technology(ICT),Mobile Edge Comput-ing(MEC)provides users with low latency and high bandwidth service by offloading computational tasks to the network’s edge.However,resource-constrained mobile devices still suffer from a capacity mismatch when faced with latency-sensitive and compute-intensive emerging applications.To address the difficulty of running computationally intensive applications on resource-constrained clients,a model of the computation offloading problem in a network consisting of multiple mobile users and edge cloud servers is studied in this paper.Then a user benefit function EoU(Experience of Users)is proposed jointly considering energy consumption and time delay.The EoU maximization problem is decomposed into two steps,i.e.,resource allocation and offloading decision.The offloading decision is usually given by heuristic algorithms which are often faced with the challenge of slow convergence and poor stability.Thus,a combined offloading algorithm,i.e.,a Gini coefficient-based adaptive genetic algorithm(GCAGA),is proposed to alleviate the dilemma.The proposed algorithm optimizes the offloading decision by maximizing EoU and accelerates the convergence with the Gini coefficient.The simulation compares the proposed algorithm with the genetic algorithm(GA)and adaptive genetic algorithm(AGA).Experiment results show that the Gini coefficient and the adaptive heuristic operators can accelerate the convergence speed,and the proposed algorithm performs better in terms of convergence while obtaining higher EoU.The simulation code of the proposed algorithm is available:https://***/Grox888/Mobile_Edge_Computing/tree/GCAGA.

Amide is essential in biologically active compounds,synthetic materials,and building blocks.However,conventional amide production relies on energyintensive consumption and activating agents that modulate processes to construct the C–N bond.Herein,for the first time,we have successfully realized the formation of amides at industrial current density via the anodic coelectrolysis of alcohol and ammonia under ambient conditions.We have proved thatmodulation of the interface microenvironment concentration of nucleophile by electrolyte engineering can regulate the reaction pathways of amides rather than acetic acids.The C-N coupling strategy can be further extended to the electrosynthesis of the long-chain and aryl-ring amide with high selectivity by replacing ammonia with amine.Our work opens up a vast store of information on the utilization of biomass alcohol for high-value N-containing chemicals via an electrocatalytic C-N coupling reaction.

Wood is an invaluable asset to human society due to its renewable nature,making it suitable for both sustainable energy production and material manufacturing.Additionally,wood derived from forest trees plays a crucial role in sequestering a significant portion of the carbon dioxide fixed during photosynthesis by terrestrial plants.Nevertheless,with the expansion of the global population and ongoing industrialization,forest coverage has been substantially decreased,resulting in significant challenges for wood production and supply.Wood production practices have changed away from natural forests toward plantation forests.Thus,understanding the underlying genetic mechanisms of wood formation is the foundation for developing high-quality,fast-growing plantation trees.Breeding ideal forest trees for wood production using genetic technologies has attracted the interest of many.Tremendous studies have been carried out in recent years on the molecular,genetic,and cell-biological mechanisms of wood formation,and considerable progress and findings have been achieved.These studies and findings indicate enormous possibilities and prospects for tree improvement.This review will outline and assess the cellular and molecular mechanisms of wood formation,as well as studies on genetically improving forest trees,and address future development prospects.

Sustainable production of high-energy-density jet fuel via cycloaddition reactions

Developing an energy supply-chain based on renewable biomass holds great potential to build a low carbon society. High-energy-density(HED) jet fuel, featuring unique fused/strained cycloalkanes, is of great significance for volume-limited military aircrafts, as their high density and combustion heat can extend flight duration and increase the payload. Therefore, the exploration of biomass-based routes towards HED fuel has drawn much attention over the past decade. Cycloaddition reaction features rapid construction of various carbocycles in an atom-and step-economical fashion. This elegant strategy has been widely applied in the manufacture of sustainable HED fuel. Here we carefully summarize the progress achieved in this fascinating area and the review is categorized by the cycloaddition patterns including [4+2], [2+2],[4+4], and [2+1] cycloadditions. Besides, the energy densities of the as-prepared biofuels and petroleumbased fuels(conventional Jet-A and advanced JP-10) are also compared. This review will provide important insights into rational design of new HED fuel with different ring-types/sizes and inspire the chemists to turn those literature studies into practical applications in military field.

Immune checkpoint inhibitors break whose heart? Perspectives from cardio-immuno-oncology

Immune checkpoint inhibitors (ICIs) are monoclonal antibody antagonists,which can block cytotoxic T lymphocyte antigen-4 (CTLA-4),programmed death-1/ligand-1 (PD-1/PD-L1) pathways,and other molecules exploited by tumor cells to evade T cell-mediated immune response.ICIs have transformed the treatment landscape for various cancers due to their amazing efficacy.Many anti-tumor therapies,including targeted therapy,radiotherapy,and chemotherapy,combine ICIs to make the treatment more effective.However,the off-target immune activation caused by ICIs may lead to a broad spectrum of immune-related adverse events (irAEs) affecting multiple organ systems.Among irAEs,cardiotoxicity induced by ICIs,uncommon but fatal,has greatly offset survival benefits from ICIs,which is heartbreaking for both patients and clinicians.Consequently,such cardiotoxicity requires special vigilance,and it has become a common challenge both for patients and clinicians.This article reviewed the clinical manifestations and influence of cardiotoxicity from the view of patients and clinicians,elaborated on the underlying mechanisms in conjunction with animal studies,and then attempted to propose management strategies from a cardio-immuno-oncology multidisciplinary perspective.
