The crane&shuttle-based storage and retrieval system(C&SBS/RS)is the first automated warehouse technology that supports pallet picking,case picking and item picking.In the C&SBS/RS,aisle-captive cranes perform pallet picking,while tier-captive shuttles handle cases and items picking.To balance picking tasks,we propose an order dividing algorithm to fulfil required specific picking sequences.The optimisation objective is to minimise the order line picking time.Therefore,we modelled and analyzed the C&SBS/RS and considered single and dual command cycles for each resource(i.***.cranes,rail-guided vehicles,shuttles and lifters)separately according to their respective operation processes.Finally,numerical experiments were conducted to analyze impact factors and a real case to verify the power of the proposed order dividing algorithm.

Premature ovarian insufficiency(POI)is a heterogeneous female disorder characterized by the loss of ovarian function before the age of 40.It represents a significant detriment to female fertility.However,the known POI-causative genes currently account for only a fraction of cases.To elucidate the genetic factors underlying POI,we conducted whole-exome sequencing on a family with three fertile POI patients and identified a deleterious missense variant in RNF111.In a subsequent replication study involving 1,030 POI patients,this variant was not only confirmed but also accompanied by the discovery of three additional predicted deleterious RNF111 variants.These variants collectively account for eight cases,representing 0.78%of the study cohort.A further study involving 500 patients with diminished ovarian reserve also identified two additional RNF111 variants.Notably,RNF111 encodes an E3 ubiquitin ligase with a regulatory role in the TGF-β/BMP signaling pathway.Our analysis revealed that RNF111/RNF111 is predominantly expressed in the oocytes of mice,monkeys,and humans.To further investigate the functional implications of RNF111 variants,we generated two mouse models:one with a heterozygous missense mutation(Rnf111+/M)and another with a heterozygous null mutation(Rnf111^(+/-)).Both mouse models exhibited impaired female fertility,characterized by reduced litter sizes and small ovarian reserve.Additionally,RNA-seq and quantitative proteomics analysis unveiled that Rnf111 haploinsufficiency led to dysregulation in female gonad development and negative regulation of the BMP signaling pathway within mouse ovaries.In conclusion,our findings strongly suggest that monoallelic deleterious variants in RNF111 can impair female fertility and induce POI in both humans and mice.

Amide is essential in biologically active compounds,synthetic materials,and building blocks.However,conventional amide production relies on energyintensive consumption and activating agents that modulate processes to construct the C–N bond.Herein,for the first time,we have successfully realized the formation of amides at industrial current density via the anodic coelectrolysis of alcohol and ammonia under ambient conditions.We have proved thatmodulation of the interface microenvironment concentration of nucleophile by electrolyte engineering can regulate the reaction pathways of amides rather than acetic acids.The C-N coupling strategy can be further extended to the electrosynthesis of the long-chain and aryl-ring amide with high selectivity by replacing ammonia with amine.Our work opens up a vast store of information on the utilization of biomass alcohol for high-value N-containing chemicals via an electrocatalytic C-N coupling reaction.

Pattern-triggered immunity(PTI) is an essential strategy used by plants to deploy broad-spectrum resistance against pathogen attacks. Heterotrimeric G proteins have been reported to contribute to PTI.Of the three non-canonical EXTRA-LARGE G PROTEINs(XLGs) in Arabidopsis thaliana, XLG2 and XLG3 were shown to positively regulate immunity,but XLG1 was not considered to function in defense,based on the analysis of a weak xlg1 allele.In this study, we characterized the xlg1 xlg2 xlg3 triple knockout mutants generated from an xlg1 knockout allele. The strong xlg1 xlg2 xlg3 triple mutants compromised pathogen-associated molecular pattern(PAMP)-triggered activation of mitogen-activated protein kinases(MAPKs) and resistance to pathogen infection. The three XLGs interacted with MAPK cascade proteins involved in defense signaling, including the MAPK kinase kinases MAPKKK3 and MAPKKK5, the MAPK kinases MKK4 and MKK5, and the MAPKs MPK3 and MPK6. Expressing a constitutively active form of MKK4 restored MAPK activation and partially recovered the compromised disease resistance seen in the strong xlg1 xlg2 xlg3 triple mutant. Furthermore, mutations of all three XLGs largely restored the phenotype of the autoimmunity mutant bak1-interacting receptor-like kinase 1. Our study reveals that all three XLGs function redundantly in PAMP-triggered MAPK activation and plant immunity.

