Hepatic HDAC3 Regulates Systemic Iron Homeostasis and Ferroptosis via the Hippo Signaling Pathway

Histone deacetylases(HDACs)are epigenetic regulators that play an important role in determining cell fate and maintaining cellular ***,whether and how HDACs regulate iron metabolism and ferroptosis(an iron-dependent form of cell death)remain ***,the putative role of hepatic HDACs in regulating iron metabolism and ferroptosis was investigated using genetic mouse *** lacking Hdac3 expression in the liver(Hdac3-LKO mice)have significantly reduced hepatic Hamp mRNA(encoding the peptide hormone hepcidin)and altered iron *** profiling of Hdac3-LKO mice suggests that the Hippo signaling pathway may be downstream of ***,using a Hippo pathway inhibitor and overexpressing the transcriptional regulator Yap(Yes-associated protein)significantly reduced Hamp mRNA *** a promoter reporter assay,we then identified 2 Yap-binding repressor sites within the human HAMP promoter *** also found that inhibiting Hdac3 led to increased translocation of Yap to the nucleus,suggesting activation of ***,knock-in mice expressing a constitutively active form of Yap(Yap K342M)phenocopied the altered hepcidin levels observed in Hdac3-LKO ***,we show that iron-overload-induced ferroptosis underlies the liver injury that develops in Hdac3-LKO mice,and knocking down Yap expression in Hdac3-LKO mice reduces both iron-overload-and ferroptosis-induced liver *** results provide compelling evidence supporting the notion that HDAC3 regulates iron homeostasis via the Hippo/Yap pathway and may serve as a target for reducing ferroptosis in iron-overload-related diseases.

Highly selective and remotely communicable nitrogen dioxide(NO_(2))sensing may contribute to future Internet of Things in environmental ***,room-temperature NO_(2)sensing materials such as carbon materials is still less than satisfactory due to their insensitive interaction with target ***,polyethylene imine functionalized three-dimensional(3D)carbon framework(PEI/C framework)has been developed for enhanced selective NO_(2)sensing,via combined template synthesis and subsequent ***,the 3D PEI/C framework is observed porous shape with irregular ***,the response of C framework to NO_(2)increases while those of interfering gases decrease after being functionalized with ***,the sensor prototypes show a 100 ppb-concentration detection limit at room ***,such excellent NO_(2)sensing is attributed to the large specific surface ratio of porous 3D PEI/C framework,in which PEI serves as an active layer for target NO_(2),while a passivated one for interfering ***,such PEI/C framework sensor prototype is simulated for NO_(2)sensing device and communicated with a smartphone,showing great potential in future intelligent environmental monitoring.

PI4KIIα regulates insulin secretion and glucose homeostasis via a PKD-dependent pathway

Insulin release by pancreatic b cells plays a key role in regulating blood glucose levels in humans, and to understand the mechanism for insulin secretion may reveal therapeutic strategies for diabetes. We found that PI4KIIα transgenic(TG) mice have abnormal glucose tolerance and higher serum glucose levels than wild-type mice. Glucose-stimulated insulin secretion was significantly reduced in both PI4KIIα TG mice and PI4KIIα-overexpressing pancreatic b cell lines. A proximity-based biotin labeling technique, Bio ID, was used to identify proteins that interact with PI4KIIα, and the results revealed that PI4KIIα interacts with PKD and negatively regulates its activity. The effect of PI4KIIα on insulin secretion was completely rescued by altering PKD activity. PI4KIIα overexpression also worsened glucose tolerance in streptozotocin/high-fat diet-induced diabetic mice by impairing insulin *** study has shed new light on PI4KIIα function and mechanism in diabetes and identified PI4KIIα as an important regulator of insulin secretion.

Comprehensive Summary,Regulating spin crossover(SCO)behavior,especially controlling the spin transition steps,is an important scientific issue,mainly because people aim to control spin bistability and ***,SCO bistability can be regulated by changing the ligand-modifying species,non-coordinated anions,guest molecules,and metal-ion ***,the control of multistability is extremely challenging,especially in Fe(III)SCO ***,we report that[FeIII(H-5-Br-thsa)(5-Br-thsa)]·H2O(5-Br-thsa=(5-bromo-2-hydroxybenzylidene)hydrazinecarbothioamide),a compound we have reported before,exists in two polymorphic forms:polymorph 1 exhibits three-step SCO,and polymorph 2 shows five-step SCO,with multi-step SCO behavior effectively regulated by *** to single-crystal and powder X-ray diffractometry,polymorphs 1 and 2 crystallize in different space groups during their spin transitions,with two-step symmetry breaking observed(Pbcn→Pnc2→Pbcn for polymorph 1;P21/n→Pn→P21/n for polymorph 2).We realized that the behavior of these two polymorphs depends significantly on the structure,including(i)the average Fe—N bond distance,(ii)deformation of octahedral FeIII atoms,and(iii)distinct crystal packing,which account for the large differences observed in magnetic properties.

Fine particulate matter(PM_(2.5))and ozone(O_(3))double high pollution(DHP)events have occurred frequently over China in recent years,but their causes are not completely *** this study,the spatiotemporal distribution of DHP events in China during 2013–20 is *** synoptic types affecting DHP events are identified with the Lamb–Jenkinson circulation classification *** meteorological and chemical causes of DHP events controlled by the main synoptic types are further *** show that DHP events(1655 in total for China during 2013–20)mainly occur over the North China Plain,Yangtze River Delta,Pearl River Delta,Sichuan Basin,and Central *** occurrence frequency increases by 5.1%during 2013–15,and then decreases by 56.1%during 2015–*** main circulation types of DHP events are“cyclone”and“anticyclone”,accounting for over 40%of all DHP events over five main polluted regions in China,followed by southerly or easterly flat airflow types,like“southeast”,“southwest”,and“east”.Compared with non-DHP events,DHP events are characterized by static or weak wind,high temperature(20.9℃ versus 23.1℃)and low humidity(70.0%versus 64.9%).The diurnal cycles of meteorological conditions cause PM_(2.5)(0300–1200 LST,Local Standard Time=UTC+8 hours)and O_(3)(1500–2100 LST)to exceed the national standards at different periods of the DHP *** pollutant conversion indices further indicate the rapid secondary conversions during DHP events,and thus the concentrations of NO_(2),SO_(2) and volatile organic compounds decrease by 13.1%,4.7%and 4.4%,*** results of this study can be informative for future decisions on the management of DHP events.

Comparison of two and three dimensional quantitative coronaryangiography to intravascular ultrasound in the assessment of leftmain coronary artery bifurcation lesions

Background Angiographic evaluation of left main coronary artery(LMCA) bifurcation lesions is often limited. two dimensional(2D) quantitative coronary angiography(QCA) with segmental analysis provides accuracy for quantification of the degree of stenosis in the main vessel and side branch ostium but can be affected by foreshortening and variable magnification. The accuracy of three dimensional(3D) QCA has recently developed to overcome 2D QCA limitations, however, accuracy and precision of 3D bifurcation QCA measurements in LMCA bifurcation lesions has not been *** We investigated whether such 3D and 2D bifurcation QCA measurements differ in their accuracy in assessing significant LMCA bifurcation lesions defined by intravascular ultrasound(IVUS) as a minimum luminal area(MLA) <6 mm2 of LMCA and MLA <4 mm2 of proximal left anterior descending(LAD) and/or proximal left circumflex(LCX)Results LMCA bifurcation lesions were assessed in 44 patients undergoing elective percutaneous coronary intervention. From 2D QCA measurements, MLA correlated moderately with threshold intravascular ultrasound MLA for LMCA(r=0.81, P <0.000 1), LAD(r=0.54, P=0.000 1) and LCX(r=0.58, P <0.000 1). Severity of lesion as MLA by derived 3D QCA, correlated moderately with threshold intravascular ultrasound MLA for LMCA(r=0.84, P <0.000 1), LAD(r=0.53, P=0.000 2); LCX(r=0.66, P <0.000 1). Overall, the C statistics tended to be slightly higher for 3D QCA and 2D QCA measurements in LMCA segment compared with proximal LAD and LCX segments, and there were no significant predictive power of percent diameter stenosis and percent area stenosis on 3D QCA for LCX IVUS MLA <4 mm2(percent diameter stenosis: area under curve 0.55, cutoff 23%, sensitivity 88%, specificity 37%, P=0.618 6; percent arer stenosis: area under curve 0.56, cutoff 41%, sensitivity 83%, specificity 38%, P=0.518 4, respectively).Conclusions The accuracy of 3D bifurcation QCA in detecting significant LMCA bifurcation les