保暖织物在人类生活中扮演着不可或缺的重要角色.传统保暖织物通常由天然纤维(棉、麻、蚕丝)或人造纤维(聚酯、尼龙、聚丙烯腈、聚氨酯)构成.随着时代发展,传统保暖织物的性能已经不能满足日益提高的特别是面对极端复杂环境的保暖需求.气凝胶纤维是一种具有三维多孔网络结构的纤维,具有极高的孔隙率、极大的比表面积和极低的密度,是一种理想的绝热纤维,具有用于高性能保暖织物的潜力.然而,气凝胶纤维通常强度低、韧性差,被加工应用的能力较差,限制了其应用于保暖衣物[1].在过去10年里,研究者基于各种材料,包括碳纳米管、凯夫拉、聚酰亚胺、纤维素和石墨烯等,通过溶胶凝胶转变和特定的干燥方法,并通过结构设计成功制备了具有一定力学柔韧性(可弯曲、打结)的气凝胶纤维[2].但是,上述方法获得的气凝胶纤维的力学性能仍然无法满足织造(编织和针织)的要求,更无法满足耐水洗、耐磨等实用性织物的要求.

Metal clusters with photoswitching ligands can undergo magnetic changes under light radiation,which makes them potential optical-response magnetic-switching materials.Herein,a photoswitchable Mn-oxo cluster[Mn^(II)_(15)Mn^(III)_(4)(L)_(18)(OH)_(12)(N_(3))_(6)](ClO_(4))_(2)(C_(6)H_(5)CN)_(8),(Mn19azo,HL=1-(hydroxymethyl)-3,5-dimethyl-4-(phenyldiazenyl)-pyrazole)has been designed and synthesized to realize the photoswitching behavior.Mn19azo shows quick trans-to-cis transformation upon 365 nm light irradiation in CH2Cl2 solution and solid state as confirmed by UV-Vis spectra.After the powder sample was irradiated for 80 min,the Curie constant of Mn19azo increases by about 7%.The change of magnetic properties can be considered as the result of the change of intermolecular interactions.

The gut microbiota plays a pivotal role in the maintenance of health for amphibians,and it has been fully recognized,but the effectiveness of various influencing factors has not yet been fully clarified.Although this association should be considered while the amphibian order Caudata is facing a severe situation of population decline and extinction,there is little understanding of the association between diets and the diversity of gut microbiota in the amphibian order Caudata.Here,we conducted an extensive analysis of the gut microbiota of Cynops orientalis fed different diets using functional prediction and 16S rRNA amplicon sequencing techniques.First,we found that wild individuals had greater gut microbial diversity and richness in comparison to captive individuals.Second,we identified the bacterial taxa associated with diets and observed differences in the relative abundance of gut microbiota among people on various diets.Finally,we have a predictive comprehension of the selection and adaptative significance of shared core ASVs in the gut microbiota in maintaining the healthy survival of C.orientalis in a large-scale spatiotemporal map.Our study emphasizes how diets alter the gut microbiota of Caudata and offers fresh perspectives on the conservation and captive management of species in Caudata.

Oxidative stress may be the unifying factor for the injury caused by hyperglycemia in diabetic peripheral neuropathy. Puerarin is the major isoflavonoid derived from Radix puerariae and has been shown to be effective in increasing superoxide dismutase activity. This study sought to investigate the neuroprotective effect of puerarin on high glucose-induced oxidative stress and Schwann cell apoptosis in vitro. Intracellular reactive oxygen radicals and mitochondrial transmembrane potential were detected by flow cytometry analysis. Apoptosis was confirmed by TUNEL and oxidative stress was monitored using an enzyme-linked immunosorbent assay for the DNA marker 8-hydroxy-2-deoxyguanosine. The expression levels of bax and bcl-2 were analyzed by quantitative real-time reverse transcriptase-PCR, while protein expression of cleaved caspase-3 and -9 were analyzed by means of western blotting. Results suggested that puerarin treatment inhibited high glucose-induced oxidative stress, mitochondrial depolarization and apoptosis in a dose-dependent manner. Furthermore, puerarin treatment downregulated Bax expression, upregulated bcl-2 expression and attenuated the activation of caspase-3 and -9. Overall, our results indicated that puerarin antagonized high glucose-induced oxidative stress and apoptosis in Schwann cells.
